[Comparative study of expression levels of the major human heat shock proteins in cancer and normal tissues].

OBJECTIVE: To investigate and compare mRNA expression levels of the major human HSPs (including HSP90 alpha, HSP90 beta, HSP70, HSC70 and HSP27) in 25 cases of laryngeal carcinomas and normal controls. METHODS: mRNA slot blot hybridization technique was used and cDNA probes of HSP27, HSC70 and HSP90 beta were prepared with PCR method. RESULTS: HSP90 alpha and HSP70 expression was significantly enhanced approximately 5 fold in laryngeal cancer tissues compared to that in the normal larynx tissues while HSP27, HSC70 and HSP90 beta expression was at similar levels. CONCLUSION: The results suggest that HSP90 alpha and HSP70 may play more important role in the carcinogenic process of laryngeal cancer. To find out mechanisms involved will pave the way for the application of HSPs on the biologic therapy of laryngeal cancer.

[Total body irradiation etoposide followed by autologous hematopoietic stem cell transplantion for non-Hodgkin's lymphoma].

OBJECTIVE: To evaluate the therapeutic effectiveness and safety of total body irradiation(TBI) and Etoposide(Vp-16) as a preparative regimen for autologous hematopoietic stem cell transplantation(AHSCT) with non-Hodgkin's lymphoma(NHL). METHODS: Twenty-four patients with intermediate and high grade NHL underwent AHSCT. They achieved complete remission (CR) or partial remission (PR) after induction chemoradiotherapy. Twenty-three patients had first CR or PR, and one third CR. Ten patients underwent autologous bone marrow transplantation (ABMT) and 14 autologous peripheral blood stem cell transplantation (APBSCT). The preparative regimen was TBI 800(700-850) cGy/Vp-16 757(323-1140) mg/m2. RESULTS: At a median follow-up of 22.5(2-92) months, the one-year disease free survival(DFS) was 86.7% (13/15), 3-year, 5-year and 7-year DFS were 80.0% (12/15) in patients with CR before AHSCT. The DFS was 66.7% (4/6) in patients with PR before AHSCT. The patients who had relapsed before AHSCT(3 cases) did not reach DFS. The hematopoietic function recovery was rapid in APBSCT than ABMT. CONCLUSION: The clinical results of AHSCT for intermediate and high grade NHL who achieved CR or PR after induction therapy are satisfactory. The TBI/Etoposide is an effective and safe preparative regimen for AHSCT in NHL patients.

Cathepsin B on invasion and metastasis of gastric carcinoma.

OBJECTIVE: To investigate the expression of cathepsin B (CB) in human gastric carcinoma tissue. METHODS: The expression of CB in human gastric tissue was studied by using monospecific polyclonal rabbit antibody raised against human liver CB for immunohistochemistry, and full length cDNA of CB for in situ hybridization and dot blot. RESULTS: CB overexpression in gastric carcinoma was found when compared with non-neoplastic gastric tissue at both mRNA and protein levels. Diffuse cytoplasmic CB staining of mRNA and protein were identified in malignant cells of 53.3% and 69.1% of gastric adenocarcinoma respectively. The increased staining of CB in malignant cells was associated with the depth of the invasiveness and growth pattern as well as metastasis of lymph nodes, but not with the histological classification. It was also found that there were the expression of CB in stromal cells of the tumor and the expression localized mainly in the endothelial cells of the microvessels which correlated with angiogenesis. CONCLUSION: These results indicate that the expression of CB in gastric carcinoma is related to tumor progression, and leads to development of the invasive phenotype.

Detection of gsp oncogene in growth hormone-secreting pituitary adenomas and the study of clinical characteristics of acromegalic patients with gsp-positive pituitary tumors.

OBJECTIVE: To investigate the incidence and clinical characteristics of gsp oncogene positive growth hormone-secreting adenomas of Chinese acromegalic patients. METHODS: Continuously 40 patients were studied. Serum hormone levels of pituitary and target glands were measured and growth hormone (GH)-TRH stimulating tests were done before transsphenoidal or transfrontal hypophysectomy. Deoxyribonucleic acid (DNA) was extracted from the frozen tumor tissue, and the DNA fragment encompassing codon 201 and 227 of the Gs alpha gene was amplified by polymerase chain reaction (PCR). Point mutations at codon 201 and 227 were detected using PCR direct sequencing method in order to get the incidence of gsp oncogene in GH secreting adenomas. RESULTS: Of 40 tumors studied, 22 (55%) were gsp positive. The point mutation from CGT (Arg) to TGT (Cys) at codon 201 was detected in 21 pituitary tumors, but the point mutation from CAG (Gln) to CTG (Leu) at codon 227 of the Gs alpha gene was found in only 1 tumor. All of the point mutations are heterozygous. The number of gsp positive patients which have 30% or more decrease of serum GH concentration after glucose inhibition is less than that of gsp negative patients (P = 0.042). Compared to gsp negative patients, most of gsp positive patients showed paradoxical response to TRH stimulation (P = 0.002). There were more gsp positive patients with the tumor diameter less than 25 mm (P = 0.029) and with normal GH levels in OGTT after surgery (P = 0.007). CONCLUSIONS: Gsp mutation is one of the major intrinsic defects in the pathogenesis of growth hormone-secreting pituitary tumors and the identification of gsp mutation can be a reference for classification and prognosis of GH tumors.

[Mobilization of autologous peripheral blood stem cells by cyclophosphamide and recombinant human granulocyte colony-stimulating factor(rhG-CSF)].

OBJECTIVE: To observe the effect of cyclophosphamide (CTX) and recombinant human granulocyte colony-stimulating factor(rhG-CSF, Filgrastim) on autologous peripheral blood stem cells (APBSC) mobilization. METHODS: CTX (3.7 +/- 0.2) g/m2 was intravenously injected the first day. rhG-CSF (4.5 +/- 0.6) micrograms.kg-1.d-1 was injected subcutaneously from the day of white blood cell (WBC) nadir to the day before the end of APBSC harvest. APBSC harvest was started when WBC > 2.5 x 10(9)/L and finished when accumulated mononuclear cells (MNC) of APBSC > 5 x 10(8)/kg. CFU-GM, BFU-E culture and CD34+ cells detection of the APBSC was performed. RESULTS: Twenty cases underwent the APBSC mobilization. The nadir of WBC was (1.1 +/- 0.5) x 10(9)/L at day (9 +/- 1). rhG-CSF was injected from day (10 +/- 1) and continued for (6 +/- 1) days. APBSC harvest began on day (13 +/- 1) and continued for (4 +/- 1) days. Accumulated MNC harvest was (8.4 +/- 1.9) x 10(8)/kg, CFU-GM (18.7 +/- 10.3) x 10(4)/kg, BFU-E (18.5 +/- 8.7) x 10(4)/kg, and CD34+ cells (20.9 +/- 5.7) x 10(6)/kg. No severe toxicity was observed. Hematopoietic reconstitution was very well in 18 patients received the APBSC transplantation. CONCLUSION: CTX combined with rhG-CSF was a safe and highly effective method for APBSC mobilization.

[Clinical study on acute respiratory infection caused by chlamydia pneumoniae].

OBJECTIVE: To find out the infected rate and the clinical features of patients with acute respiratory infection caused by chlamydia pneumonia(C. pneumoniae). METHOD: A prospective study for chlamydia pneumoniae infection was conducted in 110 patients with respiratory tract infection from January to December 1995 in Nanjing. Sputum and throat swab specimens were taken and C. pneumoniae DNA were detected by polymerase chain reaction (PCR) with the HM-1-HR-1 primer pair. At the same time, serum samples were taken and immunoglobulins G and M (IgG and IgM) fractions of antibodies to C. pneumoniae were studied by microimmunofluorescence test. RESULT: Prevalence of specific IgG was 70% in patients with respiratory tract infection. Seventeen patients (16%) were serologically diagnosed as having recent C. pneumoniae infection and 12 patients (11%) had the positive PCR in sputum and/or swab specimens. The total positive rate was 23% (25/110) combining PCR with serology. Acute infection of C. pneumoniae was common in the patients with asthma (57%), pneumonia (35%), COPD (26%) and bronchitis (25%). The clinical feature between C. pneumonia infection (n = 5) and non-C. pneumonia infection (n = 85) was not significanty different. CONCLUSION: Data suggest that Chlamydia pneumonia is an important pathogen that causes infections of the human respiratory tract and attention should be drawn to this special illness.

[The role of nitric oxide and nitric oxide synthase in the pathogenesis of asthma].

OBJECTIVE: To investigate the role of nitric oxide (NO) and nitric oxide synthase (NOS) in the pathogenesis of asthma. METHOD: 52 guinea pigs were randomly divided into four groups of 13 each: (1) asthmatic group (Group A): Dunkin-Hartley guinea-pigs were injected celiacly with 1 ml of 10% ovalbumin (OA). After 14 days, the animals were inhaled with an aerosol of 1% OA for 40-60 seconds for 10 days every other day; (2) Corticosteroid prevention group (Group CT): As above, just before the animals were inhaled with an aerosol, 0.5 mg/kg dexamethasone were injected celiacly; (3) N-nitro-L arginine prevention group (Group L): As Group A, just before the animals were inhaled with an aerosol, 0.4 mg/kg LNNA were injected celiacly; (4) Controls (Group C): and nitrate (NO2.-/NO3.-) levels in plasma, nitrite bronchoalveolar lavage fluid (BALF) and lung tissues were examined. At the same time, inducible nitric oxide synthase (iNOS) and constitute nitric oxide synthase (cNOS) activity levels in the lung tissues were examined, and the changes of cNOS in the guinea pig asthma model lung tissues were observed using histochemical detection. RESULT: All groups had no significant alteration of NO2.-/NO3.- in the plasma (P > 0.05). Group A had increased amounts of NO2.-/NO3.- in the BALF and in the lung tissues compared with the other groups (BALF: Group A 10.2 +/- 1.3, Group CT 7.2 +/- 1.1, Group L 7.3 +/- 1.3, Group C 6.2 +/- 0.8 mumol/L respectively, all of P < 0.01; the lung tissues: Group A 0.89 +/- 0.07, Group CT 0.16 +/- 0.09, Group L 0.24 +/- 0.09, Group C 0.18 +/- 0.05 nmol/mg respectively, all of P < 0.01). Group A also showed increased amounts of iNOS levels in the lung tissues than the other groups (Group A 59 +/- 18, Group CT 10 +/- 5, Group L 12 +/- 7, Group C 10 +/- 5 pmol/mg respectively, all of P < 0.01). Group L showed decreased amounts of cNOS levels in the lung tissues than the Group C (0.8 +/- 0.4, 1.2 +/- 0.4 fmol/mg, P < 0.05). While there were no significant alterations in the other groups (P > 0.05). Elevation of iNOS in the lung tissues was correlated with NO2-/NO3- in the BALF and in the lung tissues (r = 0.714, 0.842, respectively, P < 0.05, 0.01 respectively). NADPHd was found to be a histochemical marker reflecting cNOS activity. It was found that there was no marked alteration of cNOS activity in the Group A, Group CT and Group C, but lower in the Group L. CONCLUSION: There is increased production of iNOS in asthmatic guinea pigs, the iNOS produced could cause increased production of NO, and probably cause cytotoxicity and mediate airway hyperresponsiveness. NO and NOS may play an important role in the pathogenesis of asthma.

[Effect of ligustrazine on nitric oxide contents in cerebrospinal fluid and plasma of patients with cerebral infarction].

OBJECTIVE: To probe into the significance of Nitric Oxide (NO) in ischemic cerebral damage and effect of ligustrazine on it. METHODS: The NO contents in cerebrospinal fluid (CSF) and plasma of 20 controls and 62 patients with arteriosclerotic thrombotic cerebral infarction (ligustrazine group and common treatment group) were determined with Griess method before and after treatment. RESULTS: The NO content in CSF before treatment was higher in severe type, it was higher than that in moderate type, and than that in mild type, again higher than that in control group (all P < 0.05) and was positively correlated with the size of infarction (P < 0.01). There was no significant difference between the plasma NO content of patients and that of control group (P > 0.05), but there was a trend that plasma NO content decreased as the patient's condition worsened. After treatment, the curative effect and plasma NO content of ligustrazine group were both markedly higher than those of common treatment group (P < 0.05). There were no significant differences between CSF NO contents of the two groups, respectively, and that of control group (P > 0.05). CONCLUSIONS: Excessive NO produced in brain tissue and insufficient plasma NO participate the course of ischemic brain damage. Ligustrazine could increase the contents of plasma NO selectively.

[Biological effect of space flight on edible fungi onboard recoverable scientific satellite].

In order to breed good variety of edible fungi and provide data of cultivation of edible fungi in Controlled Ecological Life Support System (CELSS) in the future, 2 species of the edible fungi (Pleurotus ostreatus and Flammulina velutipes )were carried into space by a recoverable scientific satellite. A series of biological tests and cultivation experiments were done with the mycelia and sporocarp of these edible fungi. It was demonstrated that the edible fungi recovered from the satellite have superior characters such as: earlier sporocarp production, higher yield, higher transformation rate of raw fiber and organic matter and higher content of polysaccharide and endogenous hormones than the control.

[Antivirus effect of polysaccharides of brewer yeast in vitro].

The antivirus effect of polysaccharides of brewer yeast from yeast mud on 13 kinds of viruses including DNA and RNA virus along with their mechanisms were studied. The result showed that this effect was remarkable on the infections with poliovirus III, adenovirus III, ECHO6 virus, enterovirus 71, vesicular stomatitis virus, herpesvirus I, II, coxsackie A16 virus and coxsackie B3 virus. The polysaccharides of brewer yeast could also inhibit the development of cytopathic effect(CPE) and protect cultural cells from being infected with the above viruses.

[Ileocolonic pouch with appendix in situ through umbilicus as a continent conduit].

OBJECTIVE: To try to use appendix in situ without any special treatment as a conduit of the continent cutaneous ileocolonic reservoir. METHODS: An isolated ileocolonic segment, about 20 cm of terminal ileum and 20 cm of ascending colon, was detubularized by antimensenteric splitting and a pouch was completed by side-to-side anastomosis of the colon and the ileum loop. Both ureters were implanted into two separate submucosal tunnels of 3 cm long into the colon wall of the pouch. The appendix after tail was amputated was passed through the umbilicus hole with which the base of the cecum was sutured. RESULTS: As a conduit of the pouch, the appendix had a good continent ability when 8 patients were followed up for 10 to 30 months after operation. It was easy and convenient for the patients to make self-catheterization. The mean capacities of the pouch were 500-700 ml and then the pressures were 20-45 cm H2O while it was filled. There were no definite complications such as urine leakage, stoma stenosis, ureter reflux, and hydroureteronephrosis. CONCLUSION: The operation of the ileocolonic pouch with the appendix as a continent umbilical stoma is simple and easy to perform and has such advantages as good continent, convenient self-catheterization and provides a quality life for the patients undergoing total cystectomy.

[Expression of heat shock proteins in laryngeal carcinoma].

OBJECTIVE: To determine the expression of HSPs(HSP90 alpha, HSP90 beta, HSP70, HSP27) in laryngeal carcinoma and normal laryngeal mucosa. METHODS: Total RNA were extracted from 21 laryngeal cancer tissue samples and the normal mucosa of the corresponding larynx. The expression of HSPs mRNA was examined by Slot blot analysis. RESULTS: 1. HSP90 alpha and HSP70 were selectively overexpressed 5 times more in laryngeal carcinoma than in normal mucosa of larynx. 2. HSP90 beta and HSP27 were constitutively expressed at low level in these two groups. There was no significant difference between the levels of expression of HSP90 beta and HSP27 in laryngeal carcinoma and normal controls. CONCLUSION: The results that HSP90 alpha and HSP 70 mRNA were selectively overexpressed in laryngeal carcinoma suggested that HSP90 alpha and HSP70 might play a specific role in the pathogenesis of laryngeal cancer. To clarify the mechanism of HSPs action could help to provide theorical basis for the biological therapy of laryngeal cancer.

[Analysis of blood flow in ophthalmic arteries in patients with middle and late stages of glaucoma with Doppler sonography].

OBJECTIVE: To understand the actual blood flow in cases with middle and late stages of glaucoma and the role of vascular factor in the deterioration of visual function in such patients. METHODS: The blood flow in ophthalmic arteries was determined with color Doppler sonography (CDS) in 156 cases of patients with middle and late stages of glaucoma as well as in 75 cases of normal individuals as controls. RESULTS: The average peak systolic velocity (PSV) and end diastolic velocity (EDV) of ophthalmic artery in normal control are 35.98 +/- 9.13 cm/s and 11.98 +/- 3.90 cm/s; the PSV and EDV in primary open-angle glaucoma (POAG) group are 24.13 +/- 4.65 cm/s and 7.10 +/- 1.85 cm/s; and in primary close-angle glaucoma (PCAG) group are 22.89 +/- 5.43 cm/s and 7.06 +/- 1.92 cm/s, respectively. No significant difference was found between POAG group and PCAG group (P > 0.05). However, significant differences were shown between normal group and patients with glaucoma for both PSV and EDV (P < 0.01). CONCLUSION: The blood flow in ophthalmic arteries in patients with middle and late stages of glaucoma decreases significantly and the disorder in local blood circulation is evident, that is highly related to the deterioration of visual function and supports the vascular factor theory in the pathogenesis of glaucoma.

[Phacoemulsification for mature or hypermature cataracts].

OBJECTIVE: To discuss how to choose the methods of capsularhexis and phacoemulsification in cases with mature or hypermature cataract. METHODS: On fifty-nine mature or hypermature cataract patients, continuous circular capsularhexis or can-opener capsulotomy was performed. The nucleus was emulsified by the method of divide and conquer or phaco chop or rotatory carousel in the bag or at the plane of iris. RESULTS: Continuous circular capsularhexis was successful in 51 cases, failed or shifted to can-opener in 8 cases. There were posterior capsular rupture in 1 cases, partial dehiscence of the zonule in 1 cases and iris bitten by the phaco tip in 1 cases. Postoperatively, there were mild corneal edema in 6 cases and iris posterior synechia in 1 cases. CONCLUSION: Continuous circular capsularhexis is the key point to obtain a successful phacoemulsification for mature or hypermature cataract. Hydrodissection is not important before the phacoemulsification. Skillful phaco chop can reduce the surgical manipulation and the postoperative reaction.

[Synthesis of primaquine analogues and their antimalarial activity in mice].

Ten 4-methyl-5-substituted-phenoxy-6-methoxy-8-[(1-ethyl-4-amino) butylamino] quinolines (10a-j) have been synthesized and evaluated preliminarily for both suppressive and causal prophylactic antimalarial activities. The results of preliminary screening test showed that three of these compounds exhibited significant activity against Plasmodium yoelii in mice, among which 10c was 4-8 times as effective as primaquine. Moreover, 10c was superior to primaquine in suppressive test against Plasmodium berghei K173 strain in mice. On the basis of ED50 and ED90, 10c was about 2 times as active as primaquine. Most of other compounds showed similar activity and the minority of them exhibited lower activity compared with primaquine.

[The development of medical infusion monitoring and controlling network system].

The paper provided a medical infusion monitoring and controlling network system, in which the mainframe and its extensions can realize communication with each other by means of asynchronous serial port. At the same time, every extension can test infusion speed and adjust it to the presetting infusion speed gradually. The extensions also can alarm and stop infusion automatically when infusion will be over or there is a infusion malfunction.

[Experimental studies on dynamic changes of circulating antigens of Schistosoma japonicum in infected rabbits].

AIM: To observe the dynamic changes and disappearance of three kinds of schistosome circulating antigens(CAgs) in infected rabbits before and after praziquantel treatment. METHODS: Dot-ELISA assays were developed by using 3 McAbs to determine the dynamic changes of the gut-associated antigen (GAA), membrane-associated antigen (MAA) and soluble egg antigen (SEA) respectively in sera of rabbits infected with S. japonicum. RESULTS: By the 4th week after infection 39% of the rabbits infected with S. japonicum were GAA positive and only 8.6% were MAA or SEA positive. All the infected rabbits becOme positive with GAA, MAA and SEA by the 8th week after infection. The titers of the GAA, MAA and SEA in sera of the infected rabbits reached their peaks by the 10th week after infection. Titers from rabbits of the untreated group rabbits remained high level until the 27th week after infection. SEA titers were found negative in 10% of the infected rabbits in the treatment group by the 6th week after treatment. The titers of GAA of, MAA' and SEA were found negative in 20%, 80% and 70% of the rabbits in the treatment group, respectively the 8th after treatment. GAA, MAA and SEA in sera except one of the treated rabbits were all negative by the 14th week after treatment. CONCLUSION: The determination of GAA is useful for the early diagnosis of schistosomiasis, while the determination of, MAA and SEA might be used for the efficacy evaluation.
