[Isolation of 28 new STSs at Xq27.3].

A plasmid sublibrary of the 475 kb insert of YAC209G4 was constructed by using pBS II KS vector. The library of 3,500 clones having 100-600 bp inserts was screened with the probe of the 475 kb insert blocked with competitor DNA. Sixty unique single copy clones were found and sequenced. Checking with GenBank, 28 new STSs were obtained. Genbank accession numbers are U26560-26587. Three STSs were tested by PCR with appropriate primers using human genomic DNA as template and showed specific amplification bands as expected.

[Study on optimal conditions for the determination of glucose by measurement of oxygen consumption rate].

We studied optimal conditions for the determination of oxygen consumption rate (OCR). The assay, with a working range up to 35 mmol/L, had an intra-assay coefficient of variation of 0.7%-3.52%, and the inter-assay coefficient of variation was 4.86%. In spiking experiments, the recovery range was 98.9%-101%. The assay was compared with CX3 (Beckman Co.). A linear regression analysis yielded a slope of 1.05, an inercept of 3.82 and a correlation coefficient of 0.999. The serum of turbidity above +2 had a negative interference on the assay (> 3.4%). The obvious interference of hemolysis, icterus and vitamin C to the assay were not observed.

[Alternative splicing of the FMR1 gene in human fetal tissues].

OBJECTIVE: To gain more insight into biological role of the alternative splicing of the FMR1 gene, isoforms of the FMR1 mRNA from human fetal heart, spleen, liver and kidney were analyzed. METHODS: RNAs were isolated from the tissues of an abortion fetus (approximately 6 months postconception). Alternative splicing patterns of the FMR1 mRNA were analyzed by RT-PCR and cloning strategy. RESULTS: One isoform of the FMR1 mRNA was found to be dominant in all of the four tissues. The major isoform was as same as the dominant one in the fetal cortex but was different from the major isoform in adult brain. The difference between the major isoforms in the fetal tissues and adult brain was the splicing out or retaining the peptide encoded by exon 12 and exon 17. CONCLUSION: This result suggested a developmental switch of alternative splicing of the FMR1 gene. The difference between the major isoforms in the fetal tissues and adult brain suggested the two peptides may have special roles in related developmental stages.

[Survey of botanical origin of tongcao (medulla Tetrapanacis) and identification of its commercial products].

Through the investigation on botanical origin, output and sales of Togcao in producing areas, it has been proved that there are twenty-two species of six families used as Tongcao. The medicinal parts are the pith of stems or petioles. The provinces featuring more species and larger output of Tongcao are Sichuan, Yunnan, Guizhou, Guangxi, Hunan and Shaanxi. A hundred and two pieces of commercial samples collected from twenty-six provinces in China, Hongkong area, Japan, Malaysia, Thailand, Singapore and Republic of Korea have been identified. The result shows that both Xiaotongcao and Datongcao are called by the same name Tongcao. The main species is Xiaotongcao, which takes a proportion of 70% in Tongcao. And the Tongcao(Tetrapanax papyriferus, taking a proportion of 20%) as recorded in the Chinese Pharmacopoeia (1995 edition) is seldom used.

[Determination of chlorosilanes by gas chromatography].

Chlorosilanes are the essential raw material of SiC fibre made by "CVD (chemical vapour deposition)" method. The purity of chlorosilane affects the quality of SiC fibre obtained. Therefore, a simple and reliable method for the determination of the constituent of chlorosilane is needed. A gas chromatographic method for quantitative analysis of chlorosilane is described in this paper. The gas chromatographic conditions were as follows: column, 10% diethyl phthalate on 6201 support (60-80 mesh), 3m x 5mm i.d. stainless-steel column; column temp., 60 degrees C; injection temp., 110 degrees C; thermal conductivity detector temp., 140 degrees C carrier gas H2 with flow rate of 60 mL/min. The results show that the method is simple and rapid, and has good precision and reproducibility. The coefficient of variation of the method was less than 1% (n = 6).

[Determination of furanoterpenoid toxins from sweet potato by thin-layer chromatography].

Sweet potato root tissue, when infected with different pathogens, invaded by insects, or irritated by certain chemicals, turns black-brown and produces and accumulates a large amount of furanoterpenoids such as ipomeamarone, ipomeamaronol, ipomeanine and 4-ipomeanol, which are toxic to animals. The furanoterpenoids from sweet potato root tissues infected with Ceratocystis fimbriata were extracted with ether. The extract solution was washed with 5% Na2CO3 and followed with distilled water and dried with dehydrated sodium sulfate (Na2SO4). The resulting ether extract was evaporated and the residue (crude sample) was dissolved in a small volume of chloroform. The determination of furanoterpenoid toxins was made on the plates at 20 degrees C and 30 degrees C respectively. The chloroform-soluble fraction was spotted on a silica gel G thin-layer plate and developed using different solvent systems, n-hexane and ethylacetate (4:1, V/V) , benzene and methanol (9:1, V/V) and petroleum ether and ethylacetate (2:1, V/V), until the solvent front migrated 10 cm from the starting spots. After development, Ehrlich's reagent was sprayed on the plate to detect the compounds. In the experiment, pure ipomeamarone and ipomeamaronol were used as standards. Thin-layer chromatograms on silica gel plates showed the furanoterpenoids were resolved effectively. The repeatability of Rf value was good. The resolution was the best in the solvent systems of benzene-methanol and petroleum ether-ethylacetate at 20 degrees C. After spraying Ehrlich's reagent, ipomeamarone (C) appeared as a light pink spot which soon changed to a dark gray, ipomeamaronol (A) as a purple red spot, compound B and D as a purple red and a gray spot respectively. Compound A, B, C and D had Rf values of 0.05, 0.14, 0.55 and 0.64 (n-hexane-ethylacetate), of 0.15, 0.29, 0.61 and 0.71 (petroleum ether-ethylacetate), of 0.40, 0.55, 0.71 and 0.79 (benzene-methanol), respectively. The separated spots were scanned by CS-930 TLC scanner at 527 nm and quantitative determination by external standard calibration. Ipomeamarone was detected by thin-layer chromatography and the limits of detection were 0.005 microg for pure and 0.02 microg for crude sample.

[Collisional energy transfer of the Rb (7 2D) state by ground state K atoms].

In the K-Rb vaper mixtures, the two-photon stepwise excitation of rubidium atoms from the ground state to the state Rb 7 2D is obtained by the use of a rubidium lamp and a cw dye laser. The cross sections for the collisional process Rb (7 2D) +K (4S) -->Rb (5S) +K (7S. 5D) have been measured by observation of the resulting fluorescence. The ratio of K (7S. 5D) to Rb7D fluorescence contains a contribution from K 7S<==>5D mixing. The contribution could effectively be subtracted out using a second experiment in which a potassium lamp and a cw dye laser were used to pump the K7S or 5D state, and the ratio of 7S to 5D or 5D to 7S fluorescence was monitored. The cross sections (in 10(-15)cm2) for Rb7D-->K7S. 5D excitation transfer are 25.8 +/- 10. 3 and 1.5 +/- 0.6 respectively.

[FT-IR characterization of extracted organic phase containing D2EHPA-lanthanide complexes and effects of adding DMHPA].

FT-IR spectroscopy was used to characterize the extracted organic phase of di (2-ethylhexyl) phosphoric acid-n-heptane containing lanthanide ions Ln3+ (Ln: Y, Pr, Nd, Eu ). It was found that the coordination interaction of heavy lanthanide ions with D2EHPA is stronger than that of light lanthanide ions, and saponification percentage of D2EHPA affects both the absorption frequency and the intensity of P=O bond in the extracted organic phase. Furthermore, the addition of di (1-methylheptyl) phosphoric acid to the D2EHPA extraction system leads to the variation of both peak position and intensity of P=O bond.

[Study on the properties and microstructure of the gel in the extracted organic phase containing DMHPA-RE(III)].

The content of Na+, RE3+ and the swelling characteristic of dried gel containing DMHPA[di-(1-methylheptyl)phosphoric acid]-RE(III) have been studied by FTIR and ICP/AES spectroscopy methods. The result demonstrates that (1) the mechanism of microemulsion extracting lanthanide ions is complicated and it is difficult to be explained with simple chemical equations, (2) The dried gel can swell, but the aggregation states and microstructure change as the used solvents and its content vary, (3) The far infrared spectroscopy shows that there are ring structures in the gel and differences of the ability in coordination with DMHPA between light and heavy lanthanide ions.

[Energy pooling collisions between excited Na and K atoms].

Two different atomic species, sodium and potassium, are resonantly excited in a ceel by a CW dye laser and a potassium lamp, respectively. The density of the excited Na (3P) atoms in the cell is measured directly using the absorption-equivalent-width technique on the lines of the diffuse series. To isolate the heteronuclear contribution in the fluorescence spectrum, a modulation detection technique is adopted. The energy-pooling collisions between Na (3P) and K (4P) are observed and cross sections are obtained.

[Examination of scanning technique for lung cancer screening with helical CT].

As a new application of helical CT scanning we evaluated the parameters for lung cancer screening with an extremely low doses and large helical pitch. On the phantom studies, the image quality obtained with the low dose parameter was not inferior to that of the usual screening technique but artifacts were increased with the large helical pitch. A scanning technique using 120 kv, 40-60mA, 10mmth, 20mm/ sec, and a reconstruction pitch of 2 was used for lung cancer screening (screening parameters) (50 cases), and comparison was made between the detectability of the screening parameters and the routine parameters (120 kv, 200mA, 10mmth, 10-13mm/sec, and a reconstruction pitch of 1-1.3). Detectability with the screening parameters was as follows: nodular lesions (< 5mm in size: 76%. 5-10mm: 90-93% 10mm < 100%), linear lesions: 94-95%, infiltrations: 93-100%. There were no false negative lesions, when the reconstruction pitch of the screening parameters was changed from 1 to 0.25. In conclusion, reconstruction pitch had the most influence on lesion detectability.

[Improvement of longitudinal resolution by deblurring technique in helical CT of the head and neck].

We assessed the improvement of longitudinal resolution in helical CT by deblurring techniques in a phantom and 8 head and neck patients. After helical scanning with 3- or 5-mm-collimation and 3- or 5 mm/sec table feed, overlapped axial images were reconstructed at every 0.3 or 0.5 mm pitch. Deblurring techniques were applied to these data. The slice profile and longitudinal resolution were improved by deblurring. Longitudinal resolution increased with the strength of deblurring. Image sharpness was improved in reconstructed images of MPR, MIP and 3D, and better visualization of the peripheral vessels was obtained in CTAs.

[A new helical CT approach with ECG gated reconstruction].

We proposed a new helical CT approach with ECG-gated reconstruction for obtaining high quality 2D, 3D, and 4D images. Original helical images were acquired with a single helical scan with overlapping reconstruction and ECG information. Post-processing to improve temporal resolution and 4D volume set reconstruction were performed, and 2D, 3D and 4D displays were obtained. Using this new method, 2D and 3D images without cardiac motion artifacts were obtained by selecting images in diastolic phase. 2D images with cardiac motion were obtained using images of the same position and different cardiac phases. Finally, 4D images (3D images with phase information) were obtained.

Lipoprotein lipase deficiency with pancreatitis in mink: biochemical characterization and pathology.

A severe hyperlipemia in mink, with a pattern that suggested recessive inheritance, was observed at a farm in Norway. On a normal mink diet, affected animals had grossly elevated levels of plasma triglycerides which decreased towards normal on a low-fat diet. Normal minks had the main part of their plasma cholesterol in the HDL fraction. Affected minks, although severely hypertriglyceridaemic, had almost normal levels of both LDL and HDL. Affected minks frequently had lipogranulomas in the mesentery and the pancreas. The lipogranulomatous tissue contained spaces filled with an amorphous, sudanophilic substance with many foamy macrophages in the fibrous tissue between the lesions. Separation of postheparin plasma on heparin-agarose revealed that the affected minks had no detectable lipoprotein lipase activity but normal activity of hepatic lipase. Both normal and affected minks had inactive lipoprotein lipase protein in pre- and post-heparin plasma. This protein, which eluted before the active lipase from heparin-agarose, probably corresponds to lipase monomers. The presence of lipoprotein lipase mass in the affected minks, but no activity, indicates that there might be a point mutation in the lipase gene. The minks provide a new animal model for studies on pancreatitis induced by hypertriglyceridemia and on lipoprotein metabolism in the lipoprotein lipase-deficient state and show features similar to those found in human hyperlipoproteinemia type I.
