[Changes in glucocorticoid receptor and phospholipase A2 in guinea pig asthma models].

In this study we determined the glucocorticoid receptor (GR) level (using 3H-dexamethasone as a ligand) and phospholipase A2 (PLA2) activity of lung tissue in guinea pig asthma models at 1st, 3rd, 7th and 14th day of asthma attacks. The results showed that the GR maximal binding capacity (Bmax, fmol/mg pro.) were 54.36 +/- 26.57, 30.99 +/- 10.01, 19.40 +/- 10.06, and 29.16 +/- 11.35; GR dissociation constant (Kd, nmol/L) were 12.73 +/- 4.06, 8.28 +/- 3.23, 5.70 +/- 2.91, and 6.40 +/- 2.64 respectively; and PLA2 actisis (U) were 181.58 +/- 41.50, 239.80 +/- 61.15, 243.08 +/- 31.35, and 235.71 +/- 86.13 respectively. The GR Bmax, Kd and PLA2 of actisis control group were 61.48 +/- 27.60 (fmol/mg pro.), 13.11 +/- 2.88 (nmol/L), 81.06 +/- 15.97 (U). The GR Bmax and Kd of asthmatic animals decreased significantly since the 3rd day, but the PLA2 activity increased significantly since the 1st day as compared to the control group (P < 0.01). The changes of GR Bmax had significant negative correlation with those of the PLA2 activity (r = -0.9023, P < 0.05). The mechanism of the GR and PLA2 actisis changes of asthma animal models were discussed.

[The genetic mechanism of intersexuality in milk goats of Saanen breed of Xinong].

Intersexual goats of Saanen breed of Xinong at Northwestern Agricultural University were studied in anatomy, histology, cytogenetics and genetics. The different types of intersexuality were seen, ranging from psuedohermaphroditic female to male types, including testicular hypoplastic and sex reverse male. All of them were genetic female type (60, XX), the karyotype, normal, so the intersexual bodies caused by the gene, which located at autosomes, having masculinizing effect, and in homozygote made female turn to male incompletely. It was pointed that intersexuality and polledness were controlled by different genes, the half century standing problem on intersexual inheritance of goats has been solved. The normal: the intersexual = 3:1, but the intersexual came from genetic female, this is a very special segregative ratio.

[The effect of a promoter P28-1 of Bacillus subtilis on Streptomyces differentiation].

A 1.1kb promoter P28-1 was inserted into pUC19. After then, the P28-1 was subcloned into the HindIII-EcoRI sites of the high copy number Streptomyces promoter probe plasmid pIJ4083 containing xy1E reporter gene. This recombinant plasmid was designated as pIJ4498. When pIJ4498 was introduced into Streptomyces coelicolor J1501 protoplasts, transformants conferred a white phenotype, whereas the vector pIJ4083 gave rise to colonies of normal, dark grey appearance which is the same as that of J1501 itself. After confirming pIJ4498 structure with some restriction enzymes, it was also introduced into whiG mutant (C71). Crude enzyme extracts were isolated from J1501/pIJ4498, J1501/pIJ4083 and C71/pIJ4498 respectively, the crude enzyme extract from J1501/pIJ4498 could oxidize catechol (colourless) to 2-hydroxy muconic semialdehyde (yellow colour), but crude enzyme extracts from J1501/pIJ4083 and C71/pIJ4498 could not oxidize catechol to 2-hydroxymuconic semialdehyde. The results indicated that P28-1 might be recognised by sigma whiG RNA polymerase, and activated the xylE reporter gene expression and reduced J1501 sporulation.

Transfer of shuttle vectors containing Bacillus thuringiensis toxin gene into wild-type B. cereus, B. brevis and B. subtilis by electroporation.

Gram positive and negative bacterial shuttle vectors carrying Bacillus thuringiensis (B.t.) toxin gene were introduced by electroporation into wild-type Bacillus cereus, B. brevis and B. subtilis. The transformation efficiencies for these bacteria were around 10(1)-10(4) transformants per micrograms DNA based on the numbers of neomycin- and ampicillin-resistant colonies produced. The structure of the transferred plasmids proved identical with the original ones both in size and restriction pattern. Toxicity assays showed that the transformants gave a mortality of 90-100% against caterpillar of Heliothis assulta, indicating that the gene function was not changed by electroporation.

Neurochemical evidence that 5-hydroxytryptaminergic neurons tonically inhibit noradrenergic neurons terminating in the hypothalamus.

The medial zona incerta (MZI) and dorsomedial nucleus of the hypothalamus (DMN), which contain cell bodies and terminals of incertohypothalamic dopaminergic (DA) neurons, are densely innervated by both noradrenergic (NE) and 5-hydroxytryptaminergic (5-HT) neurons. In view of emerging anatomical and pharmacological evidence suggesting possible interactions between 5-HT and catecholaminergic neurons, the effects of experimental procedures that inhibit or disrupt 5-HT neurons on the activities of catecholaminergic neurons terminating in these regions were examined in the present study. Catecholaminergic neuronal activity was estimated by measuring catecholamine synthesis (accumulation of 3,4-dihydroxyphenylalanine [DOPA] after administration of a decarboxylase inhibitor) and metabolism (concentrations of the dopamine metabolite 3,4-dihydroxyphenylacetic acid (DOPAC) and the norepinephrine metabolite 3-methoxy-4-hydroxyphenylethyleneglycol (MHPG)) in the MZI and DMN of both male and female rats. Inhibition of 5-HT neurons following administration of the 5-HT1A autoreceptor agonist 8-hydroxy-2-(di-n-propylamino)tetralin (8-OH-DPAT) increased the accumulation of DOPA in the DMN and the concentrations of DOPAC in the MZI and DMN, indicating an activation of catecholaminergic neurons in these regions. Concentrations of MHPG were increased in the MZI and DMN by 8-OH-DPAT or 5,7-dihydroxytryptamine-induced lesions of 5-HT neurons, revealing that NE neurons terminating in these regions were activated following procedures that decrease 5-HT neuronal function. Following destruction of NE neurons projecting to the MZI and DMN, 8-OH-DPAT no longer increased DOPAC concentrations in these brain regions.(ABSTRACT TRUNCATED AT 250 WORDS)

Effects of copper coordinate complexes on the scavenging of reactive oxygen species.

Effects of a 20-membered macrocyclic bicopper complex and dioxotetramine copper complexes with linear or cyclic structure on the scavenging of ROS have been studied by ESR. The results indicated that all three copper coordinate complexes have SOD mimetic activity. The dioxotetramine copper complex with linear or cyclic structure did not have the scavenging effect on .OH produced by H2O2-Fe++ and 1O2 produced by the H2O2-NaOCl system, but the 20-membered macrocyclic bicopper complex was found to be capable of depressing the intensity of the ESR spectrum of the adduct formed from TMP and 1O2 as well as making the spectrum of DMPO-OH adduct splittings. This fact may suggest that there are interactions between the ROS and 20-membered macrocyclic ring in the SOD mimic.

Synthesis of new amino acid and peptide derivatives of estradiol and their binding affinities for the estrogen receptor.

A series of amino acid and peptide derivatives of estradiol have been synthesized by coupling 17 beta-aminoestra-1,3,5(10)-trien-3-ol, 17-hydrazonoestra-1,3,5(10)-trien-3-ol with amino acids or peptides, using tetrahydrothiazole-2-thione, N-hydroxy-1,4-epoxycyclohex-5-ene-2,3-dicarbonylimide, benzotriazolyloxy-tris(dimethylamino)phosphonium hexafluorophosphate, and p-nitrophenol as reagents. N-protected peptidyl steroids were deprotected by traditional methods. The relative binding affinities of the deprotected derivatives to the estrogen receptor were determined by competitive radioligand binding assay.

Genomic concatemerization/deletion in rotaviruses: a new mechanism for generating rapid genetic change of potential epidemiological importance.

Three variants of group A rotavirus with large changes in their gene 5 structures have been analyzed at the molecular level. The first of these, P9 delta 5, was obtained during plaque purification undertaken as part of the biological cloning of a field isolate of virus. The gene 5 homolog in this isolate migrated just ahead of the normal segment 6 RNA, giving an estimated size of 1,300 bp. Molecular cloning and sequencing of this homolog revealed it to have a single 308-bp deletion in the center of the normal gene 5 sequence extending between nucleotides 460 and 768 of the normal gene sequence. This deletion caused a frameshift in the gene such that a stop codon was encountered 8 amino acids downstream of the deletion point, giving a predicted size for the protein product of this gene of 150 amino acids compared with the 490 amino acids of its normal-size counterpart. Attempts to detect this shortened protein in virus-infected cells were not successful, indicating that it was much less stable than the full-length protein and/or had suffered a large change in its antigenicity. The second two variants, brvA and brvE, were generated in an earlier study following the high-multiplicity passage of the UKtc strain of bovine rotavirus. Polyacrylamide gel electrophoresis analysis of these nondefective variants showed that brvA had a gene 5 homolog approximately equal in size to the normal RNA segment 2 (approximately 2,700 bp) and that brvE had a size of approximately 2,300 bp. Both variants showed changes in their gene 5 protein products, with brvA mimicking P9 delta 5 in failing to produce a detectable product whereas brvE produced a new virus-specific protein approximately 80 kDa in size. Full-length cDNA clones of the brvE gene 5 homolog were isolated, and analysis of their structure revealed a head-to-tail concatemerization of the normal gene 5 sequence with the first copy of the concatemer covering nucleotides 1 to 808 and the second covering nucleotides 92 to 1579, giving a total length of 2,296 bp. Sequencing across the junction region of the two copies of the gene showed that they were joined in frame to give a predicted combined open reading frame of 728 amino acids with the amino-terminal region consisting of amino acids 1 to 258 fused at the carboxy terminus to amino acids 21 to 490.(ABSTRACT TRUNCATED AT 400 WORDS)

Morphometry of the dog pulmonary venous tree.

The biophysical approach to the study of blood flow in the pulmonary vasculature requires a detailed description of vascular geometry and branching pattern. The description of the pulmonary venous morphometry in the dog is the focus of this paper. Silicone elastomer casts of a dog lung were made and were used to measure the diameters, lengths, and branching pattern of the pulmonary venous vasculature. The anatomic data are presented statistically with a diameter-defined Strahler ordering scheme, a rule for assigning the order numbers of the vessels on the basis of a diameter criterion. The asymmetric branching pattern of the pulmonary venous vasculature is described with a connectivity matrix. Results show that for the dog's right pulmonary venous tree 1) a total of 11 orders of vessels lay between the left atrium and the capillary bed; 2) the average ratios of the diameter, length, and number of branches of successive orders of veins were 1.701, 1.556, and 3.762, respectively; and 3) a fractal description of the tree geometry resulted in diameter and length fractal dimensions of 2.49 and 2.99, respectively. The morphometric data were used to compute the cross-sectional area, vascular volume, and Poiseuillean resistance in the venous vessels.

Neurochemical evidence that the inhibitory effect of galanin on tuberoinfundibular dopamine neurons is activity dependent.

The purpose of the present study was to examine the effects of galanin on the basal and stimulated activity of tuberoinfundibular dopaminergic (TIDA) neurons in male and female rats. TIDA neuronal activity was estimated by measuring dopamine (DA) synthesis [accumulation of 3,4-dihydroxyphenylalanine(DOPA) after administration of a decarboxylase inhibitor] and metabolism [ratio of 3,4-dihydroxyphenylacetic acid (DOPAC) to DA concentrations] in terminals of these neurons in the median eminence. Central administration of galanin (2 micrograms/rat, i.c.v.) produced a rapid (by 15 min) increase in plasma prolactin concentrations, but had no effect on the ratio of DOPAC to DA in the median eminence of either male or female rats. In contrast, galanin decreased the ratio of DOPAC to DA in the median eminence of both male and female rats whose TIDA neuronal activity was stimulated following administration of the DA antagonist haloperidol (1 mg/kg, s.c., 12 h). The galanin receptor antagonist galanin-(1-13)-bradykinin-(2-9)-amide had no effect on the accumulation of DOPA in the median eminence of male rats per se, but blocked the inhibitory effects of either exogenous or endogenous galanin on median eminence DOPA accumulation in haloperidol-treated rats. These results indicate that in both male and female rats, galanin-induced activation of prolactin secretion is not mediated by changes in tonic inhibition of hormone release by TIDA neurons, and that TIDA neurons are responsive to the inhibitory effects of galanin only under activated conditions.

Changes of pulmonary glucocorticoid receptor and phospholipase A2 in sheep with acute lung injury after high dose endotoxin infusion.

In a sheep model of acute lung injury induced by an Escherichia coli endotoxin (5 micrograms/kg) with chronic lung lymph fistula (n = 15), we measured the changes in glucocorticoid receptor (GCR) binding capacity in lung tissue by means of radioligand binding assay. The content of cortisol and the activity of phospholipase A2 (PLA2) were also measured. The results showed that the maximal binding capacity (Bmax) of GCR in lung cytoplasma decreased continuously 2 h (113 +/- 3 versus 66 +/- 2 fmol/mg protein, p < 0.01), 4 h (105 +/- 6 versus 52 +/- 3 fmol/mg protein, p < 0.01), and 6 h (105 +/- 5 versus 37 +/- 2 fmol/mg protein, p < 0.01) after endotoxin infusion. Its affinity decreased markedly (p < 0.05) at 6 h after the infusion. The contents of cortisol in plasma elevated at 0.5 h and remained at a high level until 4 h after the infusion. PLA2 activity rose from 97 +/- 25 to 188 +/- 12 U (p < 0.05), 99 +/- 13 to 285 +/- 25 U (p < 0.01), and 106 +/- 14 to 354 +/- 32 U (p < 0.01) at 2, 4, and 6 h after endotoxin infusion, respectively. There was a negative correlation between the Bmax of GCR and PLA2 activity (r = -0.87, p < 0.01). The findings indicate that there was a secondary GCR abnormality and a higher PLA2 activity during endotoxin-induced lung injury. The glucocorticoid hypofunction caused by reduced GCR binding capacity may accelerate the pathologic response of acute lung injury.(ABSTRACT TRUNCATED AT 250 WORDS)

Insect tolerance of transgenic Populus nigra plants transformed with Bacillus thuringiensis toxin gene.

Leaves and stem segments of Populus nigra were transformed with A. tumefaciens LBA4404 harboring a binary vector containing chimeric genes of NPT and 35S-omega-B.t. toxin-Nos. Nineteen regenerated kanamycin resistant plants were analyzed by DNA hybridization, out of which 10 were shown to be the candidates of transgenic plants. Insect tolerance tests showed that the transgenic plants were toxic to two lepidopteran pests, Lymantria dispar L. and Apocheimia cinerarius Erschoff. Based on the results of Southern blot of PCR products and the cluster analysis of their growth and insect resistance, three independent transgenic plants were selected, which were then propagated in the nursery field. This field test is currently in progress.

[Nucleotide sequence of the toxic domain of an insecticidal protein gene from B. thuringiensis subsp. kurstaki HD-1].

Two crystal protein genes, the 5.3kb and 6.6kb class respectively, from Bacillus thuringiensis subsp. kurstaki HD-1 (B. t HD-1) had been cloned previously. Based on the classification system of Hofte and Whiteley, these two genes should belong to Cry I A (b) and Cry I A (c) gene type respectively. The nucleotide sequence of the toxic domain of this Cry I A (c) gene from B. t kurstaki HD-1 is firstly reported here and compared with that of Cry I A (c) gene from B. t HD-73 and Cry I A (b) gene from B. t HD-1.

[Evaluation of pulmonary arterial pressure by pulsed Doppler echocardiography compared with cardiac catheterization].

Twenty-six patients undergoing right cardiac catheterization were examined by pulsed doppler echocardiography (PDE), spectrum analysis of the shape of flow velocity curve of the pulmonary arteria during systole was made. Pulmonary arterial pressure (PAP) was qualitatively and qualitatively evaluated by right ventricular systolic time interval (RSTI) derived from pulmonary doppler blood flow velocity pattern. The correlation between PAP and the values of RSTI was evaluated, multiple stepwise regression analysis was performed. The pulmonary flow pattern and the values of RSTI measured from right ventricular outflow tract (RVOT) and main pulmonary artery (MPA) were compared. The results showed that: 1. Qualitative evaluation of PAP: (1) the triangular configuration of the pulmonary flow pattern was the feature of the pulmonary ejection flow in pulmonary hypertension (PH). (2) AT < 100ms AT/ET < 0.30 and PEP/AT > 1.1 indicated PH (PAMP > or = 2.7 kPa, AT < 100 ms, sensitivity 92.3%, specificity 100%). 2. Quantitative evaluation of PAP: (1) simple regression analysis: AT, AT/PEP especially AT showed a good inverse correlation with PAP, which was improved when a logarithmic function was applied, but not improved by the square root of heart rate. The liner regression equation and the correlation coefficient were: log PASP = -6.3617 AT +2.825 (r = -0.7925, P < 0.001); log PADP = -0.4569 AT +74.182 (r = -0.8513, P < 0.001); log PAMP = -0.0073 AT +2.198 (r = -0.8635, P < 0.001). (2) multiple stepwise regression analysis: the stepwise analysis of six parameters (PEP, ET, AT, AT/ET, AT/PEP, square root of R-R) showed that AT, PEP were the best parameters correlated with PAP.(ABSTRACT TRUNCATED AT 250 WORDS)

[Changes of adrenoceptors and glucocorticoid receptor in the lungs of rats with experimental respiratory distress syndrome].

With the radioligand binding assay, the maximal binding capacity (Bmax) and affinity (Kd) of alpha, beta-adrenoceptors(alpha AR,beta AR) in the lung membrane and glucocorticoid receptor (GCR) in the lung cytoplasma of rats with experimental respiratory distress syndrome (RDS) induced by oleic acid have been measured. The results demonstrated that the content of alpha AR in rat lungs increased continuously during the experiment, the Bmax at 1st, 4th and 6th hour after oleic acid injection were 139 +/- 40, 127 +/- 12, 116 +/- 25 fmol/mg protein, significantly higher than normal value (83 +/- 7, n = 8-10, P < 0.01). Meanwhile, the content of beta AR and GCR decreased continuously, the Bmax at the same time were 364 +/- 18, 307 +/- 55, 240 +/- 66 and 146 +/- 28, 153 +/- 37, 150 +/- 32 fmol/mg protein respectively, significantly lower than their normal value (490 +/- 61, 227 +/- 14 fmol/mg protein, n = 6-10, P < 0.01). The results indicate that the changes of these receptors may be of significance in the pathogenesis of ARDS.

Characterization of opioid receptor-mediated regulation of incertohypothalamic dopamine neurons: lack of evidence for a role of 5-hydroxytryptaminergic neurons in mediating the stimulatory effects of morphine.

The purpose of the present study was to characterize opioid receptor-mediated regulation of incertohypothalamic dopaminergic (DA) neurons in the rat brain by examining the acute effects of selective mu or kappa opioid receptor agonists and antagonists on concentrations of 3,4-dihydroxyphenylacetic acid (DOPAC) in the medial zona incerta (MZI) and the dorsomedial hypothalamic nucleus (DMN) which contain cell bodies and terminals, respectively, of these neurons. Morphine caused a dose- and time-related increase in concentrations of DOPAC in MZI and DMN; this stimulatory effect was blocked by the mu opioid receptor antagonist naltrexone. In contrast, activation or blockade of kappa opioid receptors following administration of U-50,488 or nor-binaltorphimine, respectively, had no effect on DOPAC concentrations in either the MZI or DMN. The basal activity of incertohypothalamic DA neurons and their response to morphine was similar in male and female rats. Morphine also increased the concentrations of 5-hydroxyindoleacetic acid in MZI and DMN, indicating that morphine increases the activity of 5-hydroxytryptamine (5HT) neurons projecting to these regions. This might suggest that morphine-induced activation of incertohypothalamic DA neurons is mediated by 5HT neurons; but 5,7-dihydroxytryptamine-induced lesions of 5HT neurons did not alter the ability of morphine to increase DOPAC concentrations in MZI and DMN. These results indicate that the stimulatory effects of mu opioid receptor activation on incertohypothalamic DA neurons is not dependent upon the presence of 5HT neurons.

Sexual differences in the activity of periventricular-hypophysial dopaminergic neurons in rats.

The activities of periventricular-hypophysial dopaminergic (DA) neurons were compared in male and female rats by measuring dopamine synthesis (accumulation of 3,4-dihydroxyphenylalanine [DOPA] after inhibition of L-aromatic amino acid decarboxylase) and metabolism (concentrations of 3,4-dihydroxyphenylacetic acid [DOPAC]) in terminals of these neurons in the intermediate lobe of the pituitary. For comparison, the synthesis and metabolism of dopamine in the neural lobe of the pituitary and median eminence were also determined. The concentrations of DOPAC and accumulation of DOPA were higher in females than in males in both the intermediate lobe and median eminence, revealing a sexual difference in the basal activity of periventricular-hypophysial and tuberoinfundibular DA neurons. In contrast, there were no differences between male and female rats in activity of DA neurons terminating in the neural lobe. One week following gonadectomy, DOPA accumulation in the median eminence was decreased in females and increased in males, but remained unchanged in the intermediate lobe. These results indicate that sexual differences in the activity of periventricular-hypophysial DA neurons terminating in the intermediate lobe are not dependent upon the presence of circulating gonadal steroids, and in this respect, these neurons differ from tuberoinfundibular DA neurons.
