Effect of copper deficiency on oxidative DNA damage in Jurkat T-lymphocytes.

The micronutrient copper is a catalytic cofactor for copper, zinc superoxide dismutase and ceruloplasmin, which are two important antioxidant enzymes. As such, a lack of copper may promote oxidative stress and damage. The purpose of this study was to determine the effect of copper deficiency on oxidative damage to DNA in Jurkat T-lymphocytes. To induce copper deficiency, cells were incubated for 48 h with 5-20 microM 2,3,2-tetraamine (2,3,2-tet), a high affinity copper chelator. Such treatment did not affect cell proliferation/viability, as assessed by measuring mitochondrial reduction of WST-1 reagent (4-[3-(4-Iodophenyl)-2-(4-nitrophenyl)-2H-5-tetrazolio]-1,3-ben zen e disulfonate). Furthermore, the induction of copper deficiency did not promote oxidative DNA damage as evaluated by the comet assay. Comet scores were 15 +/- 0 and 16 +/- 1 for control and copper-deficient cells, respectively. However, the copper-deficient cells sustained greater oxidative DNA damage than the control cells (comet scores of 175 +/- 15 and 50 +/- 10, respectively) when both were oxidatively challenged with 50 microM hydrogen peroxide (H(2)O(2)). Supplemental copper but not zinc or iron prevented the potentiation of the H(2)O(2)-induced oxidative DNA damage caused by 2,3,2-tet. These data suggest that copper deficiency compromises the antioxidant defense system of cells, thereby increasing their susceptibility to oxidative DNA damage.

Comparative immunohistochemical distributions of carboxy terminus epitopes from the mu-opioid receptor splice variants MOR-1D, MOR-1 and MOR-1C in the mouse and rat CNS.

The present study examined immunohistochemically the CNS distributions of a splice variant of the mu-opioid receptor, MOR-1D, in both rats and mice. In MOR-1D, exon 4 of MOR-1 is replaced by two additional exons that code for seven amino acids. Using rabbit antisera, we compared immunohistochemically the regional distribution of a C-terminal epitope of MOR-1D to that of a C-terminal epitope from MOR-1 and a C-terminal epitope from another splice variant, MOR-1C. The general distribution of MOR-1D-like immunoreactivity was similar in both mouse and rat. MOR-1D-like immunoreactivity was seen in the dentate gyrus and in the mossy fibers of the hippocampal formation, the nucleus of the solitary tract and the area postrema, the inferior olivary nucleus, the nucleus ambiguous, the spinal trigeminal nucleus and the spinal cord. MOR-1D-like immunoreactivity was not observed in some regions containing dense MOR-1-like immunoreactivity, such as the striatum or the locus coeruleus. In regions containing MOR-1, MOR-1C and MOR-1D, the pattern of each variant was unique.MOR-1D and MOR-1C are splice variants of the cloned mu-opioid receptor MOR-1. Although they differ only at the tip of the carboxy terminus, they show marked differences in their regional distributions, as determined immunohistochemically by epitopes in their unique carboxy termini. Since the splice variants are derived from the same gene, these differences in regional distribution imply region-specific messenger RNA processing.

Natural history of Streptococcus sanguinis in the oral cavity of infants: evidence for a discrete window of infectivity.

The heterogeneous group of oral bacteria within the sanguinis (sanguis) streptococci comprise members of the indigenous biota of the human oral cavity. While the association of Streptococcus sanguinis with bacterial endocarditis is well described in the literature, S. sanguinis is thought to play a benign, if not a beneficial, role in the oral cavity. Little is known, however, about the natural history of S. sanguinis and its specific relationship with other oral bacteria. As part of a longitudinal study concerning the transmission and acquisition of oral bacteria within mother-infant pairs, we examined the initial acquisition of S. sanguinis and described its colonization relative to tooth emergence and its proportions in plaque and saliva as a function of other biological events, including subsequent colonization with mutans streptococci. A second cohort of infants was recruited to define the taxonomic affiliation of S. sanguinis. We found that the colonization of the S. sanguinis occurs during a discrete "window of infectivity" at a median age of 9 months in the infants. Its colonization is tooth dependent and correlated to the time of tooth emergence; its proportions in saliva increase as new teeth emerge. In addition, early colonization of S. sanguinis and its elevated levels in the oral cavity were correlated to a significant delay in the colonization of mutans streptococci. Underpinning this apparent antagonism between S. sanguinis and mutans streptococci is the observation that after mutans streptococci colonize the infant, the levels of S. sanguinis decrease. Children who do not harbor detectable levels of mutans streptococci have significantly higher levels of S. sanguinis in their saliva than do children colonized with mutans streptococci. Collectively, these findings suggest that the colonization of S. sanguinis may influence the subsequent colonization of mutans streptococci, and this in turn may suggest several ecological approaches toward controlling dental caries.

Physiological functions of the regulatory domains of the cardiac Na(+)/Ca(2+) exchanger NCX1.

Physiological functions of the intracellular regulatory domains of the Na(+)/Ca(2+) exchanger NCX1 were studied by examining Ca(2+) handling in CCL39 cells expressing a low-affinity Ca(2+) regulatory site mutant (D447V/D498I), an exchanger inhibitory peptide (XIP) region mutant displaying no Na(+) inactivation (XIP-4YW), or a mutant lacking most of the central cytoplasmic loop (Delta246-672). We found that D447V/D498I was unable to efficiently extrude Ca(2+) from the cytoplasm, particularly during a small rise in intracellular Ca(2+) concentration induced by the physiological agonist alpha-thrombin or thapsigargin. The same mutant took up Ca(2+) much less efficiently than the wild-type NCX1 in Na(+)-free medium when transfectants were not loaded with Na(+), although it appeared to take up Ca(2+) normally in transfectants preloaded with Na(+). XIP-4YW and, to a lesser extent, Delta246-672, but not NCX1 and D447V/D498I, markedly accelerated the loss of viability of Na(+)-loaded transfectants. Furthermore, XIP-4YW was not activated by phorbol ester, whereas XIP-4YW and D447V/D498I were resistant to inhibition by ATP depletion. The results suggest that these regulatory domains play important roles in the physiological and pathological Ca(2+) handling by NCX1, as well as in the regulation of NCX1 by protein kinase C or ATP depletion.

Long-term results of functional hemispherectomy for intractable seizures.

OBJECTIVE: From May 1989 to April 1997, functional hemispherectomy was performed in 8 cases of intractable seizures. We retrospectively analyzed our experience to evaluate the seizure control and complications of this surgical technique. METHODS: Following Dr. Rasmussen's model of functional hemispherectomy or performing a modification of this operation, we removed the sensorimotor cortex and temporal lobe associated with disconnection of the remaining portions of the frontal lobe and parieto-occipital lobe. RESULTS: All the patients were followed up for 3-11 years (mean 6.7 years). Satisfactory seizure control was obtained in all the cases. Life quality improved and patients worked or studied well after the operations. No cases of superficial cerebral hemosiderosis were found. CONCLUSION: Modified functional hemispherectomy may allow the patients to lead more independent lives by leading to a cessation or reduced frequency of seizures.

Chromosomal numerical aberrations detected by fluorescence in situ hybridization on bladder washings from patients with bladder cancer.

OBJECTIVE: Previous studies on touch biopsy specimens have determined numerical or structural changes involving many different chromosomes in bladder cancer. The aim of this study was to evaluate the use of fluorescence in situ hybridization (FISH) assay in bladder washings as an objective technique to detect chromosomal numerical aberrations in bladder cancer. The main advantages of bladder washings are that they can be easily collected during the clinical follow-up of patients with superficial bladder cancer and they do not contain so many degenerate cells as urine samples. METHODS: We collected specimens from 25 patients who underwent transurethral resection of bladder tumors. Double target FISH assays with centromeric labeled probes for chromosomes 7, 8, 9 and 11 were used on the bladder washings and on the touch biopsy slides. The results were compared to flow cytometry and tumor grade and stage. RESULTS: We found monosomy 9 and trisomy 7, 8, 9 and 11 in 28, 32, 36, 28 and 25% respectively of the patients. FISH analysis of bladder washing versus touch biopsy specimens were concordant in approximately 90% of the slides. Total DNA aneuploidy correlated well with numerical aberrations of chromosomes 7, 8 and 11, but not with chromosome 9. CONCLUSION: Although better hybridization efficiency was obtained on touch biopsy slides, the results in bladder washings were in high concordance. FISH analysis on bladder washing samples may become a simple tool to improve the accuracy of cytology.

An Internet-based telediagnosis system for Chinese medicine.

The Internet is becoming an increasingly popular medium for communication and is being used more and more for telemedicine. Telediagnosis is one of the most developed components of telemedicine. We have designed and partly implemented a telediagnostic system, using the Internet as the means of communication, to enhance access to specialists and health providers in Chinese medicine.

Clinical observation and experimental study on compound hypoglycemic decoction for hyperglycemia.

PURPOSE: To observe the clinical efficacy of Compound Hypoglycemic Decoction (CHD) and its effect on serum total cholesterol in model mice. METHOD: The paired t test was used to analyze the data recorded before and after administration of drugs for hyperglycemia induced by intraperitoneal injection of 75% egg yolk emulsion in experimental mice. CHD and Fenofibrate were administered as prevention measures. RESULTS: The total effective rate of serum total cholesterol (TC) decrease in 60 cases of hyperglycemia was 86.66% and that of serum total triglyceride (TG) decrease was 81.81%. The total effective rate of high-density lipoprotein-cholesterol (HDL-C) increase was 75%. The decrease in TC and TG, and the increase of HDL-C after treatment by in-group comparison were all significant (P < 0.05). 21.9% and 22.2% decrease in the total cholesterol was respectively found in the CHD and Fenofibrate groups (both P < 0.05), with no significant difference. CONCLUSION: The hypoglycemic action of Compound Hypoglycemic Decoction was remarkable in clinical practice, and it is very effective in preventing hyperglycemia in experimental mice.

Dopamine receptor gene polymorphisms in Guangzhou Hans.

OBJECTIVE: To determine the distribution of dopamine D2, D3, and D5 receptor(DRD2, DRD3, DRD5) gene polymorphisms in Guangzhou Hans. METHODS: A total of 141 healthy Guangzhou Hans were studied by the use of polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) and PCR-amplification of specific allele (PASA) techniques. Current results were compared with the data on other ethnic groups. RESULTS: Within the 141 individuals tested, the frequencies of the A1(Taq I-) and A2(Taq I+) alleles of the TaqI A mutation site in the 3'non-coding region of the DRD2 gene were found to be 48% and 52%, respectively. The observed frequencies of the A1A1, A1A2 and A2A2 genotypes were found to be 17%, 52% and 31%, respectively, which met Hardy-Weinberg equilibrium. The frequencies of the A1 and A2 alleles of the Bal I mutation site in the exon 1 of DRD3 gene were found to be 73% and 27%, respectively. The observed frequencies of the A1A1, A1A2 and A2A2 genotypes were found to be 53%, 39% and 8%, respectively, which met Hardy-Weinberg equilibrium. The frequencies of the 1 and 2 alleles of the Msp I mutation site in the intron 5 of DRD3 gene were found to be 62.5% and 37.5%, respectively. The observed frequencies of the 1-1, 1-2 and 2-2 genotypes were found to be 35%, 55% and 10%, respectively, which met Hardy-Weinberg equilibrium. No linkage disequilibrium was observed between the Bal I and Msp I polymorphism in Guangzhou Hans(chi(2)=0165, P>0.05). The frequencies of the T and C alleles of the T978C mutation site of DRD5 gene were found to be 51% and 49%, respectively. The observed frequencies of the T/T, T/C and C/C genotypes were found to be 23.6%, 54.6% and 21.8%, respectively, which met Hardy-Weinberg equilibrium. CONCLUSION: The polymorphisms of DRD2, DRD3, DRD5 gene in Guangzhou Hans were high and different from those in other populations.

[The application of meshed acellular dermis xenograft (pigskin) with split thickness skin autograft in 23 patients].

OBJECTIVE: An effective and inexpensive coverage, which can prevent scar formation and fill up scar tissue depression for full thickness skin burns in 23 patients was presented. METHODS: A deepithelized and acellular basement membranes of dermis from freshly harvested pigskin was obtained as meshed acellular dermis xenograft. The patients were treated in one or two stages. Two stages, 7 day postoperative the meshed acellular xenograft, a split thickness skin autograft was applied. One stage, the meshed acellular xenograft and split thickness skin autograft were applied at same time. RESULTS: The take rate of two staged treatment was (87.0 +/- 25.4)% and one stage was (60.3 +/- 3.4)%. All grafts showed in pink or near normal skin color and felt soft and smooth. CONCLUSIONS: The combination of meshed acellular dermis xenograft and split thickness skin autograft demonstrated an ideal way for the coverage of full thickness skin burn, which is effective and inexpensive.

[Expression of matrix metalloproteinases and tissue inhibitor of metalloproteinases in colorectal neoplasm].

OBJECTIVES: To explore the relationship of matrix metalloproteinases (MMPs) and tissue inhibitor of metalloproteinases (TIMPs) to the oncogenesis and development of colorectal neoplasm. METHODS: RT-PCR was used to assay the level of MMP-2, MT1-MMP, MMP-7, TIMP-2, TIMP-3 mRNA in 28 cases of colorectal cancer, including tumor tissue and surrounding normal tissue. RESULTS: MMP-2, MT1-MMP, TIMP-2 and TIMP-3 were over-expressed in tumor and normal tissues, and MMP-7 was strong-expressed in tumor tissue but was weak-expressed only in one case of normal tissue. The expression of MMP-7 in tumor tissue was correlated to Dukes stage (P < 0.01). The expression of TIMP-2 in tumor tissue with positive-node was much higher than that with negative-node (P < 0.01). The expression of TIMP-3 was decreased with the Dukes stage and the depth of invasion (P < 0.01). There were no correlations between MMPs and TIMPs (P > 0.1). CONCLUSIONS: The expression of MMP-7 mRNA has a high specificity in colorectal cancer. MMP-7 may become a sensitive tumor marker. Inducing TIMP-2, TIMP-3 or suppressing MMP-2, MT1-MMP, MMP-7 mRNA's expressions may inhibit the invasion and metastasis of cancer.

[Lymph node metastasis and the extent of lymph node dissection for gastric cancer: report of 326 cases].

OBJECTIVE: To investigate the pattern of lymph node metastasis and the extent of lymph node dissection for gastric cancer. METHODS: 326 patients with gastric cancer admitted from 1990 to 1999 were analyzed retrospectively after D(2), D(3) or D(3) plus para-aortic lymphadenectomy (D(3) + PAL). RESULTS: The total incidence of lymph node metastasis was 69.9%; node involvement was 15.4% and 77.4% respectively for early gastric cancer and advanced gastric cancer. Depth of invasion, tumor size and histology affected lymph node metastasis significantly (P < 0.05). For T(1) patients, node involvement was mainly confined to N(1) and one patient had N(2) metastasis; 8.1% of T(2) patients and 28.7% of T(3), T(4) patients had N(3), M(1) lymph node metastasis. Among 107 patients who received D(3) + PAL, 16a2b1 lymph node metastasis was found in 15.0%. The patients who had 16a2b1 node involvement were all suffered from advanced gastric cancer and N(1)-N(2) node metastasis. In the patients with serosal invasion-positive tumors or tumors size over 5 cm, the incidence of 16a2b1 metastasis was high, and that of entire stomach cancer was up to 38.5%. The 3-year and 5-year survival rates for D(3) + PAL group were 60.7% and 50.0% respectively. After D(3) + PAL, the 1-and 2-year survival rate, of patients with 16a2b1 metastasis were 60.0%, 50.0% respectively. CONCLUSIONS: D(2) lymphadenectomy should be taken for patients with early gastric cancer and D(3) lymphadenectomy for those with relatively early' advanced gastric cancer. For advanced gastric cancer with suspect or confirmed N(1)-N(2) node metastasis, tumor size over 5 cm and/or serosa invasion, D(3) plus para-aortic lymphadenectomy appears to be a necessary surgical procedure.

[A study on correlation between the structural parameter of optic disc and visual field in primary open-angle glaucoma].

OBJECTIVE: To study the consistency between the optic disc neural rim depth (NRD) and visual field defect in patients with primary open angle glaucoma (POAG). METHODS: A system of computerized image analysis was used to measure the relative NRDs of temporal-superior and temporal-inferior area of the optic disc in patients with primary open angle glaucoma. The consistency between the area with relatively larger NRD and the location of severer visual field defect was investigated. RESULTS: In cases with early and middle stage POAG, the value of NRD corresponding to the severer location of the visual field defect was significantly larger than that corresponding to the location without or with mild such defect. Among 54 eyes, there were 46 eyes whose location with larger NRD value was consistent with the position of severer visual field defect, the rate of consistency being 85.2%. CONCLUSION: In cases with early and middle stage POAG, the increase of NRD value at temporal-superior and temporal-inferior area is asymmetrical, and the consistency between the area with larger NRD value and the corresponding position of severer visual field defect is relatively good. NRD is helpful to the early diagnosis of POAG.

[Stress-corrosion test of laser welded ceramic alloys].

OBJECTIVE: The corrosion properties of dental material are very critical. This study is to investigate the corrosion properties of laser welded basemetalalloys. METHODS: The precise stainless rod molds had been made in size of phi 3 mm x 4 mm, with which 20 wax patterns were made. They were invested with inner investment (Bellavest) and outer investment (phosphate and gypsum). Then they were casted with CW-PA (Ni-Cr-Nb) alloys. All casted alloy rods were ultra-sonic washed in distilled water. Every two rods were contacted longitudinally by a special welding jig. Laser welding machine (DL-2002) was used to weld them at 305 V, 20 ms. The specimens were divided into 2 groups as following: 1. The group of stress-corrosion: 5 laser welded samples were subjected to the stress-corrosion test under 261.44 MPa in the 37 degrees C artificial saliva for 3 months in a special stress-corrosion test jig. After that, the ultimate tensile strength (UTS) was tested, the surface microcrack at the fusion zone (FZ), the heat affecting zone (HAZ) and the interface zone was observed with SEM (ARMY-1845, USA), the elements were also analyzed with EPMA (ARMY-1845, USA) at the fusion zone (FZ), the parent metal zone and the interface zone. 2. The other 5 laser welded samples were made as the controlled group. No special treatment were done to them, and the UTS was also tested, the surface of the micro crack and the elements at the FZ, the HAZ, parent metal zone and the interface zone were observed and analyzed with SEM, EPMA as well. RESULTS: The study showed us that the UTS of the stress-corrosion group was 502.48 +/- 12.49 MPa, while that of the controlled group was 446.05 +/- 82.50 MPa, there was no significant difference (P > 0.05). There was no microcrack on any sample's surface at the FZ, the HAZ and the interface zone. The main elements of the FZ were Ni(56.63%), Cr(19.36%), Nb (6.57%), Si(6.01%), Al(0.32%) and Mo(11.11%), while the elements of the parent metal zone were Ni(60.77%), Cr (20.59%), Nb(5.03%), Si(3.95%), Al(0.37%) and Mo(9.29%) respectively. CONCLUSION: The results suggest that the corrosion properties of laser welded alloys are excellent.

[Transgenic mice model of human hepatitis B virus x gene].

OBJECTIVE: To set up a human hepatitis B virus x (HBx) gene transgenic model. METHODS: The model was set up by microinjection, the integration and expression of the founders were identified by molecular hybridization. RESULTS: A 1,800 bp DNA fragment containing the human HBx promoter and genomic DNA was microinjected into the male pronucleus of the fertilized eggs of mice. Three hundred and thirty-eight eggs survived from microinjection were transplanted into the oviduct of 16 pseudopregnant female, 12 pseudopregnant mice got pregnant and gave birth to 74 pups. Genomic DNA were extracted from the tails of the pups, Southern blot hybridization was used for determination of the integration of human HBx gene. The 1,800 bp microinjected fragment was used as the probe, 17 mice (founders) were identified of the integration of human HBx gene. Total RNA were extracted from the livers of the founders, all 17 founders were identified of the expression of human HBx gene by using Northern blot hybridization. CONCLUSIONS: These results may provide animal models for studying on the correlation between chronic HBV infection and the development of hepatocellular carcinoma.

[Investigation of deciduous teeth caries in children under 14-year-old].

OBJECTIVE: To investigate the caries prevalence of children under 14-year-old in three districts (Nanshi,Luwan,Xuhui) of Shanghai. To provide the first-hand data for prevention of children caries. METHODS: The standard of second whole national investigation regime of oral epidemic disease was used. RESULTS: When the children grow up,the caries incidence and dft increased. In 7-year-old group the values were 83.33% and 3.78,respectively. After that the values decreased for the alternation of deciduous teeth and permanent teeth. The caries incidence was almost the same in boys and girls. The caries incidence in lower jaw was higher than that in upper jaw. The caries incidence was highest in lower jaw D and E. The lowest caries incidence was found in lower jaw A and B. The incidence and dft decreased compared with before. At the same time, the filling rate of deciduous teeth also decreased. CONCLUSION: The oral hygiene knowledge for children is disseminated. It is important for children to find a solution to improve the filling rate of deciduous caries.
