Cardiovascular teratogenicity of ephedrine in chick embryos.

The cardiovascular teratogenicity and embryotoxicity of ephedrine was studied in chick embryos treated after 2.5-6 days of incubation (Hamburger-Hamilton developmental stages 17-28). The embryos were examined on day 14. Cardiovascular malformations were observed in 29% (29/101) of treated embryos. Complicated anomalies, such as double-outlet right ventricle (DORV), truncus arteriosus communis (TAC) or overriding aorta with ventricular septal defect (VSD) were seen frequently in embryos exposed to the agent on day 3 of incubation. Malformations were induced by ephedrine at a dose as low as 1 mumol/egg.

Diagnosis of familial amyloidotic polyneuropathy by recombinant DNA techniques.

An amino acid substitution of Met for Val at position 30 of plasma prealbumin is known to be closely related to heredo-familial amyloidotic polyneuropathy(FAP). As a first step in development of a direct method for diagnosis of the disease, cDNA for normal human prealbumin was cloned and its nucleotide sequence was determined. Our results showed that the nucleotide substitution responsible for the Val----Met change results in formation of new restriction sites for BalI and NsiI. By Southern blot hybridization analysis, the expected restriction sites were actually detected in the prealbumin locus of patients. Thus, a method was developed for diagnosis of the disease presymptomatically and prenatally.

Cloning and sequence analysis of a cDNA for the alpha-globin mRNA of carp, Cyprinus carpio.

A cDNA for alpha-globin mRNA of the carp, Cyprinus carpio, was cloned by the method of Okayama and Berg (Mol. Cell. Biol. 2 (1982) 161-170) and its complete nucleotide sequence was determined. The 5' non-coding region contained 23 nucleotides. Following this region, there was an open reading frame encoded with an alpha-globin polypeptide consisting of 142 amino acids. The 3' non-coding region was 88 nucleotides in length, including two copies of the hexanucleotide AATAAA and a poly(A) site of the GC dinucleotide. There were 16 discrepancies between the reported amino acid sequence of the carp alpha-globin chain and the amino acid sequence predicted from the DNA sequence of the clone. The possible explanations for these differences in amino acid sequence are discussed.

One base substitution in IVS-2 causes a beta +-thalassemia phenotype in a Chinese patient.

Two beta-globin genes derived from a Chinese patient with a beta +-thalassemia phenotype have been cloned and sequenced. A four-nucleotide deletion in codon 41 and 42 was found in one of the clones and a C to T substitution at position 654 of IVS-2 was detected in the other clone. The former change, which causes premature termination at codon 59, was found previously in another Chinese patient with homozygous beta 0-thalassemia. The latter change, which has not been reported so far, creates a sequence much like the consensus sequence of the 5' splice site. These results suggest that the new base change found in IVS-2 causes aberrant splicing of the beta-globin mRNA precursor. Thus, this patient is doubly heterozygous for beta 0- and beta +-thalassemia.

A fusion gene in man: DNA sequence analysis of the abnormal globin gene of hemoglobin Miyada.

An abnormal globin gene from a patient heterozygous for Hemoglobin Miyada was cloned and sequenced. The results indicated that the 5' flanking region and the 5' side of the gene were identical to those of a beta-globin gene and that the 3' side was identical to that of a delta-globin gene. The part of the gene identical to a beta-globin gene shifted to the part identical to the delta-globin gene somewhere in a homologous sequence region between the third nucleotide of the 17th codon and the second nucleotide of the 22nd codon of these two genes. Thus, results of analysis of the nucleotide sequence support the idea that the abnormal globin gene of Hemoglobin Miyada was generated as a fusion gene by unequal crossing over between a beta- and a delta-globin gene.

Herpes simplex virus type 1 restriction fragment polymorphism determined using southern hybridization.

Regions of herpes simplex virus type 1 (HSV-1) DNA with variation in the size of restriction endonuclease fragments were identified by comparison of the BamHI, KpnI or SalI restriction endonuclease digestion patterns among 15 HSV-1 isolates after hybridization with specific 32P-labeled cloned HSV-1 DNA fragments. Of the types of restriction fragment polymorphism identified, one was a strain with a distinctly different restriction fragment than the prototype (loss or gain of restriction sites). Another type, the specific fragment varied only in size among strains. Thirteen distinct variations were identified. Ten were mapped to the unique sequence of the L component; two to the inverted repeat of the L component and one to the inverted repeat of the S component. The presence of a common ancestor from which some isolates of HSV-1 might derive was deduced from an analysis of the distribution of the thirteen variations among the 15 HSV-1 isolates.

Isolation of phosphophoryn from human dentin organic matrix.

Normal human dentin was demineralized in 0.6 N HCl and then extracted in 1.0 M NaCl, 0.5 M Tris/HCl, pH 7.6 in the presence of neutral protease inhibitors. All of the soluble phosphorus-containing proteins were extracted directly in the 0.6 N HCl demineralizing solution; none were collected in the 1.0 M NaCl neutral pH extraction. The principal phosphoprotein was precipitated from solution by 1.0 M CaCl2 and subjected to further chromatographic purification. This fraction proved to be a typical phosphophoryn with Asp and Ser + PSer, in near equimolar amounts, accounting for approximately 75 residue percent of the protein. The second major organic phosphate-containing component was a peptide, Mr approximately 2,000. It was calcium precipitable and its amino acid composition showed a relationship to phosphophoryn. The residual collagenous matrix, which also contained organic phosphate, was digested with CNBr and the phosphate-containing moiety isolated. This had a composition indicative of a complex of collagen and phosphophoryn. Thus, in spite of the reports by Leaver and colleagues that human dentin contains neither soluble nor matrix-bound phosphophoryns, these data show that human dentin, like rat, hamster, rabbit, porcine, and bovine dentins, does contain a phosphophoryn as a major noncollagenous protein.

The migration function and the Todaro paradox.

"The purpose of this paper is to present a microeconomic foundation of the migration function and to discuss the impact of an increase in the job creation rate on migration and urban unemployment. Each rural worker must estimate his expected urban income on the basis of his own expected numbers of both newly created jobs and migrants during the coming period. Workers whose expected urban income is greater than the rural one decide to migrate, while those who estimate smaller urban income stay on." The geographical focus is on developing countries.

Isolation of human DNAs homologous to the BamHI-Z fragment of herpes simplex virus type 1 DNA.

Human DNA hybridizes with the BamHI-Z fragment (map coordinates 0.936 to 0.949) of Herpes simplex virus type 1 (HSV-1) DNA. To characterize the BamHI-Z homologue of human DNA, we isolated six independent hybrid phages with a sequence homologous to the BamHI-Z fragment from a human genomic DNA library. Three of the six had a common 1.2-kb BamHI-EcoRI fragment homologous to the BamHI-Z, and this fragment existed as 10-60 copies per human haploid genome. A 0.29-kb MboII segment of the BamHI-Z fragment was found to be responsible for the homology with the 1.2-kb BamHI-EcoRI fragment.

Effect of temperature and Zn2+ on isometric contractile properties and electrical phenomena of frog (Rana) and Xenopus skeletal muscle fibers.

Effects of temperature and Zn2+ on the isometric contractile properties of toe muscle fibers of Rana catesbeiana and Xenopus laevis were studied. The maximum twitch tension almost doubled when the temperature was lowered from 20 to 4 degrees C in Rana muscles but not in Xenopus muscles, although the duration of action potential in Xenopus muscle was increased slightly more than that seen in the Rana species. The maximum rate of rise of tension was greater in Xenopus muscle than in the Rana muscle, at 20 degrees C. The prolongation of the time-to-peak tension following exposure to low temperature (4 degrees C) was more pronounced in Rana than in Xenopus muscles. These results suggest that the speed of release and reuptake of Ca2+ by the sarcoplasmic reticulum (SR) differs in Rana and Xenopus muscles and that these factors may be related to differences in the SR and the T-tubular morphology. In Rana muscles, Zn2+ prolonged the falling phase of the action potential and potentiated the twitch tension. In Xenopus muscles, Zn2+ marginally prolonged the duration of action potential and the twitch tension was not markedly potentiated. These results indicate that Zn2+ potentiates the twitch by prolonging the action potential and that Rana muscles are more sensitive to the effects of Zn2+.

[Maximum tolerated dose of tegafur in rats].

Maximum Tolerated Dose of Tegafur was estimated by per-oral administration in male Donryu rats. Tegafur was administered for 24 weeks at respective doses of 90 mg, 120 mg and 150 mg/kg/day. Six of 10 (60%) died during the treatment in those groups administered with 120 mg and 150 mg/kg/day, respectively. The cause of death of these rats was severe pneumonia. All 10 of the group given 60 mg/kg/day survived until end of the administration and were sacrificed for histological examination of organs. These rats showed apparent suppression of body weight gain compared with controls (p less than 0.001). Slight inflammation of the lungs was observed in all rats, congestion of the liver and local degeneration and necrosis of the liver cells in a few rats and cellular degeneration of bone marrow in one rat. There was no remarkable change in the digestive tract, kidney, thymus or reproductive organs. Accordingly, the Maximum Tolerated Dose of Tegafur was determined to be 90 mg/kg/day.

[Total parenteral nutrition in inflammatory bowel disease. An evaluation of its clinical response].

We studied the effects of total parenteral nutrition (TPN) on the course of inflammatory bowel disease (IBD) which evaluation was still conflicting. Since 1972, in our department 32 courses of TPN were done for 27 patients with IBD. Definite improvement on nutritional status and clinical symptoms such as abdominal pain, diarrhea, vomiting was observed in most courses of TPN, especially for the patients with intestinal tuberculosis, non-specific multiple intestinal ulcers, and Crohn's disease. Clinical remission was obtained in 26 out of 32 courses of TPN (81.3%) on the short-term follow-up. But on the long-term follow-up ranging from 6 months to 11 years, 11 out of 20 patients (55%) were symptom-free without any medical treatment. Clinical relapse occurred in 6 patients and another TPN was required. These results suggest that TPN is useful as an adjunctive therapy for IBD patients requiring bowel rest and nutritional repletion. Two patients requiring prolonged TPN are returning to work with home parenteral nutrition (HPN) for 2.5 and 5 years respectively. HPN is safe and effective for improving life of quality in the patients receiving prolonged TPN.
