[Young children's representational theory of mind in understanding masked facial expression].

In this study, development of young children's understandings of masked facial expression was examined from the perspective of the development of "theory of mind". In Experiment 1, short stories in which one character masks facial expressions to make another character have false beliefs were presented to 3-, 4-, and 6-year-old. They were asked questions to assess their judgements on various mental states (desire, belief, and intention) of story characters. And, to assess children's "representational theory of mind", they performed a "Smaties Task". The results showed that recognition of masked facial expression was developed from 3 to 4 years. Scores on "Smaties Task" was also improved during the same period. An additional analysis showed that the major determinant of to determine the understanding of masked facial expression was not the age but the possession of "representational theory of mind". In Experiment 2 more sophisticated but similar type of procedures were presented to 3- and 4-year-old children. Results replicated the results in Experiment 1 and the understanding of the intention of characters was also found to develop from 3 to 4 years.

Fattiviracin A1, a novel antiviral agent produced by Streptomyces microflavus strain No. 2445. II. Biological properties.

Fattiviracin A1, showed potent antiviral activities against herpes simplex virus type 1 (HSV-1), varicella-zoster virus (VZV), influenza A virus and human immunodeficiency virus type 1 (HIV-1). It showed no cytotoxicity against Vero cells. Fattiviracin A1 exhibited no significant antibacterial or antifungal activities. Treatment of HSV-1 with fattiviracin A1 decreased its infectivity to Vero cells. The mechanism of its antiviral activity may be due to inactivation of the viral particles and inhibition of viral entry into host cells.

Fattiviracin A1, a novel antiherpetic agent produced by Streptomyces microflavus Strain No. 2445. I. Taxonomy, fermentation, isolation, physico-chemical properties and structure elucidation.

A novel antiherpetic agent, fattiviracin A1, was isolated from the culture broth of strain No. 2445 identified as Streptomyces microflavus. It was purified through 1-butanol extraction, column chromatographies on Diaion HP-10 and silica gel and HPLC using a reverse phase column. The structure of fattiviracin A1 was determined by several spectroscopic experiments and chemical degradations. It is a new macrocyclic diester consisting of four D-glucose units and two (C24 and C33) hydroxy fatty acids. It is closely related to cycloviracins B1 and B2, but differs from these known compounds in both the length of its side chain and the sugar moiety.

Histological analysis of the aortic nerve in the rat raised in space (Rapid communication on Neurolab Project).

To study development of the aortic nerve baroreflex under conditions of microgravity, we examined the cross section of the left aortic nerve (LAN), which is the afferent of the baroreflex, in the neonate rats aged 25 days raised in microgravity on the space shuttle Columbia (flight:FLT group) for 16 days. In this paper, we report a part of the result obtained from the data of the myelinated fibers of LAN analyzed with an electron microscope. Two kind of ground control groups were compared to the FLT group; one was asynchronous ground control (AGC) group where the rats were housed in the same cage as that on the shuttle, and the other was vivarium(VIV) group where the rats were housed in a commercial cage. The LANs in each group were extirpated the from rats perfused with a fixative and embedded for histological analysis. We observed the transverse sections of LAN and took pictures of several areas (magnified to x 2K to x 200K). No irregular myelination was found in all fibers of FLT group when they were compared with two control groups. The thickness of myelin of the maximally myelinated fibers were 0.55 +/- 0.17 micrometer in FLT(n=5), 0.45 +/- 0.10 micrometer in AGC(n=5), and O.47 +/- 0.06 micrometer meter in VIV(n=5). There was no significant difference among three groups (unpared t-test). The results suggest that there is no effect of space environment on the myelin formation of each nerve fiber in the aortic nerve.

Involvement of dopaminergic system in phencyclidine-induced place preference in mice pretreated with phencyclidine repeatedly.

In the conditioned place preference test, phencyclidine (PCP) produces place aversion in naive rats, whereas PCP produces place preference in rats treated with PCP repeatedly. Although the PCP-induced place aversion is thought to involve the serotonergic system, the mechanisms of the PCP-induced place preference are unclear. We investigated whether the dopaminergic system is involved in place preference induced by PCP in mice repeatedly treated with PCP, because it is well known that the dopaminergic system plays an important role in the rewarding effect of drugs. PCP (2-8 mg/kg s.c.) induced a dose-dependent place aversion in naive mice, whereas PCP (2-8 mg/kg s.c.) induced a dose-dependent place preference in mice pretreated with PCP (10 mg/kg/day s.c.) for 28 days. The place preference induced by PCP (8 mg/kg s.c.) was attenuated significantly by alpha-methyl-rho-tyrosine (100 mg/kg i.p.), a tyrosine hydroxylase inhibitor, 6-hydroxydopamine (100 micrograms/mouse i.c.v.), a dopaminergic neurotoxin, and R-(+)-7-chloro-8-hydroxy-3-methyl-1-phenyl-2,3,4,5-tetrahydro-1H-3-benza zepine (0.5 mg/kg s.c.), a dopamine D1 receptor antagonist. These agents themselves produced neither the place preference nor aversion. In contrast to the attenuating effects of these agents, N-(2-chloroethyl)-N-ethyl-2-bromobenzylamine (30 mg/kg i.p.), a noradrenergic neurotoxin, ritanserin (1 mg/kg i.p.), a serotonin2 receptor antagonist, and (-) sulpiride (50 and 100 mg/kg i.p.), a dopamine D2 receptor antagonist, failed to affect the PCP-induced place preference. In mice pretreated with methamphetamine (1 mg/kg/day s.c.) for 14 days, PCP (8 mg/kg s.c.) induced the place preference, but not aversion. These results demonstrate that the PCP-induced place preference depends on dopaminergic, but not on serotonergic and noradrenergic, neuronal systems and suggest a role for D1 receptors in the mediation of the PCP-induced place preference.

[Complete remission of liver metastasis from rectal cancer following continuous oral administration of tegafur-uracil after intrahepatic arterial single injection of CDDP and 5-FU].

An intrahepatic arterial injection of CDDP, 5-FU, followed by ten months of oral tegafur-uracil administration (2g/day), induced remission for 3 months or more in a 72-year-old male with rectal cancer and synchronous liver metastasis subsequent to anterior resection of the rectum. Tegafur-uracil showed an excellent anticancer effect against colorectal metastatic liver cancers without loss of QOL because a single-low dose of intraarterial anticancer injection was followed by continuous oral administration of tegafur-uracil, and the chemotherapy could be managed to obtain complete remission of the hepatic lesion.

[Results of preoperative high-dose-rate intraluminal brachytherapy for resectable rectal cancer].

One hundred and fifteen patients with middle and lower rectal cancer were treated with preoperative High-dose-rate intraluminal brachytherapy (HDRIBT) and radical operation. Patients were divided into a middle-dose group (group A; 30-40 Gy; n = 94) and a high-dose group (group B; 60-80 Gy; n = 18). A control group of 115 rectal cancer patients who received no irradiation prior to radical surgery was used for comparison (group C). The rate of sphincter-saving resection was 71% in group A, 61% in group B, and 42% in group C (group A vs. group C; p < 0.0001). The local recurrence rate at 5 years was 10% in group A, 6% in group B, and 26% in group C (group A vs. group C; p = 0.005). The 5-year survival rate was similar among the three groups. These results suggest that preoperative HDRIBT contributed to the improvement of local control but not to survival after radical resection of rectal cancer. The application of HDRIBT might be useful to restore intestinal continuity for rectal cancer.

Obstructive jaundice due to multiple hepatic abscesses.

A 63-year-old Japanese male with diabetes mellitus developed obstructive jaundice following the onset of multiple hepatic abscesses. Percutaneous transhepatic cholangiography showed intrahepatic bile duct irregularity and dilatations accompanied by a complete obstruction of the right branch of the intrahepatic bile duct. Three kinds of organisms were cultured from the blood and the drained bile. The cholangiographic changes returned to the normal after the liver abscesses subsided following biliary drainage and the administration of intravenous antibiotics.

Examination of prognostic factors after resection of pulmonary metastasis of osteosarcoma by multivariate analysis.

Prognostic factors for the case of pulmonary metastasis of osteosarcoma was examined using the multivariate analysis. 1) The 3-year survival rate after operation of primary focus in 23 cases was 34.8% and the 5-year survival rate was 30.4%. The 3-year survival rate after the first operation of pulmonary metastasis was 30.4% and the 5-year survival rate was 12.2%. No deaths attributed to operation were found. 2) Gender, age, histological type, solitary or multiple pulmonary metastasis, laterality, number of pulmonary metastatic tumors, TDT and DFI were examined, and the laterality, number of pulmonary metastatic tumors and DFI were considered to be useful prognostic factors. DFI among them was most closely associated with postoperative survival days. 3) A significant difference in the number of excised nodules was found between the DFI less than 90 days group and the DFI not less than 90 days group, and the number in the former group was significantly larger. 4) It was suggested that postoperative observation and close examination for pulmonary metastasis at about 3 months intervals after of primary focus are important, and also, positive or negative pulmonary metastasis as the time of examination, particularly, the number metastatic nodules of less than 5 or not less than 5, are reflected in the prognosis. 5) Expected survival time after the first operation pulmonary metastasis appeared possible to estimate as 3.0 x DFI days.

A case of pulmonary metastases of synovial sarcoma surviving for a long period after active surgery.

The patient was a 24-year-old woman. She underwent marginal excision of synovial sarcoma originating in the right femoral soft tissue at the age of 18, followed by local radiation therapy and chemotherapy. At the age of 20, abnormal shadow was indicated in bilateral lungs on plain roentgenography of the chest, and she underwent open-chest surgery for excision of bilateral pulmonary metastatic tumors. She was subjected to bilateral thoracotony repeatedly thereafter for a total of 5 times, and all of the metastatic foci detected in each operation were resected. Consequently, she has been surviving for a long period of about 7 years. The therapeutic outcome of tumor of bone soft tissue is greatly affected by its pulmonary metastasis. Although the therapeutic outcome has recently been improved by large dose chemotherapy and active excision of metastatic foci in the lung, survival time in the case of synovial sarcoma, giant cell sarcoma and Ewing's sarcoma is not very long yet. In the present study, we observed a patient who had lung metastasis of synovial sarcoma of which prognosis is generally considered poor has been surviving for a long time of 7 years owing to active excision of pulmonary metastatic foci. The relation of the present prognostic factors to those in the past were compared.

Flight coverall microclimate evaluation using a Japanese type sweating mannequin.

BACKGROUND: It is important to examine the microclimate of a pilot's clothing to evaluate the heat stress the clothing imparts on a pilot, but problems arise with regard to individual variation (phenotype of the pilot, sweating dose, wear condition of the flight coveralls, etc.). HYPOTHESIS: The present study used a Japanese type sweating mannequin (TOM-III) to evaluate the microclimate of flight coveralls. METHODS: TOM-III (TOYOBO, Co., Ltd., Tokyo, Japan) has a characteristic Japanese human body type, and the sweat rate and body temperature can be regulated (240 g of sweat per hour, 36.0 degrees C). The microclimate of the clothing was evaluated by means of 20 temperature/humidity sensors located on the surface of the mannequin skin. TOM-III wore four different assemblies of clothing (CWU-66/P USAF chemical defense flight coverall; JASDF summer-type flight coverall which is similar to the nonchemical defense USAF flight coverall; ordinary 100% cotton underwear; and CWU-66/ P with cotton underwear). We put TOM-III in climatic chamber (21.2 +/- 0.5 degrees C, 50.0 +/- 3.0% relative humidity (RH), 0.1 m air flow x s(-1), dressed in test clothing, and measured the inside temperature (IT) and inside relative humidity (IRH) of the clothing for 60 min (10 min non-sweating, 30 min sweating and 20 min non-sweating). RESULTS: Approximately 5 min after the onset of sweating, the IRH of the CWU-66/P (38.0%) was lower than JASDF flight coverall (42.1%; p < 0.01). At the end of the sweating period, the IRH of the CWU-66/P and the JASDF flight coverall were 46.2% and 52.6%, respectively (p < 0.01). The results indicated that the CWU-66/P flight coverall was better suited for heat stress than the JASDF summer-type flight coverall. CONCLUSIONS: TOM-III may be useful for microclimate evaluation and/or the development of clothing without considering individual variation against various climatic conditions.

Tissue response to fast-setting calcium phosphate cement in bone.

Fast-setting calcium phosphate cement (FSCPC) is a promising new bioactive cement with a significantly short setting time (approximately 5-6 min) compared to conventional calcium phosphate cement (c-CPC) (30-60 min) at physiologic temperatures. As a result of its ability to set quickly, it is applicable in surgical procedures where fast setting is required. In this study, FSCPC was implanted in rat tibiae to evaluate tissue response and biocompatibility. FSCPC was converted to hydroxyapatite (HAP) in bone faster than c-CPC in the first 6 h. By 24 h, significant amounts of both FSCPC and c-CPC had been converted to HAP. The conversion of FSCPC into HAP further proceeded gradually, reaching 100% within 8 weeks. Infrared spectroscopic analysis disclosed the deposition of B-type carbonate apatite, which is a biological apatite contained in human dentin or bone, on the surface of the FSCPC. Histologically, FSCPC showed a tissue response similar to that of c-CPC. A slight inflammatory reaction was observed in the soft tissue apposed to both cements in the early period, and new bone was formed along the surface of the FSCPC at the adjacent bone. However, no resorption of either cement by osteoclasts or macrophages was observed within 8 weeks. We conclude that FSCPC is superior to c-CPC in clinical applications in oral and maxillofacial, orthopedic, plastic, and reconstructive surgery, since it shows a faster setting time and higher mechanical strength in the early period that are required in these surgical procedures, as well as osteoconductivity and excellent biocompatibility similar to that of c-CPC.
