Immunocytochemical characterization of S-100 beta-positive human T-lymphocytes by a double immunostaining method.

The antigenic properties of S-100 beta-positive human T-lymphocytes (S-100 beta+ T-cells) were investigated by a double immunostaining technique employing an indirect immunoperoxidase method for cytoplasmic S-100 beta subunit and an immunoalkaline phosphatase method for cell surface antigens detected by various monoclonal antibodies to human lymphocytes. S-100 beta+ T-cells recognized by their diffuse intracytoplasmic immunoperoxidase reaction, also expressed CD2, CD3, CD8 antigens demonstrated by surface blue alkaline phosphatase reactivity, but not CD4, CD1, CD25 (interleukin-2 receptor), or HLA-DR antigens. However, they displayed a blastic change to T-cell mitogens, such as Concanavalin A(Con-A) and PHA, followed by the expression of CD25 and HLA-DR antigens. Under normal conditions, S-100 beta+ T-cells comprised approximately 5-22.8% of CD8+ cells amongst human peripheral blood mononuclear cells.

Long-term-cultured mouse B-lymphocyte line. I. Establishment and characterization of mouse B-lymphocyte line.

Mouse B-cell line was established by culturing anti-Thy-1 antibody and complement-treated splenic B cells with the conditioned medium of concanavalin A-stimulated spleen cell-culture supernatant. At the eighth week of culture, it was revealed that 100% of the long-term-cultured cells had both cytoplasmic and surface immunoglobulin. These cells were then maintained in the conditioned medium together with T-cell-depleted splenic and then splenic adherent feeder cells. Flow cytometric studies of the B-cell line showed that they had surface mu, delta, and kappa but no gamma, lambda, Lyt-1, or Lyt-2. The growth of the B-cell line was dependent on the factor(s) derived from concanavalin A-stimulated conditioned medium. It was found that IL-2 was the major factor supporting the B-cell growth. The B-cell line did not secrete immunoglobulin spontaneously, but it could differentiate into antibody-forming cells through the stimulation of bacterial lipopolysaccharides. The technique for obtaining mouse B-cell lines are reproducible in our laboratory and one of those lines has been propagated and maintained for 16 months to the present.

SCE levels in Bloom-syndrome cells at very low bromodeoxyuridine (BrdU) concentrations: monoclonal anti-BrdU antibody.

A stable staining procedure of sister-chromatid differentiation (SCD) using a monoclonal antibromodeoxyuridine (BrdU) antibody was newly established by combining it with the immunoperoxidase reaction (3,3'-diaminobenzidine, DAB reaction). This procedure permitted detection of SCD and SCE at very low BrdU concentrations. SCD was not usually observed below 2.0 micrograms/ml BrdU with flame-dried chromosome slides. When chromosome slides were prepared by air-drying over 37 degrees C warm water, SCD was detected at 10.0, 5.0, 1.0, 0.5, 0.3 and 0.2 micrograms/ml BrdU with FPG and even at 0.1 microgram/ml BrdU with the antibody technique. SCE levels were evaluated using the antibody technique and endomitotic analysis with FPG at low BrdU concentrations (1.0, 0.5, 0.3, 0.2 microgram/ml) in two BS B-lymphoblastoid cell lines (LCLs). Even though the BS SCE level was approximately 70 per cell at 10 micrograms/ml, the value decreased to the level of 20-30 SCE per cell at 0.1 microgram/ml with the antibody technique. In BrdU-labelled BS endomitoses, single SCEs highly decreased with BrdU concentrations (130-140 level at 10 micrograms/ml: 38-60 level at 0.2 microgram/ml), when compared to the rare twin SCE values (3-6 SCE level) at all BrdU concentrations. These findings conclusively indicate that the spontaneous baseline SCE in BS B-lymphoblastoid cells is low and most BS SCEs are caused by BrdU.

Electron microscopic study of mucoepidermoid carcinoma in the pancreas.

An autopsy case of extremely rare mucoepidermoid carcinoma of the pancreas in a 58-year-old male was reported. The main tumor in the pancreatic tail associated with wide-spreading metastases, was histologically composed of squamous cancer cell nests intermingled with mucin-containing cells, but not true glandular structures except for metastatic foci in the liver. Electron microscopic findings of the main tumor revealed roughly three kinds of cancer cells, namely undifferentiated cells, squamous cells, and squamous cells with mucin-containing intracytoplasmic lumina, accompanied by variety of transitional forms. The mucin was similar to that of the intra-or interlobular duct epithelium of pancreas in mucin stains. Immunohistochemically, positive immune reaction of the cancer cells was observed by anti-keratin, -epithelial membrane antigen and -carcinoembryonic antigen sera. These findings suggested that the cancer cells originated from undifferentiated cells of pancreatic duct, which showed multipotency to differentiate predominantly into squamous cells, but also into mucin-producing cells.

Malignant Brenner tumor with peritoneal metastasis.

A case of malignant Brenner tumor with peritoneal metastasis in a 67-year-old woman was reported. The multilocular cystic tumor of right ovary was 420 g in weight, and their cystic walls were covered with multilayered tumor cells showing papillary pattern very frequently. The tumor was histologically transitional cell carcinoma with occasional glandular structures but no squamous differentiation corresponding to grade 2 or 3 urinary bladder carcinoma. The pattern of benign Brenner tumor was not identified, but there was some area of proliferating Brenner tumor. Immunohistochemically, carcinoembryonic antigen was detected in several tumor cells, especially in the intercellular spaces among them, and cytokeratin was detected only in some tumor cells. Ultrastructurally, the malignant Brenner tumor shared many common features with the benign one and also bladder tumor. Intercellular spaces with microvilli were frequently found and thought to be important for diagnosis. The morphologic criteria of this rare tumor are discussed.

Multiple carcinoid tumor combined with mucosal carcinoma in the stomach. A case report.

A case of gastric multiple carcinoid tumor combined with mucosal carcinoma in a 63-year-old female is reported. The carcinoid tumors, larger than 0.5 mm in diameter, and endocrine cell micronests, smaller than 0.5 mm in diameter, were mostly located in areas of chronic atrophic gastritis. Their distribution was coincident with that of multiple carcinoid tumor in type A gastritis. The majority of the tumor cells were positive for human chorionic gonadotropin (HCG) and Leu 7 by immunohistochemistry and contained various numbers of intracytoplasmic secretory granules as revealed by electron microscopy. Two mucosal carcinoma foci did not show endocrine characteristics. These carcinoid tumors were thought to have originated in HCG-producing argyrophil cells which had been stimulated by hypergastrinemia accompanying chronic atrophic gastritis. The mucosal carcinomas were regarded as incidental.

Ultrastructural and immunological studies of virus particles observed in a T-cell line derived from a mouse spontaneous lymphoma.

Intracytoplasmic type A virus particles in clusters, associated with budding, immature type B and mature type B-like particles were observed in a T-cell line derived from a spontaneous lymphoma of an A/J mouse. These particles shared the antigenic determinants with murine mammary tumor virus from RIII, C3H, and GR strains as revealed by both indirect immunofluorescence and immunoperoxidase methods at ultrastructural level.

Combined hepatocellular and cholangiocarcinoma arising in a cirrhotic liver. Report of an autopsy case.

A rare case of combined hepatocellular and cholangiocarcinoma arising in a 56-year-old female is reported. The autopsy disclosed the presence of two different kinds of tumors in the right lobe of the liver, which showed advanced cirrhosis; a massive rubbery, ill-defined and whitish-yellow cholangiocarcinoma and a nodular soft, encapsulated and dark green hepatocellular carcinoma. They were adjacent to each other, but showed no intermingling. Only the massive cholangiocarcinoma had invaded the portal vein and showed several intrahepatic metastatic foci and hepatic, pancreaticoduodenal and perigastric lymph node metastases. Immunohistochemically, carbohydrate antigen 19-9 was strongly positive only for the cholangiocarcinoma component, explaining the high titer of this antigen in the serum on admission. On the basis of these findings, the possible morphogenesis of the tumor observed in the cirrhotic liver is discussed.

Craniopharyngioma in the third ventricle: necropsy findings and histogenesis.

A case of craniopharyngioma confined within the third ventricle with necropsy is reported. A stalk-like structure in this tumour was present in the wall of the third ventricle at its base. It is suggested that this tumour might have arisen from the remnants of Rathke's pouch persisting in the tuber cinereum.

[An autopsy case of triple primary cancers consisting of glioblastoma multiforme of the pons, colon cancer and rectal carcinoid--a statistical analysis of cases of brain tumor combined with other primary cancers in Japan autopsy annuals].

An autopsy case of glioblastoma multiforms of the pons with a colon cancer, a rectal carcinoid, a renal adenoma and three gastric leiomyomas in a 81-year-old-woman is reported with a statistical analysis on multiple primary cancers associated with primary brain tumors as reported in the Japan autopsy annuals. Out of 329, 705 autopsy cases from 1975 to 1984 in the Japan autopsy registry, double cancers and triple cancers that included a primary brain tumor amounted to 123 cases (0.037%) and 12 cases (0.0036%), respectively. Other sites for primary cancers were the thyroid (23%), the stomach (15%), the lungs (12%), and the colon (10%) in that order of frequency.

Mother-to-offspring transmission of human T cell leukemia virus type I in rabbits.

We studied the mode of natural transmission of human T-cell leukemia virus type I (HTLV-I) in rabbits. Four virus-infected rabbits (2 males and 2 females) were individually mated with 4 noninfected rabbits. Two virus-infected females mated with noninfected males gave birth to 7 offspring, and 2 noninfected females mated with infected males delivered 5 offspring. Four of the seven offspring born to the virus-infected mothers seroconverted for HTLV-I when aged 6 to 13 weeks with antibody titers of 1:40 to 1:160. None of the five offspring born to the noninfected mothers became seropositive during the observation period of 6 months, however. Peripheral lymphocytes were cultured with T cell growth factor, and HTLV-I-carrying lymphoid cell lines were established from the four seroconverted rabbits. All four cell lines were of T cells positive for Ia antigens. In addition, none of five newborn rabbits killed immediately after birth to a virus-infected rabbit was infected with HTLV-I. These findings provide an experimental support for the milkborne transmission of HTLV-I from mother to child in humans and indicate that the virus is tropic for T cells in rabbits as well.

Establishment of a new human B cell line carrying t(11;14) chromosome abnormality.

A new human cell line, SP-49, was established from the peripheral blood of a patient with leukemic conversion of diffuse lymphoma, medium-sized cell type. SP-49 cells were shown to have intracytoplasmic and surface immunoglobulin and Leu-12 antigen, but were negative for Epstein-Barr virus nuclear antigen. Chromosome analysis demonstrated that SP-49 had t(11;14) chromosome translocation involving 11q13 and 14q32, where oncogene bcl-1 and immunoglobulin heavy chain gene, respectively, were reportedly located.

S-100 beta positive human T lymphocytes: their characteristics and behavior under normal and pathologic conditions.

The characteristics of human S-100 beta-positive T lymphocytes (S-100 beta+ T cells) and their fluctuation in peripheral blood under normal and various pathologic conditions were investigated. S-100 beta+ T cells were small lymphocytes with no particular subcellular structures and showed a proliferative response to mitogens. They were present mainly in peripheral blood under normal conditions but accumulated in T zones of lymph nodes with nonspecific T-zone hyperplasia, where numerous interdigitating reticulum cells existed. In healthy adults approximately 1% to 4% (mean 3.4%) of peripheral blood mononuclear cells were S-100 beta+ T cells. The proportion of S-100 beta+ T cells in peripheral blood tended to significantly decrease (less than 0.5%) in patients with neoplastic diseases; this tendency was apparently related to tumor progression.

Characterization of a new human lymphoma cell line (RC-K8) with t(11;14) chromosome abnormality.

A human cell line (RC-K8) that produces plasminogen activator was established from the peritoneal effusion of a patient with histiocytic lymphoma. The RC-K8 cell line grew mainly in single cell suspension and consisted of primitive cells with pleomorphic morphology. RC-K8 cells were positive for alpha-naphthyl butyrate esterase, acid phosphatase and periodic acid-Schiff stainings. Immunologic and molecular biological studies showed that RC-K8 cells reacted with monoclonal antibodies to B cell antigens (B1 and Leu12) and Ia antigen (OKIa1) and possessed immunoglobulin gene rearrangement in the absence of surface and cytoplasmic immunoglobulin. Neither Epstein-Barr virus nuclear antigen nor terminal deoxynucleotidyl transferase was detected in the cells. Chromosome analysis of RC-K8 disclosed 46 XY with complex abnormalities including t(11;14)(q23;q32). Intraperitoneal inoculation of RC-K8 cells to immunosuppressed newborn hamsters produced massive metastatic tumors in the lungs. The RC-K8 cell line will be of considerable value for the study of lymphomagenesis associated with t(11;14) and pulmonary metastasis of lymphoma cells.

A virion-like particle (VLP) released from an Epstein-Barr virus-producing cell sub-line of P3HR-1: identification of a specific cellular DNA contained within VLPs.

A virion-like particle (VLP) was isolated from the culture fluid of P3HR-I cells. VLPs were separable from the enveloped Epstein-Barr virus (EBV) through rate-velocity sedimentation by virtue of their greater density. VLPs comprised electron-dense nucleoid material and the surrounding round or oval envelope which ranged from 100 to 150 nm in diameter. The polypeptide constitution of VLPs was very similar to that of enveloped EBV by SDS-polyacrylamide gel electrophoresis. VLPs contained reasonably specific cellular DNA (VLP DNA) with a sedimentation coefficient of 48S, which was cleaved into 5, 8, and at least 30 specific DNA fragments by digestion with BamHI, SalI, and EcoRI, respectively. Blot hybridization data suggested a limited heterogeneous genomic organization of VLP DNA and the presence of a VLP DNA-related molecule(s) in human cord lymphocytes.
