[Histologic study on complication reducing effects of the tumescent technique in liposuction].

The purpose of this experiment was to investigate how the tumescent technique reduces complications in liposuction. Histologic examinations revealed that the septa of fat are attached to the deep surface of the dermis and to the fascia superficialis. It is important to retain the septa that contain the perforating vessels and nerves during liposuction. The correct choices for instrument and suction pump can minimize trauma to tissues and reduce complications.

[Endoscopic subperiosteal facial rejuvenation].

It is just recently that video endoscopic techniques began to be applied to facial rejuvenation in China. Since July 1995, the authors have performed 15 operations of subperiosteal rejuvenation under the video endoscope, combined with liposuction of the sub-mandible region and the nasolabial fold. The operation uses small incisions, produces satisfactory results with less edema, less numbness and without alopecia. The indications, instrumentation, surgical technique and results and presented and discussed. According to our clinical experience, the best candidates for this operation are those aged from 40 to 55. For the patients about sixty years old, a mini multiplane facelift should be carried out in the cheek and neck region following the subperiosteal facelift. The more elderly patients are not candidates for this operation. We emphasize that the dissected scalp flap should be uplifted to the maximum and anchored on the cranial bone to ensure a satisfactory result.

[Study of mechanism of PTH modulating cytosolic free calcium in rat DCT].

OBJECTIVE AND METHODS: In order to understand the mechanism of PTH in regulating cytosolic free calcium concentration ([Ca2+]i), we differentiated rat renal proximal and distal convoluted tubules (PCT&DCT) by Percoll density gradient centrifugation, and successfully gauged the effect of hPTH (1-84), dibutyryl cAMP (db cAMP, a kind of cAMP activator) and prostaglandin E2 (PGE2) on [Ca2+]i of DCT using fluorescent Ca2+ indicator Fluo 3-AM. We found that PTH, db cAMP and PGE2 all could increase [Ca2+]i. RESULTS: The results suggest that the rise in [Ca2+]i induced by PTH in DCT was through activation of cAMP, and PGE2 might be also involved in regulation of [Ca2+]i as a messenger of PTH.

[Observations on before and after employ of screw of implant denture with SEM].

The loss of screw is one of the clinic complication in MDIC implant denture cases. The purpose of this study is to find the cause of loss of screw. The authors has studied MDIC implant denture with SEM before and after employ of the screw and found that the causes of screw loss were crevice corrosion and friction pressure. Crevice corrosion may promote the loss of screw. Base the above condition, it is necessary to select Ti-alloy screw and associat with anticorrosive measure in MDIC implant denture construction.

[Experimental study on the effect of extremities ischemia reperfusion inducing anterior tibial compartment pressure].

We observed the changes of compartment pressure after extremities were reperfused at different intervals of ischemia and its relationship with the changes of serum MDA and CPK content. The protective effect of mannite on extremities ischemia reperfusion was also studied. 18 New Zealand rabbits were separated into three groups: group A (ischemia 4 h); group B (ischemia 8 h); group C (ischemia 4 h). The rabbits of group A and B were intravenous injected with normal saline (NS), and group C was injected with 20% mannite. The changes of compartment pressure of serum MDA and CPK content and the histological changes of skeletal muscles in every group were noted at different times: preoperation; the beginning of ischemia; the end of ischemia and reperfusion for 4 h; 24 h; 72 h, respectively. The longer ischmia lasted, the more serious the tissue damage was. Reperfusion made tissue damaged further. After reperfusion for 24 h, the damage was the most serious. Meanwhile, the serum MDA and CPK content also reached the peak value. These changes were in accord with those of compartment pressure. On the other hand, the damage of mannite group was relatively mild. Extremity ischemia reperfusion would led to the increase of compartment pressure, then intensified the damage. The damage was the most serious at 24 h of reperfusion. Using mannite could reduce the tissue damage and compartment pressure. Mannite should be recommended as the first-selected drug for extremity ischemia reperfusion injury.

[Ahmed valve implantation for refractory glaucoma].

OBJECTIVE: To evaluate the short-term and long-term efficacy of Ahmed valve implantation for refractory glaucoma and discuss its intraoperative and postoperative complications and management. METHODS: 20 cases of refractory glaucoma underwent Ahmed valve implantation, including 9 cases of neovascular glaucoma, 7 cases of aphakic or pseudophakic glaucoma, 3 cases of congenital glaucoma and one primary chronic angle-closure glaucoma. RESULTS: One month postoperatively, the intraocular pressure (IOP) was less than or equal to 2.8 kPa (1 kPa = 7.5 mmHg) in fifteen cases, with a total success rate of 75%. For cases with neovascular glaucoma, the success rate was 55.6%, while for the others, the rate was 90.9%. After six months of follow-up for 12 patients, the total success rate was 58.3%, for neovascular glaucomas and other glaucomas, the rate was 40.0% and 71.4%, respectively. The complications included transient hyphema, early postoperative hypotony, obstruction of the tube tip, tube touch to lens or cornea, tube exposure, exudative choroidal detachment and dropout of the plate. CONCLUSION: Ahmed valve implantation is an effective method in the management of refractory glaucoma in spite of its unnegligible complications.

[The expression of tumor suppressor gene p53 and Rb gene in nasopharyngeal carcinoma].

In order to observe the relation of tumor supperssor gene p53 and Rb gene to nasopharyngeal carcinoma, we investigated p53 gene mutations in exon 7-8 in 33 cases of nasopharygeal carcinoma (NPC), using single-strand conformation ploymorphism analysis of PCR (PCR-SSCP). The data showed p53 mutations in 7 of 33 specimens at exon 8 (21.2%). No mutation in exon 7 was detected. Analysis of Rb gene in NPC tissues by southern blot revealed the deletion and loss of activity of Rb gene in 11 of 13 cases (86.6%). Our results suggest that Rb gene and p53 gene are closely associated with the tumorigenesis of NPC.

[Clinical study on myxoma and chordoma in the skull base].

OBJECTIVE: The clinical manifestation, neuroimaging properties, and pathologic changes of myxoma and chordoma were studied. METHODS: Their neuroimaging properties and pathologic changes of myxoma 7 cases and chordoma 13 cases were analyzed. RESULTS: (1) Clinical manifestation: the injury of oculomotor nerve and exorbitism is dominant in myxoma, but the injury of posterior cranial nerves is dominant in chordomas. (2) Plain skull X-ray film: local irregular punctate calcification and osseous necrosis were common in myxoma. Clivus invasion moruloid calcification and cauliflower-like calcification are always seen in chordoma. (3) CT and MRI: Location of the tumor and their relations with the adjacent structure were clear. (4) Pathologic changes: Cells are punctately arranged in myxoma, and vesicularly arranged in chordoma. CONCLUSION: The clinical study of myxoma and chordoma is not only useful for their differiental diagnosis, but can provide valuable clues for their surgical approach as well.

Reflux esophagitis and airway hyperresponsiveness.

OBJECTIVES: To observe the effects of reflux esophagitis(RE) on the lung function and airway reactivity, and study the mechanism of airway hyperresponsiveness(AHR) in patients with RE. METHODS: Lung function measurements and airway provocation tests were performed in 31 RE patients and 35 control subjects. TXB2 and PGF1a were determined in 20 cases of each group. RESULTS: In RE patients the lung function was lower and the rate of AHR was higher than control subjects (P < 0.05). Among RE patients 25% had higher airway sensitivity (Dmin < 3 u). The TXB2 of RE patients with AHR was higher than those without AHR. Dmin correlated significantly with TXB2 (r = -0.653, P < 0.05). CONCLUSIONS: RE could damage the lung function. The rate of AHR was 61%, the high airway sensitivity was probably potential asthma, and TXB2 may play a role in the pathogenesis of AHR.

[Determination on the electric-mate order of prosthodontic materials in artificial saliva].

Electric-mate corrosion of the different prosthodontic materials has been noticed in oral cavity. The aim of this investigation was to determine corrosion potential of prosthodontic materials, and to acquire the electric-mate order in artificial saliva. That will conduct us to select the materials of resistance corrosion. For resistance electric-mate corrosion, it's better to use same materials, or to use close materials of the electric-mate order.

[Hepatitis B virus DNA detection by means of polymerase chain reaction in patients with chronic hepatitis and hepatocellular carcinoma].

HBV immunological detection and HBV DNA detection by polymerse chain reaction (PCR) or molecular hybridization with 32P labeled probe were done in 61 patients with chronic hepatitis (CH) and 47 patients with hepatocellular carcinoma (HCC) to investigate the relationship between HBV replication and serum markers. The HBV DNA was detected in 90.50% of CH patients and 50.00% of HCC patients with HBsAg, HBeAg and anti-HBc positive; in 45.40% of CH patients and 7.14% of HCC patients with HBsAg, anti-HBe and anti-HBc positive; in 60.00% of CH patients and 40.00% of HCC patients with HBsAg and negative HBeAg, anti-HBe positive; in 20.00% of CH patients and 22.22% of HCC patients with HBsAg negative and anti-HBe or anti-HBe or anti-HBs positive; in 0 of CH or HCC patients lack of HBV serum markers. Our data suggested that the most active HBV replication was correlated to the presence of both HBsAg and HBeAg, there were some extent of HBV replication in CH or HCC patients with positive HBsAg, the inhibition of HBV replication was related to anti-HBe, the less active HBV replication was seen in HCC patients when compared with CH patients.

[Determinaion of artificial synthetic pigment in the drug by reflection spectrophotometry].

Solid phase spectrophotometry is a new, simple, fast and sensitive trace analytical method. In this report, it is presented a method that the polyamide was used for the absorbence and the reflectances absorbances were measured directly with remodelled the colorimetric cell. Not only the trace inorganic ion can be analysed by the solid phase method, but also the artificial synthetic pigment in the drug can be determined, the results are satisfied.

Regulation of ceruloplasmin by retinoic acid in the developing rat.

Ceruloplasmin (Cp), the major copper-binding protein in the plasma, is an acute phase protein with ferrioxidase activity. Both its oxidase activity and hepatic mRNA abundance increase during the developmental period. To test the possible role of retinoic acid, a derivative of vitamin A known to participate in cellular differentiation and development, on the developmental regulation of Cp, neonatal rat pups were injected with 2 micrograms 13-cis-retinoic acid (RA)/g body weight on postnatal day 1. Serum Cp activity and hepatic Cp mRNA were measured over the next 3 weeks in RA-treated and vehicle-treated controls. Serum Cp activity increased 2.5-fold 24 h after RA administration. However, hepatic Cp mRNA abundance was not elevated during this time period, suggesting that the action of RA on Cp activity was the result of post-transcriptional changes.

2,2'-Dithiobisbenzamides and 2-benzisothiazolones, two new classes of antiretroviral agents: SAR and mechanistic considerations.

Substituted 2,2'-dithiobisbenzamides and 2-benzisothiazolones were prepared and shown to possess low microM activity with high therapeutic indices against HIV-1, HIV-2 and SIV in cell culture. The mechanism of antiviral action was determined to be directed toward the nucleocapsid protein (NCp7), which contains two zinc fingers and plays vital roles in the viral life cycle. The "active sulfides" of this study cause the extrusion of zinc from these zinc fingers. Structure-activity relationships of the 2,2'-dithiobisbenzamides reveal that the disulfide bond and the ortho benzamide functional groups are essential for activity, with the best compounds having a carboxylic acid, carboxamide, or sulfonamide substituent. The 2-benzisothiazolones are formed from the disulfides both chemically and in vivo and their SAR mimics that of the 2,2'-dithiobisbenzamides. The antiviral activity of the disulfides may require cyclization to the isothiazolones. Two agents, PD 159206 and PD 161374, which showed good antiviral activity, physical properties, and excellent pharmacokinetics in mice, were selected for advanced studies.

Purification of mouse H1 histones expressed in Escherichia coli.

We amplified the coding regions of the previously cloned H1 genes H1-1, H1 zero and H1t and inserted them into the expression vector pET-11d. The synthesis of the H1 histones can be induced in the appropriate strains of bacteria, and the H1 histones can be readily purified. We report detailed protocols for the purification of the expressed proteins using combinations of ion-exchange and reverse-phase HPLC. Sufficient amounts of each pure variant protein can be obtained for use in physical studies of H1-DNA interactions.

[Glycosylated high density lipoprotein in diabetes related to pregnancy induced hypertension].

OBJECTIVE: To investigate the glycosylation of high density lipoprotein (glc-HDL) and its specific binding to human lung fibroblasts (HLF) receptors in gestational diabetes woman and to study its relationship with pregnancy induced hypertension (PIH). METHODS: The glc-HDL was measured by fluorimetry and the specific binding of HDL to HLF receptors was measured with enzyme linked immunoreceptor assay in 32 gestational diabetic woman group 1, 34 group II, 36 normal pregnant (NP) and 32 normal non-pregnant (NNP) women. RESULTS: The levels of glc-HDL and the intracellular total cholesterol in diabetic pregnant woman group I and II were significantly higher than those in the NP and the NNP (P < 0.01). The cell binding, internalization and degradation of glc-HDL were higher than that in the NP and the NNP. CONCLUSION: The change of HDL level and lipid deposition during the process may be one of the causes for arteriole spasm in PIH.

[Role of nitric oxide in pathogenesis of pregnancy induced hypertension].

OBJECTIVE: To investigate the role of nitric oxide (NO) in pathogenesis of pregnancy induced hypertension (PIH). METHODS: N(G)-nitro-L-arginine-methyl-ester (L-NAME) was used to inhibite the synthesizing of NO in the third trimester of pregnancy rats. Indices including HGB, PLT, HCT, protein uria, serum endothelin (ET), thromboxane B2(TXB2), 6-keto-PGF1 alpha and blood pressure in pregnant rats were measured. The length, weight of body, liver and brain of full-term newborn rats and the weight and pathological changes of placenta were observed. RESULTS: After inhibition of NO synthesis, blood pressure were consistently high. The protein uria, HCT, ET, TXB2/6-keto-PGF1 alpha were significantly higher and PLT was significantly lower than that of control group. The length, weight of body and placenta in neonatal rats were lower than those of the control group (P < 0.01). Severe ischimic lesion, thrombosis of vessels and endothelial proliferation in placenta were observed. CONCLUSION: NO plays an important role in pathogenesis of PIH.
