Polymorphic variation of the ARP gene on 3p21 in Japanese esophageal cancer patients.

Allelic loss of the short arm of chromosome 3 is common in esophageal squamous cell carcinoma (ESC) and its premalignant lesions. The ARP gene (Arginine Rich Protein) was mapped to 3p21, and frequent variations of the triplet, AGG, repeat around codon 50 of the ARP gene were reported in a variety of human cancers. To examine the involvement of the ARP gene in esophageal cancer, we screened mutations around codon 50 in 35 ESC tumours and matched normal tissues. Sequence variants were observed in four ESC tumours; two (AGG)2 insertions, one ATG50-to-AGG substitution and one AGG deletion. However, they were also found in its corresponding normal tissues, suggesting that variation of the ARP gene found in ESC is polymorphic. We next analyzed sequence changes in 48 unrelated Japanese healthy individuals. They consist of 33 wild-type homozygotes, nine (AGG)2 insertion/wild-type heterozygotes, two (AGG)2 insertion homozygotes, one AGG deletion/wild-type heterozygote and three ATG50-to-AGG substitution/wild-type heterozygotes. Allele frequencies for wild-type, (AGG)2 insertion, ATG50-to-AGG substitution and AGG deletion are 0.82, 0.14, 0.03 and 0.01, respectively. Observed genotype frequencies fit well with the Hardy-Weinberg's law. Significant difference was not observed between allele distributions in normal and cancer patient populations.

Carbohydrate antigens as a risk factor for hematogenous recurrence of esophageal squamous cell carcinoma patients.

The patients with hematogenous recurrence have a poor prognosis in esophageal squamous cell carcinoma (SCC). Recent developments have suggested the important role of sialyl Lewis A and X in the step of hematogenous metastasis. To evaluate the role of two carbohydrate antigens on hematogenous recurrence in esophageal SCC, we examined clinicopathological features and the expression of sialyl Lewis A and X in 125 patients retrospectively. Thirty-three out of 125 patients had hematogenous recurrence after curative esophagectomy. sLeA and sLex expression correlated with hematogenous recurrence (p=0.026, p=0.043 respectively), and sLeA expression was correlated with pM (lymph). Cox proportional hazards model and logistic regression analysis revealed that pN and pM lymph were significant prognostic factors and predictive factors of hematogenous recurrence in esophageal SCC. Although, neither sLeA nor sLeX were significant factors affecting survival in esophageal SCC, logistic regression analysis demonstrated that increased tumoral expression of sLeA and sLeX were risk factors of hematogenous recurrence in esophageal SCC. In conclusion, patients with lymph node metastasis, in particular those with distant lymph node metastasis, the expression of sLeA and sLeX may suggest a higher incidence of hematogenous recurrence in esophageal SCC. New strategies for specific inhibition of cancer cell attachment to vascular endothelial cells should be considered for the patients with the increased tumoral expression of either sLeA or sLeX.

The biological and clinicopathological characteristics of right-sided colon cancer.

The biological characteristics of right-sided colon cancer were examined in 74 colon cancers with regard to clinicopathological parameters, proliferative activity and p53 expression. Significant differences between right-sided and left-sided colon cancer were found in age, tumor size, histological type and lymphatic vessel invasion. The proliferating cell nuclear antigen labeling index did not significantly differ between the two cancers. The frequency of p53 overexpression was lower in the former than in the latter. The results suggested that right-sided colon cancer did not differ from left-sided colon cancer in proliferative activity except for its tendency to be locally advanced and to differ in carcinogenesis.

Progression of esophageal carcinoma by loss of EGF-STAT1 pathway.

PURPOSE: In only a very limited number of cultured cell lines, epidermal growth factor (EGF), a potent mitogen for many kinds of cells, was shown to activate STAT1 (signal transducer and activator of transcription 1) protein, which can transmit signals that cause cell growth arrest and apoptosis. The purpose of this work is to elucidate the physiologic and/or pathological significance of this EGF-STAT1 pathway. MATERIALS AND METHODS: A series of cultured cell lines that had been established from surgical specimens of esophageal squamous cell carcinoma was studied for the existence of the EGF-STAT1 pathway. Normal esophageal squamous epithelial cells either explanted from non-neoplastic portions of surgically removed human esophageal tissue or in bovine esophageal epithelium in situ were examined as well. RESULTS: EGF treatment leads to a strong growth arrest in three of the 30 esophageal squamous cell carcinoma cell lines. STAT1 was found to be activated by EGF in the three cell lines but not in the others. EGF can also activate STAT1 in cultured normal esophageal squamous epithelial cells. STAT1 is at the activated state in the basal cell layer of the bovine esophageal epithelium. Notably, patients who had harbored the cancer cells with the EGF-STAT1 pathway had a dramatically better prognosis. DISCUSSION: The EGF-STAT1 pathway may be intrinsic to esophageal epithelial lineage of cells and is lost in a considerable fraction of the carcinomas. This loss appears to cause a significantly more malignant clinical course. These findings may point out a critical step in the progression of esophageal cancer and could lead to the development of useful clinical applications.

Clinical and immunomodulatory effects of fun-boi, an herbal medicine, on collagen-induced arthritis in vivo.

OBJECTIVE: Crude preparations of Fun-boi (Stephania tetrandra), a traditional antirheumatic herb, have been reported to have immunomodulatory effects on both cell-mediated and humoral immunity in vitro, but little is known about the mode of action in vivo. The objective of this study was therefore to evaluate the efficacy of Fun-boi against arthritis and its effect on the immune system. METHODS: Mice were divided into the following 3 groups of 7 mice each: 1) a normal group, not treated to cause collagen-induced arthritis (CIA), received water orally; 2) a control group with CIA received water orally; and 3) the Fun-boi group with CIA, received Fun-boi (3 mg/g body weight/day) orally. We analyzed the arthritis score, the serum anti-type II collagen (CII) antibody level, and the percentage of the following lymphocyte subsets from lymphoid organs: B220, CD3/CD4, CD3/CD8 and CD40L/CD4 lymphocytes from blood or lymph nodes; and CD4-CD8-, CD4+CD8+, CD4+CD8- and CD4-CD8+ from the thymus. RESULTS: Fun-boi therapy markedly reduced the severity of arthritis (p < 0.001) and tended to reduce the serum anti-CII antibody level (p = 0.06). Whereas CII immunization of DBA/1J mice caused a significant redistribution of CD3/CD8 lymphocytes from blood or lymph nodes, Fun-boi therapy caused significant normalization of the same types of lymphocyte subsets from lymph nodes, but did not affect the CD4 or CD4/CD40L lymphocyte subsets. CONCLUSION: These results demonstrate that Fun-boi therapy exerts therapeutic effects in CIA mice, possibly by causing immunomodulatory effects at specific sites.

Interfacial morphology of an adhesive composite resin and etched caries-affected dentin.

The interfacial structure between an adhesive composite resin and dentin after caries removal and acid conditioning was investigated. The hybrid layer between the adhesive resin and surface-demineralized dentin was found to be wider in the area of empty tubules compared with that of the occluded tubules, and was found to be the thinnest at the lateral walls of the cavity where the dentinal tubules run parallel to the cavity surface. Thus, the extent of demineralization by acid conditioning as well as impregnation of resin into the intertubular dentin forming the hybrid layer is probably related to the degree of closure and direction of the dentinal tubules.

Activation of the systemic production of tumor necrosis factor after exposure to acute stress.

Tumor necrosis factor (TNF), which was originally identified as a tumoricidal factor, is now regarded as one of the main regulators of inflammation and various immune systems. Thus it has been considered to be mobilized in case of emergency. However, we assume that TNF and the cytokine network driven by the monokine also function under normal condition for homeostasis of the animal body which is exposed to various kinds of physiological stress. To test this possibility, we exposed C3H/He mice for up to 3 days to five types of acute stress: food deprivation, drinking water deprivation, sleep deprivation, swimming, and physical restraint. After release from the stress, the level of priming for systemic production of TNF was examined using OK-432 (a streptococcal preparation) as a trigger. Priming of TNF production was not observed immediately after 2-day exposure to most of the stressors. Sleep deprivation alone tended to induce a primed state especially when the stress period was lengthened to 3 days. On the other hand, by keeping mice in a normal condition for a 2-day restorative interval after 2-day exposure to the stressors, systemic production of TNF was consistently primed for all the stress examined. The time course of the priming effect was examined in detail for water-deprivation stress. The effect was detected as early as 3 hours after release from stress, was sustained for 2 days, and returned to the basal level by 4 days after the release. Based on these results, we discussed the role of the TNF-driven cytokine circuit in adaptation to stress.

Shear bond stability of current adhesive systems to enamel.

This study evaluated the micro-shear bond strength of two commercially available resin bonding systems (Single Bond, 3M, USA and SE Bond, Kuraray, Japan) to human enamel. One hundred and twenty enamel sections were prepared from extracted non-carious human molars by cutting with a slowly rotating blade. The enamel surfaces were polished with #280-grift SiC paper under running water. These surfaces were randomly divided into two groups and treated with either Single Bond or Clearfil SE Bond according to manufacturers' instructions. After the bonding procedures, a micro tygon tubing with an internal diameter approximately 0.7 mm and 0.5 mm in height was placed on the enamel surface and Clearfil AP-X resin filled the tube. After the resin was photo-irradiated, the tygon tube was removed. The specimens were further divided into 12 groups according to storage time. All specimens were stored in isotonic sodium chloride solution at 37 degrees C. The bond strength was then measured by means of microshear bond testing at one day, one week, one month, three months, six months and one year. After testing, the fractured surfaces and interfaces were observed using scanning electron microscopy (SEM). The data was analyzed by two-way ANOVA and Fisher's PLSD tests at 95% level of confidence. The result was that both factors (storage time and material) affected bond strength and there was a statistically significant interaction between them. In general, bond strength decreased with time for both materials. The highest bond strengths were achieved at one day with SE Bond and Single Bond. For Single Bond, from one month to one year, the bond strength dramatically decreased. For SE Bond, the decrease in bond strength was gradual. After one-year storage, the bond strength drastically decreased and this value was not significantly different from the results of the one-year storage of Single Bond.

Effective bond strength of current adhesive systems on deciduous and permanent dentin.

PURPOSE: To evaluate the bond strength of a total-etching, self-priming system (Single Bond) and a self-etching system (Clearfil SE Bond) to deciduous and permanent human dentin. METHODS AND MATERIALS: Buccal dentin discs were prepared with a diamond disc from permanent first premolars, permanent third molars and deciduous second molars. The flat dentin surfaces were obtained by polishing with wet 600 grit silicon carbide papers. The specimens of each group were further divided into two groups for bonding to either Single Bond or Clearfil SE Bond. After 24 hours, the microshear bond strength testing was executed on a universal testing machine. Statistical analysis was performed at alpha = 0.05. RESULTS: No significant differences in bond strength were found between materials. However, deciduous dentin demonstrated significantly lower bond strengths than permanent premolar dentin when Single Bond was applied (p < 0.05). CONCLUSION: The difference in bonding substrate (permanent or deciduous dentin) had a significant effect on bond strength when the total-etching, self-priming system was applied.

Low milk fat syndrome and magnesium oxide supplementation.

The low milk fat syndrome most commonly occurs in dairy cows fed diets with a high ratio of carbohydrate to roughage. It is characterized by markedly depressed yields of milk fat. Rumen buffers, such as sodium bicarbonate (NaHCO3) and magnesium oxide (MgO), are known to correct the milk fat depression observed on restricted roughage rations, although by apparently different mechanisms. The mode of action of NaHCO3 can be explained by control of the proportion of ruminal propionate via control of rumen pH. Cows fed diets supplement with NaHCO3 showed increased feed intake and improved digestibility of dry matter or fibre. The mechanism by which Mg increases milk fat concentration may operate at both rumen and tissue level. Most studies with MgO have suggested that it acts as a buffer or neutralizer in either the rumen or the intestine. During digestion, the addition of MgO increases digestibility of dry matter, as does NaHCO3. Among many different tissue effects, MgO supplementation increases triglyceride uptake by the mammary gland. Feeding MgO tends to increase lipoprotein lipase activity in the mammary gland and to decrease the linoleic acid content of milk fat. The percentage uptake of triglycerides from lipoproteins and milk fat by the mammary gland was greater in cows fed MgO-supplemented rations than in cows fed restricted roughage rations. Triglyceride-rich lipoprotein concentrations are uniquely low in cow plasma compared to other species, but still account for a major portion of the arteriovenous difference in plasma lipids across the mammary gland.(ABSTRACT TRUNCATED AT 250 WORDS)

Pulpal response to a new light-cured composite placed in etched glass-ionomer lined cavities.

This study evaluated the pulp biocompatability of a new light-cured composite resin which was placed in etched glass-ionomer-lined cavities of monkey teeth. The pulpal response to this material was less than that to zinc-oxide eugenol cement in each observation period. Therefore this material seems to meet acceptable biocompatability standards in nonhuman primates.

Factors mediating a regional difference in vasorelaxing effects of human natriuretic polypeptide in dogs.

Factors mediating a regional difference in vasorelaxing effects of atrial natriuretic polypeptide (ANP) were investigated, using the canine pulmonary and renal arteries. Isolated preparation was suspended in an organ bath, and the isometric tension was recorded. ANP relaxed both arteries dose-dependently under precontraction with methoxamine (3 x 10(-5) M). The maximum relaxation was extremely different between the two arteries: 78.5% and 8.6% of maximum relaxations caused by papaverine in the pulmonary and renal arteries, respectively. Removal of the endothelium affected neither the relaxation of pulmonary artery nor that of renal artery. Methylthionine (10(-5) M) greatly diminished the maximum relaxation in the pulmonary artery, but not in the renal artery, indicating that some free radical species may mediate the relaxation of pulmonary arteries to ANP. Methoxamine-induced contractions were inhibited by isosorbide dinitrate (10 and 30 nM) in both pulmonary and renal arteries, in a noncompetitive manner. Nifedipine (10 and 30 nM) inhibited the contractions competitively in the renal artery, but not in the pulmonary artery. Even in the Ca++-free solution, ANP diminished the methoxamine-induced contractions of the pulmonary arterial strips more effectively than those of the renal arterial strips. From these results, we concluded that the regional difference in the ANP-induced relaxation between the pulmonary and renal arteries was derived, partly, from a difference in the mechanism of intracellular Ca++ mobilization.

Direct effects of IL-4 on the in vitro differentiation and proliferation of hematopoietic progenitor cells.

The purpose of this study was to analyze the effects of recombinant human interleukin 4 (IL-4) on the differentiation and proliferation in vitro of human granulocyte/macrophage (GM) and erythroid progenitors. IL-4 was added to either fetal bovine serum (FBS)-supplemented or to FBS-deprived cultures of unfractionated human marrow cells or marrow cells depleted of adherent and/or T cells. Paradoxical effects similar to those reported in the murine system were detected in these experiments. In FBS-supplemented cultures, IL-4, which had no effect on the growth or erythroid bursts (from burst-forming cells; BFU-E) detected in the presence of Epo alone, decreased by 46% the number of erythroid bursts detected in the presence of Epo and phytohemagglutinin-stimulated leukocyte-conditioned medium (PHA-LCM). In contrast, in FBS-deprived cultures, IL-4 increased by 30-700% the number of erythroid bursts in cultures containing Epo alone or containing Epo, IL-3, and GM-CSF. The stimulatory effect of IL-4 on erythroid burst growth under FBS-deprived conditions was particularly evident when adherent cells were removed. Under the conditions investigated, IL-4 had little effect on the growth of GM colonies. In FBS-deprived suspension cultures of nonadherent, T-cell-depleted marrow cells, IL-4 maintained both the number of BFU-E and CFU-GM for at least 8 days. In these cultures, IL-4 antagonized the capacity of IL-3 to increase the number of BFU-E but IL-4 and IL-3 acted together to maintain the number of CFU-GM. To determine if IL-4 acted directly or indirectly, its effects on the growth of factor-dependent subclones of the murine progenitor cell line 32D were analyzed. Three subclones were studied: the original IL-3-dependent clone 32D cl.3, the Epo-dependent erythroid clone 32D Epo-1, and the G-CSF-dependent myeloid clone 32D G-1. IL-4 alone failed to induce colony growth from these cell lines. However, IL-4 inhibited by 25% the number of colonies formed by 32D cl.3 in the presence of IL-3 while increasing by 25% and 25-50% the number of colonies formed by 32D Epo-1 and 32D G-1 in the presence of Epo or G-CSF, respectively. These results indicate that human IL-4, as its murine counterpart, is a multilineage growth factor with paradoxical effects which are mediated by the direct action of IL-4 on progenitor cells.

Monkey pulpal response to adhesively luted indirect resin composite inlays.

Monkey pulpal responses to resin-bonded indirect resin composite inlays were histopathologically evaluated by placing them in either etched-enamel, total-etched, or adhesive-resin-lined cavities. Initial pulpal responses caused by re-exposure of the cut dentin surfaces and luting procedure subsided if a tight marginal seal was secured by final cementation of the inlay. The adhesive resin coating of freshly cut dentinal walls/floors seems to provide a new technique to protect the dentin and pulp in indirect restorations requiring temporary sealing.

Adhesive interface between resin and etched dentin of cervical erosion/abrasion lesions.

The interfacial structure between an adhesive composite resin and the dentinal walls of cervical erosion/abrasion lesions etched with 37% phosphoric acid gel for 60 seconds was investigated. Almost all dentinal tubules were occluded with rod-like structural depositions that remained undissolved even after acid conditioning. The hybrid layer between the adhesive resin and surface-demineralized dentin was found to be 0.3 to 3 microns, much thinner than that routinely found in either normal dentin or cariously affected dentin. The hybrid layer was thinnest at the occlusal walls of the cavity where the dentinal tubules run parallel to the cavity surface. The bond strength of adhesive resin to these areas may differ from that to intact normal dentin.

In vivo adhesive interface between resin and dentin.

V-shaped cervical cavities prepared in monkey teeth were restored with several dentin bonding systems, and the in vivo resin-dentin interfacial structures were observed under the scanning electron microscope using an argon-ion etching technique. The hybrid layer could be clearly observed; its depth was dependent on the conditioner/primer used and tended to be thinner at the deep part of the cavity. Resin tags were also clearly observed, and their structure at the inner part was noted to be rougher than that closer to the tubule orifices. When 37% phosphoric acid gel was used, the tags in the deeper parts of the cavity were much rougher. Hemispherical and spherical roughened structures were observed directly above the tubule orifices, which were thought to be a mixture of dentinal fluid and bonding resin that had flowed out from the tubules. The acidic primer containing maleic acid and HEMA could not remove the smear plugs, and undissolved smear particles were observed in the tubules.
