AIM: Extensive neurophysiological investigations were carried out in 100 healthy subjects and 84 patients with penile erectile dysfunction. METHODS: Following examinations were performed, spinal and scalp somatosensory evoked potentials (SEPs) to stimulation of the dorsal nerve of penis, motor evoked potentials (MEPs) from bulbocavernosus (BC) in response to scalp and spinal root stimulation, and measurement of sacral reflex latency (SRL) from anal sphincter (AS). RESULTS: In the healthy subjects, the mean sensory total conduction time (sensory TCT), as measured at the peak of the scalp P1 (P40) wave was 39.73 ms. The mean sensory central conduction time (sensory CCT = spinal-to-scalp conduction time) was 28.98 ms. The mean peripheral conduction time (PCP) was 9.40 ms. Transcranial brain stimulation was performed by using a magnetic stimulator during voluntary contraction of the examined muscle. Spinal root stimulation was performed at rest. Motor total conduction time (motor TCT) to BC muscles was 20.48 ms. Motor central conduction time (motor CCT) to sacral cord segments controlling BC muscles was 14.42 ms at rest. The mean SRL was 35.13 ms. CONCLUSION: Combined or isolated abnormalities of SEPs, MEPs, and SRL were found in patients with erectile dysfunction.

OBJECTIVES: To explore the association between schizophrenic symptoms and superoxide dismutase (SOD), and to investigate the effect of classic antipsychotic haloperidol plus the extract of Ginkgo biloba (EGb) on SOD. METHODS: In 54 patients with chronic refractory schizophrenia, 27 were treated with haloperidol plus EGb (group 1), and the rest received haloperidol plus placebo (group 2). Superoxide dismutase (SOD) levels of these patients were measured before and after treatment and compared with the levels of 25 healthy volunteers. Therapeutic efficacy was equated with a change in clinical rating scores assessed by standardized measurement tools including the Scale for Assessment of Positive Symptoms (SAPS) and the Scale for Assessment of Negative Symptoms (SANS). RESULTS: Patients in group 1 improved significantly as demonstrated by scores from both SAPS and SANS, while those in group 2 only by scores from SANS. Assessed by SAPS, the response of patients receiving haloperidol plus EGb was more significant than those receiving haloperidol only. SOD levels before treatment in all patients were significantly higher than those in normal controls. After treatment, SOD levels decreased significantly in group 1 but not in group 2. In addition, before treatment, SOD levels in all patients correlated significantly with SAPS score. The levels of SOD measured before treatment were also correlated with the improvement of patients as measured by SAPS and SANS after 12 weeks. CONCLUSIONS: EGb may enhance the efficacy of classic antipsychotic haloperidol on schizophrenia, especially on positive symptoms. It may work through an antioxidant efficacy that is involved in the therapeutic mechanism.
OBJECTIVE: To illustrate the possible role of thiols (SH-groups) in arsenic trioxide (As2O3)-induced apoptosis. METHODS: Leukemia (NB4, HL60, U937) and lymphoma (Jurkat and Namalwa) cell lines were treated with As2O3 together with buthionine sulfoximine (BSO), a selective inhibitor of gamma-glutamylcysteine synthetase. The mitochondrial transmembrane potentials (delta psi m) and the percentage of cells in apoptosis were measured by means of flow cytometry. RESULTS: One mmol/L BSO significantly enhanced As2O3-induced delta psi m collapse and apoptosis in NB4 cells. One mumol/L As2O3 did not induce the delta psi m collapse and apoptosis in Namalwa, HL60, U937 and Jurkat cells, while these effects were induced significantly by the co-treatment of 1 mumol/L As2O3 and 1 mmol/L BSO. CONCLUSION: Thiols are important chemosensors of As2O3-induced delta psi m collapse and apoptosis.
OBJECTIVE: To explore the relationship between platelet activation and enhanced coagulability and the occurrence unstable angina. METHODS: Plasma levels of a-granule membrane protein (GMP-140) and fibrin D-dimer were measured in 22 patients with unstable angina during the acute phase and after relief. The same procedure were compared with those in 20 control subjects and 18 patients with stable angina. RESULTS: Plasma levels of GMP-140 and D-dimer were significantly higher in patients with unstable angina both during the acute phase (P < 0.01) and after the relief (P < 0.05) as compared with those in control subjects and patients with stable angina, but there was no significant difference between the latter two groups (P > 0.05). There was also no significant difference between the patients with unstable angina during the acute phase and after the relief (P > 0.05). CONCLUSION: It is shown that activation of platelet and coagulation system combined with endothelial injury may play an important role in the development of unstable angina.
OBJECTIVE: To study the changes and clinical significance of plasma thyroid hormones were during cardiopulmonary bypass (CPB). METHODS: Plasma thyroid hormones was studied in 36 patients during open heart surgery under CPB. Blood samples were taken preoperatively, and 1, 2, 3, 4, 5 days postoperatively. Doppler echocardiography was performed in 16 patients before and after surgery for evaluating heart function. RESULTS: Before CPB, the levels of thyroid hormones were within normal range. At early stage after CPB, the levels of FT(3), FT(4), TT(3) and TT(4) were significantly decreased and increased. Both TSH and rT(3) were increased significantly after CPB (P < 0.05) and reached their highest levels at the first postoperative day. TSH and rT(3) were gradually dropped subsequently, but TSH returned to the basic level and the level of rT(3) was still high. A positive relation was seen between plasma thyroid hormones and cardiac function. CONCLUSIONS: The changes of plasma thyroid hormones are closely related to cardiopulmonary bypass and could result in postoperative cardiac disfunction.

OBJECTIVE: To explore if there was relationship between small body size at birth and type 2 diabetes and impaired glucose tolerance (IGT) during adulthood in China. METHODS: Six hundred and twenty-eight singletons born in the Peking Union Medical College Hospital in Beijing during July 1948 to the end of 1954 were followed-up and their medical records at birth were abstracted. Anthropometry and standard oral glucose tolerance test were carried out for all of them. Plasma insulin level was measured with radioimmunoassay for them. RESULTS: Prevalence of type 2 diabetes and IGT decreased with the increase in their birth weight, Ponderal index (PI) and head circumference at birth (for trend test, chi(2) = 6.7, P = 0.01; chi(2) = 4.8, P = 0.03; and chi(2) = 5.8, P = 0.02; respectively), with the highest of 43.8% in those with thin body size (PI < 24 kg/m(3)) at birth and obesity (BMI < 75 percentile) during adulthood, and the lowest of 8.3% in those with more fat at birth (PI >or= 28 kg/m(3)) and keeping relatively thin (BMI < 25th percentile) during adulthood. BMI of mothers in their early and late pregnancy correlated reversely with blood glucose levels two hours after sugar load for their children in adulthood. In addition, body weight, PI and head circumference at birth correlated reversely with their plasma insulin levels fasting and two hours after sugar load and levels of 32 -- 33 split pro-insulin during their adulthood. After adjustment for current BMI, lifestyle and economic status, body size at birth mentioned above still associated with their glucose tolerance and level of plasma insulin during adulthood. CONCLUSION: There was relationship between small body size at birth and prevalence of type 2 diabetes and IGT during adulthood. Prevalence of type 2 diabetes and IGT depended on the synergic effect of thin body size at birth and obesity during adulthood.
OBJECTIVE: To observe the antioxidative effect of four kinds of Tongcao and Xiao-Tongcao polysaccharides. METHOD: Aged mice (nine months) were abdominal administrated with four kinds of polysaccharides (Tongtuomu, Ximashanjingjiehua, Xinanxiuqiu, Ditanghua) at dosages of 160 mg/kg and 80 mg/kg for 45 days. RESULT: Four kinds of polysaccharides could obviously decrease the content of serum and liver lipoperoxide(LPO) as well as the content of lipofuscin(LF) in brain and cardiac muscle of aged mice, and also increase the activity of superoxide dismutase(SOD) of red cells in aged mice. CONCLUSION: The polysaccharide components of Tongtuomu and three kinds of Xiao-Tongcao have antioxidative effect, suggesting that these substances have antiageing function.
OBJECTIVE: To investigate the effect of hairy root of Astragalus membranaceus(HRA). METHOD: HRA were given 10 g/kg per day for 50 days to aged mice treated with D-galactose, and the effect on memory and antioxidant functions were estimated. After administration of HRA 10 g/kg for four days, anti-ischemia-reperfusion kidney model of rat was prepared. The kidney function, activity of superoxide dismutase (SOD) and content of malondialdehyde(MDA) in kidney were examined. Mice with immunosuppression induced by cyclophosphamide were given orally HRA 10 g/kg for 12 days. The activity of natural killer (NK) cells was measured. RESULT: HRA improved the memory, raised SOD activity in brain and liver, decreased the MDA content in the liver of aged mice, reduced the MDA content in ischemia-reperfusion kidney, decreased the creatinine level in blood of rats, and promoted the activity of NK cells in immunosuppressed mice. CONCLUSION: Similar to the natural A. membranaceus, HRA has senility-preventing, antioxidizing and immunomodulating functions.
It is an important issue whether and how the response kinetics, including activation, desensitization and deactivation, of gamma-aminobutyric acid (GABA) receptors plays roles in rapid inhibitory postsynaptic transmission. With the development of molecular biology, and the use of patch clamp techniques and rapid solution changers, the structural basis, underlying mechanisms at single channel level and functional roles of the characteristics of GABA receptors have been fully explored. It is revealed that the response kinetics of GABA receptors plays a crucial role in modulating inhibitory postsynaptic currents and has a great importance for rapid inhibitory synaptic transmission.
Fifty cases of male sexual dysfunction were analyzed in order to investigate the incidence and causes of the erectile dysfunction in the different injuries. The results showed that the incidence of erectile dysfunction of the patients with brevic fracture and urethra injury is 62.50%, main caused by the damage of pudendal nervous and arterial system. The incidence in the patients with the spinal fracture and light spinal cord injury is 50%, caused by the damage of nervous system. The erection in the 17 cases of the patients with only one testis injury is normal. Two cases with bilateral testis injury have suffered from erectile dysfunction.
A bacterium producing lactate oxidase was re-screened from five strains based on previous researches. The lactate oxidase activity was the highest in strain L1 and this stain was chosen as the enzyme source. Morphological and physiological studies revealed that the bacterial strain L1 belongs to the Edwardsiella tarda Biogroup I. This stain is different from the reported strains Mycobacterium and Pediococcus, which produce lactate oxidase. The enzyme producing conditions were studied in shaking cultures, and the effects of initial pH, riboflavin, lactate and ammonia sulphate concentrations on the production were carried out respectively. The bacteria resource of enzyme is significant to pyruvate production by enzymatic method, and to the enzyme assay of lactate for medical diagnosis, and the application of enzyme electronic probe.

OBJECTIVE: Human interleukin(hIL)-15 expression of lung cancer cell lines transfected by rhIL-15 cDNA in vitro was observed. METHODS: The recombinant plasmid pL-IL-15-SN was established by inserting IL-15 cDNA into vector pLXSN at sites of EcoR I and BamH I. The pL-IL-15-SN was transfected into human lung squmosuse carcinoma (PG) cell line and murine lung adenocarcinoma (LA795) cell line, respectively. Positive clones were obtained by the selection in G418 conditioned culture. Bioactivity on rIL-15 was detected by dependant-proliferation of CTLL-2 cells in vitro. RESULTS: Three positive PG and four positive LA795 cell clones were obtained respectively. The results of the assays for hIL-15 bioactivity showed that the expression levels of rhIL-15 ranged from 142 to 201 or from 138 to 178U/(ml.10(6)) for positive PG cells or LA795 cells, respectively. CONCLUSIONS: Human and murine lung carcinoma cells transfected with IL-15 cDNA can express hIL-15 with bioactivity.
OBJECTIVE: To clone the novel full-length cDNA related to mouse embryonic development. METHODS: Starting from a new expression sequence (clone 2 #) out of a mouse embryonic cDNA library, its full-length cDNA were obtained by rapid amplification of cDNA ends with polyA RNA isolated from mouse 8-9 day embryonic tissue as template, combined with screening of mouse embryonic cDNA library by PCR. Based on the sequences at the two ends of the cDNA, the primers for long range RT-PCR were designed to confirm its factuality. RESULTS: The cDNA isolated in our hands was one of the alternative splicing isoforms of mouse de novo DNA cytosine 5'-specific methyltransferase gene (Dnmt3b) reported in July, 1998. A new alternative splicing region upstream of its initiate codon was found 5' forwards 152 bp over the Dnmt3b cDNA sequence. Eight alternative splicing isoforms of Dnmt3b from mouse embryonic tissue were cloned (Genbank accession number are AF151969, AF151970, AF151971, AF151972, AF151973, AF151974, AF151975, AF151976 respectively). CONCLUSIONS: The 4 kb full-length Dnmt3b cDNA was isolated. Dnmt3b may have novel exons at its 5' upstream and present a complex alternative splicing pattern during mouse embryonic development.
