Determination of double decker sandwich structured La-substituted chlorophyll a by EXAFS.

As a representative natural porphyrin derivative, it is interesting to know the chemical form of lanthanide in chlorophyll a. We select the natural fern dicranopteris dichotoma, the most lanthanide-concentrated plant known, to isolate the chlorophyll a for our study. It is found by EXAFS that lanthanum substitutes in the magnesium position in chlorophyll and coordinates with the porphyrin ring. The lanthanum is seen to have a similar coordination structure to a sandwich-type lanthanide complex, with the La surrounded by eight nitrogen atoms from two porphyrin rings with average La-N bond length of 2.65 A.

A single peripheral injection of basic fibroblast growth factor (bFGF) stimulates granule cell production and increases cerebellar growth in newborn rats.

The control of neuronal number is critical for coordinating innervation and target organ requirements. Although basic fibroblast growth factor (bFGF) is known to regulate neuron number in the developing embryonic cortex, its potential role during postnatal brain development remains undefined. To address this issue, the cerebellum, a site of postnatal neurogenesis, was used. Previously, we found that a single peripheral injection of bFGF in newborn rats elicited mitosis of neuronal precursors in the external germinal layer (EGL) 8 h after administration. We now define the sustained effects of bFGF treatment on postnatal granule cell production and cerebellar growth. Seventy-two h after a single injection of bFGF (20 ng/g) in newborn rats, the fraction of BrdU-labeled cells in the EGL increased by 46% without altering apoptotic cell number, consistent with enhanced precursor proliferation. Moreover, bFGF increased mitotically labeled cells by 100% and total cell density by 33% in the internal granular layer (IGL), the final destination of the EGL precursors. Because cerebellar volume also increased by 22%, bFGF-induced proliferation enhanced generation of total IGL neurons and increased cerebellar growth. These morphometric measures were corroborated independently by using DNA quantitation: cerebellar DNA content increased 16% after bFGF injection, consistent with increased neuron number. Furthermore, using DNA quantitation as an index, increased total cerebellar cell number elicited by bFGF injection persisted beyond the neurogenetic period, until P35. We conclude that a single postnatal injection of bFGF increases granule neuron number and enhances cerebellar growth following mitotic stimulation.

Cryoelectron-microscopy image reconstruction of symmetry mismatches in bacteriophage phi29.

A method has been developed for three-dimensional image reconstruction of symmetry-mismatched components in tailed phages. Although the method described here addresses the specific case where differing symmetry axes are coincident, the method is more generally applicable, for instance, to the reconstruction of images of viral particles that deviate from icosahedral symmetry. Particles are initially oriented according to their dominant symmetry, thus reducing the search space for determining the orientation of the less dominant, symmetry-mismatched component. This procedure produced an improved reconstruction of the sixfold-symmetric tail assembly that is attached to the fivefold-symmetric prolate head of phi29, demonstrating that this method is capable of detecting and reconstructing an object that included a symmetry mismatch. A reconstruction of phi29 prohead particles using the methods described here establishes that the pRNA molecule has fivefold symmetry when attached to the prohead, consistent with its proposed role as a component of the stator in the phi29 DNA packaging motor.

Genetic factors related to racial variation in plasma levels of insulin-like growth factor-1: implications for premenopausal breast cancer risk.

The oral contraceptive pill is associated with a modest increase in the risk of early-onset breast cancer in the general population, but it is possible that the risk is higher in certain subgroups of women. The relative risk of breast cancer associated with oral contraceptive use has been reported to be higher for African-American women than for white women. African-American women also have a higher incidence of premenopausal breast cancer than white women. Circulating levels of insulin-like growth factor-1 (IGF-I) vary between ethnic groups and are positively associated with the risk of premenopausal breast cancer. In general, the plasma level of IGF-I is lower in women who take oral contraceptives than in women who do not. In an attempt to explain the observed ethnic difference in IGF-I levels with oral contraceptive use, we sought to identify polymorphic variants of genes that are associated with IGF-I levels and estrogen metabolism. We measured IGF-I and IGFBP-3 plasma levels in 503 nulligravid women between the ages of 17 and 35. All women filled out a questionnaire that included information about ethnic background and oral contraceptive use. Samples of DNA were used to genotype the women for known polymorphic variants in the IGF1, AIB1, and CYP3A4 genes. Black women had significantly higher mean IGF-I levels than white women (330 ng/ml versus 284 ng/ml; P = 0.001, adjusted for age and oral contraceptive use). IGF-I levels were significantly suppressed by oral contraceptives in white women (301 ng/ml versus 267 ng/ml; P = 0.0003), but not in black women. Among oral contraceptive users, the IGF-I level was positively associated with the absence of the IGF1 19-repeat allele (338 ng/ml versus 265 ng/ml; P = 0.00007), with the presence of the CYP3A4 variant allele (320 ng/ml versus 269 ng/ml; P = 0.01), and with the presence of the AIB1 26-repeat allele (291 ng/ml versus 271; P = 0.08). After adjusting for genotypes, ethnic group was no longer a significant predictor of the IGF-I level. IGF-I levels are higher among black than white women. Polymorphic variants in the CYP3A4, IGF1, and AIB1 genes are associated with increases in the plasma levels of IGF-I among oral contraceptive users and the variant alleles are much more common in black women than in white women. The high incidence of premenopausal breast cancer among black women may be mediated through genetic modifiers of circulating levels of IGF-I.

Rare earth elements in naturally grown fern Dicranopteris linearis in relation to their variation in soils in south-Jiangxi region (southern China).

Ferns (Dicranopteris linearis) and soils, sampled from four rare earth mining areas and one non-mining area locating in South-Jiangxi region, were chosen for analysis of 15 rare earth elements (REEs) by inductively coupled plasma-mass spectrometry (ICP-MS). The fern samples were divided into 4 parts: root, stem, petiole, and lamina. The soils of the same sites were also sampled, and 4 soil layers were observed at the profile: A (0-20 cm depth), B (20-150 cm depth), C (150-400 cm depth), and D (rock). The distribution patterns and transportation characteristics of REEs of different soil layers and of different parts of D. linearis were studied. The results showed that in the soil layers, the sigma REE (the sum of the concentrations of 15 REEs) of A layer, as well as B layer were lower than that of C layer, but Ce was relatively concentrated in A layer. The sigma REE of different parts of D. linearis were lamina, root > stem > petiole. The REE distribution patterns of D. linearis root were similar to the total REE distribution patterns of the A layer of their host soils, especially to the soluble REE distribution patterns of the A layer of their host soils. The REEs were fractionated in their transportation in D. linearis. The abundances of heavy rare earth elements (HREEs, Gd to Lu and Y) in the stem, petiole, lamina are lower than those in the root.

Polyurethaneurea/vinyl polymer hybrid aqueous dispersions based on renewable material.

A series of polyurethaneurea-vinyl polymer (PUA) hybrid aqueous dispersions were synthesized from castor oil and/or difunctional poly(oxypropylene) polyol (GE-210, M(n) = 1000), butyl acrylate, and styrene. The effect of hybrid between the polyurethaneurea (PUU) and vinyl polymer (PA) on the morphologies and the mechanical properties for these PUA films was examined and studied. The experimental results showed that there was appreciable miscibility between the PUU and PA phases for the PUA prepared with GE-210, giving rise to less ordered hard segments in PUU and resulting in lowering of some mechanical properties compared with those of pure PUU synthesized with GE-210. For PUA based on castor oil, the compatibility between the PUU and PA phases was also observed, although there was a network structure in the system. Moreover, the particle-accumulation morphology during the film formation process for this PUA system disappeared, but this behavior was found for a similar PUU. In this case, a great improvement of mechanical properties for such a PUA specimen was observed. The other PUA specimens with high content of castor oil exhibit excellent comprehensive mechanical properties, resulting from the reinforcement of cross-linked PUU phase existing in the systems.

Positive and negative TAF(II) functions that suggest a dynamic TFIID structure and elicit synergy with traps in activator-induced transcription.

Human transcription factor TFIID contains the TATA-binding protein (TBP) and several TBP-associated factors (TAF(II)s). To elucidate the structural organization and function of TFIID, we expressed and characterized the product of a cloned cDNA encoding human TAF(II)135 (hTAF(II)135). Comparative far Western blots have shown that hTAF(II)135 interacts strongly with hTAF(II)20, moderately with hTAF(II)150, and weakly with hTAF(II)43 and hTAF(II)250. Consistent with these observations and with sequence relationships of hTAF(II)20 and hTAF(II)135 to histones H2B and H2A, respectively, TFIID preparations that contain higher levels of hTAF(II)135 also contain higher levels of hTAF(II)20, and the interaction between hTAF(II)20 and hTAF(II)135 is critical for human TFIID assembly in vitro. From a functional standpoint, hTAF(II)135 has been found to interact strongly and directly with hTFIIA and (within a complex that also contains hTBP and hTAF(II)250) to specifically cooperate with TFIIA to relieve TAF(II)250-mediated repression of TBP binding and function on core promoters. Finally, we report a functional synergism between TAF(II)s and the TRAP/Mediator complex in activated transcription, manifested as hTAF(II)-mediated inhibition of basal transcription and a consequent TRAP requirement for both a high absolute level of activated transcription and a high and more physiological activated/basal transcription ratio. These results suggest a dynamic TFIID structure in which the switch from a basal hTAF(II)-enhanced repression state to an activator-mediated activated state on a promoter may be mediated in part through activator or coactivator interactions with hTAF(II)135.

Composition, characteristic and activity of rare earth element-bound polysaccharide from tea.

The compositions and structural characteristics of rare earth elements-bound polysaccharides from tea (REE-TPS) were studied with the methods of Inductively Coupled Plasma Mass Spectrometry (ICP-MS), Gas Chromatography (GC) and Extended X-ray Absorption Fine Structure (EXAFS) spectroscopy. The results show that polysaccharide from tea (TPS) was a sort of glycoprotein and coordinated with Rare Earth Elements (REE) closely. The sugar fraction was composed of Rha, Ara, Xyl, Fuc, Glc, and Gal. There existed almost all natural amino acids with Glx, Asx, and Hyp as the major parts in the protein fraction. The REEs in REE-TPS were mainly composed of La, Ce, and Nd, especially, more than 75% of them was La. The coordination atom of the first coordination shell of La in REE-TPS was oxygen, the coordination number of which was 6, and the average distance between the atoms was 2.52 A. The second shell was formed from sulfur atoms, the coordination number and the average distance were 3 and 2.91 A, respectively. The bio-experiments show that REE-TPS could decrease the content of blood glucose in mice significantly.

Effect of lanthanide chloride on photosynthesis and dry matter accumulation in tobacco seedlings.

To investigate the physiological effects of rare earth ions, we have studied the effect of LaCl3 on the photosynthetic light reactions in tobacco (Nicotiana tobacum). When treated with 5-20 mg/L LaCl3 in Hoagland solution by water culture, the dry matter accumulation of different parts in tobacco, the content of chlorophyll increased gradually, but decreased when the concentration of LaCl3 was > or = 50 mg/L. The optimum concentration for growth appeared to be about 20 mg/L of LaCl3 in nutrient solution. La3+ promoted the activities of the Hill reaction, Mg2(+)-ATPase, and stimulated the rate of photophosphorylation in chloroplast at low concentrations, but inhibited them at high concentrations. It is concluded that La3+ stimulated the growth of tobacco seedlings and accelerated the photosynthetic light reactions at suitable concentration in vivo.

The EXAFS study on the local structure of lanthanum in spinach PSII.

The complex of photosystem II (PSII) had been prepared from spinach by treatment with Triton X-100. The PSII, which had been depleted of the extrinsic 17- and 23-kDa polypeptides, was obtained by exposing the solution to a high concentration of NaCl, and the complex of PSII-La3+ was prepared by treatment with LaCl3. The result indicated that La3+ could inhibit the oxygen-evolution activity of PSII by replacing the Ca2+. The local structural environment of La in PSII has been also studied by using extended X-ray absorption fine structure (EXAFS). The primary result of EXAFS indicated that La coordinated with eight oxygen and/or nitrogen atoms, with the distance of the La-O/N bond being 2.5 A. In addition, La coordinated with four carbon atoms, with a distance of 3.5 A in the second shell. In the third shell, La coordinated with two manganese atoms, with the distance of La-Mn bond being 4.49 A, and it was also found that the La-Mn distance (4.49 A) was longer than that of Ca-Mn (3.3 A) (1) in PSII.

Genetic and nongenetic factors associated with variation of plasma levels of insulin-like growth factor-I and insulin-like growth factor-binding protein-3 in healthy premenopausal women.

Circulating levels of insulin-like growth factor-I (IGF-I) and insulin-like growth factor-binding protein 3 (IGFBP-3) vary considerably between normal individuals. Recent epidemiological studies have provided evidence that these levels are predictive of risk of several common cancers. To evaluate possible sources of variation of the levels of circulating IGF-I and IGFBP-3 in females, we studied specific candidate genetic and nongenetic factors in 311 nulliparous, premenopausal Caucasian women, 17-35 years of age. Women who used oral contraceptives (OC) had reduced levels of IGF-I (269 versus 301 ng/ml; P = 0.001 adjusted for age) and increased levels of IGFBP-3 (4213 versus 4009 ng/ml; P = 0.002, adjusted for age) compared with nonusers. The ratio of IGF-I:IGFBP-3 was associated with the dose of estrogen contained in the OC (P(trend) = 0.006, adjusted for age). We identified a novel single bp polymorphism in the promoter region of the gene encoding IGFBP-3. This polymorphism was related to the level of IGFBP-3 in the circulation. Mean IGFBP-3 levels were 4390, 4130, and 3840 ng/ml for the AA, AC, and CC genotypes, respectively (P(trend) = 0.006, adjusted for age and OC use). We observed no effect of a recently described polymorphism in the promoter region of the gene encoding IGF-I on the plasma IGF-I level, but there was evidence for a modifying effect of this locus on the influence of OC on the IGF-I level. Our results support the view that circulating IGF-I levels and IGFBP-3 levels are complex traits and are influenced by a number of interacting genetic and nongenetic factors.

[Effect of lead exposure in prenatal and postnatal duration on infant growth].

The blood lead (PbB) levels of pregnant women and their babies were followed up since the third month of pregnancy until their babies were 2 years old. The general condition of delivery, family history of the pregnant women, feeding and development of infants were investigated. Prenatal and postnatal PbB levels and their effects on infant growth were analysed. The result showed a significant negative correlation of PbB level of pregnant women near delivery, and in umbilical cord with the growth of infants. Multiple regression analysis showed a negative correlation between PbB level and height, weight of infant at 1 and 2 years old.

[A study on the relationship between respiratory tract viruses antigen in peripheral blood mononuclear cells and their antibody from children with steroid-sensitive and simple nephrotic syndrome].

OBJECTIVE: To explore whether respiratory tract viruses infection was involved in the pathogenesis of sterioid-senstive and simple nephrotic syndrome(SSSNS). METHODS: Using APAAP and ELISA, the authors assayed the expression of respiratory tract viruses antigen in peripheral blood mononuclear cells (PBMC) and the production of viruses antibody in serum from 20 cases of children with steroid-sensitive and simple nephrotic syndrome. RESULTS: The positive rate of viruses in the active stage group of SSSNS children increased (P < 0.05), compared with that in the remission group and the healthy control group. The positive rate of virus detection in SSSNS children was closely related with the quantity of urinary protein(P < 0.05). The positive rate of virus detection in the group with urinary protein > or = 50 mg/(kg.d) was higher than that in the group with urinary protein < 50 mg/(kg.d). CONCLUSION: The onset of SSSNS may be related to the infection of respiratory tract viruses.

[Correlation of p16 mutation and biological behavior in Chinese laryngeal cancer].

OBJECTIVE: To investigate the correlation between p16 gene mutation and laryngeal cancer biological behavior as well as its prognosis in laryngeal cancer. METHOD: 24 specimens of primary laryngeal cancer and 10 speciments with benign lesion in larynx were examined for mutations in exon2 of p16 by using PCR-SSCP silver stainning technique. RESULT: Mutations frequency of laryngeal cancer was 62.5% (15/24). Nothing was found in 10 cases with laryngeal benign lesion. CONCLUSION: There is a strong correlation between p16 gene mutation and the biological behavior of chinese laryngeal cancer, such as histologic differentiation, invasion stage, and regional lymph nodes metastasis (P < or = 0.05). PCR-SSCP silver stainning technique is one of the most sensitive and simplest measure for detecting genetic mutation. It is worth using in clinical laboratory because of its readiness, repetition and lower cost.

[Effect of high glucose condition on the secretion of TGF beta by cultured macrovascular endothelial cells of the diabetic rat].

OBJECTIVE: This study was designed to examine the secretion of TGF-beta by cultured rat thoracic aortic endothelial cells of the diabetic rats under high gluocose condition. METHODS: First we created diabetic rat models, then the thoracic aortic endothelial cells of the diabetic rats were isolated and cultured in 10% FBS-RPMI 1640 media with various high glucose concentrations (12.5 mmol/L, 25 mmol/L or 50 mmol/L D-glucose), high glucose and TNF-alpha or high glucose and insulin for 48 hours, and the supernatant obtained from the cultured aortic endothelial cells was collected. The level of TGF-beta 1 in the supernatant was measured using a standard bioassay, which utilizes the TGF-beta 1 sensitive Mv1Lu (CLL-64) Mink lung epithelial cell line, and the production of endothelin (ET) in the supernatant was also assessed by RIA (radio immol/Lunoassay). RESULTS: Both total TGF-beta 1 (406 +/- 29 vs 189 +/- 23 pg/ml, P < 0.05) and active TGF-beta 1 (161 +/- 24 vs 39.5 +/- 8 pg/ml) secreted by the cultured aortic endothelial cells under high glucose condition (25 mmol/L) were significantly increased as compared with control. TNF-alpha could enhance the effect of high glucose on the secretion of TGF-beta 1, whereas insulin could inhibit it. With various high glucose concentrations, the levels of ET were increased. Also TNF-alpha enhanced it and insulin inhibited it. CONCLUSION: TGF-beta 1 may be involved in the development of diabetic macrovascular complication.

[Clinical study of the sentinel lymph node of patients with laryngeal and hypopharyngeal carcinomas].

OBJECTIVE: To investigate the methods of detecting the sentinel lymph node of laryngeal and hypopharyngeal carcinomas and its predictive value in the cervical metastasis of the carcinoma. METHODS: In 29 patients who suffered from laryngeal or hypopharyngeal carcinoma with NO neck, the patent blue was injected into the surrounding tissue of the tumor during the operation to identify the sentinel lymph nodes. The sentinel lymph nodes were dyed blue. The frozen histopathology was done during the operation, the lymph nodes at the ipsilateral side of the neck were dissected completely, and the routine histopathology was done as the gold standard to study the predictive value of the sentinel lymph node in the metastasis of the cervical node. RESULTS: Of 29 patients, 28 patients' sentinel lymph nodes were detected successfully during operation. There was an average of 2.5 lymph nodes per side per patient. Most of the sentinel lymph nodes were in the level II and level III regions of the ipsilateral side of the neck, and there were bilateral sentinel nodes in patients suffered from superglottic carcinoma. Three patients' sentinel lymph nodes were found to be positive in the frozen inspection, and the routine histopathology confirmed the result. The micrometastasis rate was 10.7% (3/28). There were no metastatic lymph nodes found in patients who were negative for the sentinel lymph nodes during the operative frozen histopathology. The predicted value of the sentinel lymph nodes to the cervical lymph node metastasis was 100 per cent. CONCLUSION: There is a very important predicted value of sentinel lymph nodes in the cervical metastasis of patients suffered from laryngeal and hypopharyngeal carcinomas. It could reduce the neck dissection in patients with laryngeal and hypopharyngeal carcinomas.

Structure of the bacteriophage phi29 DNA packaging motor.

Motors generating mechanical force, powered by the hydrolysis of ATP, translocate double-stranded DNA into preformed capsids (proheads) of bacterial viruses and certain animal viruses. Here we describe the motor that packages the double-stranded DNA of the Bacillus subtilis bacteriophage phi29 into a precursor capsid. We determined the structure of the head-tail connector--the central component of the phi29 DNA packaging motor--to 3.2 A resolution by means of X-ray crystallography. We then fitted the connector into the electron densities of the prohead and of the partially packaged prohead as determined using cryo-electron microscopy and image reconstruction analysis. Our results suggest that the prohead plus dodecameric connector, prohead RNA, viral ATPase and DNA comprise a rotary motor with the head-prohead RNA-ATPase complex acting as a stator, the DNA acting as a spindle, and the connector as a ball-race. The helical nature of the DNA converts the rotary action of the connector into translation of the DNA.

Mapping of glucocorticoid receptor DNA binding domain surfaces contributing to transrepression of NF-kappa B and induction of apoptosis.

Glucocorticoids (GCs) function, in part, through the ability of the glucocorticoid receptor (GR) to activate gene expression and in part through the transrepression of AP-1 and NF-kappaB. Here we characterize the effect of GR DNA binding domain (DBD) mutations, previously analyzed for changes in the ability to activate gene expression or transrepress AP-1. We have identified a GR mutant capable of distinguishing between transrepression of NF-kappaB and AP-1. Using circular dichroism spectroscopy, we show that this mutation does not appreciably alter GR DBD conformation, suggesting that functional differences between the mutant and wild type protein are the result of an alteration of a specific interaction surface. These data suggest that transrepression of NF-kappaB and AP-1 occurs through distinct protein-protein interactions and argue against the hypothesis that transrepression occurs through competition for a single coactivator protein. Introduction of these mutations into GC-resistant CEM lymphoblastic T cells restored dexamethasone (DEX)-mediated apoptosis as did wild type GR regardless of whether these mutants were transrepression or activation defective. Thus, DEX-mediated apoptosis in transformed T cells is more complex than originally appreciated.
