Neonatal tuberous sclerosis with heart and brain tumors.

A 6-day-old neonate with tuberous sclerosis having brain and cardiac tumors is reported. The patient presented with a seizure 20 minutes after birth. Cranial ultrasonography and computed tomography on admission showed a large and round high-density tumor lesion in the left frontal lobe. Echocardiography also showed a large tumor lesion beside the left ventricle wall, and a diagnosis of tuberous sclerosis was promptly made. Two months later, a periventricular calcified lesion was found on the demonstrated portion, and the tumor lesion initially observed in the brain had disappeared. These findings suggest that the initial tumor lesion found in the brain developed into a typical calcified lesion.

Development of the human putamen: a morphometric study.

The development of the human putamen was studied using complete serial sections from the brains of 6 fetuses at 21, 23, 27, 30, 33 and 34 weeks of gestation (WG). Morphometric analysis revealed the following data. (1) The total number of putaminal neurons increased gradually from 21 to 33 WG, suggesting that the migration of putaminal neurons continues at least up to 33 WG, and the neuronal number increased markedly after 33 WG. (2) No neuronal death was observed from 21 to 34 WG. (3) Large-sized neurons 4-5 times the area of small-sized neurons appeared from 30 WG onward. The ratio of large neurons to the total putaminal neurons increased by up to 1.4% at 33 and 34 WG.

[Development of the human fetal brain: volumetric analysis of the component parts].

Development of human fetus brain was morphometrically evaluated through measuring volumes of the cerebrum and its component parts (i.e. cerebral cortex, subcortical white matter, basal ganglia, diencephalon, ventricles, embryonal periventricular cell layer, etc.) in the second trimester of gestational period. For the measurement of volumes, authors employed a unique method which consisted of a combination of serial celloidin sections, electric image-analyzer and computer. We found that each component part of fetus brain developed heterochronously from the view point of volumetric analyses. In the component parts of cerebrum, diencephalon developed first, followed by subcortical white matter and ganglia, and cerebral cortex developed last. It is concluded that a combined method of serial sections, image-analyzer and computer analyses is reliable and useful to obtain more precise and objective information for the morphometrical evaluation of brain development.

Neuron-specific enolase in comatose children.

Sequential examination of neuron-specific enolase in cerebrospinal fluid and serum was performed in 20 comatose children with acute encephalitis, acute encephalopathy, or Reye's syndrome. Neuron-specific enolase activities corresponded to the degree of brain damage. As neuron-specific enolase levels increased to greater than 80 ng/mL, patients had more severe impairment or died. Neuron-specific enolase may be a useful marker for evaluating the degree of neuronal damage and prognosis.

Simultaneous histochemical assay of two dehydrogenases in the same cell.

A new approach has been developed for the simultaneous assay of the activities of two enzymes (lactate and succinate dehydrogenases) in the same cell in sections of unfixed liver. The sections, mounted on coverslips, were placed on top of 0.6-mm thick 0.8% low gelling-temperature agarose films containing the substrates of both enzymes (70 mM lactate and 50 mM succinate, respectively) plus 80 mM Tris-HCl buffer (pH 7.5), 5 mM EDTA, 10 mM NaN3, 1.5 mM NAD+, 1.2 mM Nitro BT and 0.26 mM phenazine methosulphate. The integrated absorbance (A) at 585 nm of the final reaction product formazans deposited by the two enzymes in a selected hepatocyte was measured continuously at 37 degrees C as a function of incubation time, using a Vickers M85 microdensitometer. The intercept A0 on the A-axis of the linear regression line of A on time was determined. After a known incubation time t, the absorbance A1, was noted and the section placed on another gel film lacking the substrates in order to estimate the final reaction product either formed in the gel film or lost from the cell. The absorbance A2 of the hepatocyte was remeasured. The reaction velocities (activities) vL and vS of lactate and succinate dehydrogenases, respectively, were calculated from the following equations: vL = [(A1-A2) - A0(1- alpha L)]/(1-alpha L)t and vS = (A2-alpha LA1)/(1-alpha L)t where alpha L = A2/A1 for hepatocytes incubated on gel films containing only lactate as the substrate. Histochemical modification of the active site of succinate dehydrogenase with N-acetylimidazole.

The kinetics of acetylation of mitochondrial succinate dehydrogenase [EC 1.3.99.1] in the two fibre types (A and C) of rat gastrocnemius with N-acetylimidazole was studied by a newly modified histochemical technique. Acetylimidazole partially inactivated the enzyme, but subsequent deacetylation with hydroxylamine restored the enzyme activity completely. Inactivation of the enzyme by acetylimidazole was prevented by malonate, which is a competitive inhibitor of the enzyme. The value of the inhibition constant (Ki = 34 microM) for malonate, obtained from the dependence of the pseudo-first order rate constant of acetylation of the enzyme with acetylimidazole on the malonate concentration, was in good agreement with the Ki value (33 microM) obtained by a different method, the dependence of the initial velocity of succinate oxidation by the dehydrogenase on the substrate concentration in the presence of malonate. These findings suggest that a tyrosyl residue is located in the malonate binding site (the active site) of succinate dehydrogenase in the gastrocnemius and plays a role in substrate binding, but is not a catalytic group.

Quantitative double-staining methods for tryptophyl and cysteinyl residues and protein in model films.

The reliability of three new staining methods developed by the authors were examined with polyacrylamide gel films containing concanavalin A and ovalbumin. The methods were a 2-nitrophenylsulphenyl chloride technique for staining tryptophyl and cysteinyl residues together, a nitrophenylsulphenyl-2-mercaptoethanol method for tryptophyl residues only, and the combination of both these techniques with Coomassie Brilliant Blue for staining total protein. The relative contents of tryptophyl and cysteinyl residues of concanavalin A and ovalbumin in the films, calculated from absorbance values at 370 and 650 nm of double-stained films, were in good agreement with the theoretical values calculated from their amino acid compositions. Thus, these quantitative double-staining methods are reliable and should be useful for histochemical investigations of changes in the amount and composition of tissue proteins.

Kinetic behaviour of succinate dehydrogenase of three fibre types in skeletal muscle. I. Effects of temperature and a competitive inhibitor.

The kinetic behaviour of succinate dehydrogenase [EC 1.3.99.1] in three fibre types of rat gastrocnemius was examined by a quantitative histochemical method without disruption of the cellular structure. 2-(2-Benzothiazolyl)-3-(4-phthalhydrazidyl)-5-styryl-t etrazolium chloride (BPST) and phenazine methosulphate were used as electron acceptors. On measurement of the absorbance value at 530 nm of BPST formazan, produced by the succinate dehydrogenase reaction in sections, it was found that the staining intensity of succinate dehydrogenase was linearly proportional to both the incubation time and the thickness of the slice therefore, the initial velocity of the staining could be calculated. By Michaelis-Menten (1913) treatment of the dependence of the initial velocity on the substrate concentration in the absence and the presence of a competitive inhibitor, malonate, the Km and Vmax values for succinate and the Ki value for malonate were obtained. The Km and Ki values of the three fibre types were similar. The ration of the Vmax values of type A, B and C fibres was 1.0:2.0:3.3. The temperature dependence of the kinetic parameters was very similar in the three fibre types. These findings confirm that the differences in the staining intensity of the three fibre types reflect differences in the amounts, but not the properties, of succinate dehydrogenase.

Combined quantitative cytophotometric method for staining indolyl and sulfhydryl groups and protein.

Sulfenylation with 2-nitrophenylsulfenyl chloride (NPS-Cl), which is specific for tryptophyl and cysteinyl residues in protein, was applied to quantitative histochemistry. By measurement of the absorbance values at 370 nm of sections stained with NPS-Cl, Beer-Lambert's law was found to hold for NPS staining. Treatment of NPS-stained sections with 2-mercaptoethanol (ME) (NPS-ME staining) resulted in sulfenylation of tryptophyl residues only. For determination of the amounts of tryptophyl and cysteinyl residues per unit of protein, protein staining with Coomassie Brilliant Blue (CB) was combined with NPS and NPS-ME staining. CB and NPS-CBB staining also followed Beer-Lambert's law. By measuring the absorbance values at 370 and 650 nm of doubly stained sections, the relative contents of tryptophyl and cysteinyl residues in various tissue proteins were calculated. This method will be useful for the investigation of changes in both protein amount and composition.

Colitis cystica profunda: report of a case in which differentiation from rectal cancer was difficult.

Colitis cystica profunda is a benign condition that has been confused with carcinoma. This rarely recognized condition is characterized by submucosal cysts. Macroscopically, the ulcerated character of the mass suggests carcinoma. Microscopically, attenuation of the innocuous-appearing cyst linings may give the false impression of a mucus-secreting carcinoma. A case of colitis cystica profunda in which the macroscopic appearance was so suggestive of malignancy that operation was carried out is reported.
