Transnuclear transport and axon collateral projections of the mamillary nuclei in the rat.

Transnuclear transport of horseradish peroxidase (HRP) and wheat germ agglutinin-HRP conjugate (WGA-HRP) and the retrograde transport of fluorescent tracers were used to study axon collaterals of neurons in the mamillary nuclei. Tracers were injected into the thalamus or brain stem and after 18 hour-5 day survival periods, the brains were processed for fluorescence microscopy or for light and electron microscopic HRP histochemistry. Neurons in all divisions of the ipsilateral mamillary nuclei projected to both the thalamus and tegmentum. After HRP and WGA-HRP injections, anterogradely labeled axon terminals were observed in the known projection fields of the mamillary nuclei. Mamillary neurons were characterized by deeply invaginated, eccentrically located nuclei. Most labeled terminals of axon collaterals in the contralateral anterodorsal thalamic nucleus and dorsal and ventral tegmental nuclei contained round vesicles and formed asymmetrical synapses with somata and dendrites. The present results demonstrate that transnuclear transport of HRP and WGA-HRP can be used to study the connectivity and ultrastructure of axon collaterals and their cells of origin in the central nervous system in a manner comparable to that of transganglionic transport in the peripheral nervous system.

Amitrole: strain differences in morphological response of the liver following subchronic administration to mice.

Strain differences in the induction of hepatocellular preneoplastic lesions by amitrole were examined in NOD, ICR and DS mice. Amitrole was administered to mice in drinking water at a dose of 1% for 6 months. After 3 months, hyperplastic nodules (HN) and severe fibrosis were prominent in NOD mice but not in other strains. On examination at 6 months, both number and size of HN were greatest in the NOD strain. Furthermore, a hepatocellular carcinoma was found in a NOD mouse, suggesting that this strain is more susceptible to amitrole-induced hepatocarcinogenesis than are ICR or DS mice.

Tuberous sclerosis: report of two cases studied by computer-assisted cranial tomography within one week after birth.

Two neonates with tuberous sclerosis are reported. The patients presented convulsive seizures 30 mins or 5 days after birth. Computer-assisted cranial tomography (CT) on admission showed typical paraventricular calcifications in Case 1 and a calcified mass in Case 2. In addition to the CT findings, the diagnosis was made on the basis of the depigmented skin, and the mother with tuberous sclerosis in Case 1 and the pathological findings after surgery in Case 2. The importance of CT as a diagnostic procedure for this disease within 1 wk after birth is emphasized.

Cross-subtype protection in humans during sequential, overlapping, and/or concurrent epidemics caused by H3N2 and H1N1 influenza viruses.

A total of 663 pupils at four schools were studied serologically and clinically during a period of large sequential and/or mixed epidemics of infection with two subtypes of influenza A virus, H3N2 and H1N1. Of 91 middle-school pupils infected with H3N2 virus shortly before and 82 pupils not previously infected with this subtype, 59% and 91% became infected with H1N1 virus, respectively; this difference was significant. Similar results were obtained at the two primary schools studied. At a high school where epidemics due to the H3N2 and H1N1 subtypes occurred concurrently, the rate of infection of individual pupils with both viruses (2%) was significantly lower than those at the other three schools (21%, 23%, and 31%, respectively), where an epidemic caused by the H3N2 subtype appeared first and was then partially overlapped and succeeded by an epidemic caused by the H1N1 subtype. These findings suggest the existence of cross-subtype protection in humans during sequential and/or concurrent epidemics caused by two viral subtypes.

Immunohistochemical studies of the serotonergic supraependymal plexus in the mammalian ventricular system, with special reference to the characteristic reticular ramification.

Distributional and morphological features, especially characteristics of the ramification of serotonin-containing supraependymal fibers (SEF), were studied in the ventricular systems of mammals (mouse, rat, guinea pig, rabbit, cat, dog, monkey) by means of a modified peroxidase antiperoxidase technique, using antiserotonin antiserum prepared in our laboratory. SEF were present in all ventricular systems, except on the third ventricle floor and in the choroid plexus. The density of SEF was higher in the smaller species. In the rat, light- and scanning electron microscopical SEF were almost completely abolished 1 week after intraventricular administration of 5,6-dihydroxytryptamine. Ramification of SEF was complicated; the SEF formed a true network with frequent anastomosing. In the ventricular system of rats rendered hydrocephalic by kaolin administration, the mode of axonal branching in the supraependymal plexus could best be analyzed by the scanning electron microscope because the meshes of the plexus were spread out.

Treatment of malignant pleural effusion.

Two hundred consecutive patients with malignant pleural effusion were reviewed. The pathologic etiology of malignant pleurisy was: primary lung cancer in 123 cases; five, mesothelioma; and 72 cases secondary to metastatic tumors. Adenocarcinoma of the lung and mammary cancer were the most frequent tumors causing malignant pleural effusion. The modalities employed in local treatment consisted of thoracocentesis in 62 patients, tube thoracotomy in 111 cases with local instillation of adriamycin, MMC, CQ, 5FU, OK432 or talc. Surgical procedures including pleuropneumonectomy or reduction surgery of the tumor with decortication were performed in ten patients. Tube drainage with local instillation of drugs was more effective than thoracocentesis with or without local therapy. Excellent initial results were obtained in patients who received reduction surgery with decortication and pleurodesis. Results of cytologic investigation were positive in 157 cases (78.5 percent). The tumor cells disappeared in 79.4 percent of primary cancer pleurisy cases and 81.1 percent of patients with metastases while disappearance or significant decrease in pleural effusion following treatment was obtained in 75.2 and 77.8 percent respectively. The median survival was 11.3 months in primary cases, and 11.7 months in patients with metastases.

[Extraneural metastasis of malignant glioma through a ventriculo-peritoneal shunt: growth in peritoneal cavity as ascitic form].

This is the first report of extraneural metastasis of malignant glioma through V-P shunt tube and growth in peritoneal cavity as ascitic form. The patient was a 43-year-old man who was admitted to our hospital with occipital headache. CT scan showed enhanced cystic tumor mass at left temporal lobe. Craniotomy and partial excision of the tumor was done and the histology of tumor tissue showed a malignant astrocytoma. Following this treatment, the patient received the adjuvant therapies of radiation, chemotherapy and immunotherapy with interferon, and also recraniotomy three times. In the mean time, a ventriculo-peritoneal shunt was set up for internal hydrocephalus. One month later, abdominal bulging appeared and yellowish ascites could be obtained with peritoneal tap. In the ascite, tumor cells with glial fibrillary acidic protein were observed at the concentration of 5-10 x 10(4) cells/ml. The patient died three months after extraneural metastasis to the abdominal cavity as ascitic form. At autopsy, solid metastatic mass lesion was not found in extraneural region include abdomen.

[The clinical significance of OFA-I in the immunological rejection mechanism in trophoblasts].

Oncofetal Antigen-I is a membrane antigen of melanoma cells that cross-reacts with human fetal brain tissues. In this paper, from the point of view of fetal antigen, we investigated the immune response of pregnant women and the role of OFA-I in pregnancy. The results obtained are as follows. A higher prevalence of anti-OFA-I with the IA method was obtained in sera from pregnant women (50% in the 1st, 48% in 2nd and 55% in 3rd trimester). The prevalence of anti-OFA-I with the IA method was 78% (21/27) in total hydatidiform mole, 80% (8/10) in invasive mole and 86% (28/32) in choriocarcinoma patients. OFA-I was not detected in thirteen normal trophoblasts and five total mole ones by means of the IA absorption assay. With the indirect immunofluorescence method, OFA-I was found in 40% of incomplete abortions (4/10) and 28% of inevitable ones (5/18) but not found in normal trophoblasts (0/5). Specific fluorescence of OFA-I was found mainly in the syncytiotrophoblast. From these findings, we are tempted to conclude that OFA-I is not expressed on normal trophoblasts but expressed on aborted ones and that OFA-I may induce a rejection reaction in vivo to the trophoblast cells of the immunological front in aborted pregnancy.
