Studies into hormonal imprinting in intact and transformed 3T3 fibroblast cell lines.

The cells of the NIH 3T3 fibroblast line responded to primary interaction with insulin by a positive imprinting, i.e. by an increased binding capacity for the hormone on re-exposure. Positive imprinting, although to a lesser degree, was also induced by thyrotropin. However, oncogenic transformation by polyoma virus oncogens resulted in decreased imprinting in both the middle-T-antigen (MT3) and small-T-antigen-expressing (N4) cells.

Increased production and/or secretion of pulmonary surfactant in rats by long term sulfur dioxide exposure.

Influence of long term SO2 exposure on the pulmonary surfactant in rats was studied by means of chemical analysis and microscopic verification. At a time after termination of the exposure period, the general symptom in rats was similar to that of bronchitis. The content of disaturated phosphatidylcholine, a main functional component of the pulmonary surfactant, significantly increased not only in broncho-alveolar lavage fluid but also in pulmonary microsomal fraction by long term SO2 exposure. Microscopic verification of alveolar type II cells from the bronchitic rats demonstrated the development of rough surface endoplasmic reticulums and an increase of the number of osmiophillic bodies. The results suggest that pulmonary surfactant production and/or secretion are activated in rats with bronchitis caused by long term SO2 exposure.

Protective effect of surface-active phospholipids against the acid-inducing inhibition of the tracheal mucociliary transport.

Little is known about the physiological role of surface active phospholipids (SAP) in the central respiratory tract. In the present study, the effect of SAP on acid-inducing inhibition of particle transport by the tracheal mucociliary function was investigated in unanesthetized pigeons. SAP and acids were directly nebulized to the surface of the trachea. The composition of SAP was based on that of the pulmonary surfactant. The experiment was carried out under application of acetylcholine providing a constant mucociliary transport. SAP significantly diminished HCl- and H2SO4-inducing inhibitions of the mucociliary transport. Benzalkonium chloride also reduced the inhibition. The results suggest that SAP may be a protecting factor of the mucociliary clearance and that SAP may be connected with hydrophobicity not only of the alveolar surface but of the tracheal epithelium.

[Chemotherapy with Cisplatin-Phosphatidyl-choline-Lipiodol (CPL) suspension in unresectable hepatocellular carcinoma].

Twenty-seven patients with unresectable hepatocellular carcinoma (HCC) were treated with Cisplatin-Phosphatidyl-choline-Lipiodol (CPL) suspension. PR was obtained in two of ten cases (20%) by one shot therapy. AFP decreased in 9 of 10 patients by one shot therapy with a 62.1% rate of decrease. In all of 13 patients by TAE, AFP decreased and the rate of decrease was 64.8%. The concentration of CDDP in the peripheral venous blood was lower and continued longer than that of CDDP on the market. Nausea, vomiting and fever were noted in most cases as adverse effects, but they were slight. These results suggest that CPL agents were very chemotherapeutic for unresectable HCC.

Polymorphic DNA haplotypes at the phenylalanine hydroxylase (PAH) locus in Asian families with phenylketonuria (PKU).

DNA polymorphisms at the phenylalanine hydroxylase (PAH) locus have proved highly effective in linkage diagnosis of phenylketonuria (PKU) in Caucasian families. More than 10 RFLP sites have been reported within the PAH structural locus in Caucasians. With information from affected and unaffected offspring in PKU families it is often possible to reconstruct complete RFLP haplotypes in parents and to use these haplotypes to follow the segregation of PKU within families and to determine the distribution of PKU chromosomes within populations. To establish the utility of these RFLPs in characterizing Asian families with PKU, we typed eight DNA sites in 21 Chinese families and 12 Japanese families with classical PKU. The eight RFLPs were chosen for their informativeness in Caucasians. From these families we reconstructed a total of 91 complete PAH haplotypes, 44 from non-PKU chromosomes and 47 from PKU-bearing chromosomes. Although all eight marker sites are polymorphic in both Chinese and Japanese, there is much less haplotypic variation in Asians than in Caucasians. In particular, one haplotype alone, haplotype 4, accounts for more than 77% of non-PKU chromosomes and for more than 80% of PKU-bearing chromosomes. Haplotype 4 is also relatively common in Caucasians. The next most common Asian haplotype is 10 times less frequent than haplotype 4. By contrast, in many Caucasian populations the sum of the frequencies of the five most common haplotypes is still less than 80%, and several of the most common haplotypes are equally frequent. Even though the extent of haplotypic variation in Asians is severely limited, the few haplotypes that are found often differ at a number of RFLP sites.(ABSTRACT TRUNCATED AT 250 WORDS)

[Piperidine in the brain: its neurobiological significance].

Piperidine (Pip) is a normal constituent in mammalian brain, affects synaptic mechanism in the CNS, and influences neural mechanisms governing regulation of emotional behavior and extrapyramidal function. In addition, there are enzyme systems within the brain that synthesize and metabolize Pip, and uptake and storage mechanisms for Pip are found in the nerve endings. Pip is highly concentrated in the pituitary and pineal glands, hippocampus and caudate nucleus among the regions of the brain. Levels of Pip in the brain show physiological variations associated with environmental changes. The levels increase significantly under deep anesthesia. The study on the time relations of the change in brain levels of Pip and the anesthetic activity demonstrates that the level increases prior to the loss of the righting reflex and that the elevated level declines prior to the reappearance of the reflex. Furthermore, Pip levels in the lower brainstem reticular formation show sleep-related changes during REM sleep deprivation and REM sleep rebound that followed. Direct administration of Pip into the hippocampus and amygdala of cats with chronically implanted electrodes and a cannula caused resting and calmness in small doses, and seizure discharge accompanied by hyperemotionality in large doses. Administration into the pontine reticular formation induced REM and NREM sleep. Iontophoretic application produced the excitation and inhibition of single neuron activities in the cerebral cortex, hippocampus, caudate nucleus, cerebellum, and pituitary in anesthetized rats. With no anesthesia, Pip caused the inhibitory action in a higher percentage of the neurons studied, compared with the result obtained under anesthesia. Pip-induced excitation and inhibiton were blocked by tetramethylammonium but little affected by scopolamine. The kinetic study of Pip-induced Cl- current in internally perfused neurons of Aplysia, by using the 'concentration camp' and voltage clamp techniques, revealed that Pip acted on at least two components of nicotinic receptor-Cl- channel complex, and further that Pip could discriminate between the transient and the persistent components of ACh-induced Cl- current. These findings suggest that Pip may have close connections with neuroendocrine as well as neuronal functions, and further, with the mechanisms underlying sleep-consciousness and emotional function. Because of piperidine's multiplex pharmacological activities, the study of piperidine may provide a clue to the discovery of new active drugs and to the elucidation of causes of pathological states relating to the brain function.

Molecular genetics of phenylketonuria in Orientals: linkage disequilibrium between a termination mutation and haplotype 4 of the phenylalanine hydroxylase gene.

Phenylketonuria (PKU) is a common metabolic disorder among Chinese, with a prevalence of about 1 in 16,500 births. This frequency is very similar to that among Caucasians. Individual exons of the phenylalanine hydroxylase (PAH) gene with flanking introns were amplified by polymerase chain reaction and cloned into M13 for sequence analysis. An Arg111-to-Ter111 mutation has been identified in exon 3 of the PAH gene in a Chinese PKU patient. The mutation is in linkage disequilibrium with the mutant haplotype 4 alleles which are the most prevalent haplotype among the Orientals. The mutation accounts for about 10% of the Chinese PKU alleles and is absent from the Caucasians, demonstrating that independent mutational events have occurred in the PAH locus after racial divergence.

Bradykinin-induced generation of inositol 1,4,5-trisphosphate in fibroblasts and neuroblastoma cells: effect of pertussis toxin, extracellular calcium, and down-regulation of protein kinase C.

The net content of inositol 1,4,5-trisphosphate [Ins(1,4,5)P3] was measured in bradykinin (BK)-stimulated NIH3T3 fibroblasts and neuroblastoma-glioma hybrid cells (NG108-15). BK-mediated production of Ins(1,4,5)P3 was not affected by replacing the medium with Ca2+-free medium, but addition of EGTA (1mM) to Ca2+-free medium markedly prevented production of Ins(1,4,5)P3. Although pertussis toxin (PT) treatment caused ADP-ribosylation in both NIH3T3 cells and NG108-15 cells, the BK-induced Ins(1,4,5)P3 formation was considerably reduced in the former cells but not in the latter cells, suggesting that PT-sensitive and PT-insensitive GTP-binding proteins are involved in phosphoinositide phospholipase C (PI-PLC) activation in fibroblasts and neuroblastoma cells, respectively. In NG108-15 cells down-regulated in protein kinase C (PKC) by long-term exposure to phorbol 12-myristate 13-acetate (PMA), BK-stimulated Ins(1,4,5)P3 accumulation was significantly enhanced compared to control cells.

Pyloric stenosis in a patient with progressive systemic sclerosis.

A 64-year-old Japanese woman with progressive systemic sclerosis (PSS) who developed severe pyloric stenosis is described. The conservative treatments brought only the temporary symptomatic relief, and pyloroplasty became necessary. No ulcerative lesions or tumors were found in the resected stomach or duodenum specimens implicated for stenosis. The histological examinations revealed edema and replacement fibrosis in the pyloric ring. The possible mechanisms of pyloric stenosis are discussed.

The effect of metoclopramide on the absorption and pharmacology of chlorpromazine in the rat.

The mechanism of interaction between metoclopramide (MCP) and chlorpromazine (CPZ) has been examined in rats. MCP given intraperitoneally 30 min before orally administered CPZ significantly enhanced the cataleptic and hypothermic effects of CPZ, and also initially increased its plasma and brain concentrations. However, MCP had no effect on the plasma and brain concentrations of CPZ given as an intravenous bolus, indicating that MCP interacts with CPZ during its intestinal absorption. Furthermore, co-administration of MCP (i.p.) with CPZ (p.o.) markedly accelerated gastric emptying compared with CPZ alone, and MCP (i.v.) did not alter the uptake of CPZ by the intestinal membrane. Therefore, it is concluded that MCP causes an increase in the rate of CPZ absorption, by accelerating the gastric emptying.
