Preparation and characterization of neutralizing monoclonal antibodies to feline calicivirus.

Fourteen neutralizing monoclonal antibodies (N-MoAbs) were prepared against the F4 strain of feline calicivirus (FCV), the prototype strain of FCV in Japan, and examined for their ability to neutralize FCV isolates. Neutralization-resistant variants of the F4 strain were selected under the presence of 4 individual N-MoAbs in cell culture systems and used in cross-neutralization tests and enzyme-linked immunosorbent assay with all of the 14 N-MoAbs. The results revealed the identification of at least two antigenic determinants on FCV F4: one being more broadly conserved among FCV isolates than the other. Usefulness of antigenic variants resistant to N-MoAbs for analysis of neutralization determinants on FCV was also demonstrated.

Peripheral neuropathy associated with osteosarcoma in a Japanese monkey.

Peripheral neuropathy associated with osteosarcoma of the humerus was reported in a Japanese monkey (Macaca fuscata). The monkey developed osteosarcoma in the right humerus. Postmortem morphometric analyses on lower limb nerves revealed loss and size reduction of myelinated nerve fibers. Pathological findings including various demyelinative processes were found with electron microscopy. These findings were more predominant in the distal part than in the proximal part of the nerves, which is compatible with axonal degeneration of peripheral nerve fibers. This is the first case found in a monkey of dying-back neuropathy associated with malignancy, which is not infrequent in humans.

[An experimental study of sensory innervation of the ureter in the dog using wheat germ agglutinin-horseradish peroxidase conjugates].

The sensory innervations of the ureter in the dog were studied using the anterograde and retrograde axonal transport of wheat germ agglutinin-horseradish peroxidase conjugates (WGA-HRP). Following upper ureteral injections, labeled cells were observed in the ipsilateral T7-L3 spinal ganglia to the injection site, with the greatest concentration at the T12-L2 levels. And labeled cells were seen in the contralateral T7--L3 spinal ganglia to the injection site with the greatest concentration at the T10 and L3 ganglia. Lower ureter injections resulted in the labeling of ipsilateral T11-Co1 and contralateral T9-Co1 spinal ganglia, with highest concentration at the ipsilateral L3 and S2 levels. Following thoracic and lumbar spinal ganglia T12, L1-L3 injections labeled fibers bundles were observed in the adventitia of the upper and lower ureter. Some labeled fibers were bifurcated from these bundles and passed through the two layers of the smooth muscles. In tunica submucosa and tunica propria mucosae, many labeled fibers were observed. A few labeled fibers were seen in the epithelium. After injections into the sacral and coccygeal spinal ganglia S1-Co1, labeled fibers were not observed in the upper ureter. Course and distribution of labeled fibers in the lower ureter were similar to those of the case in which injection was done into the thoracic and lumbar spinal ganglia.

Biological and immunological properties of Sugi basic protein-pullulan conjugate. II. Is the reduced ability to elicit the Arthus reaction based on the poor activation of complement by immune complex consisting of anti-Sugi basic protein and Sugi basic protein-pullulan?

Ability of Sugi basic protein (SBP)-pullulan conjugate to elicit the Arthus reaction was found to be markedly reduced, about 100 times lower than that of native SBP. To analyze this reduced ability, activation of complement by immune complex consisting of SBP-pullulan and anti-SBP antibodies was studied. Tests for complement consumption, C3 conversion and cleavage of factor B revealed that immune complex formed with SBP-pullulan is incapable of supporting efficient activation of the complement system. Previously, we have shown data suggesting that SBP-pullulan conjugate would be a good candidate for desensitization therapy against cedar pollinosis. The results presented in this paper provide additional support for the suggestion.

[Cytoprotection by 16,16-dimethyl prostaglandin E2--role of gastric content and mucus gel layer].

The mechanism of cytoprotection by prostaglandins (PGs) is still unknown, although many hypotheses have been proposed. We postulated a hypothesis that increased gastric content and thickened mucus gel layer by PGs may protect the gastric mucosa against damage from necrotizing agents. Two series of experiments were performed on Wistar male rats, weighing 250-300 g. (1) 16,16-dimethyl prostaglandin E2 (dmPGE2) in doses of 20 micrograms/kg was given orogastrically. Fifteen minutes later, the stomachs were emptied and/or the mucus gel layer removed, and several concentration of ethanol applied. After ten minutes, the stomachs were removed and lesions of the gastric mucosae were evaluated macroscopically and histologically. (2) Volume and pH of the gastric content and mucus thickness were measured 15 minutes after dmPGE2 administration. DmPGE2 did not protect the gastric mucosa against 40% ethanol in the emptied stomach. This agent had no cytoprotective action on the emptied and mucus gel-removed stomach in 30% ethanol application. These results had no significant difference with control group (saline 1 ml p.o.) about the extent of erosion. In histological study of the erosive region by scanning and light microscopy, we also found no differences in the depth of erosion between dmPGE2 group and control. In addition, dmPGE2 increased the gastric volume and mucus thickness significantly. These data suggest that following two effects by PGs play major role in cytoprotection of the gastric mucosa; (1) dilution of necrotizing agents by increased gastric content, and (2) thickening of the mucus gel layer.

[Generalized pustular psoriasis in siblings].

Generalized pustular psoriasis in siblings, 31-year-old male and 26-year-old female, is presented. In both cases, pustular lesions appeared in childhood and typical eruption of psoriasis vulgaris was not observed during their clinical course. Cholecystitis and chronic tonsillitis of the brother and periodontitis and chronic tonsillitis of the sister were considered to be possible provocative factors. HLA-A24, Bw52-, DR2, as the common HLA haplotype in our cases, was estimated.

[Clinical study of two cases of traumatic cerebellar injury].

Two cases of traumatic cerebellar injury complicated with a traumatic medial longitudinal fasciculus (MLF) syndrome or cerebellar mutism were reported, and the cause of these mechanisms was discussed: Case 1: A 9-year-old boy who struck his head in the occipital region during an automobile accident was operated on for a delayed traumatic intracerebellar hematoma. The operation improved the level of his consciousness but MLF syndrome was noticed. The mechanism of traumatic MLF syndrome was discussed in relation to vascular injury and to neurovascular friction. The outcome of the syndrome including our case, which recovered spontaneously, seemed to support the theory of neurovascular injury. Case 2: A 6-year-old boy who struck his head in the temporooccipital region during an automobile accident was admitted to our hospital without conciousness. On admission, contusion of the temporal lobe and left cerebellar hemisphere was demonstrated by a computerized tomography (CT) and magnetic resonance imaging (MRI). A mute state (cerebellar mutism) was recognized after his recovery of consciousness. The cause of the cerebellar mutism was thought to be an injury of the cerebellar vermis or left cerebellar hemisphere. The findings of CT scan and MRI in our case suggested that the cause of the cerebellar mutism was the contusion of these areas.

Polypoid squamous cell carcinoma of the esophagus.

Since 1971, 111 patients with esophageal carcinoma have undergone esophagectomy at the affiliated hospitals of Nippon Medical School. Of the 101 patients with ordinary squamous cell carcinoma, seven (7%) had intraluminal polypoid masses, endoscopically, and radiologically. The criteria used to select cases were as follows: (1) a size greater than 3 cm, (2) an intraluminal polypoid or pedunclated tumor, and (3) absence of wall construction and ulceration. Four of the seven polypoid cases have survived more than 5 years. One has lived more than 3 years. The 5-year survival rate of those patients with the polypoid type was 71%, but it was only 11% for the other types (P less than 0.05). Age, sex, size or location of the tumor, histologic grade, or lymph node metastasis did not affect survival. Only the incidence of adventitial involvement was significantly lower for the polypoid type. These results indicate that polypoid-type squamous cell carcinoma of the esophagus has a fairly good prognosis after surgery.

Functional changes of aorta with massive accumulation of calcium.

The present study was undertaken to elucidate pathophysiological changes and functional alterations of the calcified artery. For this purpose, rats were treated with 500,000 units/kg vitamin D3, and tension development of isolated rat aortae was examined. Treatment of rats with vitamin D3 resulted in an increase (approx. 64-fold) in the tissue calcium. Light microscopic examination of the aorta after staining with hematoxylin-eosin and von Kossa indicated numerous plaques in the aortic media. The results indicate a massive accumulation of calcium in the aortic media. Responsiveness of the calcified tissue to norepinephrine, epinephrine, serotonin, prostaglandin F2 alpha was found to be 11-66% when compared to that of the control. Furthermore, the calcified tissue responded minimally to isoproterenol and acetylcholine, which elicited a relaxation in control aortae. Isoproterenol-induced relaxation of the calcified aorta after 100 mM KCl contracture was also diminished. In the present study we have demonstrated poor responsiveness of the calcified aorta to physiological and pharmacological substances relative to normal tissue, which implies a functional damage of the artery upon massive calcium accumulation.

Prognostic implication of sequential bone marrow cultures in the myelodysplastic syndromes.

Twenty-five patients with newly-diagnosed myelodysplastic syndromes were studied by the clonal culture method at least three times during the clinical course. Clinical outcomes of the patients were classified into: a stable disease (ten patients); subsequent leukemic transformation (eight patients) and nonleukemic death (seven patients). The growth of the marrow granulocyte-macrophage progenitors (CFU-GM) at the time of diagnosis was significantly related to the survival. In addition, sequential changes in the CFU-GM growth patterns correlated with the different clinical outcome of myelodysplastic syndromes patients.

Monoclonal antibodies to chicken iodopsin.

The protein moiety of chicken iodopsin, R-photopsin, was purified from the chicken retina using a sucrose flotation method followed by two steps of column chromatography. Apparent molecular weights of R-photopsin and scotopsin (the protein moiety of chicken rhodopsin), which was partly purified in the process of purification of R-photopsin, were estimated to be 34,000 and 36,000, respectively, by sodium docecylsulfate-polyacrylamide gel electrophoresis. Using the purified R-photopsin as an antigen, four kinds of hybridoma cells which secreted monoclonal antibodies specific for R-photopsin and iodopsin were prepared. The antibodies thus obtained reacted with neither other chicken cone visual pigments nor rhodopsin as analyzed by immunoblots and immunoprecipitation methods. All the monoclonal antibodies stained the majority of the cone outer segments in chicken retina, while an antiserum raised against cattle rhodopsin stained the rod outer segments as well as some cone outer segments in the retina.

Chronically streptozocin-diabetic monkey does not closely mimic human diabetic neuropathy.

In order to evaluate the value of diabetic Japanese monkeys (Macaca fuscatus) as an animal model for studying the pathogenesis of diabetic neuropathy, morphological examinations were performed on myelinated nerve fibers and endoneurial microvessels at three levels of the lower limb nerve in eight streptozocin (STZ)-diabetic monkeys with the duration of diabetes up to 36 months and in four roughly age-matched control monkeys using a computer-assisted image analyzer. Nerve fiber loss was not found, although a tendency for nerve fiber atrophy was found in diabetic monkeys. Endoneurial microvessels did not show either endothelial or pericyte proliferation or basement membrane thickening. The results suggest that chronically STZ-diabetic Japanese monkeys with the duration of diabetes up to 36 months might be useful for studying diabetic axonopathy, but do not closely mimic the nerve pathology found in human diabetic neuropathy.

Biological and immunological properties of Sugi basic protein-pullulan conjugate. I. Suppressive effect on IgE antibody production and on IgE-mediated reactions.

Sugi basic protein (SBP), a major allergen of Japanese cedar pollen, conjugated to pullulan acquires suppressive activities for IgE, but not for IgG antibody production, and for IgE-mediated PK reactions in a SBP-specific manner. When either normal or IgE anti-SBP antibody-producing mice were treated with the conjugate, IgE anti-SBP antibody levels scarcely increased even after immunization with SBP + alum. This suppression was shown to be SBP specific and IgE selective. The ability of SBP-pullulan conjugate to elicit PK reactions on passively sensitized rat skin was markedly reduced. Furthermore, the conjugate inhibited the reactions mediated by the IgE antibodies and native SBP when it was injected into the IgE antibody-sensitized sites 2 h prior to SBP challenge. The reduced ability of the conjugate to elicit PK reactions was not due to loss of antigenic determinants during the conjugation procedures because binding activity of the conjugate to anti-SBP antibodies was as strong as that of native SBP. These results indicate that SBP-pullulan conjugate may be useful for 'desensitization therapy' against the pollinosis.

Neurokinin A-induced bronchial hyperresponsiveness to methacholine in Japanese monkeys.

The present study was designed to investigate whether neurokinin A (NKA) and substance P (SP) increase bronchial responsiveness to methacholine (MCh). Before and 1, 2, 3 and 4 weeks after three Japanese monkeys had inhaled 10(-8) M NKA and 10(-8) M SP for 2 min, bronchial responsiveness to inhaled MCh was evaluated. Pulmonary flow resistance (R1) and dynamic lung compliance (Cdyn) were used as pulmonary function tests. Dose-response curves of R1 to inhaled MCh were shifted to the left 1 to 4 weeks after NKA treatment, compared with that obtained before treatment. However, SP treatment did not change the R1 dose-response curve. Thus, bronchial responsiveness to MCh was increased by inhalation of 10(-8) M NKA in Japanese monkeys.

[Factors affecting the setting time of zinc oxide-eugenol impression materials--the influence of humidity and temperature to the reaction velocity].

Zinc oxide-eugenol paste are widely used in clinical dentistry, principally for impression of non-undercut edentulous ridges, or bite taking materials. However it has been realized by those who use these paste that different products, direction for mixing, or different conditions for mixing, have different characteristics. An experiment have been made to observe the influence of different conditions for mixing to the setting time. Zinc oxide-eugenol impression pastes were mixed in accordance with the manufacturer's instruction when supplied. The condition for mixing was at temperature of 15 +/- 1, 20 +/- 1, 25 +/- 1, 30 +/- 1 degrees C and at humidity of 40 +/- 5, 70 +/- 5, 90 +/- 5%. The setting time (a initial setting time, a final setting time) was defined in the A.D.A. specification No. 16. The following results were obtained. 1. A higher temperature and humidity shortened the initial setting time of all paste. 2. A higher temperature and humidity shortened slightly the time interval between a initial and a final setting time of all paste. 3. In clinical use, sample C is better than other ones, so that a setting time of the sample is property for impression making.

A polymorphic DNA marker genetically linked to congenital malformations.

Unusual restriction fragments were detected by DNA blot hybridization with PCNA (DNA polymerase-delta auxiliary protein) probe in one of seven cases of congenital malformations. Chromosomal in situ hybridization localized PCNA gene to region q31-35 of human chromosome 2. To discover the locus more closely associated with congenital malformations, a cloned DNA segment which has been mapped to chromosomal region 2q33-36 was tested for restriction fragment length polymorphisms (RFLPs) in these patients. The 2q33-36 probe hybridized with 2.1-kb, 1.9-kb and 1.7-kb fragments in ten normal control samples. In seven cases of congenital malformations examined, however, the band of 2.1 kb is absent in six cases and the band of 1.7 kb in one case. These results indicate that the locus closely linked to congenital malformations is present in the proximity of PCNA locus.
