Gene expression of fibroblast growth factors in human gliomas and meningiomas: demonstration of cellular source of basic fibroblast growth factor mRNA and peptide in tumor tissues.

The growth autonomy of human tumor cells is considered due to the endogenous production of growth factors. Transcriptional expression of candidates for autocrine stimulatory factors such as basic fibroblast growth factor (FGF), acidic FGF, and transforming growth factor type beta were determined in human brain tumors. Basic FGF was expressed abundantly in 17 of 18 gliomas, 20 of 22 meninglomas, and 0 of 5 metastatic brain tumors. The level of mRNA expression of acidic FGF in gliomas was significant. In contrast, transforming growth factor type beta 1 was expressed in all the samples investigated. The mRNA for basic FGF and its peptide were localized in tumor cells in vivo by in situ hybridization and immunohistochemistry, showing that basic FGF is actually produced in tumor cells. Our results suggest that tumor-derived basic FGF is involved in the progression of gliomas and meningiomas in vivo, whereas acidic FGF is expressed in a tumor origin-specific manner, suggesting that acidic FGF works in tandem with basic FGF in glioma tumorigenesis.

Vascular abnormalities in congenital cutis laxa--report of two cases.

Two cases of Japanese girls with congenital cutis laxa associated with cardiovascular abnormalities are described. Case 1: A 12-year-old girl has been under our observation from the age of 6 months. Cardioangiogram revealed dilatation of the ascending aorta, meandering of the descending aorta and the coronary arteries, coiling of the carotid and innominate (brachiocephalic) arteries, and hypoplasia of the pulmonary arteries. Case 2: A 2.8/12-year-old girl died after our follow-up from the age of 3 months. The cause of death was congestive heart failure secondary to peripheral stenosis of the pulmonary arteries. In both cases, skin biopsy revealed a decreased number of elastic fibers and an increased amount of acidic mucopolysaccharides. The same histological features were observed in the pulmonary arteries and other arteries as well. Electron microscopic findings were diffuse thinning of elastic fibers and reduced elastic content. The high blood level of elastase (167.8 micrograms/l) in case 1 may cast a light on the unknown etiology of the disease.

[Changes in lipid peroxides content and antioxidant enzyme activities on airway surface in SO2-induced bronchitic rats].

The changes in lipid peroxides (LP) content and antioxidant enzyme activities were investigated on airway surface during the aggravating process of bronchitis induced by SO2 exposure in rats. LP content in broncho-alveolar lavage fluid (BALF) has gradually increased from 3 weeks after starting of SO2 exposure. Whereas, the activities of antioxidant enzymes, superoxide dismutase and glutathione peroxidase, increased at 1 week and then gradually reduced from 3 weeks. The ratio of LP to each antioxidant enzyme activity in BALF of the exposed rats was higher than that of normal rats. Morphological changes of the lung, a decrease of PaO2 and an increase of PaCO2 of blood depended on the increase of LP on airway surface. These findings indicate that LP may be involved in the development of bronchitis.

Dissociation between sympathetic purinergic response and ATP response in the mesenteric artery of the dog.

In the presence of prazosin and propranolol, electrical transmural stimulation of isolated dog mesenteric artery produced a sympathetic purinergic contraction, which was followed by a relaxation in PGF2 alpha-contracted arteries. Such purinergic responses were mimicked by brief exposure to alpha, beta-methylene ATP (alpha, beta-Me ATP) and were completely inhibited after desensitization of P2x-purinergic receptors. However, exogenous ATP predominantly evoked a relaxation in PGF2 alpha-contracted artery. These results suggest that the sympathetic purinergic response may be caused by a P2x-purinergic receptor-selective mechanism or substance, rather than ATP.

[Relationship between aging and renal function: a positron emission tomographic study].

Blood volume (V), blood flow (F) and glomerular filtration rate (GFR) per 100 g (100 ml) of kidney were calculated with C15O, H(2)15O and 68Ga-EDTA, to study age-related changes per unit tissue in 19 subjects using positron emission tomography. Age was negatively correlated with V, F and GFR. The aging-related decrease in V was less marked than those in F and GFR. Analysis by regression formula induced that F, GFR for subjects in their 70s are 76%, 46% and 55% for those in their 20s, respectively.

[Evaluation of renal blood volume by PET--the aorta-kidney equilibrium monitored with C15O].

Twenty subjects inhaled C15O and its dynamics were monitored to determine when equilibrium between the aorta and kidney occurred. One minute after inhalation, the increase in radioactivity in the aorta paralleled that in the kidney, but thereafter, both declined slowly. The ratio of aorta/kidney radioactivity become constant 1 min after inhalation and remained unchanged for 4 min. Thus renal blood volume can be measured 1 min after inhalation of C15O its distribution in the aorta and kidney has attained equilibrium.

[Preoperative adjuvant therapy for urinary bladder cancer--preoperative intra-arterial anticancer drugs infusion using artificial liver, and preoperative radiotherapy].

One hundred patients with carcinoma of the urinary bladder were treated with preoperative intra-arterial anticancer drug infusion in combination with an active carbon-based artificial liver or preoperative radiotherapy (400 rad X 5 days, total 2000 rad) in order to alleviate systemic side effects after preoperative adjuvant therapy and to perform radical cystectomy as soon as possible. Five-year survival was 69.8% in the artificial liver combined preoperative intra-arterial anticancer drug infusion group, and that of the no-pretreatment group and the preoperative irradiated group was 48.6% and 45.8%, respectively. In particular, the CDDP treated group had a higher 5-year survival of 77.4% than did the without CDDP treated group. This was attributable mostly to improved prognosis of T2, whereas either of these preoperative adjuvant therapies failed to ameliorate the prognosis of T3 or T4 stage patients. Therefore, systemic preoperative anticancer chemotherapies such as M-VAC are necessary to destroy "micrometastases" and to improve the prognosis of highly advanced bladder cancer.

The pattern design for evaluation of castability of nickel-chromium alloys.

To find a pattern design that would provide a higher degree of discrimination for castability of nickel-chromium alloys using a polyester sieve cloth pattern, four shaped patterns were chosen. The results as calculated by t-test indicated that patterns No. 2 and No. 3 can discriminate the castability of nickel-chromium alloys over almost the total range of the casting temperatures, but that pattern No. 3 is a better discriminator for the castability. Pattern No. 1 can only discriminate the castability at lower casting temperatures. Pattern No. 4 can only discriminate the castability at higher casting temperatures. The percent castability value calculated from the number of complete square segments and the areas of the four patterns are highly linear correlations, so it is reasonably accurate to calculate the castability only from counting the number of complete square segments. Beryllium-containing nickel-chromium alloy is more castable than non-beryllium-containing alloy and the higher casting temperatures appear to result in better castability.

A study of the method of making dental prosthetic appliances by sintered titanium alloys: effect of copper powder content on properties of sintered titanium alloy.

The effects of added copper powder to the properties of the sintered titanium alloys were investigated by measuring the compressive strength and densities of the green and sintered compacts, the thermal expansion curves and dimensional changes in the sintered compacts, and the accuracy of the crown-type restorations. The compressive strengths of green compacts ranged from 55 to 75 MPa. The expansion of green compacts increased with increased copper content. The sintered density was lower than the green density. The compressive yield strength of sintered compacts ranged from 260 MPa to 410 MPa. The sintered compacts expanded from 0.35% to 1.03% and the expansion increased with increased copper content. The dimensional accuracy of crown-type restorations showed the same dimensional change tendencies as did the sintered compacts. These results showed that the fit and the strength of sintered titanium alloy restorations could be improved.

[Corrosion of dental titanium alloys binary system of Ti-Al, Ti-Cu, Ti-Ni].

Anodic polarization, natural electrode potential, polarization resistance, tarnish, weight change and metal component dissolution in artificial saliva, of Ti-Al, Ti-Cu and Ti-Ni binary alloys were investigated to determine the effects of the added elements on the corrosion characteristics. In the case of Ti-5 at % Al to Ti-50 at % Al system alloys, increase in the aluminum content of the alloys raised the corrosion resistance. However pitting corrosion presumed at an aluminum content of more than 10 at %. In the case of the Ti-5 at % Cu to Ti-70 at % Cu system alloys, increase in the copper content of the alloys decreased the corrosion resistance and the decrease was marked at a copper content of more than 50 at %. In the case of Ti-5 at % Ni to Ti-70 at % Ni system alloys, 50 at % Ni was estimated to be the maximum content to keep corrosion resistance of the titanium alloys.

localization of type VI collagen in the lining cell layer of normal and rheumatoid synovium.

Types I to VI collagens were immunolocalized in normal and rheumatoid synovium using monospecific antibodies. Immunofluorescence studies showed type VI in the extracellular matrix of the lining cell layer, whereas positive staining for type III collagen was observed in both the lining and sublining cell layers. All other collagens could not be detected in the lining cell layer. Immunogold staining of the rheumatoid synovium localized type VI collagen to filamentous material, which was the major extracellular structure of the lining cell layer. Type III collagen was associated with thin cross-striated collagen fibrils. A brief treatment of rheumatoid synovial tissue with bacterial collagenase produced in the lining cell layer numerous broad-banded fibrils with 100-nm periodicity; these fibrils could be labeled with the antibody against type VI collagen. This suggests that type VI collagen filaments have the potential to form periodic structures under certain conditions. We further studied the susceptibility of type I to VI collagens to matrix metalloproteinase 1, 2 and 3 (collagenase, gelatinase of molecular weight 72,000, stromelysin), which are secreted by synovial lining cells in rheumatoid synovium, and found only type VI collagen to be completely resistant to all these metalloproteinases. These data indicate that type VI collagen, which has the ability to bind to cells and to interstitial collagens, plays an important role in supporting the synovial lining cells in the normal and rheumatoid synovium.

Lidocaine metabolism by human cytochrome P-450s purified from hepatic microsomes: comparison of those with rat hepatic cytochrome P-450s.

The metabolism of lidocaine by human hepatic microsomes and purified human cytochrome P-450s, P-450NF (P450IIIA4), P-450MP (a P450IIC form) and P-450PA (P450IA2) was examined and their metabolism was compared with that by rat hepatic cytochrome P-450s. Human hepatic microsomes produced monoethylglycinexylidide (MEGX) and 3-hydroxylidocaine (3-OH-LID) from lidocaine. In a reconstituted system with dilauroylphosphatidylcholine, P-450NF efficiently produced MEGX. P-450PA was not efficient in lidocaine N-deethylation (formation of MEGX) but produced 3-OH-LID. P-450NF and P-450MP did not produce 3-OH-LID. Lidocaine N-deethylation activity of P-450NF was enhanced in a modified reconstituted system with a phospholipid mixture and sodium cholate. P-450NF appears to be an ortholog to rat P450 PB-1 (P450IIIA2). Anti-P450 PB-1 antibody cross-reacted with P-450NF and efficiently inhibited lidocaine N-deethylation in human hepatic microsomes. The correlation of lidocaine N-deethylation activity with the concentration of P-450NF determined immunochemically with anti-P450 PB-1 antibody was good (r = 0.81). In addition, correlation between P-450NF content estimated with anti-P450 PB-1 and anti-P-450NF preparations was good (r = 0.96). These results suggest that rat P450 PB-1 and human P-450NF have closely related properties and P-450NF is the major enzyme involved in lidocaine N-deethylation in human hepatic microsomes.

[Pheochromocytoma combined with malignant lymphoma and prostatic cancer--a case report].

A rare case of pheochromocytoma associated with a malignant lymphoma and a prostatic cancer is reported. An 80-year-old male had had his terminal ileum resected one year earlier due to a malignant lymphoma. A year later, postoperative follow-up study by ultrasound revealed a solitary retroperitoneal tumor. The resected tumor was found to be a pheochromocytoma, which had provoked intraoperatively an intractable hypertension and ventricular arrhythmia. One year following this, a urinary disturbance was noted. On examination, a hard and irregular prostata was palpate and a subsequent biopsy revealed an adenocarcinoma. As far as we have been able to ascertain after a perusal of the Japanese literature, we believe that this case represents the first reported case of such a malignant lymphoma, combined with prostatic cancer and a pheochromocytoma.

[Effectiveness of fibrinogen to avoid serous fluid leakage through polytetrafluoroethylene (PTFE) tubular graft].

Serous fluid leakage is not infrequently an annoying complication of systemic-to-pulmonary artery shunt constructed with polytetrafluoroethylene (PTFE) tubular graft. Various causative factors have been postulated by many clinical and investigative studies, but still remain to be controversial. During the period from 1981 to 1989, 26 patients underwent PTFE shunt operations (16 modified Blalock-Taussig shunts and 10 central aortopulmonary shunts), among whom serous fluid leakage occurred in 10, an incidence of 38.5%. Possible roles of the operative methods, length and diameter of the graft, bodyweight, age and sex of the patients were examined but none of these demonstrated consistent correlation with this complication. The first 3 of the 10 patients in the series required prolonged period of drainage of the fluid. Meanwhile, unequivocally low serum values of fibrinogen in these patients presented a clue for the new therapeutic method. Intravenous administration of fibrinogen to the fourth patient results an almost instantaneous cessation of leakage and was applied to the subsequent 6 patients with excellent results. The duration of leakage was 2.7 +/- 2.6 days in the patients with fibrinogen treatment in contrast with 19.7 +/- 2.3 days in those without. In succeeding 4 patients, fibrinogen was used for the purpose of prevention instead of treatment, and the luminal surface of PTEF graft was coated with fibrinogen prior to the usage. Pretreatment of the graft was absolutely effective and did not allow leakage in any of the 4 patients.(ABSTRACT TRUNCATED AT 250 WORDS)

Metabolism of lidocaine by purified rat liver microsomal cytochrome P-450 isozymes.

The metabolism of lidocaine was studied using rat liver microsomes or a reconstituted lidocaine monooxygenase system with one of eight forms of cytochrome P-450 purified from liver microsomes from untreated- (P450 UT-2 and UT-5), phenobarbital- (P450 PB-1, PB-2, PB-4, and PB-5) or 3-methylcholanthrene- (P450 MC-1 and MC-5) treated rats. A reverse phase high-performance liquid chromatography system capable of simultaneously assaying four major lidocaine metabolites, namely, monoethylglycinexylidide (MEGX), 3-hydroxylidocaine (3-OH LID), methylhydroxylidocaine (Me-OH LID) and glycinexylidide (GX), was employed to determine the rate of formation of each metabolite. Untreated microsomes generated MEGX, Me-OH LID, and 3-OH LID, but the formation of GX was not detected. In male rat liver microsomes, MEGX was the major metabolite of lidocaine when a concentration of 1 mM was employed. The formation of MEGX and Me-OH LID was increased significantly (P less than 0.01) by microsomes from phenobarbital-treated rats, and the formation of 3-OH LID was increased with 3-methylcholanthrene. The study with the reconstituted system with purified cytochrome P-450 isozymes revealed that all eight forms of cytochrome P-450 used have an ability to N-deethylate lidocaine to form MEGX. Among these isozymes, cytochrome P450 PB-4 and P450 UT-2 showed a higher turnover number for the formation of MEGX. Me-OH LID was formed exclusively by P450 PB-5, and 3-OH LID exclusively by P450 MC-1. Selectivity of cytochrome P450 PB-5 for aromatic methyl hydroxylation of lidocaine was confirmed by an inhibition study; formation of Me-OH LID by microsomes of rats treated with phenobarbital was inhibited completely by antibody against P450 PB-5. It was concluded that different cytochrome P-450 isozymes metabolize lidocaine with a different rate and different position selectivities. Since a specific substrate of cytochrome P450 PB-5 (P-450e) is not known, lidocaine may be a useful substrate for the identification of P450 PB-5.

[Efficacy and adverse effects of beta-interferon in the treatment of malignant glioma].

Sixteen patients with malignant glioma were treated by the intravenous administration of beta-interferon together with chemoradiotherapy. Five of the cases were recurrent gliomas. Seven of the 11 fresh cases were treated with beta-interferon, more than two months after cessation of the radiotherapy. Of the 16 cases, 12 cases showed partial regression of tumors, or no regrowth of tumors on CT scan, 2 cases showed no improvement, and 2 cases were unevaluable due to the short follow-up periods. IFN-beta is often administered, in combination with antineoplastic agents and radiotherapy, to patients with malignant glioma. Some patients have shown sufficient suppression of the growth of the malignant glioma only through administration of IFN-beta 1 x 10(6) IU once or twice a week. Some patients, however, have developed severe bone marrow suppression due to the combination therapy of IFN-beta and antineoplastic agents. Therefore, the blood of patients should be tested twice a week, and the data should be analyzed within the same day to determine the subsequent treatment. IFN-beta administration should be stopped if the platelet count drops below 1.0 x 10(5)/mm3 or half of the initial figure, and the course of disease of the patient should be carefully observed.

Interleukin 5 enhances interleukin 2-mediated lymphokine-activated killer activity.

IL-5 expresses various biologic effects on several types of lymphocytes, including B cells, eosinophils, and T cells. We demonstrated that the incubation of resting splenocytes from C57BL/6 mice in murine rIL-5 enhances IL-2-mediated lymphokine-activated killer (LAK) activity against various tumor cells. IL-5 alone, however, does not induce killer activity. IL-2-mediated LAK activity increases in proportion to the dose of IL-5. During the late phase of the culture period, IL-5 seems to have some effect on the induction of IL-2-mediated LAK activity. We expect that IL-5 will prove useful for adoptive immunotherapy.
