Using monoclonal antibodies and flow cytometry percentages and absolute number of lymphocyte populations and T cell subsets were enumerated in peripheral blood collected between 8:00 and 10:00 a.m. from 210 healthy adults. Although absolute numbers of total lymphocytes did not differ depending on age and sex, the numbers of T (Leu-4+) and natural killer (Leu-7+) cells as well as their percentages showed negative and positive correlations, respectively, with age. In Leu-7+ cells, the percentage was male-dominant irrespective of age, and the absolute number was male-dominant only in older subjects. Absolute numbers of suppressor/cytotoxic T (Leu-2a+) and helper/inducer T (Leu-3a+) cells and percentages of Leu-2a+ cells were negatively correlated with age in females. In males, only the percentage of Leu-2a+ cells was age-dependent. Leu-3a/Leu-2a was positively correlated with age in females, and was female-dominant depending on age. These results indicate that peripheral lymphocyte populations and T cell subsets vary remarkably in healthy adults even at a fixed time of day.
A metal ion-activated acid ATPase was present in chicken liver lysosomes. We used Zn2+ as an activator. Lysosomal extract containing octylglucoside from chicken liver was centrifuged at 100,000 xg for 60 min. The supernatant was analyzed by gel filtration on a Sepharose 6B column. Two peaks of metal ion-activated acid ATPase activities were obtained according to the distribution patterns. Each of the two active fractions was incubated with phosphatidylinositol-specific phospholipase C at 37 degrees C for 60 min. The resulting solution was analyzed by gel filtration on a smaller size column of Sepharose 6B again. Molecular weight of the major peak was altered from approx. 1,600,000 to 130,000, whereas that of the minor one, 700,000, remained unchanged.
An inbred strain of stroke-prone spontaneously hypertensive rat, the M-SHRSP, was established by brother-sister breeding of selected SHRSP for 24 generations while administering apresoline. Compared with SHRSP, the M-SHRSP shows an earlier rise in blood pressure, plasma renin activity (PRA) and cerebrospinal fluid (CSF) pressure, and somewhat changed cerebrovascular lesions. Crosses and back-crosses, using M-SHRSP, Wistar-Kyoto rats (WKY), spontaneously hypercholesterolaemic (SHC) rats and their hybrids produced colonies with various blood pressure levels and hypercholesterolaemia. Continued successive selective brother-sister breeding of M-SHRSP and SHC hybrids produced a colony with severe hypertension and marked hypercholesterolaemia. Streptozotocin diabetes was induced in an M-SHRSP and SHC hybrid (TC), from which diabetic TC was successively bred to the fifth generation. While each generation was hypertensive and showed a decrease in islet B-cells, symptoms of lasting glycosuria were first observed in the fourth generation among those given a high alpha-corn starch (75.7%) diet.

Parameters for blood concentrations of the peroral antibiotics talampicil in and bacampicillin, in NZW rabbits were determined using a two compartment model. On the basis of these pharmacokinetic parameters, a three compartment model was prepared for the concentrations of these drugs in the tongue, gingiva, submandibular gland, parotid gland, cervical lymph-node and mandibular bone. Simulation curves based on the parameters of tissue concentrations revealed visual conformity to the measured values.

Cerebrospinal fluid (CSF) involvement in myeloma is rarely seen. Recently we experienced a case with this lesion. A 70-year-old man developed consciousness level disorder during the course of bronchopneumonia. Neurological examination revealed stuporous consciousness, neck stiffness and Kernig's sign. Immunoelectrophoresis showed monoclonal IgG in serum. CSF which was obtained through lumbar puncture was clear and its pressure was 155 mm H2O. It contained 207 white cells/3 mm3; glucose, 54 mg/dl; and protein, 33 mg/dl. The differential count of the CSF was (in %) monocytes, 48.0; plasma cells, 25.5; neutrocytes, 15.5; and lymphocytes, 11.0. Cytoplasm and nucleus of the plasma cells were in various sizes. Some irregular multiple nuclei, flaming cells and grape cells were also observed in them. The cytoplasm of the plasma cells fluoresced with antisera against lambda chains IgG. The value of immunofluorescent technique in identifying plasma cells in the CSF is emphasized.

Passive injection of mice with preformed immune complexes (IC) made from cationized bovine serum albumin (BSA) and anti-native BSA antibody gave immune deposits along the glomerular capillary walls at predominantly subepithelial sites, while similar quantities of complexes made with native, anionized BSA did not deposit. Peripheral localization could be obtained also using low avidity antibody and a great excess of native BSA. Ultracentrifugation analysis showed that the size of IC in the animals given complexes containing cationized BSA was a little larger than 7 S, whereas those formed with the native or anionized BSA were around 19 S. The anti-native BSA antibody had a low avidity for cationized BSA in vitro, and thus all the IC which could deposit peripheral capillary walls were small and contained low avidity antibody. Chemical cationization of BSA alters the precipitability of the antibody and also the size and stability of the complexes formed. In an active model, injection of cationized BSA into mice preimmunized with cationized BSA caused localization of the BSA and its antibody in the peripheral capillary walls. Analysis of the circulating IC formed in this model also revealed low avidity of antibody and small-sized IC. From these results, it is clear that chemical cationization of antigen changes the characteristics of the antigen-antibody interaction, e.g. low precipitating efficiency and the formation of small-sized IC. Therefore, in addition to interaction of cationized IC with the polyanion layer of the glomerular basement membrane (GBM), the properties of antigen-antibody interaction play an important role in the deposition of IC along the peripheral capillary walls in a model of membranous glomerulonephritis.
