[The modification of combined subperiosteal facelift].

OBJECTIVE: To reposit all the elements of the aging midface more adequately. METHODS: We have performed combined facelift on 100 patients using extended subperiosteal dissection or sub-SMAS and subcutaneous dissection. In particular, the zygomatic arch subperiosteal dissect releases the attachment of the zygomatic ligament. RESULTS: In this group of 100 patients, postoperative follow-up showed that this technique can restore the initiate anatomical relationship of multiple tissue layers of the face, provides the patient with both an excellent nasolabial fold and jawline contour without damage of the frontal branch of the facial nerve. CONCLUSION: This technique is most suitable for the patients about 40 years old.

[Suture distraction osteogenesis for closure of cleft palate in the dog: long-term effect and its influence on facial growth].

OBJECTIVE: The purpose of the study was to evaluate the long-term effect and its influence on facial growth of suture distraction osteogenesis for closure of cleft palate. METHOD: 14 Beagle dogs were used in the study. Six of them were in the experimental group, eight served as the control. Direct measurements were taken on dry skulls of the dogs and the data were analysed statistically. RESULTS: The results showed that bony healing at the midline between the two horizontal portions of the palatine bones appeared in all the experimental dogs. There were no significant differences in the variables of facial length and height between the two groups. But the facial width and the distance between the two vertical portions of the palatine bones were significantly less in the experimental dogs than the control. CONCLUSIONS: It is suggested that bony repair of the cleft could be accomplished permanently with the technique of sutural distraction. This technique had no influence on facial length and height growth, but could led to underdevelopment of the width of the face and the respiratory tract of the palatal portion.

[A long-term study of a small-caliber arterial autograft used for repair of a large-caliber arterial defect].

OBJECTIVE: The long-term fate of a small arterial graft used to repair a large diameter artery was studied in this study. METHODS: An end segment of the tail artery (0.3-0.4 mm in diameter) was transplanted to the defect of the carotid artery (1.0-1.4 mm in diameter) in 50 rats. The vessel grafts were examined histologically at 3 weeks and 5, 10 and 12 months after implantation. RESULTS: At 3 weeks, intimal thickening of the grafts was mild and present only in regions adjacent to the anastomosis. At 5 months, severe intimal thickening was seen along the entire graft wall. At 10 and 12 months, the intimal thickening reduced. Regression of the thickening was uneven within the graft wall. The adventitia thickened at the early postoperative period and reduced at the end of this study. The patency rate of the grafts at 12 months postoperation was 95% (19/20). Angiography indicated that the grafts stopped expanding after 5 months and were only slightly smaller than the recipient arteries at 12 months postoperation. CONCLUSION: These results suggest that a small, autogenous arterial graft may effectively repair a large-calibre arterial defect.

[A serial histological study on suture expansion osteogenesis for cleft palate closure].

OBJECTIVE: The purpose of this study was to know the process of tissue response and regeneration in the palato-maxillary suture under tensile forces. METHODS: Sutural tissue changes after 3, 7, 14, 28, 56 days of suture expansion were examined histologically and fluorescently. RESULTS: It was found that a cyst-like zone appeared in the conjuncture of bony front and sutural connective tissue at the early stage of sutural expansion. Then a great amount of osteoblasts and fibroblasts proliferated. New bone deposited along the septum and the front of the cyst until the new bone front formed and the suture restored its original morphology. CONCLUSIONS: The findings suggested that sutural expansion be a process of bone tissue regeneration and fibrous tissue repair.

[Study of human tissue engineering cartilage].

OBJECTIVE: To investigate the technique of human tissue-engineered cartilage and to study the medical collagen membrane of guided tissue regeneration (GTR) as the carrier of in vitro chondrocytes culture. METHODS: Tissue engineering technique was used to make human Tissue-engineered cartilage, which was examined histologically and functionally. RESULTS: It was found that the chondrocytes seeded on the medical collagen membrane of GTR grew well. A layer of milk white and cartilage-like tissue grew on the surface of medical collagen membrane of GTR after 1 week. It was demonstrated that the cartilage-like tissue was strong enough to be transferred after being implanted for 8 weeks. The cartilage-like tissue was proved to be human tissue engineered cartilage by HE stain and Alcian blue-poncean S stain. The chondrocytes could secrete chondroitin sulfate as proved by Lev-Spicer stain. CONCLUSIONS: The medical collagen membrane of GTR has characteristics of three-dimensional structure and cell reticular function, and it has the possibility to be developed as a natural scaffold for tissue engineering. The results indicate that it is possible to make human tissue-engineered cartilage with tissue-engineering technique.

[Experimental study on revascularization of subdermal vascular network thin flap].

OBJECTIVE: To investigate the survival mechanism of subdermal vascular network thin flap (SVNF). METHODS: 24 rabbits were divided into three groups, in which the SVNF, subdermal vascular network skin graft and the traditional random flap were created respectively. Tissue hyalinization technique was used to find out the differences of vascular architecture between the three groups at the 1st-7th day, and the 14th day postoperatively. RESULTS: The results showed that the blood supply reestablished earlier in SVNF group than the other two groups. CONCLUSION: There is a zone of rapid and intensive angiogenesis in the mid-part of SVNF known as "intermediary pedicle". This is the anatomic basis of the early pedicle division and superior survival ratio of SVNF.

[Comparison of physiological characteristics of different ecotype plants].

Studies on the responses of photosynthesis, leaf water content and stoma resistance of 4 different ecotype plants to water stresses showed that their mechanism of drought-resistance was different. Mesic plants limited water loss from transpiration by increasing their stoma resistance, while xeric plants decreased water loss by keeping the high concentration of cell protoplasm. The latter had a higher efficiency of keeping water than the former. The leaf water content and stoma resistance was decreased from mesic to xeric plants, while the net photosynthetic rate per unit leaf was increased, indicating the difference of physiological characteristics among different ecotype plants.

[Long-term monitoring of artificial vagina reconstructed with sigmoid colon].

OBJECTIVE: To observe the influence of the artificial plastic vagina on overall patient health. METHODS: Length and width of the artificial vagina were measured. Histological studies were performed under light and electron microscope. Vaginal bacterial flora were analyzed using the drop-implant method. Vaginal HPV and ER were detected with ABC histochemical staining. RESULTS: According to the results of anatomic measurements, artificial vagina was similar to normal vagina. The reductions of glands in the mucosa and collagen fibers as well as interstitial tissue hyperplasia in the lower segment of artificial vagina were documented. Four kinds of bacteria indigenous to the sigmoid colon were found in artificial vagina and were studied microbiologically with only differences in the amount and detection rates of bacteria flora. The positive presentation of HPV was not significantly different from that in the control group. The positive presentation of estrogen receptor (ER) in the epithelium of artificial vagina was not found. CONCLUSION: The reconstruction of artificial vagina with sigmoid colon is a safe, effective and satisfactory procedure for the patient.

[Microsatellite instability in endometrial neoplasms correlation with clinicopathologic parameters and estrogen receptor or progesterone receptor status].

OBJECTIVE: To assess the prevalence of microsatellite instability in a series of endometrial carcinomas as well as to define the clinicopathologic features and estrogen receptor (ER) and progesterone receptor (PR) status associated with microsatellite instability (MI). METHODS: We examined 40 cases for replication error (RER) using polymerase chain reaction, polyacrylamide gel electrophoresis and silver stain at 8 microsatellite loci. Immunohistochemical staining of 27 paraffin sections was performed using antibodies to ER and PR. Finally, MI was compared with the clinicopathologic characteristics as well as ER and PR status. RESULTS: MI was observed in 9 (23%) at two or more loci, which is defined as RER positive phenotype. The 9 RER positive cases were all endometrioid type. Four non-endometrioid tumors failed to show MI in any locus. RER positive phenotype was more frequent in poorly differentiated (50%) than in well differentiated tumors (15%). We found no significant correlation of RER with stage and depth of invasion. Of 27 carcinomas, 19 (70%) showed homogeneous positive staining for ER and PR. ER and PR positive staining occured more frequently in RER negative endometrioid cancers than in RER positive cases. CONCLUSIONS: MI is one of the molecular mechanisms of a subset of endometrial carcinomas. Its frequency is associated with tumor grade, histological type and ER or PR status.

[Effects of dexamethasone on apoptosis and expression of Fas/FasL system in lung tissues of ALI rats].

OBJECTIVE: To determine whether apoptosis occurs in the lung tissues from rats with acute lung injury (ALI), and observe protective effects of dexamethasone by regulating apoptosis of lung tissues in rats with ALI. METHODS: By using TUNEL, in situ hybridization, SqRT-PCR and immunocytochemistry techniques, apoptosis and Fas, FasL expression were studied during early phase of ALI in rats. RESULTS: LPS leads to the rapid appearance of apoptosis in alveolar epithelial cells and pulmonary vascular endothelial cells at early stage of ALI in rats. The expression of Fas, FasL mRNA and protein was up-regulated in lung tissues of rats with ALI. The administration of dexamethasone suppressed apoptosis as well as expression of Fas, FasL mRNA and protein, inhibited TNF-alpha production and abated lung injury. CONCLUSIONS: The excessive apoptosis and Fas/FasL system may play a role in the pathogenesis of LPS-induced ALI in rats. The protective effects of dexamethasone include inhibiting inflammatory mediators production, suppressing the activation of Fas/FasL system and apoptosis in lung tissues of ALI rats.

[The study of tarnish induced by Actinobacillus actinomycetemcomitans serotype b (Aay4) on titanium and Ti-75 alloys].

OBJECTIVE: To study the tarnish induced by Aay4 on titanium and Ti-75 alloys. METHODS: Pure titanium and Ti-75 alloys were machined into size of 10 mm x 10 mm x 1 mm in tablets(30 of pure titanium; 60 of Ti-75 alloys). Thirty specimens sampled randomly from Ti-75 alloy together with 30 specimens of titanium were subjected to chemical passivation treatment. 30 specimens of the pure titanium, depassivation Ti-75 alloy and passivation Ti-75 alloy were randomly divided into three groups (blank control group, media control group and inoculated media group) respectively, every group contains 10 specimens. Under the anaerobic conditions, the surfaces of the modified GAM media were inoculated with Aay4. The labeled specimens surfaces were then aseptically placed on top of the media surface for 10 weeks with one generation time per week. The specimens were removed after 10 weeks of incubation, immersed in 0.05% glutaradehyde solution to eliminate Aay4, rinsed with distilled water. After 2 weeks, the specimens were removed and observed visually or examined by MINOLTA CR-100 color apparatus. RESULTS: Visual evaluation: Aay4 caused significant tarnish on specimens exposed to inoculated media, all the specimens surface changed into yellow color. Color apparatus evaluation: this result showed that comparing with the blank control, media control has no influence on all the specimens L*a*b* values (P > 0.05). Aay4 caused significant tarnish on specimens exposed to inoculated media, the result showed that there was significant difference between media control group and blank control group with inoculated media group on all the specimens L*a*b* values (P < 0.05). CONCLUSION: Aay4 caused significant tarnish on pure titanium, depassivation Ti-75 alloys and passivation Ti-75 alloys by naked eyes and color apparatus, according CIE1976L*a*b* color system reading, the color of titanium and Ti-75 alloys changed from the yellow-green ranges to red-yellow ranges.

[Study on 515 anti-tumor recipe in inducing leukemic HL-60 cells apoptosis].

OBJECTIVE: To observe the efficacy of 515 anti-tumor recipe on inducing human myeloid leukemic HL-60 cells apoptosis. METHODS: The morphological change of apoptotic cells was observed by light microscopy and transmission electromicroscopy (TEM), and flow cytometry and TUNEL reaction to further confirm the apoptosis of HL-60 cells. RESULTS: Typical apoptosis appeared such as marginal nuclei, chromatin condensation and nuclear fragmentation could be seen by light microscopy and TEM. The drug concentration was between 0.013-0.04 g/ml. The apoptosis rate is 30% in 0.04 g/ml group at the 72nd hour, but it was only 0.8% in the control group at the same time (P < 0.01). CONCLUSIONS: The 515 anti-tumor recipe can induce apoptosis in HL-60 cells. The apoptosis rate increased as the time extended. There was an enhanced efficiency of apoptosis in a dose-dependent manner.

[The potential role of growth factor in the airway wall remodeling of a chronic obstructive pulmonary disease rat model and the effects of drugs on them].

OBJECTIVE: To evaluate the expression and distribution of transforming growth factor-beta1 (TGF-beta1), epidermal growth factor (EGF) and basic fibroblast growth factor (bFGF) in the lung tissue of chronic obstructive pulmonary (COPD) rat models and the relationship between these growth factors and the airway wall remodeling. The effects of drugs on them were also investigated. METHODS: The COPD rat model (model group) was established by intratracheal instillation of lipopolysaccharide twice and daily exposure to cigarette smoking. Drug intervention groups received daily inhalation of heparin since the second week and TGF-beta1 monoclonal antibody (TB21) 0.5 mg twice through the tail veins. At the end of four weeks, the thickness of the smooth muscle and collagen in bronchi and pulmonary arterioles were measured by computer image analyzer, also the protein and gene relative content of these growth factors as well as the effects of drugs on them were observed. RESULTS: There was a significant increase in the smooth muscle and collagen thickness in the bronchi and pulmonary arterioles of the model group in comparison with that of the control group (P < 0.01), the relative contents for TGF-beta1, EGF and bFGF in the epithelial cells of the bronchi, endothelial cells of the pulmonary arterioles and alveolar macrophages of the model group were significantly higher than those of control group (P < 0.001 approximately 0.05). The relative content for TGF-beta1 in TB21 group was significantly lower than that of model group (P < 0.01). These were statistical positive relationships between the smooth muscle e thickness of bronchi and the relative contents for TGF-beta1, EGF and bFGF in the epithelial cells, between the collagen thickness of the bronchi and the relative content for TGF-beta1, between the smooth muscle thickness of the pulmonary arterioles and the relative content for TGF-beta1 and EGF in the endothelial cells (P < 0.05 approximately 0.01). CONCLUSION: TGF-beta1, EGF and bFGF may play an important role in the airway wall and pulmonary arteriole structure remodeling in COPD, the intervention against TGF-beta1 and long term inhalation of heparin mat be helpful for the inhibition of airway wall remodeling in human COPD and worth of further observation.

[Quantitative evaluation of dyspnea and the effects of M-receptor antagonist on dyspnea in patients with chronic obstructive pulmonary disease].

OBJECTIVE: To study the method of quantitative evaluation of dyspnea and to investigate the effects of M-receptor antagonist inhalation on dyspnea in patients with chronic obstructive pulmonary disease (COPD) during exercise. METHODS: Cycle exercise test was used in 27 patients with COPD and the effects of ipratropium bromide, a M-receptor antagonist, by inhalation on dyspnea and exercise capacity in the patients were observed. RESULTS: (1) Dyspnea index as Borg scale was closely correlated with oxygen uptake (V(O2)) during exercise in all the patients. Three variables for quantitative evaluation of dyspnea could be derived from the linear regression line. They were Borg scale slope (BSS), threshold load of dyspnea (TLD), and breakpoint load of dyspnea(BLD). (2) BSS was negatively correlated with forced expiratory volume in one second to forced vital capacity ratio (r = -0.519, P < 0.01) and diffusing capacity of the lungs for carbon monoxide (r = -0.497, P < 0.01) but positively correlated with residual volume/total lung capacity (r = 0.396, P < 0.05). (3) Inhalation of ipratropium bromide could significantly improve BSS [from (12.09 +/- 3.41) BS x L(-1) x min(-1) to (8.57 +/- 2.79) BS x L(-1) x min(-1), P < 0.01] and BLD [from (0.77 +/- 0.19) L/min to (0.81 +/- 0.14) L/min, P < 0.05] during exercise. CONCLUSIONS: (1) BSS, TLD and BLD could be used as reliable and convenient variables in quantitative evaluation of dyspnea in patients with COPD. BSS and BLD could to some extent reflect the severity of the disease. (2) M-receptor antagonists could alleviate dyspnea and increase exercise capacity in patients with COPD.

[The genetic instability of mismatch repair gene linked microsatellite and the evolution of CML].

OBJECTIVE: To explore the relationship between the genetic instability of microsatellite linked with mismatch repair gene and the evolution of CML to blast crisis. METHODS: The loss of heterozygosity (LOH) and microsatellite instability (MSI) of two polymorphic microsatellite markers, D2s123 and D3s1298, linked with mismatch repair gene hMSH2 and hMLH1 respectively, were detected by PCR-silver staining method on the bone marrow cells of 18 CML patients, who clinically progressed from the chronic phase to accelerated phase or blast crisis. RESULTS: Differences in microsatellite D2s123 and D3s1298 at the CML accelerated phase or blast crisis in 5 (27.8%) of the 18 patients were demonstrated compared with chronic phase. For D2s123, MSI and LOH were observed in 1 of 8 patients with accelerated phase (12.5%) and 2 of 10 patients in blast crisis (20%). For D3s1298, MSI and LOH were observed in 2 of 10 patients in blast crisis (20%). CONCLUSION: The genetic alteration of microsatellite D2s123 and D3s1298 may play a role in the progress of some CML cases.

[Allotransplantation of cultured fetal parathyroid gland cells in treating patients with hypoparathyroidism].

OBJECTIVE: To evaluate the therapeutic efficacy of transplantation of cultured human fetal parathyroid gland (chf-PTG) cells in treating patients with primary or secondary hypoparathyroidism. METHODS: Chf-PTG cells were allotransplanted into the renal adipose capsules of 6 patients with hypoparathyroidism under the ultrasonic guidance. The levels of serum parathyroid hormone (PTH) and calcium were monitored by radioimmunometric assay (RIA) and biochemical method, respectively. Both indexes of pre-and post-operation were compared and the data were analyzed. RESULTS: The levels of serum PTH and calcium were markedly elevated from three days to the first two weeks following transplantation of chf-PTG cells (P < 0.01). The PTH and calcium levels gradually stabilized from day 14 up to months 9-12, during which the symptoms of the patients alleviated or relieved. CONCLUSION: The transplantation of chf-PTG cells is a potential method for treating patients with primary or secondary hypoparathyroidism.

[Homogeneity study on the Streptococcus suis isolated from human and swine].

OBJECTIVE: To identify S. suis and to evaluate the homogeneity of isolates of S. suis from human and swine. METHODS: Culture, morphology, API biochemical tests and serum coagulate tests were used. All the 7 strains of streptococcus were confirmed to be S. suis serotype 2. Two strains and 1 strain were isolated from blood and cerebrospinal fluid (CSF) of patients, respectively, while the other 4 strains were obtained from the infected swine or their corpses. Seven strains of S. suis serotype 2 and reference strain SS2 were analyzed by randomly amplified polymorphic DNA analysis with six primers and Rep PCR with four primers and thalli fatty acid profile analysis. Cluster and principal compound analysis of results were performed with RAPD, Phylip and Treeview software. RESULTS: By analysis on RAPD patterns, a close relationship among 7 strains of S. suis serotype 2 and reference strain SS2 was discovered. Human-born strains and swine-born strains exhibit similar RAPD patterns. There was a same clonal relationship between strains originated from the blood and CFS of the patients. These results were confirmed by thalli fatty acid profile to have genotypic and phenotypic identity. CONCLUSION: Homogeneity exists among the 7 strains and reference strain SS2.
