Breeding of cynomolgus monkeys through successive generations by indoor cage system.

Vital statistics on the breeding through successive generations were presented for the cynomolgus monkey colony of NIH, Tokyo. The results of this retrospective survey clearly demonstrated the third (F2) and the fourth (F3) generations could be bred and reared successfully by the indoor caged-breeding system in which either individual timed-mating or group mating procedure was adopted. Several important and difficult problems involved in the production of successive generations of the cynomolgus monkey by our breeding system were discussed from the standpoint of laboratory animal science.

Effects of cytochalasin B and dibutyryl cyclic AMP on intraleukocytic bactericidal activity.

Cytochalasin B (CB) is known to regulate the movement of intracellular microfilaments system. In this experiment, the effect of CB on the intraleukocytic bactericidal activity was first studied and concluded that CB inhibited the intracellular bactericidal activity remarkably. Addition of CB resulted in increased nitroblue tetrazolium (NBT)-dye reduction, while the specific activities of glucose-6-phosphate dehydrogenase (G-6-PD) and reduced nicotinamide-adenine dinucleotide phosphate (NADPH) oxidase of CB-treated neutrophils exhibited within the normal range. But the uptake of Glucose-1-14C or -6-14C was markedly inhibited by CB treatment. However, the following substances inhibited NBT-dye reduction in decreasing order: cyclic adenosine 3', 5'-monophosphate (cAMP), N6-O2' dibutyryl cyclic adenosine 3', 5'-monophosphate (DBcAMP), prostaglandin F2alpha (PGF2alpha) and prostaglandin E1 (PGE1). These drugs also decreased hexose monophosphate shunt (HMS) enzyme activities. Addition of CB and DBcAMP decreased the intracellular bactericidal activity of neutrophils. But CB had no effect on intracellular levels of cAMP. From the results obtained, it is likely that intracellular bactericidal phenomena of human neutrophils are controlled by cyclic AMP cascades and by the microfilaments system, separately.

[A clinical evaluation of the effect of cephacetrile on bacterial pneumonia. A comparative test with cefazolin by a double blind method (author's transl)].

The therapeutic efficacy of cephacetrile (CEC) in bacterial pneumonia was evaluated in contrast with that of cefazolin (CEZ) by a double blind method. Both drugs were administered via intravenous route at a dose of 1 g twice daily for 14 days. 1) Of 81 patients, each 2 from both groups were eliminated from the study because of unknown results. In CEC group, 36 out of 38 obtained a slightly effective or better results (94.7% of effectiveness). In CEZ group, 31 out of 39 showed a similar result and there was no significant difference between the two groups. 2) In more detail, CEC achieved significantly better results in AaDo2 and cardiac insufficiency than CEZ, and this trend was also seen in dyspnea. 3) Regarding background factors, pretreatment severity was slightly in favor of CEC. However, so long as supplementary analysis is concerned, we could not find any relation between the pretreatment severity of symptom and drug efficacy or improvement of symptom. 4) Since there was a slight bias in the background factors, it is difficult to conclude that CEC is better than CEZ in terms of effectiveness. However, we consider CEC is superior to CEZ if compared in details. 5) Both drugs had the same incidence of side effect (6.25%, 3/48 in both groups). When clinical efficacy of CEC in bacterial pneumonia is evaluated together with the incidence of side effect, we may consider that CEC is an effective antibiotic agent equal to or better than CEZ.

Intraleukocytic bactericidal activity in patients receiving corticosteroid and radiation therapy, and in patients with diabetes mellitus.

Bactericidal activities of peripheral white blood cells obtained from patients and from healthy persons were examined in vitro. The results obtained are summarized as follows. 1. Peripheral white blood cells from patients receiving corticosteroid and radiation therapy showed decreased levels of intracellular bactericidal activities against Staphylococcus aureus. The leukocytes from almost all patients examined displayed intense activities of intracellular bacterial killing against Streptococcus pyogenes. 2. Only polymorphonuclear leukocytes (PMNs) and macrophages obtained from patients in severe stages of diabetes mellitus exhibited decreased levels of intracellular bactericidal activities against S. aureus. 3. The leukocytes from all patients examined exhibited the same levels of intracellular bactericidal effects against S. pyogenes as leukocytes from healthy persons. 4. Pseudomonas aeruginosa, which was phagocytized by PMNs obtained from healthy persons, demonstrated a remarkable degree of resistance to any intracellular bactericidal effect.

The metabolic and phagocytic activities of leukocytes from patients receiving corticosteroid and radiation therapy, and patients with bacterial infections.

Peripheral blood leukocytes from patients given corticosteroid or radiation therapy, as well as patients with bacterial or viral infections, were studied with regard to the selected enzyme activities of the hexose monophosphate shunt (HMS). Glucose-6-phosphate dehydrogenase (G-6-PD), 6-phosphogluconate dehydrogenase (6-PGD) and reduced nicotinamide-adenine dinucleotide phosphate (NADPH) oxidase were assayed spectrophotometrically on mixed leukocyte suspensions in isotonic glycerol. Enzyme activities of G-6PD and NADPH oxidase in patients receiving corticosteroid or radiation therapy were significantly lower than the enzyme activity of 6-PGD. In patients with bacterial infections, activities of the three enzymes increased but in patients with viral infections, only the activities of NADPH oxidase and G-6PD were slightly decreased. Nitroblue tetrazolium (NBT) dyereducing activities of neutrophils from patients receiving corticosteroid or radiation therapy were attenuated which coincides with the reduced activities of HMS enzymes. From these results, it is likely that the reduced activities of intraleukocytic HMS enzymes of patients receiving corticosteroid or radiation therapy are correlated with intracellular bactericidal activities which might result from the attenuated level of hydrogen peroxide production.

[Laboratory studies on doxycycline intravenous (author's transl)].

The results of laboratory studies on doxycycline intravenous are summarized as follows: 1. The standard curves of doxycycline in five dilution systems were compared. Zones of growth inhibition were largest in the phosphate buffer solution at pH 7.2 and smallest in human serum. Lung system was close to the buffer system, liver system close to the serum system, and kidney system intermediate. 2. The doxycycline concentrations in blood of healthy male adults were compared by cross-over method. The concentration following intravenous injection of 200 mg, dissolved in 40 ml of 20% glucose and injected in 5 minutes, was high than that after oral administration of 200 mg immediately after the injection, but was same after 3 hours and thenafter. 3. No tendency of accumulation was observed in six volunteers injected intravenously with 100 approximately to 200 mg of doxycycline daily for three days. 4. In a patient with chronic broncho-bronchitis injected intravenously with 200 mg, the blood level reached a peak of 4.8 mcg/ml after 30 minutes, while the sputum level reached a peak of 1.8mcg/ml after 90 minutes. The sputum level was 30 to approximately 40% that of serum. 5. The urinary excretion of doxycycline was 94 mg/day (47.0%) after a single intravenous injection of 200 mg. and 53 mg/day (26.5%) after a single oral administration of 200 mg.

An investigation of leukocyte function and phagocytosis of immune complexes in patients with rheumatoid arthritis.

By means of the nitroblue tetra-zolium (NBT)-dye test, patients with chronic rheumatoid arthritis (RA) were studied. It was found that patients with rheumatoid factor (RF) in their sera displayed higher levels of NBT-dye reduction than those without RF did. In RF(+) serum, the authors observed a remarkable activation of NBT-dye reduction in phagocytosis of immune complexes and 1gG-coated latex particles. A fluorescent antibody technique and immuno-electron microscopy showed an increased phagocytosis of gamma-globulin in peripheral polymorphonuclear leukocytes (PMN) from patients with activated levels of NBT-dye reduction. From these results, it is likely that PMNs from patients, particularly from those possessing high values of NBT-dye reduction, displayed the activated levels of phagocytosing autologous globulin.
