Transcription of IL-2 receptor gene is stimulated by ATL-derived factor produced by HTLV-I(+) T cell lines.

In HTLV-I transformed T-cell lines established from the patients with adult T-cell leukemia (ATL), there is a constitutive activation of the normal IL-2 receptor (IL-2-R) gene. These cell lines continuously produce an ATL-derived factor (ADF), an IL-2-R inducing factor without IL-2 activity. ADF enhances the expression of the IL-2-R through the augmentation of the IL-2-R mRNA in the HTLV-I(+) T-cell line (ED) as well as the NK cell line cells (YT). In YT cells, the transcriptional initiation of the promoter of the IL-2-R gene was enhanced by ADF but not by IL-2. Production of ADF by HTLV-I(+) T-cell lines may be involved in the abnormal expression of IL-2-Rs on these cells.

Interleukin-2 receptor-inducing factor(s) in adult T cell leukemia.

The expression of the interleukin-2 receptor (IL-2-R) is regulated by transcriptional and post-transcriptional mechanisms. IL-2-R gene expression is induced by pharmacological agents including calcium ions, phorbol esters such as phorbol myristate acetate (PMA) and forskolin (FK), a direct activator of adenylate cyclase. HTLV-I(+) leukemic T cells and T cell lines from patients with adult T cell leukemia (ATL) continuously expressed IL-2-R without production of IL-2. However, there was no abnormality of the structural gene for IL-2-R in these cell lines as well as in fresh leukemic cells of ATL. We have detected that many HTLV-I(+) T4(+) T cell lines constitutively produce a non-IL-2 lymphokine named ATL-derived factor (ADF), which induced the expression of the high-affinity IL-2-R on a variety of cells including HTLV-I(+) T cells, myeloid leukemia cells and YT cells. IL-2-R-inducing agents such as ADF and FK were shown to induce elevation of the mRNA levels for IL-2-R through transcriptional enhancement of the IL-2-R gene. The possible involvement of IL-2-R-inducing cytokines in the physiological lymphocyte activation and the leukemogenesis in ATL and other T cell leukemias is discussed.

Immunohistochemical identification of somatostatin-containing neurons projecting to the median eminence of the rat.

Immunohistochemical staining of somatostatin (SRIF) and a retrograde transport method with horseradish peroxidase (HRP) were simultaneously applied to the same section of the rat brain to identify the specific SRIF-containing neurons sending their fibers to the median eminence. After HRP injection into the median eminence, SRIF-positive neurons in the rostral parts of the periventricular nucleus were shown to be labeled with HRP, SRIF neurons in the other brain areas, such as the amygdala, the ventromedial nucleus, dorsomedial nucleus and arcuate nucleus, had no HRP-positive material. The present findings appear to demonstrate that SRIF neurons in the periventricular nucleus project directly to the median eminence.

Biological features of the skin of the pika, Ochotona rufescens rufescens: histological structure and enzymatic histochemical reactivity.

To examine the usefulness of the pika, Ochotona rufescens rufescens, as an experimental animal for skin irritability tests, the histological structure and enzymatic histochemical reactivity of pika skin were investigated. The pika had a hair cycle similar to that of the rabbit. The skin and epidermis of the pika trunk were 1.16mm and 29.5 microns thick, on the average, respectively. Both of them were the thickest in the dorsal region followed by the interscapular area, while they were the thinnest in the abdominal region. In the epidermis of the pika, the strata corneum, granulosum, spinosum and basale were rather clearly distinguished. The cell arrangement in the stratum basale was more compact than that in the rabbit. Dermal mast cells, which are distributed in the stratum reticulare in rabbits and guinea pigs, were distributed in the stratum papillare right beneath the epidermis. The mast cell of the pika in the TEM images had granules of low electron density and with relatively long microvilli and rather large mitochondria. The activities of the enzymes, SDH, MDH, LDH, beta HBDH, alpha GPDH, ALD, G6PDH and GPR, in the hair follicles and sebaceous glands of the pika were similar to those of the rabbit.

Differentiation of striated muscle fibers in pituitary gland grafts transplanted beneath the kidney capsule.

Striated muscle fibers appear with regularity in pituitary isografts transplanted beneath the kidney capsule. These differentiated striated muscle fibers consist of multinucleated cells resembling skeletal muscle fibers. They appear mainly in the pars distalis, though occasionally in the pars intermedia of the grafts. Although most glandular cells disappeared due to necrosis in the center of the grafted anterior pituitary, where the striated muscle fibers occurred, the folliculo-stellate cells were well preserved in this region and formed elongated myotube-like structures. The muscle fibers appearing in the central pars distalis were in close contact to the folliculo-stellate cells and also to the marginal epithelium of the pituitary cleft. In these cases the striated muscle fibers and folliculo-stellate cells were surrounded by a common basal lamina. This close association between heterotopically differentiated muscle fibers and the normally occurring folliculo-stellate cells strongly suggests a close relationship in differentiation and function between these different cells.

[The study in the relationship between serum calcium and serum parathyroid hormone (PTH) by employing the various kits of PTH assay].

In order to evaluate the influences of serum PTH assay in the various concentrations of serum calcium, we divided into three groups which serum calcium had below 8.0 mg/dl, 8.2 mg/dl to 9.8 mg/dl and above 10.0 mg/dl at random samples and assayed PTH in serum sample, using various kits of PTH assay obtained from commercial sources. Our results suggested that the measurement of serum PTH influenced by the concentration of serum calcium and therefore, should be taken an attention of serum calcium in each sample.

[The measurement of serum human parathyroid hormone (h-PTH53-84) and effect of exercise on calcium metabolism].

This study was focussed our attention on the measurement within the upper physiological level of human serum parathyroid hormone (PTH), using kits of human PTH 53-84. This assay kit was able to detect serum PTH in sera with subtle changes of serum calcium concentrations before and after short term exercise. These serum PTH levels before and after exercise seemed to be changed within the upper physiological levels of PTH. Thus, this study suggested that the assay kit was likely to become a useful tool of the measurement of the physiological level of serum PTH in humans.

[A case of intrasellar meningioma with panhypopituitarism and hyperprolactinemia].

A case of intrasellar meningioma is reported. A 49-year-old woman was admitted to our hospital on July 22, 1985, complaining of reduced visual acuity and visual field defect. Visual acuity was 0.6 in the right eye and 0.1 in the left eye. Visual field examination revealed upper temporal quadrantanopsia on the right side and incomplete temporal hemianopsia on the left side. Ocular fundi were normal. X-ray films of the skull showed a balloon-shaped sella turcica with "double floor". CT scan showed a isodense mass with central low density occupying the intrasellar and suprasellar region. After administration of contrast medium, almost homogenous enhancement was noted. Bilateral carotid angiographies demonstrated that horizontal portion of the right anterior cerebral artery was raised. No tumor blush was evident. Endocrinologic function tests confirmed a complete deficit of the anterior lobe hormones except for elevated serum prolactin level of 110 ng/ml. She showed no galactorrhea. On August 6, 1985, the sella turcica was reached via the transsphenoidal rhinoseptal approach. The sella floor and dura matter were intact. The grey, soft and necrotic tumor tissue was encountered and bleeding was controllable. The tumor extending to suprasellar region was firm in consistency. Pathologically, the tumor was a typical meningothelial meningioma. Postoperatively, visual field defect improved and visual acuity was recovered on the right side immediately. Postoperative CT scan showed a thin residual enhanced lesion, which was the attachment of the tumor. It seemed to be the elevated diaphragma sellae. Clinical observation, radiological and endocrinological findings of intrasellar meningioma are similar to that of non-functioning pituitary adenoma.(ABSTRACT TRUNCATED AT 250 WORDS)

[Ifosfamide treatment for children with malignant tumor].

Seven drugs were administered to nude mice with transplanted TNB9 neuroblastoma. DTIC, cyclophosphamide and ifosfamide were the first-line drugs against TNB9. Chemotherapy including ifosfamide was effective to three of six children with malignant solid tumor resistant to cisplatin and/or cyclophosphamide. Two of the patients who were pretreated with radiotherapy were found to have hemorrhagic cystitis after ifosfamide administration. It therefore seems that chemotherapy with ifosfamide should be withheld after radiotherapy.

Interleukin 1 alpha mRNA in virus-transformed T and B cells.

IL-1 alpha cDNA clone was isolated from a T cell line infected by the human T lymphotropic retrovirus type-I (HTLV-I/ATLV). We found significant amounts of mRNA hybridizing to IL-1 alpha cDNA not only in HTLV-I-transformed T cells but also in Epstein-Barr Virus-transformed B cells. A part of IL-2 receptor inducing activity in Adult T cell leukemia (ATL) cell line seems to be due to IL-1 alpha.

Congenital stationary night blindness with myopia: a clinico-pathologic study.

The left eye of a 77 year old male patient was enucleated because of absolute glaucoma. This eye showed subnormal electroretinogram (ERG) and oscillatory potentials preoperatively, but the other eye showed Schubert-Bornschein type ERG and monophasic dark adaptation curve. Light and electron microscopic studies of the left eye showed a normal arrangement of discs of rod outer segments, normal synaptic ends of the photoreceptors, and complete loss of ganglion cells. From the subnormal ERG in the left eye we assumed it was originally Schubert-Bornschein type ERG but inverted to subnormal type ERG following the loss of inhibitory mechanisms. Thus we propose that the cause of night blindness in congenital stationary cases with Schubert-Bornschein type ERG may be related to the mechanisms inhibitory to cells of the bipolar layer.

Expression of interleukin 2 receptors on interleukin 3-dependent cell lines.

Several mouse IL 3-dependent cell lines, IC2, LT4, FDC-P2, and PB-3C, derived from spleen or bone marrow cells were shown to express low affinity receptors for IL 2 (Kd; 0.5 to 8 X 10(-8) M). High affinity receptors for IL 2 were not detected on the IL 3-dependent cells within the experimental limitation of this study. The clones did not respond to IL 2 at all at the concentration as high as 25 micrograms/ml. The number of the receptors expressed on those clones was estimated to be 0.2 to 2 X 10(5)/cell, which is comparable with the number of those on IL 2-dependent T cell clones. Expression of IL 2 receptor was confirmed in mRNA levels for both IC2 and LT4 cells. A relatively low level expression of one (4.5 Kb) of four IL 2 receptor mRNA species was observed with those IL 3-dependent clones compared with IL 2-dependent T cells. It seems that these low affinity receptors may be expressed on IL 3-dependent cells that undergo differentiation or maturation in mast cell and some myeloid cell lineages.

Lack of correlation between in vitro corticosteroid effect on hemopoietic colony formation and response to corticosteroid therapy in aplastic anemia.

We performed hemopoietic colony culture assays in 15 patients with aplastic anemia (AA) in order to test the effect of hydrocortisone (HC) on late erythroid colony (CFU-e) formation of the patients' marrow and to correlate the in vitro culture results with the clinical response to corticosteroid therapy. HC enhanced CFU-e growth in four patients. All four patients failed to respond to corticosteroid, but three improved with with androgens. The addition of HC did not increase CFU-e colony formation in 11 patients. However, two of them responded to corticosteroid therapy. Among the nine patients showing no HC effect in vitro, two subsequently improved with androgens and one each with anti-thymocyte globulin and anti-lymphocyte globulin. The results suggest that the in vitro corticosteroid effect may not necessarily correlate with responsiveness to corticosteroid therapy.

A Zn2+-dependent and histone H1-specific protease in sea urchin sperm.

Protease activity was extracted from sea urchin sperm with 1% Triton X-100 and partially purified by DEAE-cellulose and Sephadex G-100 chromatography. The enzyme preferentially degraded histone H1, while showing only a weak activity toward other histones. Heat-denatured casein and bovine serum albumin were not digested by this enzyme under the present experimental conditions. This protease hydrolyzed only Boc-Val-Leu-Lys-MCA among various peptidyl-MCAs. The optimal pH ranged from 7 to 11. Its molecular weight was about 41,000. Among various known inhibitors of proteases, only omicron-phenanthroline effectively inhibited the activity. The enzyme was stimulated by Zn2+ or Co2+. It was inactivated by omicron-phenanthroline but could be reactivated by the addition of Zn2+ or Co2+. Therefore, this protease seems to be a metalloprotease dependent on Zn2+ or Co2+. The insensitivity of this enzyme to phosphoramidon and its very restricted substrate specificity suggest that this enzyme is very different from other metalloproteases described hitherto.
