[Detection of human papillomavirus infection in cervical Pap smears by computer-assisted cytologic test].

OBJECTIVE: The sensitivity and accuracy of cytologic computer-assisted test (CCT) in diagnosis of human papillomavirus (HPV) infection in cervical Pap smear were evaluated. METHODS: Cervical Pap smears obtained from 158 patients with vulva condyloma acuminata were examined by CCT. The diagnostic criteria were based on The Bethesda System (TBS). Simultaneously HPV DNA in cervical mucus was examined by polymerase chain reaction (PCR) technique and cervical biopsy taken under the guidance of colposcopy for pathological examination. RESULTS: The sensitivity and accuracy of CCT in diagnosis of cervical HPV infection were 74.62% and 67.72% compared with pathological results whereas 69.11% and 70.88% compared with PCR results respectively. CONCLUSION: The results indicates that CCT, a cytologic pathological technique, is a useful method in diagnosis of clinical and subclinical cervical HPV infection.

[Genetic instability of microsatellites Mfd41 and DCC in the evolution of chronic myelogenous leukemia].

OBJECTIVE: To explore the relationship between the genetic instability of microsatellite and the evolution of chronic myelogenous leukemia(CML). METHODS: The loss of heterozygosity(LOH) and the microsatellite instability(MSI) of two polymorphic microsatellite markers, DCC and Mfd41, which located on chromosome 18q and 17p respectively, were assayed by standard PCR-silver staining analysis in bone marrow cells from 17 CML patients progressing from chronic phase to accelerated phase or blast crisis. RESULTS: LOH or MSI of the two microsatellites were demonstrated in 8/17 (47.5%) patients in accelerated/blastic phases. For DCC, genetic instability was revealed in 2 of 9 patients in accelerated phase and in 4 of 8 in blast crisis. For Mfd41, genetic instability was revealed in 1 of 9 patients in accelerated and in 1 of 8 in blast crisis. CONCLUSION: Genetic instability of DCC and Mfd41 may play a role in the evolution of CML.

[Morphological observations of tumors in the cardiac conduction system].

OBJECTIVE: To investigate the incidence of tumors in the cardiac conduction system (CCS) and to study their relation to sudden deaths. METHODS: The CCS from 149 cases of sudden death without extracardiac cause of deaths and those from 737 cases of non-cardiac death were sampled using technique designed by the authors. Routine histological examinations were carried out. RESULTS: Tumors in the CCS were found in 12 cases (1.35%). Another case with tumor in the CCS was found in a specimen received for consultation. Of these 13 tumors, 10 were primary benign tumors affecting the CCS, 3 were metastatic tumors. The benign tumors included fibroma, hemangioma, lipomatous hypertrophy of the atrial septum, mesothelioma and rhabdomyoma. 8 of the 10 cases were located in the SAN or AVN. All the metastatic tumors were in the SAN. Of the 13 cases, 3 died suddenly. CONCLUSION: Tumors in the CCS are the smallest tumors which can cause sudden death.

[Effect of HHV-6 on expression of M-CSF in patients with hematological disorders].

OBJECTIVE: To study the relationship between HHV-6 infection and abnormal expression of M-CSF in bone marrow mononucleated cells (MNC) from patients with hematological disorders. METHODS: M-CSF expression level were detected in 30 specimens of MNC from patients with hematological diseases by ABC immunocytochemical staining and reverse transcription polymerase chain reaction methods. RESULTS: The percentage and score of M-CSF antigen positive cells, and percentage of M-CSF mRNA expressing samples derived from HHV-6 infected group were 36.9% +/- 24.9%, 45.9 +/- 31.4 and 83.3%, respectively, which were significantly higher than those of control (20.3% +/- 19.4%, 25.3 +/- 24.2 and 50.0%, respectively) (P < 0.001). HHV-6 could promote M-CSF production to a large extent in MNC from leukemic patients than that from patients with benign hematological disorders. M-CSF existed mainly on membrane or in cytosol of granulocytes and monocytes. CONCLUSION: HHV-6 infection in vitro could promote M-CSF production in MNC from patients with hematological diseases.

[Clinical study on acute respiratory infection caused by chlamydia pneumoniae].

OBJECTIVE: To find out the infected rate and the clinical features of patients with acute respiratory infection caused by chlamydia pneumonia(C. pneumoniae). METHOD: A prospective study for chlamydia pneumoniae infection was conducted in 110 patients with respiratory tract infection from January to December 1995 in Nanjing. Sputum and throat swab specimens were taken and C. pneumoniae DNA were detected by polymerase chain reaction (PCR) with the HM-1-HR-1 primer pair. At the same time, serum samples were taken and immunoglobulins G and M (IgG and IgM) fractions of antibodies to C. pneumoniae were studied by microimmunofluorescence test. RESULT: Prevalence of specific IgG was 70% in patients with respiratory tract infection. Seventeen patients (16%) were serologically diagnosed as having recent C. pneumoniae infection and 12 patients (11%) had the positive PCR in sputum and/or swab specimens. The total positive rate was 23% (25/110) combining PCR with serology. Acute infection of C. pneumoniae was common in the patients with asthma (57%), pneumonia (35%), COPD (26%) and bronchitis (25%). The clinical feature between C. pneumonia infection (n = 5) and non-C. pneumonia infection (n = 85) was not significanty different. CONCLUSION: Data suggest that Chlamydia pneumonia is an important pathogen that causes infections of the human respiratory tract and attention should be drawn to this special illness.

[Biological responses of a Streptomyces strain producing-Nikkomycin to space flight].

In order to see biological responses to the production of Nikkomycins in general and Nikkomycin X and Z in particular by space conditions, Streptomyces ansochromogenus, a Nikkomycins-producing strain, was carried onboard a satellite for 15 d in 1996. Several strains were isolated from the treated sample and found that the productivity of Nikkomycins in all was increased by 13-18 percent, and the proportion of Nikkomycin X and Z increased correspondingly. Besides, some biological properties of the isolated strains varied markedly.

[Biomechanical characteristics of masticatory muscles of rat after using bite plate].

OBJECTIVE: The purpose of this study is to evaluate the changes on biomechanical characteristics of masticatory muscles of rat after using bite plate. METHODS: 30 growing rats were selected to wear the bite plate for four weeks. The changes of contractive force and time of masticatory muscles were measured by method and technique of biomechanics. RESULTS: The biomechanical characteristics of masticatory musclse changed significantly. The contractive tension and contraction time of masseter muscles increased shortened respectively, while that of the digastric muscles increased and prolonged. CONCLUSION: This suggests that the orthopedic mechanism of the bite plate is to alter the original masticatory muscle dynamics and finally to reach the goal of functional jaw orthopedics.

[Primary epididymal tumors].

OBJECTIVE: To improve the diagnosis of primary epididymal tumors. METHOD: In 23 primary epididymal tumors (22 benign and 1 malignant), 14 were adenomatoid tumors, 6 were leiomyomas, 1 was multiple fibrous pseudomonas and 1 was sclerosing hemangioma. Epididymal adenocarcinoma was seen in a 57-year-old man. RESULT: Epididymal tumors might be easily misdiagnosed as epididymal tuberculosis, chronic epididymis and spermatocele. Apart from certain benign clinical characteristics, benign epididymal tumors could be diagnosed by B-ultrasonography and aspiration biopsy. Most malignant tumors of epididymis presented as rapidly growing scrotal masses and cured by the removal of tumors or the whole epididymis of the same side. 19 of 22 cases were free from recurrence. CONCLUSION: Malignant epididymal tumors should be treated as malignant tumors of testis and their prognosis is extremely poor.

[Corneal epithelium implantation under corneal flap after excimer laser in situ keratomileusis: a clinical pathological and cytological study and management].

OBJECTIVE: To study the clinical pathological and cytological characteristics of corneal epithelial cells implanted under the cap of laser in situ keratomileusis and the method of management. METHODS: For 4 eyes with epithelial cell implantation, the corneal cap was torn from temporal side to nasal, and the implanted material was removed from the flap and the stromal base. After washing between the interface, the cap was repositioned. RESULT: No epithelial implantation occurred again in these 4 cases after follow-up for 1 similar 12 months. The clinical pathological and cytological studies showed the material removed was degenerated and necrotic epithelial cells mixed with some unknown materials. CONCLUSION: Clinically, the material formed under the cap white and yellow in color should be removed as soon as it happens. The material consists mainly of non-structural material with necrotic and degenerated epithelial cells.

[Immunohistochemical study on ANP in the atria of electrocution].

By the immunohistochemical staining for ANP with LSAB method and the analysis of areas and grays of ANP with computer image analysis, it was found that the ANP granulus in right atria of electrocution were obviously depleted, compared with that in the control groups. The results showed that depletion of ANP granules in right atria could be used as a reference for the forensic diagnosis of electrocution and might benefit for the clinical therapy to the patients suffering from electric injuries.

[The design of a novel multifunction ECG board].

In this paper an ECG amplifying circuit is comprehensively presented. This circuit has all the necessary functions which include isolated ground, programmable ECG lead and filter band, programmed gain, rapid baseline restoring, anti defibrillating electrical shock, anti interference of high frequency electrotome etc.

[Development of the IMP900 ECG & VT patient monitoring system].

A new respiration monitor was developed with this paper, which can monitor tidal volume continuously based on the impedance technique as well ad ECG at the same time. Advanced digital storage display technique and modular hardware design are adopted in this system. One by one calibration method was introduced to solve problem of individual differences in the measurement of tidal volume, which make it viable to monitor Tidal volume with impedance technique. The system possesses characteristic of high ratio of performance to cost. Clinical tests of the system have been made for nonsurgical patients, the results show that the system meets the basic clinical requirements, and its wide clinical applications are expected in the future.

[Application of three-dimension luminescence method to the identification of petroleum pollutants].

As three-dimension luminescence method can provide abundant sample information, it is applied to the identification of petroleum pollutants. In this paper, the whole analytical process of three-dimension luminescence method is described, including sample disposal, analytical conditions, analytical process, especially the automation in acquisition, disposal and differentiation of analytical data.

In vitro, in vivo, and tissue retrieval studies on particulate debris.

The biologic effects of wear debris are an important factor limiting the longevity of total joint replacements. In vivo, in vitro, and tissue retrieval studies have underlined a central role for the macrophage in the etiology of loosening and periprosthetic osteolysis. Wear particles from the materials used for total joint replacement activate macrophages to secrete proinflammatory factors. Complex interactions between macrophages and other cells stimulate bone resorption and suppress bone formation at the prosthetic interface. To improve the long term outcome of joint replacements, future research must find innovative approaches to minimize the production and biologic effects of wear debris.

Effects of novel anti-inflammatory compounds on healing of acetic acid-induced gastric ulcer in rats.

Nonsteroidal anti-inflammatory drugs often cause development of significant GI lesions. Selective inhibitors of prostaglandin G/H synthase/cyclooxygenase-2 (PGHS-2) enzyme and some dual inhibitors of PGHS/5-lipoxygenase (5-LO) enzymes have been reported to be potent anti-inflammatory compounds that carry a much lower risk of having GI irritating effects. We have evaluated the anti-inflammatory effect and the GI safety profile of three new anti-inflammatory compounds: the selective PGHS-2 inhibitors NS-398 and PD 138387 and the PGHS/5-LO dual inhibitor PD 137968. All the compounds tested showed an anti-inflammatory activity in the carragenan footpad edema test in rats. None of these compounds caused either gastric damage 4 h after p.o. administration of 100 mg/kg in rats or inhibition of PGE2 synthesis in the stomach. However, when administered p.o. at an effective anti-inflammatory dose to rats with pre-existing acetic acid-induced gastric ulcer, NS-398 caused a statistically significant delay of ulcer healing. No impairment of the ulcer healing was observed with the other compounds evaluated. Derivatives of 2,6-di-tert-butylphenol, whose members may act as PGHS-1/PGHS-2 inhibitors, selective PGHS-2 inhibitors or PGHS/5-LO dual inhibitors, are novel anti-inflammatory compounds that are devoid of GI irritating effects and do not affect the rate of pre-existing gastric ulcer healing.

In vivo NMR and MRI using injection delivery of laser-polarized xenon.

Because xenon NMR is highly sensitive to the local environment, laser-polarized xenon could be a unique probe of living tissues. Realization of clinical and medical science applications beyond lung airspace imaging requires methods of efficient delivery of laser-polarized xenon to tissues, because of the short spin-lattice relaxation times and relatively low concentrations of xenon attainable in the body. Preliminary results from the application of a polarized xenon injection technique for in vivo 129Xe NMR/MRI are extrapolated along with a simple model of xenon transit to show that the peak local concentration of polarized xenon delivered to tissues by injection may exceed that delivered by respiration by severalfold.

Characterization of a Drosophila proximal-sequence-element-binding protein involved in transcription of small nuclear RNA genes.

In a wide variety of eukaryotic organisms, transcription of small nuclear RNA (snRNA) genes is dependent upon a proximal sequence element (PSE) located upstream of position -40 relative to the transcription start site. There is little or no existent knowledge concerning the PSE-binding proteins of organisms other than human. Here, we report the purification of a fraction enriched in the Drosophila melanogaster PSE-binding protein (DmPBP). DmPBP forms a highly specific complex with the PSE. The protein stimulates transcription from the U1 gene promoter by RNA polymerase II and from the U6 gene promoter by RNA polymerase III in Drosophila nuclear extracts, and activation is dependent upon the presence of a PSE. The molecular mass of native DmPBP as measured by gel-filtration chromatography is 375 kDa. Two polypeptides (apparent molecular masses 59 kDa and 61 kDa) appear to be in close contact with the DNA in that they can be very efficiently and specifically crosslinked to the PSE sequence by ultraviolet irradiation.
