Performance evaluation of leachate treatment system using innovative sulfur circulation method.

A pilot-scale experimental study was carried out to evaluate the performance of an innovative method for early stabilization of landfill. This method employs sequential processes of: leachate collection; nitrification through aeration; sulfate addition in effluent tank; and leachate injection back to the landfill. This study focused on: 1) decomposition of recalcitrant organic matters, 2) the characteristics of denitrification, 3) sulfide oxidation for sulfate recovery, and 4) nitrification by aerating discharged leachate. It was found that, when sulfate (SO4(2-)) added leachate was introduced inside the landfill, sulfate-reducing bacteria decomposed recalcitrant organic matters like lignocellulose by using SO4(2-) as an electron acceptor and simultaneously sulfur-oxidizing bacteria denitrified the leachate using sulfides (H2S, HS-, S2-) as electron donors. These two types of bacteria existed not competitively but symbiotically on substrate utilization. Sulfate-reducing bacteria produced S2- while sulfur-oxidizing bacteria oxidized S2- to SO4(2-). During aeration, the concentration of NO3(-)-N increased from near zero up to 925 mg l(-1). Eventual denitrification efficiency in the simulated waste landfill was observed to be approximately 92.3%. Also, S2- present in the discharged leachate was converted to SO4(2-) again in the aeration process. Sulfate needed in this process could be recirculated. Therefore, the amount of sulfate required in the operation of this method could be maintained marginal after the initial addition of sulfate in the effluent tank. Decomposition of recalcitrant organic matters and denitrification hastened the stabilization of landfill. The results of this study indicated that this innovative method was effective and economic.

Dose response and temporal patterns of radiation-associated solid cancer risks.

Findings of the Life Span Study (LSS) cohort of atomic-bomb survivors are a primary source for quantitative risk estimates that underlie radiation protection. Because of the size and length of follow-up, the LSS provides considerable information on both the nature of the dose response and on how radiation-associated excess risks vary with age, age at exposure, sex, and other factors. Our current analyses extend the mortality follow-up by 7 y (through 1997) and add 8 y (through 1995) to the incidence follow-up. During the follow-up periods there have been a total of about 9,300 solid cancer deaths and almost 12,200 incident cases. As outlined in this presentation, while discussing issues related to the shape of the dose response and low dose risks in some detail, the new reports consider temporal patterns in greater detail than has been done previously. As we have reported, the LSS solid cancer dose response is well described by simple linear dose response over the 0 to 2 Sv range (with some leveling off at higher estimated doses). This remains the case with the extended follow-up. Although LSS is often referred to as a high dose study, about 75% of the 50,000 cohort members with doses in excess of 5 mSv have dose estimates in a range of direct interest for radiation protection (0-200 mSv). Analyses of data limited to this low dose range provide direct evidence of a significant solid cancer dose response with a risk per unit dose that is consistent with that seen for the full dose range. Previous LSS reports have focused on descriptions of the solid cancer excess risks in which the excess relative risk varies with age at exposure and sex. In addition to the age at exposure effects, our current analyses suggest excess relative risks also vary with age (at death or diagnosis). Excess relative risks are higher for those exposed earlier in life, with attained age-specific risks changing by about 20% per decade, but tend to decrease with increasing attained age, roughly in proportion to (1/attained-age)1.5, for any age at exposure. Despite the decreasing relative risk, excess rates have increased rapidly throughout the study period with some indication, especially for the incidence data, that attained-age-specific rates are higher for those exposed at younger ages. Simple comparisons of site-specific excess risks are used to illustrate how the interpretation of age-at-exposure effects on excess relative risks or excess rates is complicated by changes in baseline rates with birth cohort or time period.

Monte Carlo dosimetry of 60Co HDR brachytherapy sources.

Monte Carlo based dosimetry is presented for three 60Co HDR sources with a unique configuration of two active pellets in contact or spaced 9 and 11 mm apart. Results are presented in Cartesian "away and along" as well as polar coordinates following the AAPM TG-43 dosimetric formalism. Iso-dose rate contours around the sources in Cartesian coordinates reveal that significant differences between the three source designs exist only close to the source centers where dose rate distributions bear the effect of the unique source configurations. Dose rate constants of all three sources are accurately described by an equation of the form: lambda(cGyh(-1)U(-1))=lambda*pointG(1 cm,90 degrees) = 1.094*G(1 cm,90 degrees) where lambdapoint is the dose rate constant of a bare 60Co point source and G(1 cm,90 degrees) is the "exact" source geometry factor as defined by the TG-43. Radial dose and anisotropy function data extracted using the point source approximated geometry factors are tabulated for all three source designs. Finally, the dependence and variation of the above TG-43 parameters are discussed and it is demonstrated that the dosimetric properties of high-energy photon emitters are largely dependent on radionuclide source distribution.

Effect of basic fibroblast growth factor on vascularization in esophagus tissue engineering.

We carried out an experimental study to evaluate the effect of basic fibroblast growth factor (bFGF)-containing collagen gel on vascularization in esophageal tissue engineering. We compared an acellular collagen sponge scaffold and an acellular collagen gel scaffold in combination with bFGF using a canine model. The construct was implanted in the cervical esophagus and the regenerated tissue was evaluated one month after surgery. Histological analysis confirmed a significantly large amount of blood vessels in the bFGF-containing collagen gel group as compared to the collagen gel group without bFGF (bFGF (-)). However, in the collagen sponge groups, no difference was observed between the bFGF (+) group and the bFGF (-) group. These results showed that bFGF-containing collagen gel is suitable not only for an acellular scaffold for tissue engineering but also for an effective tropic factor vehicle in vivo.

Regeneration of hypogastric nerve using a polyglycolic acid (PGA)-collagen nerve conduit filled with collagen sponge proved electrophysiologically in a canine model.

The hypogastric nerve (HGN) is a sympathetic nerve in the peritoneal cavity and controls urinary and seminal functions. In this study, the regeneration of HGN was determined by using a new type of an artificial nerve conduit, polyglycolic acid (PGA)-collagen nerve conduit filled with collagen sponge in two dogs. A PGA-collagen nerve conduit (diameter=2 mm) was interposed in a 10 mm gap of the right HGN. The regeneration of the HGN was evaluated electrophysiologically 8 months after the operation. The intraluminal pressure of spermatic duct and the bladder neck were elevated 80 mmHg and 25 mmHg respectively by the stimulation across the regenerated HGN. The prostate contraction was also elicited. The responses diminished after the excision of the regenerated portion of HGN. These results proved the regeneration of HGN and this nerve conduit will be great help for patients who suffer from urinary and seminal disturbances.

The effects of humic substances on the intake of micro-organic pollutants into the aquatic biota.

Humic substances, naturally occurring highly polymerized organic compounds, exist widely in the water and soil environments. It has been known that the humic substances affect the fate of micro-organic pollutants (e.g. intake, accumulation, movement, degradation, toxicity, etc.). Of these, the effect of humic substances on the intake into biota (i.e. living cell) is one of the most important. In this research, the effects of co-existing humic substances on the intake of micro-organic pollutants into aquatic biota were experimentally evaluated. The humic acid filtrate using a 3,000 Da ultra-filtration membrane was used. Two PAHs (i.e. pyrene and phenanthrene) were used as micro-organic pollutants. Liposome for simulating living cell membrane was synthesized in the laboratory, and used for investigating the intake of micro-organic pollutants into aquatic biota precisely. The batch experiment results (PAHs onto humic acid, humic acid into liposome, and PAHs into liposome (K(lipw))) led to the fact that the sorption of PAHs into liposome is suppressed apparently by binding with humic acid in the aqueous phase. This suggests that the accumulation and/or toxicity of micro-organic pollutants is retarded by humic substances in the actual aqueous environment. Moreover, the experimental results indicated that the sorption into liposome (i.e. liposome/water partition coefficient (K(lipw))) could be a better parameter for estimating the intake of micro-organic pollutants into aquatic biota than n-octanol/water partition coefficient (K(ow)) in the aqueous environment.

[Coronary endarterectomy and on-lay patch anastomosis in a case with unstable angina due to diffusely diseased coronary artery].

A 76-year-old woman with unstable angina due to diffusely diseased coronary artery successfully underwent coronary artery bypass grafting (CABG) using endarterectomy and on-lay patch anastomosis. She had triple vessel disease in the coronary artery and all of them were diffusely stenotic. It was impossible to undertake simple CABG. We were able to perform 3 coronary artery bypass grafting using endarterectomy and on-lay patch anastomosis. The postoperative course was uneventful and postoperative angiogram revealed well patent 3 grafts. Coronary endarterectomy and on-lay patch anastomosis were effective procedure in a patient with diffusely diseased coronary artery.

A new acellular vascular prosthesis as a scaffold for host tissue regeneration.

OBJECTIVE: In situ tissue engineering using acellular xenografts is a new approach to autologous tissue regeneration. In the present study, we analyzed a new regeneration scaffold comprised of xenogenic acellular vessels in a dog model. DESIGN OF STUDY AND RESULTS: Xenogenic vascular conduits of porcine carotid arteries were acellularized by a 48-hour detergent incubation to extract all the cell components of the graft. The acellularisation procedure resulted in a complete removal of all cells. After this procedure, heparinization was performed sequentially to induce antithrombogenicity in the graft. The burst pressure of the graft was tested. The bursting pressure of the fresh vessels was 2.83 +/- 0.56 x 10(3) mmHg (377 +/- 75kPa, n = 6). After detergent treatment, the bursting pressure of the grafts was 2.50 +/- 0.48 x 10(3) mmHg (334 +/- 63 kPa, n = 6); after heparinization, it was 3.44 +/- 0.38 x 10(3) mmHg (459 +/- 51 kPa, n = 5). In vivo function was tested in a dog model by transplantation to the abdominal artery. By 18 weeks, endothelial cells were aligned as in normal, healthy artery over the surface of the entire graft. Fibroblasts and macrophages had infiltrated the graft from both inside and outside. The neointima contained normal layers of smooth muscle cells, which were identified by anti alpha-smooth muscle fiber antigen staining. CONCLUSIONS: The acellular heparinized xenograft has sufficient mechanical properties and was successfully replaced with host tissue, and is thus a promising new type of vascular prosthesis.

Sigma-nucleus potential in A=28.

We have studied the (pi(-),K+) reaction on a silicon target to investigate the sigma-nucleus potential. The inclusive spectrum was measured at a beam momentum of 1.2 GeV/c with an energy resolution of 3.3 MeV (FWHM) by employing the superconducting kaon spectrometer system. The spectrum was compared with theoretical calculations within the framework of the distorted-wave impulse approximation, which demonstrates that a strongly repulsive sigma-nucleus potential with a nonzero size of the imaginary part reproduces the observed spectrum.

Isulinotropic properties of Nigella sativa oil in Streptozotocin plus Nicotinamide diabetic hamster.

The present study was designed to investigate the possible insulinotropic properties of Nigella sativa L. (N. sativa) oil in Streptozotocin plus Nicotinamide-induced diabetes mellitus in hamsters. Nicotinamide was injected intraperitoneally 15min before injection of Streptozotocin intravenously. Oral treatment with N. sativa oil began 4 weeks after induction of diabetes. Serum insulin was measured by enzymeimmunoassay. Islets insulin was stained using anti-insulin monoclonal antibody. Significant decrease in blood glucose level together with significant increase in serum insulin level were observed after treatment with N. sativa oil for 4 weeks. Big areas with positive immuno-reactivity for the presence of insulin were observed in the pancreases from N. sativa oil-treated group compared to non-treated one using immunohistochemical staining. Therefore, our data show that the hypoglycemic effect of N. sativa oil in Streptozotocin plus Nicotinamide diabetic hamsters resulted, at least partly, from a stimulatory effect on beta cell function with consequent increase in serum insulin level. These results indicate that N. sativa oil has insulinotropic properties in type 2-like model.

The Bacillus subtilis rec-assay: a powerful tool for the detection of genotoxic substances in the water environment. Prospect for assessing potential impact of pollutants from stabilized wastes.

The Bacillus subtilis rec-assay has been specially developed to detect genotoxicity in environmental water samples. The rationale of the B. subtilis rec-assay is based on the relative difference of survival of a DNA repair-recombination proficient strain and its deficient strain, which is interpreted as genotoxicity. This assay method can be very powerful in that it has higher sensitivity for the detection of mutagens in highly polluted waters than other bacterial mutation assays. Hydrophobic fractions from various environmental waters were fractionated by using XAD-2 resins and assayed, targeting the detection of organic genotoxic substances. Genotoxic response was detected in most of them, which revealed that many unknown micropollutants with genotoxicity occur in public water bodies. Positive response was also detected from a treated municipal solid waste (MSW) landfill leachate. Genotoxicity remaining in the treated effluent suggests that genotoxic micropollutants may pass through conventional water treatment processes such as activated sludge treatment process. Without proper control of waste quality and landfill facilities, waste landfill could be a heavy pollution source.

Thymoquinone suppresses expression of inducible nitric oxide synthase in rat macrophages.

The objective of the present study was to determine the immunomodulatory role of thymoquinone (TQ) regarding its effect on the production of nitric oxide (NO) by rat peritoneal macrophages. Under certain conditions, macrophagesand certain other cells can produce high concentrations of NO from its precursor L-arginine via inducible nitricoxide synthase (iNOS)pathway. TQ has been established as the major component of the oil extracted from Nigella saliva plant seeds, which is being used frequently in herbal medicine. TQ (IC50 1.4-2.76 microM) dose- and time-dependently reduced nitrite production, a parameter for NO synthesis, in supematants of lipopolysaccharide (LPS)-stimulated (5 microg/ml) macrophages without affecting the cell viability. The protein level of iNOS in peritoneal macrophages was also decreased by TQ in a concentration-dependent manner. In addition, TQ inhibited the increase in iNOS mRNA expression induced by LPS indicated by reverse transcription-polymerase chain reaction (RT-PCR). These inhibitory effects of TQ were confirmed by immunofluorescence staining of iNOS in macrophages which showed decreased immunoreactivity for iNOS after treatment with TQ if compared with the control LPS-stimulated cells. These results suggest that TQ suppresses the production of NO by macrophages; an effect which may be useful in ameliorating the inflammatory and autoimmune conditions.

Characterization of 'adult-type' mast cells derived from human bone marrow CD34(+) cells cultured in the presence of stem cell factor and interleukin-6. Interleukin-4 is not required for constitutive expression of CD54, Fc epsilon RI alpha and chymase, and CD13 expression is reduced during differentiation.

BACKGROUND: In vitro-derived human mast cells exhibit different properties, depending in part on the source of progenitor cells. Most investigations have used fetal liver, cord blood or peripheral blood. Few have used adult bone marrow. OBJECTIVE: Human mast cells derived in vitro from the CD34(+) progenitors in bone marrow and cord blood that had been cultured with recombinant human stem cell factor (rhSCF) and recombinant human interleukin-6 (rhIL-6) were compared. METHODS AND RESULTS: After 12 weeks of culture, nearly all of the cells were mast cells, and nearly all of these had cytoplasmic granules containing both tryptase and chymase (MCTC type), stained metachromatically with acidic toluidine blue, and expressed CD117 on the cell surface. Both tryptase protein and mRNA were detected by two weeks of culture. Chymase mRNA and protein were detected at 4 weeks but not at 2 weeks of culture. By 12 weeks, chymase content per cell, measured by ELISA, was significantly higher (P < 0.05) in human bone marrow-derived mast cells (HBMMC) (5.6 +/- 0.9 pg) than in cord blood-derived mast cells (CBMC) (2.4 +/- 0.9 pg), whereas histamine and tryptase levels were not significantly different. Of the cluster designations tested, CD29, CD49d, CD51 and CD61 were strongly expressed on HBMMC. CD54 and Fc epsilon RI alpha also were expressed constitutively. Approximately half of CD34-sorted cells at day 0 were CD13(+) and this diminished as mast cell maturation occurred. Electron microscopy revealed that 12-week-old HBMMC had many secretory granules that contained spherical electron dense cores surrounded by electron lucent space, consistent with previous reports of immature MCTC cells developing in vivo. CONCLUSIONS: CD34(+) progenitors of human bone marrow are a rich source of mast cell progenitors capable of expressing granule and surface markers of mature mast cells in the presence of rhSCF and rhIL-6.

The relationship between consumption of antimicrobial agents and the prevalence of primary Helicobacter pylori resistance.

BACKGROUND: Primary and acquired resistance to the antimicrobial agents is a primary reason for the failure of Helicobacter pylori eradication therapies. We assessed the primary antibiotic resistance rates of H. pylori to three different antibiotics and its relationship due to the annual antibiotic consumption in Japan during the period prior to approval of anti-H. pylori therapy in Japan. MATERIALS AND METHODS: Antibiotic susceptibility was tested using the agar dilution method for clarithromycin, amoxicillin and metronidazole. Isolates were considered resistant when the MIC value was > 8 mg/l for metronidazole, > 1 mg/l for clarithromycin and < 0.5 mg/l for amoxicillin. RESULTS: Helicobacter pylori isolates were obtained from 593 Japanese patients from 1995 to 2000. Primary resistance of H. pylori to clarithromycin, metronidazole and amoxicillin was found in 11%, 9% and 0.3% strains, respectively. The proportion with clarithromycin resistance significantly increased from 7% in 1997-98 to 15.2% in 1999-2000 (p =.003). During the same period the metronidazole resistance rate also increased from 6.6% in 1997-98 to 12% in 1999-2000 (p =.02). The prevalence of clarithromycin and metronidazole was related to the annual consumption of these antimicrobial agents. CONCLUSION: Resistance rates for both clarithromycin and metronidazole appear to reflect the annual consumption of these agents. The high rate of clarithromycin resistance in Japan suggests that the effectiveness of clarithromycin-based therapies may be compromised in the near future.

Non-invasive quantitative monitoring of cerebral blood flow in aneurysmal subarachnoid haemorrhage with 99mTc-ECD.

The purpose of this prospective study was to detect symptomatic cerebral vasospasm in aneurysmal subarachnoid haemorrhage (SAH) by a non-invasive mean cerebral blood flow (mCBF) quantification using 99mTc-ethyl cysteinate dimer. Measurement of mCBF without blood sampling and single photon emission tomography (SPECT) were performed at 1 and 7 days after surgery in 35 consecutive SAH patients, of whom 16 were examined at day 30 as well. A decrease in mCBF of more than 10% on day 7 versus day 1 was considered to indicate vasospasm. On visual interpretation of SPECT, a perfusion decrease which appeared newly on day 7 was considered to indicate vasospasm. In total, nine of 35 patients had cerebral vasospasm confirmed by computed tomography (CT) and/or angiography. The mCBF measurement showed a 77.8% (7/9) sensitivity, a 88.5% (23/26) specificity, a 70.0% (7/10) positive predictive value, and a 92.0% (23/25) negative predictive value. SPECT yielded a 33.3% (3/9) sensitivity, a 73.1% (19/26) specificity, a 30.0% (3/10) positive predictive value, and a 76.0% (19/25) negative predictive value. On SPECT, decreased perfusion was observed in most of the patients at clipping sites, which might represent post-operative transient abnormal perfusion and should not be read as vasospasm. In conclusion, this mCBF measurement is more accurate than visual interpretation of SPECT for detecting vasospasm.

Oral beraprost sodium improves exercise capacity and ventilatory efficiency in patients with primary or thromboembolic pulmonary hypertension.

OBJECTIVE: To investigate the effect of beraprost sodium, an orally active prostacyclin analogue, on exercise capacity and ventilatory efficiency in patients with primary pulmonary hypertension and chronic thromboembolic pulmonary hypertension. PATIENTS AND DESIGN: Symptom limited cardiopulmonary exercise testing was performed before and 3 (1) months (mean (SEM)) after beraprost treatment in 30 patients with precapillary pulmonary hypertension (14 with primary pulmonary hypertension and 16 with chronic thromboembolic pulmonary hypertension). RESULTS: Long term treatment with beraprost resulted in significant increases (mean (SEM)) in peak workload (87 (4) W to 97 (5) W, p < 0.001) and peak oxygen consumption (peak VO2, 14.9 (0.7) ml/kg/min to 16.8 (0.7) ml/kg/min, p < 0.001). Beraprost decreased the ventilatory response to carbon dioxide production during exercise (VE-VCO2 slope, 42 (2) to 37 (1), p < 0.001). No significant difference in the responses of these variables to beraprost treatment was observed between patients with primary pulmonary hypertension and chronic thromboembolic pulmonary hypertension. CONCLUSIONS: Oral administration of beraprost sodium may improve exercise capacity and ventilatory efficiency in patients with both primary and chronic thromboembolic pulmonary hypertension.

Evaluation of peripheral nerve regeneration across an 80-mm gap using a polyglycolic acid (PGA)--collagen nerve conduit filled with laminin-soaked collagen sponge in dogs.

We evaluated peripheral nerve regeneration using a novel artificial nerve conduit. The conduit was made of a polyglycolic acid(PGA) - collagen tube filled with laminin- soaked collagen sponge. We implanted this nerve conduit across an 80mm gap in the peroneal nerve of dogs. Histological observation 12 months after implantation showed numerous unmyelinated and myelinated nerve fibershad regenerated beyond the gap. Neurofilaments were widely observed immunohistochemically in the regenerated nerve segments. These findings indicated that newly regenerated axons had extended across the gap and connected into the distal nerve segments. Compound muscle action potentials(CMAPs) and somatosensory evoked potentials (SEPs) were recorded in all dogs. At 12 months, the CMAPs indicated complete recovery, while the SEPs showed incomplete but substantial recovery. Walking patterns had returned to near-normal 12 months after implantation. Use of this nerve conduit can lead to peripheral nerve elongation and favorable functional recovery across a wider nerve gap.
