[Volume reduction and tritium retention factor in electrolytic enrichment of water].

Volume reduction(N), tritium retention factor (R), tritium concentration factor(Z) and apparent separation factor(beta) were measured on the large and small electrolytic cell systems. The relative variation of R was smaller than that of Z. So, it is recommended to use R in calculation of tritium concentrations in water samples. Furthermore, it was empirically revealed that R can be obtained only from N if a reliable beta-value is previously known. Therefore, it is possible to obtain R without electrolysis of the tritium standard solution. Taking into account the above facts, the so-called non-spike analysis of tritium, in which electrolytic enrichment and liquid scintillation counting are combined, becomes practicable.

[Treatment of radioactive waste phenol with Fenton's oxidation].

A new procedure was developed for the waste treatment of radioactive phenol on a laboratory scale. Waste phenol was dissolved in water (2% as phenol), and ferrous ion (10 mg/l as Fe2+) was added. Hydrogen peroxide (6% as H2O2) was added separately in three steps, 20, 30, and 50% of the volume, in order to avoid rapid reaction. The solution was heated at 80-90 degrees C for several hours in a beaker covered with a watch glass till the color of the solution changes from red purple to pale yellow. Then, the solution was neutralized with sodium hydroxide and heated for 1 h to decompose excess hydrogen peroxide. When the procedure is carried out with four 2 l beakers on a hot plate, 100 ml of phenol (1,200 ml of the aqueous solution saturated with phenol) can be oxidized simultaneously.

[A case of lateral thoracic meningocele without neurofibromatosis].

A case of lateral thoracic meningocele without neurofibromatosis is reported. CT-myelography revealed the mass has the communication with CSF space. So we could differentiate it from a posterior mediastinal tumor. It is important to think of the presence of lateral thoracic meningocele when any posterior mediastinal mass is detected.

[A case of Borrmann type 4 gastric carcinoma which disappeared by intraaortic infusion chemotherapy with ADM and MMC].

A 64-year-old woman with Borrmann type 4 gastric cancer was treated by intraaortic (celiac artery) one-shot infusion chemotherapy with adriamycin (ADM) and mitomycin C (MMC). One month after the first administration of 30 mg ADM and 20 mg MMC, the second administration of 50 mg ADM and 20 mg MMC was done. The third administration of 50 mg ADM and 20 mg MMC was performed one month after the second administration. Clinical symptoms of back pain, lumbago and anorexia had almost disappeared soon after the third administration. Although the findings of the X-ray examination of stomach, gastroscopy and celiac angiography showed marked improvement, total gastrectomy with lymphadenectomy was performed 2 months after the third administration because residual cancer cell nests were highly suspect. Histological examination of the resected stomach and lymphnodes revealed no cancer cells. The patient has been in good health without any recurrent signs for 16 months after operation.

[A case of pulmonary arteriovenous fistula treated by subsegmentectomy with CUSA].

A 35-year-old female who had no symptom was admitted to our hospital because of an abnormal shadow in the middle area of the right lung in a chest X-ray film. The chest tomography, CT and the angiography revealed a pulmonary arteriovenous fistula. It located in the right upper lobe, and was successfully resected by subsegmentectomy of S3b using CUSA. The results indicated that the CUSA was useful in the surgical treatment of the pulmonary arteriovenous fistula which had many vascular anastomoses.

[Clinicopathological analysis of leiomyosarcomas of the small intestine and a review of the Japanese literature].

An analysis has been made of 9 cases of a leiomyosarcoma of the small intestine that were treated from 1974 through 1989. In two patients (22.2%), a perforation was seen, and a case report of a 59-year-old woman is presented. Further, case reports of leiomyosarcomas in the Japanese literature from 1980 through 1989 were clinico-pathologically analyzed. The frequency of a perforation was found to be 8.6%, and the distribution of the perforated leiomyosarcomas was the same as for the non-perforated cases. The five-year postoperative survival rate was 41.2%. Of note is that 3 out of 10 patients that had survived for 5 years subsequently died of sarcomatosis.

Characteristics of rat lateral hypothalamic neuron responses to smell and taste in emotional behavior.

Single unit activity in the lateral hypothalamus (LHA) of the rat was recorded while the animal learned to discriminate cue signals. Normally preferred potables (glucose, orange, or grape solution) or intracranial self-stimulation (ICSS) were used as rewards. Electric shock or tail pinch were used as aversive stimuli. The same behavior, licking, was the response required to either obtain the rewarding stimuli or avoid the aversive ones. For positive reinforcement a rat was rewarded with fluid or ICSS upon licking a spout presented in front of its mouth. In negative reinforcement experiments, an aversive stimulus, electric shock or tail pinch, was applied if the rat did not lick the spout. Solutions having smell only, taste only, or smell-plus-taste, were prepared from oranges or grape extract. Of 392 neurons analyzed, 256 responded differentially to rewarding and aversive stimuli, and 138 of these were tested with the 3 different solutions. Similar LHA neural responses occurred during actual drinking of the 3 kinds of solutions, as well as on recognition of the cue signal. Responses to smell only had shorter latency than responses to taste only. Neural activity in response to solutions that could be both smelled and tasted was the sum of activity in response to taste-only solutions plus that in response to smell-only solutions. Cue signal responses were rapidly acquired, usually within 2-5 trials, for both taste-only and smell-only solutions. The results indicate the integration of both taste and olfactory information by the same LHA neurons, and these neurons are involved in cue signal learning. Present results of LHA neuronal responses to taste and smell suggest that the intensity of gustation and olfaction may add together to enhance instinctive hedonic sensations. These neurons are involved in the formation of stimulus-reinforcement association in learning, and in elicitation of conditioned emotional responses.

Initiation activity of endogenously synthesized N-nitrosobis(2-hydroxypropyl)amine in the rat liver.

The initiation potential of N-nitrosobis(2-hydroxypropyl)amine (NDHPA) endogenously synthesized from bis(2-hydroxypropyl)amine (DHPA) or tris(2-hydroxypropyl)amine (THPA) in the presence of sodium nitrite (NaNO2) was investigated in the rat liver by quantitation of hepatocellular foci showing phenotypic expression of glutathione S-transferase placental form (GST-P). The investigation consisted of two experiments. In the first, male Wistar rats were divided into six groups as follows: group 1 was non-treated; groups 2 and 3 received 0.15% and 0.3% NaNO2, respectively; group 4 received 1% DHPA; groups 5 and 6 received 1% DHPA together with 0.15% and 0.3% NaNO2, respectively. In experiment 2, the same protocol was used except that 2% THPA was substituted for 1% DHPA. The treatments were continuous until sacrifice at week 94 in experiment 1 and week 104 in experiment 2. As a result putative preneoplastic GST-P-positive foci observed in the liver and increased dose-dependently in rats from groups 5 and 6 which had received DHPA and NaNO2, but not in rats administered THPA and NaNO2. The results indicate that endogenously synthesized NDHPA from DHPA and NaNO2 is capable of initiating neoplastic development in the rat liver.

Avian spinal cord chimeras. Further studies on the neurological syndrome affecting the chimeras after birth.

These experiments bring new information concerning the immunological status after birth of quail----chick spinal cord chimeras. Such birds have been produced using recipient flocks of chickens different from those in our previous experiments. The breakdown of tolerance after hatching has been recorded and found to vary with the origin of the embryos. Chickens of broiler JA 657 strain and of a white leghorn strain raised in Japan started to exhibit signs of neural graft rejection later in life than the white leghorn chickens from a French breeder used in our previous studies. As previously described, in two animals, long-term tolerance was observed only for allogeneic chick----chick neural tube grafts. In one chimera the neurological syndrome resulting from rejection was reversible, and no signs of immune attack of the grafted central nervous tissue could be detected at sacrifice. This and other observations reported in this article strongly support the contention that the host immune response to foreign neural tissues starts in peripheral nerves and ganglia where no blood-brain barrier exists rather than in the spinal cord. A humoral response of the host against non-polymorphic quail antigens present on fibroblasts was observed in all birds at the time of rejection.

Possible involvement of arachidonic acid metabolism in phenobarbital promotion of hepatocarcinogenesis.

The effects of inhibitors of arachidonic acid metabolism and antioxidants on the rat liver tumor promotion activity of phenobarbital (PB) were assessed using the enzyme-altered focus as the end-point lesion. Fischer 344 male rats were initiated with N-nitrosodiethylamine (200 mg/kg) and then divided into five groups placed on basal diet, diet containing 0.05% PB, diet containing 0.05% PB plus 0.75%, 1% or 1.5% levels of various inhibitors of arachidonic acid metabolism or antioxidants, or diet containing 1% or 1.5% inhibitors or antioxidants alone for 10 weeks, and then killed. p-Bromophenacyl bromide, an inhibitor of phospholipase A2, significantly inhibited the promotion activity of PB at dose levels of 0.75% and 1.5%, reaching plateau at 0.75%. Both quercetin, an inhibitor of lipoxygenase, and morin, a dual inhibitor of lipoxygenase-cyclooxygenase, significantly reduced the promotion activity of PB at the 1.5% but not 0.75% dose levels. Moreover, acetylsalicylic acid, an inhibitor of cyclooxygenase dose-dependently inhibited the promotion activity of PB. Among the antioxidants investigated, vitamin E did not affect, but n-propyl gallate and ethoxyquin exerted a dose-dependent inhibition of PB promotion. These results are strongly suggestive of an involvement of phospholipase A2, lipoxygenase and cyclooxygenase arachidonic acid metabolic pathways in the mechanisms underlying PB enhancement of hepatocarcinogenesis.

Cycles of repeated augmentation pressure in rapid production of pancreatic and cholangiocellular carcinomas in hamsters initiated with N-nitrosobis(2-oxopropyl)amine.

We previously reported a rapid production model for pancreatic carcinoma development in Syrian hamsters incorporating the principle of selection by resistance to cytotoxicity. In the present experiment, the efficacy of repeated augmentation pressure with regard to generation of pancreatic lesions in hamsters initiated with N-nitrosobis(2-oxopropyl)amine (BOP) was investigated. Forty-eight female Syrian golden hamsters were divided into four groups according to the frequency of augmentation pressure. Group 1 received 70 mg/kg body weight of BOP and three injections of 20 mg/kg BOP. Groups 2-4 received 70 mg/kg BOP followed by one, two or three cycles of augmentation pressure consisting of dl-ethionine on sugar and salt diet, l-methionine and 20 mg/kg BOP. Hamsters were killed 10 weeks after the beginning of the experiment and the resultant incidences of pancreatic carcinomas from groups 1-4 were 0, 30, 50 and 46.2% respectively, the numbers of pancreatic carcinomas increasing with the frequency of augmentation pressure. A 46.2% yield of cholangiocarcinomas was also observed in group 4. The model should be useful for investigation of potential modulating factors since large numbers of lesions can be induced within a total experimental period of only 10 weeks.

Study on absorption of indomethacin from sustained-release suppositories containing hydrogenated soybean lecithin in rabbits.

The absorption of indomethacin (IM) from suppositories containing hydrogenated soybean lecithin (HL) after rectal administration in rabbits was investigated with the aim of producing sustained-release suppositories. The suppositories were prepared by the fusion method with IM, HL and Witepsol H-15 (H-15). The IM release rate from the suppositories (IM 10 mg, HL 200 mg, total weight 1 g) was faster than that of the control suppositories without HL. The release of IM from the suppositories (IM 10 mg, HL 300 mg or 350 mg) showed slow-release profiles. In absorption studies in rabbits, sustained-plasma levels of IM were obtained when suppositories having an HL content of more than 300 mg were administered. The suppositories composed of Witepsol E-85 (melting point approximately 43 degrees C), 10 mg of IM, and 200 mg of HL, showed slow-release profiles in the release test, but did not show sustained plasma levels of IM in the absorption test. These results indicate that sustained-release suppositories able to release IM gradually from the surface of the suppositories can be obtained when HL, IM and H-15, whose melting point is lower than body temperature, are used in the preparation of the suppositories, provided that the HL content is high enough in relation to IM.

Clinical use of intermediate to high dose of N4-behenoyl-1-beta-D-arabinofuranosylcytosine in children with acute leukemia.

N4-behenoyl-1-beta-D-arabinofuranosylcytosine (BH-AC) is one of the ara-C derivatives resistant to deamination. Administration of BH-AC in an intermediate (500 mg/m2) or a high dose (1000 mg/m2) resulted in maximum BH-AC plasma levels that were 10 to 20 times as high as those obtained at the conventional BH-AC dose and prolonged maintenance of comparatively high levels. High concentrations of ara-C in the plasma were observed just after BH-AC administration and at 24 hours were proportionally far higher than the ratio between the BH-AC doses administered in this study and those usually administered. To prevent the occurrence of shock, which has reportedly occurred in other studies, because of the presence of HCO-60, hydrocortisone was administered together with BH-AC in this study. In the two patients studied, no shock was observed and the only side effects were transient nausea and vomiting. The results suggest that administration of intermediate and high doses of BH-AC, accompanied by HDC, will prove a safe, effective means of enhancing the efficacy of leukemia therapy.

Expression of the glutathione S-transferase placental form in human lung carcinomas.

Expression of glutathione S-transferase placental form (GST-pi) in human lung carcinoma tissue taken at autopsy or biopsy was investigated immunohistochemically. All of 34 cases of squamous cell carcinomas, including poorly, moderately and well-differentiated examples were shown to stain positively for GST-pi. Poorly differentiated adenocarcinomas were, however, negatively stained (0/5 cases), while moderately and well differentiated adenocarcinomas were found to stain with GST-pi at rates of 69% (9/13 cases) and 71% (5/7 cases), respectively. Six cases of small cell carcinomas examined were all negative. The results indicate that GST-pi may be a useful marker for non-small cell type lung cancer, especially squamous cell carcinoma which is in agreement with findings for rat lung neoplastic lesions reported previously.
