[The measurement of umbilical artery flow velocity waveforms in normal late pregnancy by Doppler ultrasound].

Doppler ultrasound was adopted in a study of fetal umbilical artery flow velocity waveforms in 81 cases of late pregnancies (28-41 gestational weeks). The peak systolic/end-diastolic ratio (A/B) and the Pourlot index were used as indices for assessing placental resistance. Both the A/B ratio (2.34 +/- 0.44) and Pourlot index (0.56 +/- 0.07) decreased while the gestational week was increasing, and they had a good correlation with the gestational week (r1 = -0.90, r2 = -0.93). The results indicated that the placental resistance became lower when the gestational week was increasing. This method has been documented a useful noninvasive technique for the exam of placental resistance.

Serotyping and subgrouping of some rotavirus strains in China.

This article reports for the first time in China the results of serotyping and subgrouping of some rotavirus strains obtained from faecal fluids of cases with infant diarrhoea. Infant diarrhoea rotavirus serotype 1 (Wa strain)-specific and serotype 2 (KUN strain)-specific hyperimmune sera, and infant diarrhoea rotavirus serotype 3-specific, rotavirus subgroup I-specific and subgroup II-specific monoclonal antibodies were used in serotyping and subgrouping by means of enzyme-linked immunosorbent assay of 14 rotavirus strains which had been identified by electron microscopy. Results showed that three out of four Beijing strains in 1982 belonged to serotype 2 and subgroup I rotavirus, one belonged to serotype 3 and subgroup II rotavirus; one out of nine Beijing strains in 1984 belonged to serotype 1 and subgroup II rotavirus, seven belonged to serotype 2 and subgroup I rotavirus, one belonged to serotype 3 and subgroup II rotavirus; and one Kunming strain in 1984 belonged to serotype 3 and subgroup II rotavirus.

Interaction and combination of separate A and B chains of insulin effects of D-and L-tryptophan in position A1.

The S-thiomethyl derivatives of insulin A chain with A1-Gly replaced by D- or L-Trp have been prepared and their respective interaction and combination with the S-thiomethyl B chain studied. The UV difference spectra of the mixed against the separated [Trp1]A chains with the B chain at pH 10.8 are similar to those obtained for the unmodified chains except that the 295-nm-negative peak for ionized Tyr residue appears to be less marked. Fluorescence studies show very little environmental changes at the A1-Trp residues when mixed with the B chain. The intact hormone with A1-Gly replaced by D-Trp is known to be considerably more active than the analog with L-Trp replacement. However, for both derivatives the resynthesis of the whole molecules correctly joined by disulfide bridges starting from the separated reduced chains, gives similar low yields as shown by HPLC analysis and by receptor-binding assay. The replacement of A1-Gly by D-Trp appears to affect the separated A chain more than the intact hormone and replacements at A1 by both D- and L-Trp probably lead to significant conformational changes of the A chain so as to prevent its correct pairing with the B chain.

Antibody against adult diarrhoea rotavirus among healthy adult population in China.

This paper deals with a method of using unknown serum as the first antibody to coat wells of the microtiter plate directly in application of the sandwich ELISA method with double antibodies for the assay of the antibody against adult diarrhoea rotavirus. This method was used for assay of the antibody against adult rotavirus in 1,380 sera of healthy adults obtained from some provinces and cities of China, of which 141 showed a positive result with a total positive rate of 10.2%. The infection rate varied in different regions. In order to confirm the accuracy of these results a blocking test was carried out on 25 positive specimens, and the results revealed that the P/N ratios after blocking were all lower than those before blocking. Seventeen of these 25 positive serum specimens were obtained from rural areas of Qian'an County, Hebei Province where epidemic adult diarrhoea had occurred two years ago. The antibody against adult diarrhoea rotavirus was not detected in 50 adult sera from Xizang Autonomous Region. Only one out of 50 adult sera from Hainan Island was positive for the antibody.

Urinary excretion of N-nitrosamino acids and nitrate by inhabitants of high- and low-risk areas for esophageal cancer in Northern China: endogenous formation of nitrosoproline and its inhibition by vitamin C.

A total of 238 samples of 24-h urine were collected from inhabitants of high-risk (Lin-xian) and low-risk (Fan-xian) areas for esophageal cancer in northern China, according to three protocols: (a) from undosed subjects; (b) from subjects who had ingested 100 mg L-proline three times a day 1 h after each meal; and (c) from subjects in Lin-xian who had ingested 100 mg ascorbic acid together with 100 mg L-proline three times a day 1 h after each meal. As an index of individual exposure to N-nitroso compounds or their precursors, ingested in food and/or formed endogenously, the levels of four urinary N-nitrosamino acids and nitrate were determined. The amounts of N-nitrosoproline, N-nitrosothiazolidine 4-carboxylic acid, N-nitrososarcosine, and nitrate excreted in the 24-h urine of undosed subjects in Lin-xian were significantly higher than those in Fan-xian, indicating a higher exposure of the inhabitants in the high-risk area to N-nitroso compounds and their precursors. Ingestion of L-proline resulted in a marked increase in urinary N-nitrosoproline levels in inhabitants from both areas, suggesting that endogenous nitrosation may occur to a larger extent when appropriate amine precursors are ingested in foods. Intake of moderate doses of ascorbic acid by high-risk subjects effectively reduced the urinary levels of N-nitrosamino acids to those found in undosed subjects in the low-risk area. If N-nitroso compounds formed in vivo are among the causative factors for esophageal cancer in this area of northern China, ascorbic acid appears to be effective in lowering the body burden of these carcinogenic compounds, thus offering a rational basis for long-term intervention studies in this area.

Classification of several tobamoviruses isolated in China on the basis of the amino acid composition of their virion proteins.

The amino acid compositions of the virion proteins of several tobamoviruses isolated in China were determined. A classification computed from these and published data of other tobamoviruses was compared with published data of their relatedness assessed using the cDNA:RNA molecular hybridization technique. The classifications are clearly congruent; that based on amino acid composition appears to be best for determining the hierarchical relationships of members within a taxonomic group, rather than for distinguishing between closely related members, whereas nucleic acid hybridization unequivocally places a virus into the appropriate subgroup but is less useful for determining distant relationships.

The pharmacokinetics of orally administered fudosteine in healthy Chinese volunteers.

The pharmacokinetics of fudosteine in healthy Chinese volunteers was investigated for the first time after single- and multiple-dose administration. Five male and five female volunteers were enrolled in this study. Each subject received 400 mg fudosteine capsules (the therapeutic dose) on day 1 after overnight fasting for the single-dose study and three times daily oral administration (400 mg) for 5 consecutive days until the sixth morning for the multiple-dose study. Serial blood samples were collected at specified time intervals up to 16 hours following the first and last doses of fudosteine. Plasma harvested from the blood was separated and analyzed for fudosteine levels by a validated high-performance liquid chromatography-electrospray ionization mass spectrometry (HPLC/ESI/MS) method employing percolumn derivatization with 9-fluorenylmethyl chloroformate (FMOC-Cl). Noncompartmental analysis was used for the calculation of the total area under the plasma concentration-time curve (AUC) from time zero to time infinity and the terminal half-life (t1/2) of fudosteine. The pharmacokinetic parameters for single- and multiple-dose administration were estimated as follows: Cmax amounted to 10.13+/-4.39 microg/mL and 11.75+/-6.51 microg/mL, tmax to 0.69+/-0.36 h and 0.53+/-0.12 h and t1/2 to 2.33+/-0.63 h and 2.40+/-0.37 h, respectively. No significant differences were found between single- and multiple-dose oral administration, although gender differences were observed.

Induction of micronuclei formation in preimplantation mouse embryos after maternal treatment with 2-bromopropane.

We examined effects of 2-bromopropane (2-BP), a chlorofluorocarbon replacement, on mouse embryonic mutagenicity. 2-BP was administered to pregnant mice intraperitoneally (i.p.) (300, 600, 900, and 1800 mg/kg) during the early preimplantation period. On day 3 of gestation, micronuclei (MN) frequency and embryo cell number were determined. 2-BP induced a dose-related significant increase in MN frequency and a treatment-related decrease in embryo cell number. Furthermore, the cell numbers were significantly smaller in the MN-positive embryos by two-way ANOVA taking it into account an interactive effect between 2-BP dose and the presence or absence of MN. A simultaneous decrease in cell number and increase in MN frequency may reflect an embryonic developmental disadvantage resulting from maternal treatment with 2-BP. Further study is needed to establish how 2-BP contributes to postimplantation embryonic development.
