Experimental bladder substitution using biodegradable graft of natural tissue.

A controlled experimental study has been performed using a biodegradable graft of natural tissue to achieve vesical augmentation. The graft is progressively reabsorbed while normal capacity is maintained by concomitant regeneration of the layers of the bladder remnant. No significant complications were encountered, and the graft was equally successful when applied either to a histologically normal bladder or to a pathologic bladder of intramural fibrosis. This material is potentially ideal for clinical bladder substitution in humans.

Uptake and utilization of vitamin B6 and its phosphate esters by Escherichia coli.

Escherichia coli KG980, a vitamin B6 auxotroph derived from E. coli K12, utilized the three unphosphorylated forms of vitamin B6, more or less effectively, for growth, but did not utilize the three phosphate forms at concentrations ranging from 10(-7) to 10(-5) M in the minimum medium of Davis and Mingioli. The bacterium, however, utilized the phosphate forms, although less effectively than the unphosphorylated forms, in the phosphate starving medium of Garen and Levinthal which is known to derepress alkaline phosphatase. Correspondingly, the phosphate forms of 3H-labeled vitamin B6 in the minimum medium were not taken up by the bacterial cells grown in the same medium, but were taken up when the phosphate starving medium was used for growth and uptake experiments; the unphosphorylated forms were taken up with either of the media used. After 30-min incubation of the cells grown in the phosphate starving medium with 3H-pyridoxine phosphate added to the same medium, the main extracellular 3H-vitamin B6 was found to be pyridoxine, evidently indicating an involvement of alkaline phosphatase action. It is concluded from these results that the phosphate forms of vitamin B6 can be taken up and utilized only after dephosphorylation but not taken up in their intact form. The initial rate of 3H-pyridoxal and 3H-pyridoxamine uptake in the minimum medium showed saturation kinetics. The Km and Vmax values were 1.2 X 10(-6) M and 62 pmoles/min/mg (dry cell weight) for pyridoxal; 11 X 10(-6) M and 65 pmoles/min/mg for pyridoxamine. 3H-Pyridoxine uptake apparently consisted of a high-affinity saturable component, whose Km value was tentatively estimated to be 2.2 X 10(-6) M, and an unsaturable component. The uptake rates of these three unphosphorylated vitamin B6 compounds compared at limiting concentrations for the growth of E. coli KG980 appear to be essentially in good agreement with the response pattern of this bacterium to the three compounds.

HLA antigens and susceptibility to atopic dermatitis.

The possible association between peculiar HLA antigens ant atopic dermatitis associated allergic rhinitis and/or bronchial asthma was investigated. (1) Atopic dermatitis (no deviation in HLA), complicating allergic rhinitis (HLA-BW40), and bronchial asthma (HLA-B12) are associated by different HLA antigens. (2) The diseases are associated through the same antigen (HLA-BW15), if the concept of disease resistant antigen is accepted. (3) No association is found between HLA antigens and serum IgE levels in patients.

HLA antigens and Ia antibody in the gastric cancer patient.

The possible association of 18 HLA specificities in HLA-A locus and 23 specificities in HLA-B locus with susceptibility to gastric cancer was investigated in a total of 87 patients and 355 controls in Japan. Significantly decreased antigen, HLA-B5 (17.2%) was found in gastric cancer patients compared with 40.0% in controls. X2 for this antigen difference is 15.8 (corrected P less than 0.005). The most frequent HLA haplotype was A9-B5 in controls and A2-Blank B locus antigen in gastric cancer patients. No reaction was found of HLA antiserum, Bou-Pri with cells of gastric cancer patients, compared with 29.4% in control subjects. No linkage was observed between HLA-A and -B locus antigens and gastric cancer in family study with double the frequency of HLA haplotype A2-BW40. Ia-like B cell specific antibody was found 10% in 2 out of 20 gastric cancer patients in a preliminary study. There was no clear association between B cell specific antibody and clinical implications.

HLA antigens and susceptibility to psoriasis vulgaris in a non-Caucasian population.

Fifty-four unrelated Japanese patients with psoriasis vulgaris were tissue typed using the Sixth International Histocompatibility Workshop antisera. Two control groups were included in this study: Thirty-one pustulosis palmaris et plantaris and 17 seborrheic dermatitis as a disease control and 66 normal, healthy, unrelated Japanses as a reference. HLA-Al (P = 0.0065) from the A locus and HLA-BW37 (P = 0.0164) from the B locus were found to occur with increased frequency in patients with psoriasis vulgaris. No significant difference in antigen frequencies in pustulosis palmaris et plantaris was found, however, HLA-AW30 and/or AW31 and HLA-B12 occurred with increased frequency in seborrheic dermatitis. No linkage between psoriasis and HLA was observed in eight families. Therefore our findings in Japanese do not confirm the previous observation made in Caucasians of an association between psoriasis vulgaris and HLA-B13 or BW17.

Is hollow fibre occlusion due to maldistribution of blood?

Blood flow has been shown to be very slow in the central portion of a capillary bundle where intracapillary clotting mainly occurs. It is concluded that extremely uneven blood distribution is a causative factor in intracapillary clot formation in the HFAK examined.
