Selective deposition of amyloid-beta protein in the entorhinal-dentate projection of a transgenic mouse model of Alzheimer's disease.

Early and selective deposition of amyloid beta protein(Abeta) is thought to be a pathological feature central to Alzheimer's disease (AD). It has been a great challenge to identify the mechanism(s) responsible for the selectivity of Abeta deposition and the deposition into a temporal sequence of the pathogenesis in this disorder. We now report that the transgenic mouse (PDAPP), which overexpresses the human amyloid precursor protein containing the familial AD mutation (APP717V-F), develops brain region-specific Abeta deposits along with some pathologies associated with AD. By using monoclonal antibodies that recognize multiple sites on the human Abeta peptide, we show that Abeta deposits are localized primarily to the entorhinal cortex and hippocampal/dentate gyrus regions. Abeta deposition exhibited consistent laminar distribution throughout the hippocampal formation that was confined mostly to a dense terminal field in the outer portion of the stratum moleculare of the dentate gyrus and the stratum lacunosum of the hippocampus proper of 13-month-old heterozygous PDAPP mice. Abeta deposits were also observed in the supragranular zone of the dentate gyrus. Similarly, laminar distribution of Abeta deposits were evident in the entorhinal cortex, most notably in the molecular layer and in laminar layer II-III. The defined laminar pattern of the Abeta deposition, which resembles that of AD, suggests that intrinsic factors in the perforant path, the major projection from the entorhinal cortex to the hippocampal formation, and their respective local paths contribute, at least in part, to the extracellular Abeta deposition in the transgenic mouse model of AD.

A unified theory of carcinogenesis based on order-disorder transitions in DNA structure as studied in the human ovary and breast.

Fourier transform-infrared/statistics models demonstrate that the malignant transformation of morphologically normal human ovarian and breast tissues involves the creation of a high degree of structural modification (disorder) in DNA, before restoration of order in distant metastases. Order-disorder transitions were revealed by methods including principal components analysis of infrared spectra in which DNA samples were represented by points in two-dimensional space. Differences between the geometric sizes of clusters of points and between their locations revealed the magnitude of the order-disorder transitions. Infrared spectra provided evidence for the types of structural changes involved. Normal ovarian DNAs formed a tight cluster comparable to that of normal human blood leukocytes. The DNAs of ovarian primary carcinomas, including those that had given rise to metastases, had a high degree of disorder, whereas the DNAs of distant metastases from ovarian carcinomas were relatively ordered. However, the spectra of the metastases were more diverse than those of normal ovarian DNAs in regions assigned to base vibrations, implying increased genetic changes. DNAs of normal female breasts were substantially disordered (e.g., compared with the human blood leukocytes) as were those of the primary carcinomas, whether or not they had metastasized. The DNAs of distant breast cancer metastases were relatively ordered. These findings evoke a unified theory of carcinogenesis in which the creation of disorder in the DNA structure is an obligatory process followed by the selection of ordered, mutated DNA forms that ultimately give rise to metastases.

Preparation and cytotoxic effect of ceanothic acid derivatives.

Six ceanothane and 1-norceanothane derivatives (1, 2, 8-11) were prepared from ceanothic acid dibenzyl ester. These ring-A homologues of betulinic acid exhibited cytotoxic effects. Among these, 1-decarboxy-3-oxo-ceanothic acid (2) was found to be the most cytotoxic against OVCAR-3 and HeLa cancer cell lines, with an IC50 of 2.8 and 6.6 microg/mL, respectively, and an IC50 of 11.3 microg/mL against normal cell line FS-5.

Energy expenditure of Chinese infants in Guangdong Province, south China, determined with use of the doubly labeled water method.

The doubly labeled water method was used to measure the energy expenditure of a group of 41, 4- or 6-mo-old infants with a cross-sectional design. The infants were divided into two groups according to whether they were breast-fed (11 at 4 mo, 9 at 6 mo) or formula fed (11 at 4 mo, 10 at 6 mo). Anthropometric measurements were recorded at birth and at the beginning and end of the 8-d study. Anthropometric data, which were supported by the food intake and energy expenditure results, indicated that the infants were within the norms for European and American infants of the same age. Mean energy intakes of 352 kJ (84 kcal) kg(-1) x d(-1) at 4 and 6 mo were lower than the FAO/WHO/UNU recommended value of 452 kJ (108 kcal) x kg(-1) x d(-1)) and Chinese recommendations of 502 kJ (120 kcal) kg(-1) x d(-1). However, some authors believe that values of 398 kJ (95 kcal) kg(-1) x d(-1) at 4 mo and 356 kJ (85 kcal) kg(-1) x d(-1) at 6 mo are more appropriate. At 6 mo the infants' length-for-age and weight-for-age were at the National Center for Health Statistics 55th and 47th percentiles, respectively, whereas 58% were below the 50th percentile for weight-for-length. We conclude that at 4 mo infants receive sufficient energy for their requirements. However, at 6 mo energy requirements might well be greater than the revised recommendations, when infants are being weaned to alternative foods and are more prone to the influence of diet on their growth and development.

Transient global forebrain ischemia induces a prolonged expression of the caspase-3 mRNA in rat hippocampal CA1 pyramidal neurons.

The authors recently cloned a cDNA for an ICE/CED3-related cysteine protease from rat brain, which is closely related to human CPP32 (now designated caspase-3). In situ hybridization histochemistry revealed a profound developmental regulation of the caspase-3 transcript in rat brain, with relatively high levels of caspase-3 mRNA observed in neurons of the fetal and neonatal brain and low levels of mRNA in neurons of the adult brain. The authors report that transient forebrain ischemia, which results in a delayed apoptotic death of CA1 pyramidal neurons, results in prolonged expression of caspase-3 mRNA in these same pyramidal neurons. Up-regulation of caspase-3 mRNA in CA1 pyramidal neurons is prominent 24 hours after transient global ischemia, and expression is maintained at higher levels for at least 72 hours after ischemia. However, by 96 hours after ischemia, a marked decrease in caspase-3 mRNA expression is observed in CA1 pyramidal neurons, showing severe degenerative changes (e.g., nuclear condensation). By contrast, there is no change in the expression of a closely related member of caspase family, caspase-2, in CA1 pyramidal neurons after global ischemia. Instead, caspase-2 mRNA is induced in lamina layers of cerebral cortex 24 hours after the ischemia. A selective and prolonged induction of the caspase-3 gene in committed CA1 pyramidal neurons suggests that transcriptional activation of this caspase-3 gene may be involved in the apoptotic cell death cascade of CA1 neurons after transient global ischemia.

Effect of cigarette smoke extract on nitric oxide synthase in pulmonary artery endothelial cells.

Cigarette smoking is associated with impaired endothelium-dependent vasodilation and reduced nitric oxide (NO) in the exhaled air of smokers. To explore the mechanism for the impairment of NO-mediated vasodilation, we studied the effect of cigarette smoke extract (CSE) on NO synthase (eNOS) activity and content in pulmonary artery endothelial cells (PAEC). Incubation of PAEC with CSE resulted in a time- and dose-dependent decrease in eNOS activity. The inhibitory effect of CSE on eNOS activity was not reversible. Both gas-phase and particulate-phase extracts of CSE contributed to the inhibition of eNOS activity. The protein kinase c (PKC) inhibitors staurosporine and chelerythrine did not affect the CSE-induced inhibition of eNOS activity. Catalase, superoxide dismutase (SOD), vitamin C, vitamin E, glutathione, and dithiothreitol (DTT) also did not prevent the CSE-induced inhibition of eNOS activity, and incubation of PAEC with 3 mM nicotine did not change the activity of eNOS. Treatment of PAEC with CSE also caused a nonreversible, time-dependent decrease in eNOS protein content detected by Western blot analysis, and in eNOS messenger RNA (mRNA) detected by Northern blot analysis. Treatment of PAEC with CSE had no effect on cell protein or glutathione contents or on lactate dehydrogenase (LDH) release. These results indicate that exposure to CSE causes an irreversible inhibition of eNOS activity in PAEC, and suggest that the decreased activity is secondary to reduced eNOS protein mass and mRNA. The decrease in eNOS activity may contribute to the high risk of pulmonary and cardiovascular disease in cigarette smokers.

Efficacy and safety of a topical gel formulation of recombinant human platelet-derived growth factor-BB (becaplermin) in patients with chronic neuropathic diabetic ulcers. A phase III randomized placebo-controlled double-blind study.

OBJECTIVE: To compare the efficacy and safety of topically applied recombinant human platelet-derived growth factor-BB (rhPDGF-BB) (becaplermin) with placebo gel in patients with chronic diabetic neuropathic ulcers of the lower extremities. RESEARCH DESIGN AND METHODS: This multicenter double-blind placebo-controlled phase III trial included 382 patients with type 1 or type 2 diabetes and chronic ulcers of at least 8 weeks' duration. After sharp debridement of the ulcer, patients were randomized to receive becaplermin gel 30 micrograms/g, becaplermin gel 100 micrograms/g, or placebo gel, in conjunction with a standardized regimen of good wound care until complete wound closure was achieved or for a maximum of 20 weeks. Moist saline-soaked gauze dressings were changed twice daily with study medication applied by patients or caregivers at the evening dressing change. Safety was assessed by monitoring adverse events (AEs) and by clinical laboratory evaluations. RESULTS: Compared with placebo gel, becaplermin gel 100 micrograms/g significantly increased the incidence of complete wound closure by 43% (50 vs. 35%, P = 0.007) and decreased the time to achieve complete wound closure by 32% (86 vs. 127 days; estimated 35th percentile, P = 0.013). AEs reported during treatment or during a 3-month follow-up period were similar in nature and incidence across all treatment groups. CONCLUSIONS: Becaplermin gel 100 micrograms/g, in conjunction with good wound care, significantly increased the incidence of complete wound closure and significantly reduced the time to complete closure of chronic diabetic neuropathic ulcers. The safety profile of becaplermin gel was similar to that of placebo gel.

[Lipoprotein lipids in polycystic ovarian syndrome: independent associations with androgen excess and insulin resistance].

OBJECTIVE: To investigate the relationship between androgen excess, insulin resistance and altered lipoprotein lipids in polycystic ovarian syndrome (PCOS). METHODS: Both basal and luternizing hormone-releasing hormone (LHRH, 100 micrograms) induced responses of circulating testosterone (T), triglycerides (TG), high-density lipoprotein-cholesterol (HDL-C) and apoprotein A1 (apo A1) were measured in two PCOS groups with similar T levels (LH/FSH > or = 3, Group 1, n = 15, LH/FSH < 3, Group 2, n = 15) and the control (n = 20) of matched body mass index with Group 1. Insulin resistance was assessed by fasting insulin levels. RESULTS: In basal state, the mean TG levels in women of three groups were as follows: Group 2 > Group 1 > control, while HDL-C showed the opposite. After LHRH test, mean T and TG concentrations increased and HDL-C decreased in all PCOS subjects, especially Group 2 whereas the indices remained unchanged in the controls. CONCLUSION: Our data suggest that altered lipid profiles in women with PCOS may result from the independent effects of androgen excess and insulin resistance.

[Functional states of pituitary-ovary, adrenal and thyroid axes in women with polycystic ovarian syndrome].

OBJECTIVE: To assess the role of the functional states of pituitary-ovary, adrenal and thyroid axes in the pathophysiology of polycystic ovarian syndrome (PCOS). METHODS: The stimulation tests of above-mentioned three endocrine axes by luteinizing hormone releasing hormone (LRH, 100 micrograms), adrenocorticotrophin (ACTH, 250 micrograms) and thyroid releasing hormoune (TRH, 500 micrograms), respectively, were performed in each subjects of two PCOS groups [luteinizing hormone/follicle stimulating hormone (LH/FSH) > or = 3, Group 1, n = 15; LH/FSH < 3, Group 2, n = 15] and the control (n = 20). Endocrine indices of corresponding target glands were evaluated by radioimmunoassay (RIA). RESULTS: After LRH administration, the amplitude of LH responses in three groups were as follows: Group 1 > Group 2 > control while mean testosterone levels were elevated to a similar extent in two PCOS groups and remained unchanged in the control. Following ACTH stimulation, PCOS subjects, especially of Group 2 with obvious insulin resistance, showed higher responses of cortisol, dehydroepiandrosterone sulfate and testosterone as compared with the controls. However, during TRH testing, exaggerated thyroid-stimulating hormone and prolactin responses in two PCOS groups and blunted free thyroxine response at two hour point in Group 1 were observed, as compared with the control. CONCLUSION: It appears that ovarian androgen excess in women with PCOS is mainly LH-dependent in Group 1 and insulin-dependent in Group 2. Enhanced adrenal activity may contribute to both hyperandrogenism and insulin resistance in this syndrome, and subclinical hypothyroidism may exist in affected subjects, especially of Group 1.

[Survey on breakfast-eating behavior among residents in Beijing, Guangzhou and Shanghai].

A survey of breakfast-eating behavior among residents in Beijing, Guangzhou and Shanghai was carried out in 1994-1995. The results showed that the breakfast-eating rates in three cities were 74.8%-90.5%. Subjects aged below 35 years skipped breakfast more often. "Time limiting" in the morning became the main cause of breakfast-skipping. Nearly half of subjects in three cities got their breakfast out of home. Moreover, compared with two years ago, the present rates of eating outside increased at various levels in three cities. Steamed bun, bread and congee were main foods consumed at breakfast. In addition, 39% to 56% of subjects often drank milk. Through this survey, the authors got an all-round understanding on breakfast-eating rates, eating outside rates and variety of food consumed of residents in 3 cities. This survey will also provide a background for nutrition intervention and education in the future.

[Observation of inducing effect of five kinds of soft wood dust in micronucleus test].

The authors investigated the micronucleus frequencies (MP) in peripheral lymphocytes in 584 workers exposed to five kinds of soft wood dust and found that all kinds of soft wood dust could increase the micronucleus frequency. As the control's 95% confidential upper limit > or = 0.4% is defined as the standard for positivity the micronucleus positive frequency (MPF) in the exposure group is much higher than that in the control group (P < 0.01). Meanwhile the increase of exposure years may increase the MP, and the time-response relationship is evident (r = 0.90, P < 0.01). For different exposures, the MPFs are: 36.2% for linden, 26.3% for fir. 15.7% for lacebark pine, 9.6% for birch and 2.1% for polar. The positive rate of linden and fir exposure are significantly higher than that of the control group (P < 0.01). Water extract of these five kinds of soft wood dust can increase the frequencies of micronucleus in polychromatic red blood cell in mice's sternum marrow, which also showed a dose-response. The correlation coefficients are: 0.86 for linden 0.6 for fir, 0.39 for lacebark pine, 0.76 for birch and 0.73 for polar. All but the lacebark pine have the P value less than 0.01 (P < 0.01). At the dose of 15 g/kg body weight, micronucleus frequency induced by linden was the highest (0.8%), next were fir (0.71%), birch (0.65%), lacebark pine (0.63%), and polar (0.43%). The results from animal test are consistent with those from the exposure workers. These results suggest that there are some substances in soft wood which can induce chromosome aberration.

[The LDL receptor activity of hepatocytes during cholesterol gallstone formation in rabbits].

In order to study the mechanism of cholesterol gallstone formation through rabbit model which was induced by high cholesterol diet, we investigated the LDL receptor activity of hepatocytes binding to 125I-LDL in different phases, namely 1, 2, 3 week group and 4 week, and in a control group besides. In this animal experiment, cholesterol gallstones were induced at 2 week, 3 week and 4 week groups in 4/10, 6/10, and 7/10 cases respectively. The Bmax values of LDL receptor of hepatocytes binding to 125I-LDL decreased significantly in 3 week and 4 week groups (vs 1 week and control groups, P < 0.05). The kd values became increased in 3 week and 4 week groups (vs 1 week and control group, P < 0.05), which suggested that the activity of LDL receptor decreased gradually. In conclusions owing to the intake of high cholesterol diet with the passage of time, the decreased activity of LDL receptor of hepatocytes would reduce the synthesis of bile acid.

[Treatment of malignant hematological diseases with autologous peripheral blood stem cell transplantation].

OBJECTIVE: To observe the effectiveness of autologous peripheral blood stem cell transplantation(ABSCT) in the treatment of malignant hematologic diseases. METHODS: Thirteen ABSCT and 15 autologous peripheral blood stem cells(ABSC) combining autologous bone marrow transplantation (ABMT) were performed for hematological malignancy patients. RESULTS: In polyactin A mobilization group, the mononuclear cells(MNC) collected in each case were 3.58 +/- 2.12 x 10(8)/kg, and in G-CSF mobilization group 6.68 +/- 5.31 x 10(8)/kg. Twenty-one of the 28 transplanted cases achieved continuous complete remission(CCR). The medium CCR duration was 18(5-58) month. The 3 year disease-free survival rate was 68.7% and relapse rate was 22.3%. CONCLUSION: After ABSCT or ABSC combining ABMT, there were earlier hematopoietic reconstitution and fewer complications than that after ABMT.

[Experimental study on treatment of viral myocarditis in mice by integrated traditional Chinese and Western Medicine].

OBJECTIVE: To clarify the effects of Astragalus Membranaceus (AM) combined with taurine and/or coenzyme Q10(CoQ10) on coxsackievirus B3 (CVB3) murine myocarditis. METHODS: Viral myocarditis model was created by intraperitoneal inoculation with CVB3 solution and were treated by saline, AM, taurine, CoQ10, AM + taurine, AM + CoQ10, AM + taurine + CoQ10, respectively. The mortality, ECG, CVB3-RNA in myocardium and myocardial histopathologic changes were observed. RESULTS: AM combined with taurine and CoQ10 could significantly reduce the mortality of the mice and the incidence of abnormal ECG at acute stage. CVB3-RNA was significantly reduced in AM treated group, especially in AM + taurine group. No anti-virus effect was found in CoQ10 group. All drugs could lighten myocardial histopathologic changes and the effect could be enhanced by combined treatment. CONCLUSIONS: AM, taurine and CoQ10 have some curative effects on CVB3 murine myocarditis, AM combined with taurine and CoQ10 is the best.

[Study of portable synchronous ECG recorder and its long-distance communication system].

This paper presents a convenient long-distance ECG communication system. It is made up of portable ECG recorder, hospital work station and public telephone network. This paper also introduces a kind of modulation and demodulation method in sound coupling communication-pulse edge modulation and demodulation. This system has rational structure, convenient operation and strong management power. A Philips 80C552 used as CPU in the recoder which is portable, cheap and reliable.

[Study of the excitation and diffusion processes of sample atoms in a microwave boosted microsecond-pulse glow discharge source with optical emission spectrometry].

The excitation and diffusion processes of brass sample atoms in a GD-MIP tandem source have been studied with a home-made MIP boosted micros-pulse GD device. Experimental results show that under relatively low discharge pressure (< 180Pa), the micros-pulse GD can couple quite well with MIP and emit strong radiation of analytical lines. When the GD source is operated under a pressure higher than 200Pa, two emission peaks appear independently in time for a given resonant atomic line, because sample atoms are structurally excited separately first by the micros-pulse GD and then by the MIP. According to the two emission peaks, the diffusing velocities of copper atoms and zinc atoms can be calculated, yielding values of 150 and 129m/s, respectively, and the most excited area in-the micros-pulse GD is about 1.94-2.25mm away from the sputtering surface of the sample cathode. The effects of discharge parameters on emission intensities have been also investigated.

Auto-regulation of thyroid hormone receptor genes during metamorphosis: roles in apoptosis and cell proliferation.

Amphibian metamorphosis is an excellent model system for studying postembryonic development in vertebrates. It involves specific degeneration of larval cells through programmed cell death with apoptotic morphology and selective proliferation and differentiation of adult cell types. Thyroid hormone (T3) plays a causative role in this process and the effects of T3 is presumed to be mediated by T3 receptors (TRs). Studies in other systems have suggested that TRs function as heterodimers formed with RXRs (9-cis retinoic acid receptors) and require the presence of various cofactor in transcriptional activation and repression in the presence and absence of T3, respectively. The T3-induced transcriptional activation leads to chromatin remodeling which may involve some of the cofactors. Recent investigation on receptor expression has implicated a role of TRs in T3-induced apoptosis in larval tissues and proliferation of adult cell types. Functional studies in tadpoles and developing embryos have provide strong support for such a role and further demonstrate the importance of RXR in mediating the effect of T3.
