Local structures in unfolded lysozyme and correlation with secondary structures in the native conformation: helix-forming or -breaking propensity of peptide segments.

CD spectra of reduced and S-3-(trimethylated amino) propylated lysozyme (TMAP lysozyme) have been measured in various solutions containing guanidine hydrochloride or trifluoroethanol (TFE). The CD spectra indicate that there remain residual secondary structures in protein in aqueous solution. The addition of TFE further promotes the formation of secondary structures. In order to examine whether secondary structures are evenly induced over all the polypeptide chain, or locally at particular segments, the limited proteolysis of TMAP lysozyme by trypsin has been performed, and the CD spectra of all the final and intermediate products have been observed in solutions containing TFE. As a result, the fragments vary in a helix-forming propensity. The CD spectra of peptide fragments T5, T7, T9T10, T12T13, T14T15T16, and T17T18 are not significantly affected by the addition of TFE, where T refers to the nomenclature of R.E. Canfield [(1963), Journal of Biological Chemistry, Vol. 238, pp. 2691-2697]. They are fragments of a helix-breaking propensity. On the other hand, fragment I2 composed of T1-T4, and fragments T6T7, T8, and T11, attain secondary structures with the addition of TFE. They are fragments of a helix-forming propensity. Further, it is found that the fragments of a helix-forming propensity just correspond to the helical segments in native lysozyme. We examine the interactions between neighboring fragments, which contribute to the stabilization of local structures along the polypeptide chain.

Involvement of superoxide radicals in the mouse two-cell block.

The effect of oxygen toxicity on the development of mammalian embryos was assessed by the use of superoxide dismutase (SOD), a potent scavenger of superoxide radicals. Mouse pronuclear embryos recovered 17 h after human chorionic gonadotropin (hCG) were cultured in medium BWW at 37 degrees C under an atmosphere of 5% CO2 in air. Culture of mouse pronuclear embryos in the presence of Cu.Zn-SOD (500 micrograms/ml) significantly increased the blastulation rate (44.6%) when compared with the control culture system (4.2%). Essentially the same effects were observed in SOD containing either Mn or Fe in the catalytic center. Heat treatment of the SOD preparation, and the addition of anti-SOD antibodies to the culture medium, significantly reduced the attenuation of the two-cell block by SOD, indicating that this effect is SOD dependent. SOD activity was detected in rabbit oviduct fluid (3.675 +/- 3.084 mIU/mg protein) by electron spin resonance. These results suggest that active oxygen is involved in the two-cell block phenomenon in mouse embryos exposed to air and that SOD in the oviduct may play an important role in the protection of embryos from superoxide radicals.

Developmental potentiality of embryos cultured under low oxygen tension with superoxide dismutase.

A low-oxygen culture of preimplantation embryos with superoxide dismutase (SOD) has been demonstrated to produce a marked increase in the blastocyst formation. In this study, we examined the effects of low oxygen-SOD cultures of embryos on the potentiality of embryonic development after intrauterine transplantation and the reproductive ability of the offspring. Mouse pronuclear embryos were cultured in Biggers-Whitten-Whittingham's solution containing 0.3% bovine serum albumin and 500 micrograms/ml SOD under a low-oxygen condition (5% O2, 5% CO2, 90% N2), and 37.2% (110/296) of the embryos developed to blastocysts, with a significant difference from the 1.3% (3/231) under an atmospheric oxygen condition without SOD (P less than 0.01). The blastocysts obtained under a low oxygen-SOD culture condition were transplanted by direct insertion into the uterus of pseudopregnant females 3 days after infertile coitus. The implantation rate was 65.0% (65/100), and the embryo viability rate was 46.0% (46/100). In the control group, in which in vivo developed blastocysts were transplanted into 3-day pseudopregnant females, the implantation rate was 72.5% (81/112), and the embryo viability rate was 51.8% (58/112). The differences between the two groups were not significant. Significant differences also were not observed in either the body weight of pups at birth and 35 days after birth or the mean number of pups delivered by successive mating of the offspring between the two groups.(ABSTRACT TRUNCATED AT 250 WORDS)

Involvement of central cholinergic mechanism in RU-24969-induced behavioral deficits.

The present study was undertaken to investigate the role of cholinergic mechanisms in the behavioral effects of RU-24969, a compound with serotonin1B (5-HT1B) receptor agonist properties. RU-24969 caused an increase in locomotion (2-5 mg/kg IP) and an impairment of spontaneous alternation (SA) behavior in a T-maze (0.5-2.0 mg/kg IP) in mice, effects that were also induced by the cholinergic hypofunction with scopolamine treatment (0.5-5.0 mg/kg IP), an acetylcholine (ACh) receptor antagonist. The impairment of the SA behavior by RU-24969 was enhanced by scopolamine. Both the hyperlocomotion and the SA impairment by RU-24969 were markedly reduced by propranolol (20 mg/kg IP) which has 5-HT1A/5-HT1B receptor antagonist properties, as well as by physostigmine (0.05-0.2 mg/kg IP), an ACh esterase inhibitor, and oxotremorine (0.005-0.01 mg/kg IP), an ACh receptor agonist. Moreover, these behavioral deficits of RU-24969 were diminished in mice pretreated intracerebroventricularly with AF64A (30 nmol/body), a presynaptic cholinergic neurotoxin, whereas scopolamine induced the deficits even in animals with the same treatment. These results suggest that the serotonergic behavioral deficits observed after RU-24969 treatment may be caused by an inhibition of ACh release through its action on the presynaptic receptor (particularly RU-24969-sensitive sites) localized on the cholinergic terminals.

Predominant and developmentally regulated expression of dynamin in neurons.

We have cloned a cDNA for dynamin, a 100 kd microtubule-associated motor protein whose 5' region contains a GTP-binding motif homologous to that of the Mx proteins, from a rat brain library and analyzed its expression. Dynamin mRNA is 3.6 kb and is preferentially expressed in the brain after postnatal day 7, parallel to the developmental increase of the protein. In situ hybridization revealed high levels of dynamin transcripts in neural cells in the cerebellar cortex, hippocampus (particularly in the CA3 area), and cerebral cortex. The transcripts appeared in cerebellar granular cells only after they had ceased dividing and had migrated to the inner granular layer. We show that dynamin is expressed predominantly in neural cells after elongation of their processes, suggesting a role especially in mature neurons.

Immunohistochemical localization of superoxide dismutase in the human ovary.

To elucidate the physiological function of superoxide dismutase (SOD) in the ovary, we examined the immunohistochemical distribution of CuZn-SOD in the human ovary. We also measured the CuZn-SOD concentration in human follicular fluid by enzyme-linked immunosorbent assay (ELISA). The germinal epithelium and tunica albuginea showed no or weak immunoreactivity. No or weak staining activity was also observed in the non-antral follicle. Once the follicle began to form the antral cavity, theca interna cells began to show intensive immunostaining of SOD, as compared with no staining in the granulosa and theca externa cells. In the gestational corpus luteum, theca and granulosa lutein cells showed intensive and moderate staining activity, respectively. The concentration of CuZn-SOD was 0.222 +/- 0.186 ng/mg protein (mean +/- SD) in the preovulatory follicular fluid. In the present study, the immunohistochemical distribution of SOD was confirmed in the human ovary for the first time. Taking into consideration the fact that SOD catalyses the dismutation reaction of superoxide anion radicals, the present results suggest that theca interna cells play an important role in the protection of the developing oocyte from oxygen radicals by acting as a blood-follicular barrier during follicle maturation.

Block to development in cultured rat 1-cell embryos is overcome using medium HECM-1.

Rat pronuclear embryos were cultured in hamster embryo culture medium-1 (HECM-1) or a modified Krebs-Ringer bicarbonate solution (mKRB). Embryo cultures in HECM-1 were also challenged with low oxygen concentrations. In HECM-1, 57.9% (70/121) of the pronuclear embryos developed into the 4-cell stage after 48 h of culture. The rates of 8-cell, morula and blastocyst formation were 32.2% (39/121), 17.4% (21/121) and 9.9% (12/121), respectively. On the other hand, in mKRB, rat pronuclear embryos showed developmental blockages at the 2-cell and 4-cell stages, and never developed beyond the 4-cell stage. The rate of blastocyst formation under a low oxygen concentration was 20.1% (43/214), showing a significant difference from the value of 5.5% (11/201) obtained under a standard oxygen concentration (P less than 0.005). This is the first report of successful culture of rat pronuclear embryos to the blastocyst stage. Furthermore, it is suggested that protection from oxidation stress is a prerequisite for rat embryo development in vitro.

Protection from oxidative stress by thioredoxin and superoxide dismutase of mouse embryos fertilized in vitro.

A low oxygen tension and superoxide dismutase (SOD) have been demonstrated to promote development of embryos in vitro. In this study, we evaluated the effects of thioredoxin (TRX), a powerful protein disulphide reductase, on the development of mouse embryos fertilized in vitro. Moreover, to clarify the difference between the embryo-protecting mechanisms of TRX and SOD, as well as the critical periods of their actions, we carried out experiments in which we transferred embryos among three different media: basic, TRX- and SOD-supplemented. The rates of 4-cell, blastocyst and expanded blastocyst formation in TRX- (53%, 40% and 34% respectively), or SOD- (59%, 29%, 22%) supplemented media were significantly higher than those (28%, 4%, 0%) in the basic medium. The results of the transfer of embryos using basic and TRX-supplemented media indicated an increase in the rate of expanded blastocysts when TRX was applied from 7 to 30 h, and especially from 18 to 24 h after insemination. In contrast, the results of the transfer of embryos using basic and SOD-supplemented media indicated that SOD, which was also most effective when applied from 7 to 30 h after insemination, had some effects after 30 h. These results indicate that TRX and SOD promote the in-vitro development of mouse embryos fertilized in vitro, which suggests that protection of embryos from oxidative stress is a prerequisite for their development in vitro. SOD and TRX provide protection from oxidative stress by different mechanisms.

The expression of c-kit protein during oogenesis and early embryonic development.

The c-kit proto-oncogene encodes a transmembrane tyrosine kinase receptor and was shown to be allelic with the white-spotting locus (W) of the mouse. Mutations at the W locus have pleiotropic effects on the development of hematopoietic stem cells, melanoblasts, and primordial germ cells. In order to elucidate the role of c-kit protein in gametogenesis and oocyte maturation, we have examined immunohistochemically the expression of c-kit in the ovaries of mice at late fetal and postnatal stages, and in early embryos. By the avidin-biotin-peroxidase (ABC) method using rat anti-mouse c-kit monoclonal antibody, the c-kit protein was detected in ovaries after the time of birth, but not before. The expression of c-kit was observed mainly on the surface of oocytes, but not in granulosa cells nor in interstitial regions. Oocytes of primordial to fully grown Graafian follicles showed the c-kit protein. When ovulation was induced by hCG, the expression of c-kit in ovulated unfertilized oocytes was weaker than in oocytes of Graafian follicles. In 1-cell embryos the c-kit protein was still observed, but with cell division its expression further decreased, and it was not detected in embryos of 4-cell, 8-cell, and morula stages. In summary, the highest expression of c-kit was observed on the surface of oocytes arrested in the diplotene stage of meiotic prophase. With ovulation and the resumption of meiotic maturation, its expression declined. These results suggest that the c-kit protein may play some role in meiotic arrest, oocyte growth, and oocyte maturation.

Effect of reductive alkylation on thermal stability of ribonuclease A and chymotrypsinogen A.

In order to probe changes in the structural stability induced by the introduction of hydrophobic groups into proteins, the amino groups of ribonuclease A and chymotrypsinogen A were reductively alkylated by reaction with various aliphatic aldehydes, formaldehyde, acetaldehyde, n-butylaldehyde and n-hexylaldehyde, and their thermal stabilities were investigated by differential scanning calorimetry (DSC) at different acidic pH values. Ribonuclease A was thermally unstabilized by reductive alkylation, while chymotrypsinogen A was slightly stabilized, depending on both the size of the introduced alkyl groups and the extent of modification. These observations suggest that the effects induced by alkylation involve not only steric hindrance due to the entering bulky groups but also certain other factors such as the participation of the chemically introduced alkyl groups in hydrophobic interactions.

Epstein-Barr virus activating principle in husbands' semen of cervical cancer patients.

A short-term in vitro assay for detecting tumor promoter activity has been cultivated utilizing the induction of Epstein-Barr virus early antigen (EBV-EA) in EBV genome-carrying human lymphoblastoid cell line (Raji cell) system. By using this system, we found that some of human semen samples possessed a marked capacity to induce EBV-EA, suggesting possible tumor promotor activity of semen. In the present study, 60 semen samples obtained from husbands of cervical cancer patients and 90 control samples were incubated with Raji cells for 48 h, and EBV-EA-expressing cells detected by indirect immunofluorescence were counted. The percentages of EBV-EA-positive cells treated with semen of cervical cancer patients' husbands (8.17 +/- 5.43%) were significantly higher than those of controls (5.23 +/- 5.91%). Since most of EBV-EA expressing effect on Raji cells show overlapping with tumor promotor activities, our present results implies the circumstantial association of seminal EBV-EA inducing activity with occurrence of human cervical cancer.

A study of haemolytic streptococci isolated from outpatients in dermatological clinics.

A total of 44 patients suspected of streptococcal infections were studied in outpatient clinics in Tokyo during the one year from December 1988 to December 1989. Employing bacteriological culturing and serodiagnosis, the following results were obtained. 1) There were 9 cases of impetigo and 15 cases of erysipelas with typical clinical manifestations and age distributions. 2) It seemed that some of the skin infections were caused by group A streptococci whose M-types were different from those of upper respiratory infections typically occurring in Japan. 3) The type distribution of group A streptococci found were quite similar to those isolated in Thailand or Malaysia. 4) There were found group A streptococci exhibiting unique combinations of T- and M-types, such as T11 and M9, T11 and M62 or T13-49 and MOD8 (Provisional type). 5) As for serodiagnostic method, ADNB (anti-deoxyribonuclease B) titer reflected infection by group A streptococcus only, while ASK (anti-streptokinase) and ASO (anti-streptolysin O) reflected not only group A streptococcal infections but group G infections as well.

Reassessment of gamma-ray doses in Hiroshima and Nagasaki.

Reassessment of gamma-ray doses from the atomic bombs in Hiroshima and Nagasaki has been carried out with the thermoluminescent measurements of bricks and decorative tiles which were collected from the buildings that remain as they were at the time of the explosions. The thermoluminescent measurements were performed using a dating technique generally used in archaeology. Annual background dose rates from natural radionuclides in the brick and tile samples and from environmental radiations around the samples were determined with commercially available thermoluminescent detectors. The resultant gamma-ray doses in Hiroshima and Nagasaki are given as a function of distance from the hypocenters and are compared with the DS86 (Dosimetry System 1986) and the T65D (Tentative 1965 Dose).

Fine structure of atrial natriuretic peptide(ANP)-granules in the atrial cardiocytes in the pig, cattle and horse.

In the pig, cattle and horse, the right and left atria and ventricles were examined by immunohistochemistry, and the right atrial and auricular cardiocytes were studied by transmission electron microscopy. Moreover, ANP-granules in the cardiocytes were analyzed by ultrastructural morphometry. Immunohistochemically, the most intensely ANP-reacted cardiocytes were localized in the right auricle, particularly more prominent in the pig and cattle than in the horse. Ultrastructurally, ANP-granules were located principally in the perinuclear region associated with the Golgi apparatus and throughout the sarcoplasmic layers. They were classified into 2 types, A and B. A-granule had a conspicuous electron-dense core enveloped by a membrane. B-granule had a fibrillogranular core enveloped by an indistinct membrane. By TEM equipped with a goniometer stage, a part of the limiting membrane of B-granules became visible as the stage tilt progressed. By ultrastructural morphometry, the number of each type of granules was significantly greater in the pig and cattle than in the horse. In the pig and cattle, the total number of both types of granules in the auricular cardiocytes was significantly greater than that in the atrial ones. The diameter of each type of granules was significantly larger in the pig and cattle than in the horse. The diameter of A-granules was significantly larger than that of B-granules.

Fine structure of atrial natriuretic peptide (ANP)-granules in the atrial cardiocytes of the mouse, rat and Mongolian gerbil.

Mouse, rat and Mongolian gerbil atrial and ventricular cardiocytes were examined by immunohistochemistry, and the right atrium including the auricle was examined by transmission electron microscopy. In addition the ANP granules of both right atrial and auricular cardiocytes were analyzed by ultrastructural morphometry. ANP immunoreactivity was detected in the atria of all three species, and the most intensely reacting cardiocytes were localized in the right auricular part of the atrium. These reactions were more prominent in the mouse and rat than in the Mongolian gerbil. ANP immunoreactivity was not detected in the ventricular myocardium of any of the three species, but was occasionally seen in the subendocardium of the ventricular septum. Ultrastructurally, the ANP granules in the auricular and atrial cardiocytes were observed to be variable in size and number, and these granules were located principally in the paranuclear region in association with the Golgi apparatus, and found throughout the sarcoplasmic layers in all three species. The ANP granules were classified into two types: A-granules containing a conspicuous electron-dense core possessing a membrane, and B-granules having profiles with a fibrillogranular, less electron-dense core than the A-granules and an indistinct membrane. The features of these granules were similar in all three species. When examined by ultrastructural morphometry, the number of each type granule and the total number of granules in the right auricular and atrial cardiocytes of the mouse and rat were significantly greater than in the Mongolian gerbil. The total number of granules in the right auricular cardiocytes was significantly greater than in the cardiocytes of the right atrium exclusive of the auricle, however, there was no significant difference between the number of A-granules and B-granules in the three species. The diameter of each type of granule in the right auricular and atrial cardiocytes of the mouse and rat was significantly greater than in the Mongolian gerbil, and the diameter of the A-granules was significantly greater than the diameter of the B-granules in all three species.

Fine structure of the atrial cardiocytes in the house musk shrew (Suncus murinus).

The atrial and ventricular cardiocytes of the house musk shrew were examined by immunohistochemistry, and the right atrium containing the auricle was studied by transmission electron microscopy. The atrial natriuretic peptide (ANP)-granules of the cardiocytes in the auricle and the rest of the atrium were also analyzed by ultrastructural morphometry. On immunohistochemistry, ANP immunoreactivity was detected in the atria, with the most intensely reacting cardiocytes being localized in the right auricular part of the atrium. ANP immunoreactivity was not detected in the ventricular muscles. On ultrastructure, in most of the atrial cardiocytes including the auricle, ANP-granules, well-developed Golgi apparatus and rough endoplasmic reticulum were observed, and the nuclei were characteristically situated in the periphery of the cardiocytes, being different from many other mammalian hearts. The ANP-granules were classified into two types (A and B), with most of these granules being located in the paranuclear region in association with the Golgi apparatus, and a few ANP granules being observed throughout the sarcoplasmic layers intervening between the myofibrilar bundles. On ultrastructural morphometry, the total number of granules in the right auricular cardiocytes was significantly greater than those in the atrial cardiocytes, and the diameter of the A-granules was significantly greater than that of the B-granules in both the atrial and auricular cardiocytes.

Atrial natriuretic peptide (ANP)-granules of auricular cardiocytes in aging Mongolian gerbils.

The atrial natriuretic peptide (ANP)-granules of right auricular cardiocytes were studied in aging Mongolian gerbils by immunohistochemistry and transmission electron microscopy, and analyzed by ultrastructural morphometry. Immunohistochemically, the ANP immunoreactions were weaker in 3 and 4-year-old animals than in 90-day-old, 1- and 2-year-old animals. There was no difference in the reaction among 90-day-old, 1 and 2-year-old animals, or between 3 and 4-year-old animals. Ultrastructurally, the morphological features in 1 and 2-year-old animals were similar to those of 90-day-old animals, but distinct in 3 and 4-year old animals by the presence of a few lysosomal structures in the cell. By ultrastructural morphometry, number and diameter of ANP-granules of 3 and 4-year-old animals were significantly smaller than those in the 90-day-old, 1 and 2-year-old. There was no significant difference in number and diameter among the 90-day-old, 1 and 2-year-old, or between the 3 and 4-year-old.
