Inhibitory effect of bovine brain calmodulin on calmodulin-dependent stimulation of plasma membrane-bound guanylate cyclase in Tetrahymena pyriformis.

Bovine brain calmodulin (B-CaM) was shown to inhibit the native Tetrahymena calmodulin (T-CaM)-dependent activation of guanylate cyclase in Tetrahymena at the concentrations that failed to affect the basal enzyme activity. The enzyme inhibition was completely reversed by high concentration of T-CaM, but not by Ca2+. The antagonistic interaction between T-CaM and B-CaM was not observed in the calmodulin-dependent cyclic nucleotide phosphodiesterase from bovine brain. Two calmodulins migrated independently on 15% polyacrylamide gel system. These results suggest that B-CaM exerts its inhibitory effect on the guanylate cyclase activation by interacting with the calmodulin-binding site of this enzyme.

[Primary amyloidosis associated with early gastric carcinoma(IIb like IIa type) diagnosed by preoperative gastric biopsy--a case report].

A case of primary amyloidosis associated with intramucosal adenocarcinoma of the stomach, diagnosed by preoperative biopsy, is reported. A 77-year-old man was admitted to our hospital with complaints of vertigo and general fatigue. X-ray and endoscopic examination revealed a IIb-like IIa type protruded lesion on the cardia of the stomach. A biopsy specimen from the protruded lesion disclosed well differentiated tubular adenocarcinoma and amyloid deposition. Partial gastrectomy was performed on August, 18, 1981. Upon histological study, a diagnosis of primary amyloidosis associated with intramucosal well differentiated tubular adenocarcinoma was made. We suggest that the histogenesis of this gastric carcinoma was related to the gastric lesion due to primary amyloidosis. Careful review of the Japanese literature disclosed that ours is the first case report of primary amyloidosis associated with early gastric carcinoma diagnosed by pre-operative gastric biopsy.

Histopatological findings of the liver before and during hepatic coma in fulminant hepatitis.

The histopathological findings of the liver taken by needle biopsy before and during hepatic coma of fulminant hepatitis were studied in 3 patients; two died and one survived. In the first fatal case, massive hepatic necrosis was already present 7 days before the development of hepatic coma. In the second fatal case, submassive hepatic necrosis was seen during coma grade IV. In both cases, histological findings of the biopsy liver specimens were in accord with those of the autopsied liver. In the third survived case, diffuse degeneration and multilobular liver cell necrosis were found on the biopsy specimen taken immediately after the development of hepatic coma, grade IV.

Quantitative analysis of beta-lactamase production of multiple resistance to beta-lactam antibiotics in clinical isolates of Escherichia coli.

A simple test of procedure is described for the rapid evaluation of beta-lactamase substrate profiles against all the clinically important beta-lactam antibiotics. By use of this method 100 clinical isolates of Escherichia coli were classified into four beta-lactamase types: TEM-like, 35; chromosomal-like, 56; others, 3; beta-lactamase-less, 6 strains. They were tested for their susceptibilities to cefuroxime, cefoxitin, cefamandole, cephalexin, cefazolin, cephaloridine, cephalothin, ampicillin, carbenicillin and kanamycin. The isolates of each of TEM-like and chromosomal-like beta-lactamase types showed a good correlation between the beta-lactamase activity and the minimum inhibitory concentration (MIC) determined by the agar dilution method and the least square line analysis; MIC = ax + b, where x is enzyme activity, and a and b are constants. TEM-like beta-lactamase increased the resistance to ampicillin and carbenicillin markedly and that to cephalothin, cephaloridine and cefamandole moderately. In contrast, chromosomal-like beta-lactamase increased the resistance to ampicillin, cephalexin and cephalothin only slightly.
