[Clinical bacteriological studies on cefotiam and cefsulodin in the field of otorhinolaryngology].

Clinical bacteriological studies on cefotiam and cefsulodin in the field of otorhinolaryngology were carried out and the following results were obtained. 1) Aerobic and anaerobic Gram-positive bacteria were dominantly isolated from the clinical materials sent to the center from the clinical institutes. 2) It was considered that Streptococcus pneumoniae, Haemophilus influenzae and beta-Streptococcus played an important role in the primary infections in the field of otorhinolaryngology. Staphylococcus aureus was also frequently isolated from the primary infections. Peptostreptococcus spp. was dominantly isolated from peritonsillar abscess. Gram-negative bacilli (GNB) were mainly isolated from the chronic secondary infections. Among GNB, Pseudomonas aeruginosa and Proteus spp. were more frequently isolated. Staphylococcus aureus was also constantly detected in the secondary infections together with GNB. Anaerobic bacteria were isolated from 20.1% of the patients with chronic otitis media and 27.1% of sinusitis. 3) Cefotiam showed potent antibacterial activities against most isolates of Gram-positive and Gram-negative bacteria. 4) Cefsulodin showed potent antibacterial activities against clinically isolated Pseudomonas aeruginosa. Staphylococcus aureus and beta-Streptococcus were also susceptible to cefsulodin.

Mechanism of development of bronze baby syndrome in neonates treated with phototherapy.

Comparisons of serum concentrations of unknown pigment and photobilirubin IX alpha , the two main bilirubin photoproducts, were made during phototherapy in infants with and without bronze baby syndrome who were treated similarly. The serum concentrations of unknown pigment estimated by high-pressure liquid chromatography in infants with the bronze baby syndrome were significantly increased in comparison with those in the control hyperbilirubinemic neonates during phototherapy. However, there was no difference in the serum concentrations of photobilirubin IX alpha between infants with bronze baby syndrome and the control groups. The unknown pigment separated from bilirubin photoproducts obtained from experiments in vitro by high-pressure liquid chromatography was gradually decomposed into brown products that showed the absorption spectrum similar to that of the serum of infants with bronze baby syndrome. This fact is probably due to reduction in hepatic excretory function of bilirubin photoproducts, especially in known pigment, because its main excretory pathway is the biliary route. The pigment accumulated in the body may be polymerized and forms bilifuscin-like substances following a free radical reaction. It is concluded that the brown pigment is formed via unknown pigment.

[Scotomatous defects in the central visual field, associated with pituitary tumor--a case report (author's transl)].

A rare case which showed scotomatous defects in the left central visual field was presented. A 51 year-old man was admitted to our clinic because of the visual deterioration. In neuroradiological examinations the left carotid angiography showed elevation of both A1 portions and CT suggested pituitary tumor. On February 5, 1980 radical operation was carried out, in which it was confirmed that part of the optic nerve was concave and split longitudinally with a compression of the pituitary adenoma. Postoperative course was uneventful and recovery from the defects in the central visual field was quickly. In this case longitudinal splitting of unilateral optic nerve, caused by gradual compression of supra- or parasellar tumor, was considered as one cause of scotomatous defects in the central visual field. In addition, visual status of forty six cases of pituitary adenoma which had been operated at our clinic during the past fourteen years was analysed both pre- and post-operatively, compared with the case described above.

Characterization of a radiation-resistant Acinetobacter.

For characterizing the radiation-resistant Acinetobacter strain FO-1 reported in the previous paper (1980), we investigated the structure of cell wall, the cellular fatty acid composition, and the detailed taxonomic characteristics. The results denoted that the cell division occurred by simple constriction with the formation of a slight septum and the intermediate dense layer between the outer membrane and the plasma membrane was seen, the main components of the cellular fatty acids were oleic acid, palmitic acid, and palmitoleic acid, and that the strain was tolerant to salt and could not produce acids from cellobiose, melibiose, lactose, and ribose.

Junction-like structure appearing at apposing membranes in the double cone of chick retina.

No type of junction has yet been recognized between the two entities of the retinal double cone. In the present study, a junction-like structure was observed in serial sections of the double cone of the chick retina. It is recognized from the slightly outer part of the double cone to the outer limiting membrane. The apposing membranes are virtually parallel and separated by 5 to 7 nm of extracellular space. Dense material is associated on the cytoplasmic side of the opposing membranes. The structure resembles a gap junction, but there are no cross striations between the two membranes. Further experiments are required to establish this as a new type of junction.

Mutations in the gene coding for Escherichia coli DNA topoisomerase I affect transcription and transposition.

Mutations in top, the structural gene for Escherichia coli DNA topoisomerase I, have been identified and mapped at 28 min on the chromosome, near cysB. Strains carrying deletions of the top gene are viable. The top mutations, however, do exert pleiotropic effects on transcription and transposition. Mutants lacking DNA topoisomerase I have a more rapid rate of induction and a higher level of catabolite-sensitive enzymes including tryptophanase and beta-galactosidase. This general activation of transcription by top mutations can be attributed to an increase in the negative superhelicity of the DNA in vivo when the topoisomerase activity is abolished. The frequency of transposition of Tn5, a transposon carrying kanamycin resistance, is decreased by a factor of 40 or more in top mutants. A direct or indirect role of the topoisomerase in transposition is discussed. The transposition frequency of Tn3, however, is not dependent on top. Based on the studies of the E. coli top mutants, it appears that the supX gene, which was originally studied in Salmonella typhimurium [Dubnau, E. & Margolin, P. (1972) Mol. Gen. Genet. 117, 91-112] is likely to be the structural gene for DNA topoisomerase I.

K+ selectivity and HCl secretion in Xenopus and their relation to cAMP and Ba2+.

Doses of 1, 5, and 10 mM adenosine 3',5'-cyclic monophosphate (cAMP) applied to previously resting Xenopus gastric mucosae initiated a dose-dependent acid secretory response. Increases in the K+ concentration of the submucosal solution. [K+sm], resulted in higher acid secretory rates and concomitant lower transmucosal resistances at the indicated cAMP concentration. The transmucosal potential difference, PD, had a liner slope against log [K+sm] over the range of 20-80 mM K+sm. Values of the slope (K+ selectivity, -PD/log[K+sm]) were less than 26 mV/log for resting mucosae, and in the range of 29-58 mV/log for both spontaneously secreting and cAMP-stimulated mucosae. These values were dose dependent on the cAmP concentration. In the presence of 10 mM Ba2+ together with cAMP, the K+ selectivity and the H+ secretory rate decreased compared with the case of cAMP only.
