Single-stranded DNA transcription by yeast RNA polymerase B.

Single-stranded DNA is not transcribed randomly by yeast RNA polymerase B. A denatured yeast DNA fragment, containing the gene for yeast alcohol dehydrogenase I, directs the transcription of defined RNA products visualized as discrete RNA . DNA hybrid bands following S1 nuclease treatment and agarose gel electrophoresis. Blocking the 3' end of the template by 3' deoxyadenosine did not change the band pattern but reduced the proportion of RNA covalently bound to the DNA from 20 to 4%. On the other hand, the band pattern was affected by the salt concentration, the nature of the divalent cation and the nucleoside triphosphate concentration. The four major RNA bands, found at low substrate concentration, hybridized to the same region of the template. This observation suggests the potential requirement for DNA destabilization in gene activation.

Early operations for ruptured intracranial aneurysms. Comparative study with computed tomography.

The authors have analyzed the correlation between the results of early operation for ruptured intracranial aneurysms (45 cases) and CT scan (54 cases) performed within the second week after subarachnoid haemorrhage (SAH). There was a good correlation between the results of early operation within one week after the haemorrhage and CT findings. Surgical results were closely correlated to the incidence of vasospasm in our cases. All cases with symptomatic vasospasm showed bilateral high density in the cistern or ventricles on CT scan performed within three days after initial bleeding. For the cases within 24 hours after haemorrhage, surgical operation is recommended as early as possible, including removal of blood clots in the basal cistern and Sylvian fissure, and ventriculo-cisternal drainage for improvement of cerebrospinal fluid (CSF) circulation. On the other hand, the cases in which operations were performed during the second to seventh days and where CT scan revealed bilateral high density in the cistern or ventricles resulted in poor outcome or death.

Multiple intracranial aneurysms associated with moyamoya disease. Case report.

A case of multiple intracranial aneurysms associated with unilateral moyamoya vessels is reported. The authors have reviewed the age, sex, initial symptoms, site of aneurysm, and operative indication in similar cases reported in the literature. It was found that aneurysms associated with moyamoya disease were frequently located in the vertebrobasilar system.

[Early operations for ruptured intracranial aneurysms--comparative study with computed tomography (author's transl)].

The authors have analyzed the correlation between the result of early operation for ruptured intracranial aneurysms (45 cases) and CT scan (54 cases) performed within the second weeks after subarachnoid hemorrhage (SAH). 1) Twelve patients operated on within 24 hours after SAH showed no mortality and a morbidity of 16%. On the contrary, in 12 patients operated on during 2nd-7th days after SAH, the mortality rate was 38% and the morbidity 15%. 2) There was a good correlation between the result of early operation within one week after the hemorrhage and CT findings. Surgical results were closely correlated to the incidence of vasospasm in our cases. All cases with symptomatic vasospasm showed bilateral high density in the cistern or ventricles on CT scan performed within 3 days after initial bleeding. 3) In the cases of ruptured aneurysm with giant intracranial hematoma, but no remarkable blood clot in ventricles or basal cisterns, the results of surgery in acute stage were good. 4) For the cases within 24 hours after the hemorrhage, it is recommended to carry out surgical operation as early as possible, including removal of blood clots in the basal cistern and sylvian fissure, and ventriculo-cisternal drainage for improvement of cerebrospinal fluid (CSF) circulation. On the other hand, the cases in which operations were performed during 2nd-7th days and CT scan revealed bilateral high density in the cisterns or ventricles resulted in poor outcome or death.

[Chronic subdural hematoma associated with intracranial aneurysm (author's transl)].

Four cases of chronic subdural hematoma associated with intracranial aneurysm were reported. In two cases aneurysms were found at the peripheral portion of middle cerebral artery under the hematoma, and in other two cases aneurysms were found incidentally. The causality was discussed about subdural hematoma and intracranial aneurysm especially false aneurysm found in the peripheral artery under the hematoma. Radical operation of intracerebral aneurysm associated with chronic subdural hematoma should be performed after angiography and CT scan regardless the rupture of the aneurysm.

[The association of polycystic kidneys with intracranial aneurysms (author's transl)].

A 48 year-old man is presented who died of rupture of intracranial aneurysm. Autopsy findings revealed a ruptured anterior communicating artery aneurysm associated with polycystic kidneys and polycystic liver. Epidermiological review is performed using "Annual of the Pathological Autopsy Case in Japan" (Vol. 17-20). In 92854 autopsy cases, there are 243 cases of polycystic kidney, 1542 cases of intracranial aneurysm and 10 cases of association with both of them. Several points come clear from comparing our data with foreign ones, as follows; 1) The frequency of polycystic kidney (0.25%) is almost the same between Japan and foreign country. 2) The frequency of intracranial aneurysm (1.6%) is higher in Japan. 3) The frequency of association with both (0.01%) is lower in Japan. 4) The frequency of intracranial aneurysm appears to be higher in cases suffered from polycystic kidney than in others. We suspected that cerebral aneurysm formation is caused by hypertension due to polycystic kidney, although in many reports, the association of polycystic kidney and cerebral aneurysm is explained to be maldevelopment. The operation of cerebral aneurysm with polycystic kidney is the same as that without polycystic kidney. But the control of hypertension and renal function is necessary during and after operation.

[Lipoma of corpus callosum with callosal agenesis (author's transl)].

Intracranial lipoma is rare. The case reported here was a woman aged 60. She came to our hospital with complaint of headache. Neurological examination was negative. Plain craniogram showed abnormal calcification in the callosal region. A-P view of right carotid angiogram revealed winding sinous course of bilateral anterior cerebral arteries around the calcific deposit. Lateral view showed vertical stretching of ascending portion of the anterior cerebral artery. In the frontal projection of the pneumogram, the lateral ventricles were separated and concave medially with hornlike superior angles. The third ventricle extended upward between the lateral ventricles. The plain CT scan revealed an abnormal low density area (Hounsfield's number-90- -50) in the callosal region. The calcific deposit was seen within a low density area. At operation, the callosal tumor was partially removed. Histologically, lipomatous tissue was demonstrated. In reference to literature, we discussed the roentgenographic findings, especially computed tomography of callosal lipoma.

Inhibitory effects of nucleoside triphosphates on nucleolar RNA synthesis.

Studies on the effects of substrates on RNA polymerase I [EC 2.7.7.6] in vitro showed that nucleolar RNA synthesis was inhibited by an excess of substrate nucleoside triphosphates in the presence of Mg2+. GTP and UTP were more inhibitory than CTP and ATP. These compounds specfically inhibited nucleolar RNA synthesis and a concentration of GTP that strongly inhibited nucleolar RNA synthesis did not inhibit RNA synthesis by partially purified RNA polymerase I. The inhibition of nucleolar RNA synthesis disappeared at pH 9.0 without any change in the apparent Km for GTP or the Vmax of RNA synthesis.

Hepatic sarcoma associated with hepatoma.

An autopsy case with simultaneous occurrence of hepatoma and spindle cell sarcoma in the liver was presented. These tumors arose in the left and right lobes and showed no continuity. The sarcoma metastasized in hilar lymph node of the liver and in the cervical and lumbar vertebrae, but no metastasis of the hepatoma was found except in the opposite lobe of the liver. Literature on such combinations was briefly reviewed.

Pseudosarcomatous carcinoma of the esophagus--reference to its histogenesis.

A case of pseudosarcomatous carcinoma of the esophagus with metastasis to lymph node was presented and the literature was reviewed briefly. Transition from carcinomatous to sarcomatous picture was noted and the presence of tonofibrils and desmosomes was demonstrated by electron microscope in the cells with sarcomatous features. These findings suggested an epithelial origin of the sarcomatous component in pseudosarcomatous carcinoma.

Changes of the cell cycle during carcinogenesis of hamster cheek pouch epithelium by 7,12-dimethylbenz[a]anthracene.

Changes of the cell cycle during carcinogenesis of hamster cheek pouch epithelium were studied and it was demonstrated that in hyperplastic and neoplastic stages, the cell cycle time decreased to approximately 2/3 and 1/3 of the normal control, respectively. A nominal growth fraction was found to exceed unity in carcinogen-treated epithelium, whether hyperplastic or neoplastic. This result seems to indicate that a considerable number of cells in which DNA synthesis is stimulated by the carcinogen fail to enter mitosis. A possible implication of repair synthesis is also discussed.
