[Dose estimation of Tc-99m-DTPA-galactosyl-human serum albumin (Tc-99m-GSA) in the blood with nonlinear regression method].

Tc-99m-DTPA-galactosyl-human serum albumin (Tc-99m-GSA) was applied as an agent for receptor-binding hepatic scintigraphy. A single dose of 3-10 mg/185 MBq (5 mCi) Tc-99m-GSA per body was administered in four healthy volunteers and three patients. Time-activity curves for 60 (40) min after injection were obtained from the regions-of-interest over the heart and lung (back-ground). Exponential regression curve was generated from the heart curve during 2-60 (40) min after background subtraction with nonlinear least squares method. Regression curve was well fitted to the heart curve and a significant correlation coefficient of 0.997 +/- 0.003 was obtained between these two curves. Absolute dose of GSA in the blood was decided on the assumption that extrapolated value of regression curve at time 0 represented the total injected dose. The absolute dose differences between sample blood and regression curve were 1.4-4.3% (mean). Nonlinear regression analysis is useful for estimating quantitative dose of GSA in blood without any blood samples.

A sporadic case of bulbospinal muscular atrophy of late onset.

We describe a 72-year-old male with bulbospinal muscular atrophy (BSMA) who was being treated for diabetes mellitus and congestive heart failure due to an old myocardial infarction. Although BSMA is a rare form of X-linked spinal and bulbar muscular atrophy of late onset, this case is a sporadic case. We emphasize the importance of recognizing a sporadic case of BSMA.

[The purification of high molecular weight protease from the rat's skin].

In this study, the purification of a high molecular weight protease from Wistar Rats' skin was investigated. A high molecular weight protease from Wistar Rats' skin was purified by successive (NH4)2SO4 fractionation, Phenylsepharose CL-4B, and HPLC gel filtration chromatographies. The activity of the enzyme was determined using a synthetic substrate, succinyl-leucyl-leucyl-valyl-tyrosine methylcoumarinamide (SLLVT-MCA). The molecular weight of the purified enzyme was estimated to be 750,000 by gel filtration. With SDS-PAGE the purified enzyme was dissociated into several bands of which the majority fell into the range of 20,100-36,000 daltons. These findings suggest that high molecular weight protease in rat's skin consists of various subunits.

[Skin high-molecular-weight protease. 2. Biochemical properties].

The purification of High-Molecular-Weight Protease from Wistar Rat's skin was previously investigated and reported. Some properties of this enzyme were subsequently investigated. This enzyme showed high activities toward succynyl-leucyl-leucyl-valyl-tyrosine-methylcoumarinamide (SLLVT-MCA). The addition of 0.03% sodium dodecyl sulphate (SDS) stimulated enzyme activity to 2,000% of the initial activity. The pH optimum was pH 8.5 in the presence of 0.03% SDS, but pH 6.0-8.5 in the absence of SDS. This enzyme was inhibited by one of serine protease inhibitors, diisopropylfluorophosphate (DFP) strongly. It was also inhibited by some of SH protease inhibitors, N-ethylmaleimide (NEM), iodoacetamide (IA), and leupeptin. Interestingly, chymostatin, a serine and SH protease inhibitor also inhibited enzyme activity. These results show that both serine and cysteine++ residues are related to the enzyme activity of High-Molecular-Weight Protease.

[Reaction mechanism catalyzed by skin sulfhydryl oxidase].

The mechanism involved in the reaction catalyzed by skin sulfhydryl oxidase was investigated. Skin sulfhydryl oxidase, which was purified to a 300-fold increase in specific activity over rat skin, was used. As the substrate for the reaction catalyzed by skin sulfhydryl oxidase, dithiothreitol (DTT) was used. The disappearance of DTT, consumption of O2 and production of H2O2 were measured according to Takamori's Janolino's and Kakinuma's methods, respectively. The molar ratio of the disappearance of DTT and the consumption of O2 during the enzyme reaction was 1:1.05. The molar ratio of the disappearance of DTT and the H2O2 production during the enzyme reaction was 1:086. Results therefore showed that the molar ratio of the disappearance of DTT, consumption of O2, and production of H2O2 was 1:1:1. Diffuse midperipheral acute retinal pigment epitheliopathy.

We examined a 16-year-old boy with a remarkable decrease in vision OS. Typical lesions of acute retinal pigment epitheliitis (Krill's disease) were not present in the macula but were diffuse in the midperiphery. Angiographic findings were similar to those in Krill's disease. The left fundus returned to normal two weeks later, but ten months passed before electro-oculography, light sensitivity, and dark adaptation were normal. Thus, there may be two types of acute retinal pigment epitheliitis: one is Krill's disease, and the other is a vast disturbance in the retinal pigment epithelium.

A case of tuberous sclerosis with calcification of angiomatous change in the white matter.

A 5-year-old Japanese boy had had infantile spasms at 5 months of age, and had intractable seizures, adenoma sebaceum and prominent tramtrack-like calcification in the left occipital pole at five years. A neurosurgical operation was performed for the resection of the left occipital focus. Pathological examination showed patchy glial fiber nets on the cortical surface, scattered dysgenetic giant cells both in the cortex and white matter, and calcified angiomatosis in the white matter. These findings correspond to those of tuberous sclerosis and calcified angiomatosis in the white matter.
