Oscillatory potentials in electroretinograms of the human macular region.

Using focal stimuli, we successfully recorded oscillatory potentials (OPs) in electroretinograms of the macular regions of 72 normal volunteers. The OPs consisted of three to four wavelets with a mean peak interval of approximately 6.5 msec, consistent with that recorded with conventional full-field stimuli over the entire retina. The changes of amplitude in response to the spot sizes and ring stimuli suggested that the distribution of OPs is different from that in a- and b-waves in human macular region.

Synthesis of prostaglandin E in rabbit eyes with topically applied epinephrine.

Using radioimmunoassay techniques, we measured the amounts of prostaglandin E (PGE) in the aqueous humor and vitreous body of 22 phakic and ten aphakic rabbit eyes. Either epinephrine and placebo, epinephrine and indomethacin, indomethacin and placebo, or placebo and placebo were administered topically for 5 months. Treatment of aphakic eyes was initiated 1 month after intracapsular lens extraction. Topically applied epinephrine apparently induced the synthesis of prostaglandin, manifested by elevated PGE in the aqueous and vitreous. Phakic eyes treated with epinephrine and placebo had mean PGE levels of 407.33 pg/ml in the aqueous and 177.0 pg/ml in the vitreous, whereas control eyes given only placebo had mean levels of 165.83 in aqueous and 59.17 in vitreous. Topically applied indomethacin inhibited epinephrine-induced synthesis of PGE in the aqueous humor, but had no significant effect in the vitreous. PGE levels, higher in placebo-treated aphakic eyes than in phakic ones, were elevated further by epinephrine treatment (from 388.40 to 1851.60 pg/ml in aqueous of aphakic eyes, and from 236.40 to 850.60 pg/ml in vitreous also of aphakic eyes). Our findings relate to the pathogenesis of epinephrine-induced maculopathy and to the mechanism of the ocular hypotensive effect of epinephrine.

[5-FU/ADR/MMC combined hepatic infusion in liver metastases from gastric cancer].

Twenty patients with liver metastases from gastric cancer judged non-resectable and the factors limiting their survival were treated with 5-fluorouracil (5-FU), adriamycin (ADR) and mitomycin C (MMC) combined hepatic arterial infusion chemotherapy. The catheter was placed into the hepatic artery, via the left subclavian artery in every case, and 5-FU 334 mg/m2 qw, ADR 20 mg/m2 q4w, MMC 2.7 mg/m2 q2w was administered bolusly. Myelo-suppression (WBC less than 2,000 or platelet less than 50,000) occurred in 25% of the patients, but this therapy was tolerable in all patients without major complication. The response rate in evaluable cases by CT-scan was 76.5% (4CR + 9PR/17), and 50% survival was 407 days. A significant correlation between the existence of extra-hepatic lesions and 50% survival (extra-hepatic lesions (-): 196 days vs (+): 464 days) was noted. The comparison between responder and non-responder in terms of 50% survival and the cause of death showed the importance of control of liver metastases for improved survival.

Molecular cloning and sequence analysis of cDNAs encoding porcine kidney D-amino acid oxidase.

Complementary DNAs encoding D-amino acid oxidase (EC 1.4.3.3, DAO), one of the principal and characteristic enzymes of the peroxisomes of porcine kidney, have been isolated from the porcine kidney cDNA library by hybridization with synthetic oligonucleotide probes corresponding to the partial amino acid sequences. Analysis of the nucleotide sequences of two clones revealed a complete 3211-nucleotide sequence with a 5'-terminal untranslated region of 198 nucleotides, 1041 nucleotides of an open reading frame that encoded 347 amino acids, and a 3'-terminal untranslated region of 1972 nucleotides. The deduced amino acid sequence was completely identical with the reported sequence of the mature enzyme [Ronchi, S., Minchiotti, L., Galliano, M., Curti, B., Swenson, R. P., Williams, C. H. J., & Massey, V. (1982) J. Biol. Chem. 257, 8824-8834]. These results indicate that the primary translation product does not contain a signal peptide at its amino-terminal region for its translocation into peroxisomes. RNA blot hybridization analysis suggests that porcine kidney D-amino acid oxidase is encoded by three mRNAs that differ in size: 3.3, 2.7, and 1.5 kilobases. Comparison of the sequences of the two cDNA clones revealed that multiple polyadenylation signal sequences (ATTAAA and AACAAA) are present in the 3'-untranslated region, making the different mRNA species. The efficiency of 3' processing of the RNA was quite different between the two signal sequences ATTAAA and AACAAA. Southern blot analysis showed the presence of a unique gene for D-amino acid oxidase in the porcine genome.
