Defective dietary fat processing in transgenic mice lacking aquaporin-1 water channels.

Immunocytochemistry showed expression of aquaporin-1 (AQP1) water channels at sites involved in dietary fat processing, including intrahepatic cholangiocytes, gallbladder, pancreatic microvascular endothelium, and intestinal lacteals. To determine whether AQP1 has a role in dietary fat digestion and/or absorption, mice were placed on a diet that contained 50% fat. Whereas wild-type mice (3-3.5 wk of age, 10-12 g) gained 49 +/- 5% (SE, n = 50) body weight in 8 days, and heterozygous mice gained 46 +/- 4%, AQP1 null mice gained only 4 +/- 3%; weights became similar after return to a 6% fat diet after 6 days. The null mice on a high-fat diet acquired an oily appearance, developed steatorrhea with increased stool triglyceride content, and manifested serum hypotriglyceridemia. Supplementation of the high-fat diet with pancreatic enzymes partially corrected the decreased weight gain in null mice. Absorption of [(14)C]oleic acid from small intestine was not affected by AQP1 deletion, as determined by blood radioactivity after duodenal infusion. Lipase activity in feces and small intestine was remarkably greater in AQP1 null than wild-type mice on low- and high-fat diets. Fluid collections done in older mice (that are less sensitive to a high-fat diet) by ductal cannulation showed threefold increased pancreatic fluid flow in response to secretin/cholecystokinin, but volumes, pH, and amylase activities were affected little by AQP1 deletion, nor were bile flow rates and bile salt concentrations. Together, these results establish a dietary fat misprocessing defect in AQP1 null mice.

Airway surface liquid pH in well-differentiated airway epithelial cell cultures and mouse trachea.

Airway surface liquid (ASL) pH has been proposed to be important in the pathophysiology of cystic fibrosis, asthma, and cough. Ratio image analysis was used to measure pH in the ASL after staining with the fluorescent pH indicator 2',7'-bis(2-carboxyethyl)-5(6)-carboxyfluorescein (BCECF)-dextran. ASL pH in bovine airway cell cultures grown at an air-liquid interface was 6.98 +/- 0.06 in the absence and 6.81 +/- 0.04 in the presence of HCO/CO(2). Steady-state ASL pH changed in parallel to changes in bath pH and was acidified by Na(+) or Cl(-) replacement but was not affected by the inhibitors amiloride, glibenclamide, or 4,4'-dinitrostilbene-2,2'-disulfonic acid. In response to sudden acidification or alkalization of the ASL by approximately 0.4 pH units by HCl/NaOH, ASL pH recovered to its initial value at a rate of 0.035 pH units/min (-HCO) and 0.060 pH units/min (+HCO); the pH recovery rate was reduced by amiloride and H(2)DIDS. In anesthetized mice in which the trachea was surgically exposed for measurement of BCECF-dextran fluorescence through the translucent tracheal wall, ASL pH was 7.14 +/- 0.01. ASL pH was sensitive to changes in blood pH created by metabolic (HCl or NaHCO(3) infusion) or respiratory (hyperventilation, hypoventilation) mechanisms. ASL pH is thus primarily determined by basolateral fluid pH, and H(+)/OH(-) transport between the ASL and basolateral fluid involves amiloride-sensitive Na(+)/H(+) exchange and stilbene-sensitive Cl(-)/HCO exchange. The rapid response of ASL pH to changes in systemic acid-base status may contribute to airway hypersensitivity in asthma and other airway diseases.

Determination of the enhancing action of HSP90 on neuronal nitric oxide synthase by EPR spectroscopy.

Recent studies showed that heat shock protein 90 (HSP90) enhances nitric oxide (NO) synthesis from endothelial and neuronal NO synthase (eNOS and nNOS, respectively). However, these findings were based on indirect NO measurements. Moreover, although our previous studies showed that the action of HSP90 involves increased Ca(2+)/calmodulin (Ca(2+)/CaM) binding, quantitative measurements of the effect of HSP90 on CaM binding to nNOS have been lacking. With electron paramagnetic resonance spectroscopy, we directly measured NO signals from purified nNOS. HSP90 augmented NO formation from nNOS in a dose-dependent manner. Tryptophan fluorescence-quenching measurements revealed that HSP90 markedly reduced the K(d) of CaM to nNOS (0.5 +/- 0.1 nM vs. 9.4 +/- 1.8 nM in the presence and absence of HSP90, P < 0.01). Ca(2+) ionophore triggered strong NO production from nNOS-transfected cells, and this was significantly reduced by the HSP90 inhibitor geldanamycin. Thus these studies provide direct evidence demonstrating that HSP90 enhances nNOS catalytic function in vitro and in intact cells. The effect of HSP90 is mediated by the enhancement of CaM binding to nNOS.

Selective attenuation by adenosine of arrhythmogenic action of isoproterenol on ventricular myocytes.

We examined whether adenosine equally attenuated the stimulatory effects of isoproterenol on arrhythmic activity and twitch shortening of guinea pig isolated ventricular myocytes. Transmembrane voltages and whole cell currents were recorded with patch electrodes, and cell twitch shortening was measured using a video-motion detector. Isoproterenol increased the action potential duration at 50% repolarization (APD50), L-type Ca2+ current [I(Ca(L))], and cell twitch shortening and induced delayed afterdepolarizations (DAD), transient inward current (I(Ti)), and aftercontractions. Adenosine attenuated the arrhythmogenic actions of isoproterenol more than it attenuated the effects of isoproterenol on APD50, I(Ca(L)), or twitch shortening. Adenosine (0.1-100 micromol/l) decreased the amplitude of DADs by 30 +/- 6% to 92 +/- 5% but attenuated isoproterenol-induced prolongation of the APD50 by only 14 +/- 4% to 59 +/- 4% and had no effect on the voltage of action potential plateau. Adenosine (30 micromol/l) inhibited I(Ti) by 91 +/- 4% but decreased isoproterenol-stimulated I(Ca(L)) by only 30 +/- 12%. Isoproterenol-induced aftercontractions were abolished by adenosine (10 micromol/l), whereas the amplitude of twitch shortening was not reduced. The effects of adenosine on twitch shortenings and aftercontractions were mimicked by the A1-adenosine receptor agonist CPA (N6-cyclopentyladenosine) and by ryanodine. In conclusion, adenosine antagonized the proarrhythmic effect of beta-adrenergic stimulation on ventricular myocytes without reducing cell twitch shortening.

A low level of TNF-alpha mediates hemorrhage-induced acute lung injury via p55 TNF receptor.

Acute lung injury after hemorrhagic shock (HS) is associated with the expression of tumor necrosis factor (TNF)-alpha in the lung. However, the role of TNF-alpha and its receptors in this pulmonary disorder remains obscure. This study examined the temporal relationship of pulmonary TNF-alpha production to neutrophil accumulation during HS and determined the role of TNF-alpha in neutrophil accumulation and lung leak. HS was induced in mice by removal of 30% of total blood volume. Lung TNF-alpha was measured by ELISA. Neutrophil accumulation was detected by immunofluorescent staining, and microvascular permeability was assessed using Evans blue dye. Although HS induced a slight and transient increase in lung TNF-alpha, neutrophil accumulation preceded the increase in TNF-alpha. However, lung neutrophil accumulation and lung leak were abrogated in TNF-alpha knockout mice, and both were restored by administration of recombinant TNF-alpha to TNF-alpha knockout mice before HS. Neutrophil accumulation and lung leak were abrogated in mice lacking the p55 TNF-alpha receptor, but neither was influenced by p75 TNF-alpha receptor knockout. This study demonstrates that a low level of pulmonary TNF-alpha is sufficient to mediate HS-induced acute lung injury during HS and that the p55 TNF-alpha receptor plays a dominant role in regulating the pulmonary inflammatory response to HS.

Noninvasive in vivo fluorescence measurement of airway-surface liquid depth, salt concentration, and pH.

The concentration of salt in the thin layer of fluid at the surface of large airways, the airway-surface liquid (ASL), is believed to be of central importance in airway physiology and in the pathophysiology of cystic fibrosis. Invasive sampling methods have yielded a wide range of ASL [NaCl] from 40 to 180 mM. We have developed novel fluorescent probes and microscopy methods to measure ASL thickness, salt concentration, and pH quantitatively in cell-culture models and in the trachea in vivo. By rapid z-scanning confocal microscopy, ASL thickness was 21 +/- 4 microm in well-differentiated cultures of bovine tracheal epithelial cells grown on porous supports at an air-liquid interface. By ratio imaging fluorescence microscopy using sodium, chloride, and pH-sensitive fluorescent indicators, ASL [Na+] was 97 +/- 5 mM, [Cl-] was 118 +/- 3 mM, and pH was 6.94 +/- 0.03. In anesthetized mice in which a transparent window was created in the trachea, ASL thickness was 45 +/- 5 microm, [Na+] was 115 +/- 4 mM, [Cl-] was 140 +/- 5 mM, and pH was 6.95 +/- 0.05. Similar ASL tonicity and pH were found in cystic fibrosis (CFTR-null) mice. In freshly harvested human bronchi, ASL thickness was 55 +/- 5 microm, [Na+] was 103 +/- 3 mM, [Cl-] was 92 +/- 4 mM, and pH was 6.78 +/- 0.2. These results establish by a noninvasive approach the key properties of the ASL and provide direct evidence that the ASL is approximately isotonic and not saltier in cystic fibrosis.

Effects of leucocyanidines on activities of metabolizing enzymes and antioxidant enzymes.

Procyanidolic oligomers (leucocyanidines, LCs) extracted from grape seeds (Vitis vinifera) are known to have antioxidant and antimutagenic activities, and a protective effect against cardiovascular disease. In the present study we examined the influence of LCs on the activities of phase 1 enzymes and conjugation enzymes and on antioxidant enzymes such as superoxide dismutase, catalase, and glutathione peroxidase. Administration of LCs (25, 50, and 100 mg/kg. p.o. for 7 d) markedly decreased the activities of NADPH-cytochrome P450 reductase, P4501A1, P4501A2, and P4503A4, but significantly increased the activities of glutathione S-transferase and phenolsulfotransferase in rat liver. However, the activities of antioxidant enzymes were not affected by LC administration. The inhibition of P450s and increases in phase II enzyme activities indicate a role for LCs as a chemopreventive agent against toxic or carcinogenic metabolites of P450 isozymes.

Expression of metalloproteinases and its inhibitor in later stage of rabbit neointima development.

Neointima formation after arterial de-endothelialization refers not only to smooth muscle cell (SMC) migration and proliferation, but also involves extracellular matrix (ECM) metabolism. Most studies regarding the role of matrix metalloproteinases (MMPs) and tissue inhibitors of metalloproteinases (TIMPs) in neointima have focused on the early phase of vascular remodeling. In this study, we examined the expression of MMP and TIMP in rabbit aortic neointima at a relatively late stage of lesion development, between 4 and 12 weeks after initial de-endothelialization. Northern blot analysis revealed expression of steady-state MMP-9 mRNA was increased up to the 4th week and MMP-2 mRNA to the 12th week after de-endothelialization. In situ hybridization shown that MMP positive cells were predominantly distributed in arterial neointima. Expression of TIMP-1 mRNA was continuously up-regulated up to the 12th week and TIMP-1 positive cells, primarily SMCs, were also localized to the neointimal tissue. Alteration at mRNA level was accompanied by that at protein level, as assessed by SDS-PAGE zymography for MMPs and immunoblotting for TIMP-1. The profile of alteration at protein level correlated well with that at mRNA level. These data suggest that synthesis of MMPs and TIMP is a prolonged process and arterial SMC is a major source of MMP production in arterial neointima. Enhanced synthesis of MMPs and TIMPs at late stage of neointimal development may contribute to arterial ECM metabolism.

[Study on polymorphism of D gene exons among RhD-negative Chinese Han population].

OBJECTIVE: To explore the genomic structure of 8 exons in D gene of RhD(-) Chinese Han population. METHODS: Polymerase chain reaction-sequence specific primers(PCR-SSP) were used to study genomic DNA from 50 samples of RhD(-) Chinese Han donors. Exons 2, 3, 4, 5, 6, 7, 9, 10 of RHD gene and exons 1, 2, 4, 5 of RHCE gene were specifically amplified, also intron 4 of them was amplified. RESULTS: Phenotypes of the 50 RhD(-) donors were: 22 ccdee, 22 Ccdee, 3 CcdEe, 3 CcdEe. The 8 exons of RHD gene were completely absent in 25 donors with ccdee or ccdEe phenotype, while polymorphisms of D gene exons were found in 25 donors with Ccdee or CcdEe phenotype: the presence of all 8 exons of D gene in 9 donors, the absence of the 8 exons in 7 donors, the presence of exon 2 in 5 donors, the presence of exon 6 in 3 donors and the presence of exons 2, 6, 10 in 1 donor were demonstrated. CONCLUSION: Polymorphisms of RHD gene exons were present among RhD(-) Chinese Han blood donors. The 8 exons of RHD gene were completely absent in donors with Rhesus cc phenotype, while 5 polymorphisms of RHD gene exons were found in donors with Rhesus Cc phenotype. The discrepancy of the RHD gene in RhD(-) individuals between Chinese Hans and Caucasians indicates that care should be exercised by clinicians in the use of the RhD genotyping results.

[Application of chromosome painting technique to analysis of structural aberration of human chromosomes].

OBJECTIVE: This study was aimed at using chromosome painting technique to detect translocation, especially microtranslocations, on chromosomes in comparison with G-banding analysis. METHODS: Chromosome painting technique was applied to analysis of metaphase chromosomes of patients for detecting translocations with biotin-labeled chromosomes X, Y, 14q, 10 specific probes. RESULTS: Fluorescence in situ hybridization FISH signals were shown clearly in slides even in specimen stored at room temperature for 10 years and at -80 centigrade degree. Translocations were located precisely. CONCLUSION: Microtranslocations, which are hard to analyze by G-banding, can be detected exactly using chromosome painting technique with G-band karyotype on metaphase chromosome.

[Applied researches on polylactide internal fixation devices].

Nowadays, more and more basic and clinical researches on polylactide internal fixators were carried out in China. In this paper are reviewed the researches of polylactide acid(PLA), including its physical and chemical characters, biodegradation, absorption and mechanical properties, effects on fracture healing, and clinical application. Some problems that have not been solved are high-lighted and discussed. Also dealt with are some researches of PLA in future.

[The effects of Vitreoscilla hemoglobin expression on growth and antibiotic production in Streptomyces cinnamonensis].

Streptomyces cinnamonensis A-10 is a monensin \ producing strain. pHZ1252 is an E. coli-Streptomyces shuttle vector for expressing VHb, in which vhb structural gene is controlled by thiostrepton-inducible promoter PtipA. pHZ1252 was unstable in S. cinnamonensis, occurring deletion recombination. The deletion DNA fragment of pHZ1252 included not only part of E. coli plasmid but also vhb gene. However, plasmid pHZ1252 isolated from S. avermitilis transformants, which lost part of E. coli plasmid but still contained vhb gene and PtipA was stable in S. cinnamonensis and expressed VHb with biological activity after thiostrepton induction. Shake flask fermentation experiments showed that the presence of VHb in S. cinnamonensis efficiently enhanced cell growth and improved antibiotic production under aeration-poor conditions.

[Comprehensive assessment on environmental quality in vulnerable ecotone of Tarim River Basin].

Tarim River Basin (TRB) is one of ecological environment vulnerable areas. Due to the matching of material and energy existed in some problems, the different fragility characteristics occurred obviously. According to related principles of ecological environment quality assessment, combined with ecological environment situation of Tarim River Basin, 20 indexes were selected, and the indicator system for synthetically ecological environment assessment was built. Moreover, ecological fragility index was set up, which can be used to indicate the degree of ecological environment quality in Tarim River Basin. The results were as follows: Aksu River Basin belongs to improved area. Yerkart River Basin and the upper reaches of TRB belongs to balance areas and Hetian River Basin and the middle reaches of TRB belongs to maladjustment areas. The lower reaches of TRB belongs to severe damage area. The assessment results are consonant with actual situation, which play an important function in ecological environment construction.

[Genetic structure and intraspecific genetic polymorphisms in natural populations of Psathyrostachys huashanica].

Psathyrostachys huashanica Keng, which is endemic to China, only distributes in Mt. Hua of Shaanxi Province. 5 enzyme systems coding 13 loci were analyzed in 13 subpopulations of P. huashanica from Mt. Hua. Allozyme analysis showed that high genetic variability and clonal diversity existed in the species. Mean proportions of polymorphic loci (95% criterion), average number of alleles per locus, and mean expected heterozygosity were 69.2%, 1.8 and 0.344, respectively. Simpson index was 0.998. A negative species mean fixation indice (-0.252) indicates that there is a considerable excess of heterozygotes compared to the expected proportions of heterozygote loci. Mean genetic distance among populations was 0.049 (range: 0-0.139), and 91% of the genetic variation resided within populations. Gene flow estimated by indirect methods was 2.77, which was much lower than the Nm value (5.24) of anemophytes. This suggested that gene flow of P. huashanica might be in a critical level, and genetic differentiation might occur in natural population of this species.

Protein structural domain parsing by consensus reasoning over multiple knowledge sources and methods.

Domain parsing, or the detection of signals of protein structural domains from sequence data, is a complex and difficult problem. If carried out reliably it would be a powerful interpretive and predictive tool for genomic and proteomic studies. We report on a novel approach to domain parsing using consensus techniques based on Hidden Markov Models (HMMs) and BLAST searches built from a training set of 1471 continuous structural domains from the Dali Domain Dictionary (DDD). Validation on an independent test sample of family-matched structural domain sequences from the Scop database yields a consensus prediction performance rate of 75.5%, well above the 58% obtained by simple agreement of methods.

[Effect of VAM fungi on phosphatase activity in maize rhizosphere].

The effect of VAM fungi on phosphatase activity in maize rhizosphere was examined by pot culture experiment, in which, three-compartment-pots were used, the central compartment being separated from the outer two by a nylon net with 30 microns mesh. Plants were harvested 70 days after planting. Soil acid and alkaline phosphatase were measured at different distances from root surface. The results showed that VAM increased the activities of soil acid and alkaline phosphatase in the rhizosphere. It was found that different phosphorous sources had different effects on phosphatase activity.

[Landscape dynamics of two-side lands-along main urban roads in southwestern Shanghai].

Based on the aerial photos of Shanghai in 1984, 1989 and 1994, the landscape databank of southwestern Shanghai was set up. According to the analysis of the landscape dynamics of three two-side belts along the main urban roads in southwestern Shanghai, it was showed that there was a great difference in the landscape succession of these belts with various functions. The main urban roads trended to multi-serving function with the expansion of the city. The change from increase to decrease in the diversity and evenness of landscape and the decline in landscape fragmentation were the major symbols of the landscape dynamics.
