[Residual risk research of HIV infection after blood screening in one county in China].

OBJECTIVE: To estimate the risk of HIV infections from blood transfusions, to evaluate existing blood screening measures, and to make recommendations to improve current blood screening practices. METHODS: Research was conducted in a county blood station. A statistical model and retrospective method were used to analyse the data. RESULTS: Between June 1995 and March 1998, 19 blood donors tested positive for HIV antibodies and among them, 7 were repeat blood donors. Twenty - two blood recipients were identified from these 7 donors during six months, 9 are still alive today, while one patient was infected with HIV from a blood transfusion while undergoing an operation. The residual risk of the blood station using a statistical model and retrospective methods were 1/1,773 - 1/28,867 and 1/2,008 - 1/2,730 donations respectively. CONCLUSION: The residual risk of HIV after screening in the county blood station was high. We recommend further improvements to current blood screening practices.

[The survey of HIV prevalence among drug users in Guangxi, China].

OBJECTIVE: To understand the distribution and related risk factors of HIV infection among drug users in Guangxi. METHOD: Three hundred and twenty five drug users were tested for serum HIV antibody, with enzyme linked immunosorbent assay. RESULTS: One hundred and sixteen drug users (35.7%) were found to be HIV positive. The main risk factors associated with HIV infection were intravenous drug users and needle sharing among drug users. The HIV prevalence was 40.9% (114/279) among IDUs, and only 4.3% (2/46) among oral drug users. The phenomenon of sharing injecting equipment (76.7%) among intravenous drug users in Guangxi represents a very serious problem. CONCLUSION: There is a high risk of HIV infection among drug users in Guangxi. Prevention programs urgently need to be implemented.

[Lung injury caused by passive smoking and its effects on cytokines in rats].

OBJECTIVE: A rat model with chronic bronchitis was replicated by passive inhalation of cigarette smoking fume to study its long-term effects on lung injury and nitric oxide (NO), interluekin-6 (IL-6), interleukin-8 (IL-8). METHODS: Levels of nitrogen dioxide (NO(2)) and nitrogen trioxide (NO(3)) were measured with spectrophotometry in rats indicating their level of nitric oxide (NO). Levels of IL-6 and IL-8 were determined by enzyme-linked immunosorbent assay (ELISA). RESULTS: Levels of NO in serum, bronchial alveolar lavage fluid (BALF) and lung tissue in the smoking group were significantly lower than those in the normal controls (P < 0.01). But, levels of IL-6 and IL-8 were higher in the smoking group than those in the controls. CONCLUSION: Long-term passive smoking could cause injury of lung tissue to certain extent, reduction in secretion of NO in endothelial cells and damage to pulmonary vessels.

[Clinical application of irradiated drug-containing porcine-cornea to patients with ocular burns].

OBJECTIVE: To explore a new method for the management of patients with ocular burns. METHODS: Fifty-five cases of patients with ocular burns (in 88 eyes) were randomly divided into treatment and control groups. Thirty cases in treatment group with 49 eyes were transplanted with irradiated drug-containing (ofloxacin, acetyl cysteine and reduced glutathione) porcine-cornea. 25 cases in control group with 39 eyes were treated with routine program. RESULTS: Thirty-two eyes were rescued in treatment group with the cure rate of 65.3%. But only 17 eyes were saved in control group with the cure rate of 43.59%, indicating significant difference of the cure rate between the two groups (P < 0.05). CONCLUSION: Irradiated drug-containing porcine-cornea might well be an ideal therapeutic material for the management of patients with ocular burns.

[The relationship of cellular DNA content with clinical stage and biological features of colorectal cancer].

The purpose of this study was to explore the relationship of cellular DNA content with clinical stage and biological features of cancer. Flow cytometry was performed on fresh specimens from 86 patients from 1997 to 1998. Forty-five (53.3%) specimens were found to contain cells with abnormal DNA (DNA nondiploidy). Although none of the sex, age, site, differentiation variables correlated with flow cytometric DNA ploidy, nondiploidy was associated with Dukes' stage and lymph node metastasis. Duke's A stage tumors were more frequently diploid than were more advanced tumors, but no difference between Duke's B, C and D stages were observed. These findings suggest that ploidy is associated with some pathological factors.

[Quantification analysis of vascular endothelial cells in human solid tumor].

Angiogenesis is essential for the growth and metastasis of solid tumors. Intratumoral microvessel count, which represents a measure of tumor angiogenesis, has been associated with prognosis of patients with a variety of malignancies. But till now, it is still a problem to quantitate the angiogenesis of tumors exactly. In this study, the endothelial cells in 20 solid tumors were conjugated with anti-CD34 monoclonal antibodies labelled with red-fluorescent substances, and were observed by fluorescence microscopy and quantitated by flow cytometry. The results of flow cytometry were analysed by t-test. The correlation was analysed between percentage of flow cytometry and vessel counts (light microscopy, x 200) in each tumor which was stained with anti-CD34 monoclonal antibodies and anti-VIII factor related antigen polyclonal antibodies by using immunohistochemistry. The results showed there was significant difference between the treatment group and control group in flow cytometry, and a significant correlation between the endothelial cells percentage by flow cytometry and the vessel counts by immunohistochemistry was observed, although the average vessel counts were approximately three times with CD34 staining as much as with VIII factor related antigen staining. These data indicate that flow cytometry as a novel method can quantitate angiogenesis of tumors exactly and quickly.

[Immunohistochemical study of type I and type II collagen in the remodelling of the supraosseous tissue of mandibular condyle].

OBJECTIVE: Previous studies focused on the pathological changes of the supraosseous tissue of mandibular condyle (STMC), but little is known about extracellular matrix during the remodelling activity of temporomandibular joint (TMJ). This study will examine the characteristics of type I and type II collagen during this process. METHODS: The animal model was established by extracting the left lower molars of adult rabbits. Temporomandibular joints of two sides were taken after 2 weeks, 1 month and 3 months respectively, and embedded in paraffin. Then 5 microns-thick sections were cut and processed for immunohistochemistry and haematoxylin-eosin staining. An immunohistochemical ABC method was used to determine the local changes of type I and type II collagen. The positive cells were counted in order to analyze quantitative changes. RESULTS: After loss of teeth from 2 weeks to 3 months, the number and arrangement of chondrocytes changed. Some chondrocyte clusters were also observed in the non-extraction side of TMJ. Most STMC cells stained positively with anti-type I collagen, but weakly in chondrocytes and hypertrophic chondrocytes. On the other hand, type II collagen only existed in chondrocytes and some hypertrophic chondrocytes. The expression of type I collagen decreased after 2 weeks and became stronger after 1 and 3 months, furthermore distributed unevenly in STMC. Some hypertrophic chondrocytes in the non-extraction side showed strong positive staining with anti-type I collagen. Type II collagen in two sides decreased after 2 weeks, then became stronger after 1 and 3 month, but the whole level was less than that of the control group. Expressions of two kinds of collagen were compared between two sides. Type I collagen in the non-extraction side was stronger than that in the extraction side, while type II collagen had an opposite result. CONCLUSION: Adult rabbits have a limited remodelling ability, and the synthesis and secretion of type I and type II collagen will change when the remodelling is beyond a proper extent, which will lead to impaired resistance and elasticity of fibrocartilage.

[A preliminary study on tissue culture of stem apex of Chrysanthemum morifolium].

To solve the breed degeneration of Chrysanthemum the stem apex of Chrysanthemum morifolium used as explants were inoculated on MS medium with different pH and supplemented with different ratio of plant hormone. The results showed that when pH was between 5.8 and 6.4, the rate of callus were all over 80%. While explants inoculated on MS medium supplemented with NAA 0.2 mg/L and KT 0.1-0.2 mg/L, the rate of callus was noticeably higher than that of NAA 0.2 mg/L and 6-BA 1-3 mg/L. The preferable medium which roots could be induced was 1/2 MS + NAA 0.2 mg/L.

[Influence on luminescence of Tb o-aminobenzolate complexes by second ligand].

Two Tb o-aminobenzolate complexes with different second ligands (phenanthroline and triphenyl phasphine oxide) were synthesized. Their structures and fluorescent properties were investigated by fluorescence spectroscopy and FTIR spectroscopy. The results indicate that the second ligand affected on the complexes luminescence by changing its structure.

Attenuation of temporary focal cerebral ischemic injury in the mouse following transfection with interleukin-1 receptor antagonist.

The proinflammatory cytokine interleukin-1 beta (IL-1beta) is thought to play an important role in the stimulation of the inflammatory response following ischemia and reperfusion. This study investigated the inflammatory effect of IL-1beta during transient focal cerebral ischemia and reperfusion in the mouse transduced with the interleukin-1 receptor antagonist (IL-1ra) gene. An adenoviral vector encoding, either the human IL-1ra gene (AdRSVIL-1ra) or the LacZ gene (AdRSVlacZ) or normal saline, were injected into the right lateral ventricles of adult CD-1 mice (n=96). Five days later, the mice received 1 h temporary middle cerebral artery occlusion (tMACAO) followed by 23 h reperfusion. Cerebral blood flow (CBF), infarct volume, blood-brain barrier (BBB) permeability, and the number of intracellular adhesion molecule-1 positive vessels were measured to determine the effect of IL-1beta during postischemic reperfusion. Infarct volume in the AdRSVIL-1ra-transduced mice was markedly reduced compared to the AdRSVlacZ-transduced and saline-injected mice (36.0+/-5.3 mm(3) vs. 60.0+/-6.2 mm(3), 69. 5+/-6.3 mm(3), after 23 h of reperfusion, n=6-8 per group, p<0.05). BBB disruption and intracellular adhesion molecule-1 expression (135+/-23 vs. 311+/-40 and 357+/-51, n=6-8 per group, p<0.05) in the AdRSVIL-1ra-transduced mice were also less than that of the AdRSVlacZ-transduced and saline-injected mice. Our studies demonstrated that overexpression of IL-1ra in the mouse brain can downregulate intracellular adhesion molecule-1 expression both in the cortex and basal ganglia, which suggests that IL-1beta may play an important role in the activation of the inflammatory response during focal cerebral ischemia by promoting leukocyte adhesion to endothelial cells. The decrease of BBB disruption in AdRSVIL-1ra-transduced mice suggests that the endothelial cells may be a target for IL-1beta during postischemic reperfusion.

Mutations of human topoisomerase II alpha affecting multidrug resistance and sensitivity.

Two mutations, R450Q and P803S, in the coding region of the human topoisomerase II alpha gene have been identified in the atypical multidrug resistant (at-MDR) cell line, CEM/VM-1, which exhibits resistance to many structurally diverse topoisomerase II-targeting antitumor drugs such as VM-26, doxorubicin, m-AMSA, and mitoxantrone. The R450Q mutation mapped in the ATP utilization domain, while the P803S mutation mapped in the vicinity of the active site tyrosine of human topoisomerase II alpha. However, the roles of these two mutations in conferring multidrug resistance are unclear. To study the roles of these two mutations in conferring multidrug resistance, we have characterized the recombinant human DNA topoisomerase II alpha containing either single or double mutations. We show that both R450Q and P803S mutations confer resistance in the absence of ATP. However, in the presence of ATP, the R450Q, but not the P803S, mutation can confer multidrug resistance. The R450Q enzyme was shown to exhibit impaired ATP utilization both for enzyme catalysis and for its ability to form the circular protein clamp. Interestingly, an unrelated mutation, G437E, which is also located in the same domain as the R450Q mutation, exhibited multidrug hypersensitivity in the absence of ATP. However, in the presence of ATP, the G437E enzyme is only minimally hypersensitive to various topoisomerase II drugs. In contrast to the R450Q enzyme, the G437E enzyme exhibited enhanced ATP utilization for enzyme catalysis. In the aggregate, these results support the notion that the multidrug resistance and sensitivity of these mutant enzymes are due to a specific defect in ATP utilization during enzyme catalysis.

Activation of topoisomerase II-mediated excision of chromosomal DNA loops during oxidative stress.

Hydrogen peroxide (H2O2), a reactive oxygen species (ROS), is known to induce oxidative stress and apoptosis. U937 cells treated with H2O2 were shown to produce high molecular weight (HMW) DNA fragments approximately 50 to 100 kb in size in <1 min. The formation of these HMW DNA fragments is reversible and shown to be mediated by DNA topoisomerase II (TOP2). Following this initial event, formation of irreversible HMW DNA fragments and nucleosomal ladders occurs. Our results thus demonstrate a potential role of TOP2 in oxidative damage of DNA and apoptotic cell death.

Expression of intercellular adhesion molecule 1 (ICAM-1) is reduced in permanent focal cerebral ischemic mouse brain using an adenoviral vector to induce overexpression of interleukin-1 receptor antagonist.

Our previous studies have demonstrated that overexpression of recombinant human interleukin-1 receptor antagonist protein (IL-1ra) via gene transfer can reduce ischemic brain injury. However, the mechanism of action of IL-1ra in ischemia is unclear. Since interleukin-1 can up-regulate intercellular adhesion molecules in endothelium, the present study was designed to determine whether overexpression of the IL-1ra can reduce the expression of intercellular adhesion molecule-1 (ICAM-1) after ischemic injury. Normal saline or adenovirus vector (1x109 particles) encoding the human IL-1ra gene (Ad.RSVIL-1ra) or the Escherichia coli LacZ gene (Ad.RSVlacZ) was injected into the right lateral cerebral ventricle of adult CD-1 mice. After five days, permanent middle cerebral artery occlusion (MCAO) was achieved for 24 h using an intraluminal suture. Cerebral blood flow was monitored by transcranial laser Doppler flowmetry to verify the occlusion. ICAM-1 protein was quantified using Western blot analysis and localized using immunohistochemistry. After MCAO, surface blood flow in the ischemic hemisphere was decreased to 9-11% of the baseline. There were fewer ICAM-1 positive vessels in the ischemic cortex of the Ad.RSVIL-1ra transfected mice than in the Ad.RSVlacZ transfected and saline treated mice (138+/-19 vs. 249+/-25, 284+/-22, p<0.05). Western blot analysis shows that ICAM-1 protein decreased 50-60% in the Ad. RSVIL-1ra group compared to the other two groups. There were no significant differences in the numbers of positive vessels in the ischemic basal ganglia and contralateral hemisphere among the three groups. Our studies suggest that IL-1ra overexpression can down-regulate the expression of ICAM-1 in the ipsilateral cortex in ischemic mice. Interleukin-1 may play an important role in the activation of inflammatory reaction during focal cerebral ischemia by promoting leukocyte adhesion on the endothelium cells.

Focal cerebral ischemia in the mouse: description of a model and effects of permanent and temporary occlusion.

Our study is to demonstrate the advantages and disadvantages of middle cerebral artery occlusion (MCAO) model in the mouse. CD-1 mice had permanent MCAO for 24 h, or temporary occlusion for either 1 h followed by 23 h of reperfusion or 2 h of occlusion with 22 h of reperfusion. The infarct volume and blood-brain barrier disruption were smaller in the 1-h/23-h temporary occlusion than in either the 24-h permanent occlusion group or the 2-h/22-h temporary occlusion group (p<0.05). Our study demonstrates that blood flow, infarct volume, and blood-brain barrier disruption remain important markers of focal cerebral ischemia.

Direct injection versus liquid-liquid extraction for plasma sample analysis by high performance liquid chromatography with tandem mass spectrometry.

Direct injection versus liquid-liquid extraction for post-dose human plasma sample analysis by high performance liquid chromatography with tandem mass spectrometry (LC/MS/MS) have been studied using a drug candidate compound. For the direct-injection method, an Oasis(R) HLB column (1 x 50 mm, 30 micrometer) was used as the on-line extraction column and a conventional Waters symmetry C18 column (3.9 x 50 mm, 5 micrometer) was used as the analytical column. Each plasma sample (100 microL) was mixed with 100 microL of a working solution of the internal standard in aqueous 0.05 M ammonium acetate (pH 6.9), and portions (10 microL) of these samples were then injected into the LC/MS/MS system. For the liquid-liquid extraction method, a YMC Basic C18 column (2.0 x 50 mm, 5 micrometer) was used as the analytical column. Each sample (0.5 mL) was extracted with methyl tert-butyl ether and the extract was reconstituted and injected into the LC/MS/MS system. The total analysis time for both methods was 2.0 min per sample. The accuracy, inter-day precision and intra-day precision obtained from the quality control samples were within 8% for both methods. The analysis results of post-dose human plasma samples showed that the deviations of 91% of the concentrations obtained using the direct-injection method were within +/-20% from the concentrations obtained using the liquid-liquid extraction method, and the overall average percentage deviation was -1.5%. The results showed that the two methods were equivalent in terms of total chromatographic run time, accuracy and precision. However, for a batch of 100 samples, the sample preparation time for the direct-injection method was only about 25% of the time required for liquid-liquid extraction. This decrease in sample preparation time resulted in the doubling of the overall sample analysis throughput.

Suppression of carrageenan-induced inflammation in vivo by an extract of the Chinese herbal remedy Tripterygium wilfordii Hook F.

OBJECTIVE AND DESIGN: The current studies investigated the effect of subacute administration of an ethyl acetate extract of the Chinese herbal remedy, Tripterygium wilfordii Hook F (TWHF) on the inflammatory response induced by carrageenan in the rat. MATERIALS AND SUBJECTS: F344 rats were randomly divided into 3 groups of seven animals. TREATMENT: The rats were treated orally for 5 days with either vehicle or an ethyl acetate extract of TWHF at a dose of 270 mg/kg, equivalent to 1/5 of the dose causing death in 50% of the animals. METHODS: Air pouches were induced subcutaneously on the backs of rats and injected with carrageenan on the last day of treatment. Sixteen hours after carrageenan challenge, the air pouches were removed and analyzed. Student's t-test was employed for statistical analysis. RESULTS: The number of exudate cells and the concentration of PGE2, nitrite and TNF-alpha in the exudate obtained from TWHF-treated animals were significantly reduced (by 69%, 78%, 57% and 77%, respectively) compared to that from vehicle-treated animals. mRNA for cyclooxygenase-2 was markedly suppressed in the air pouch lining tissue of TWHF treated rats (p < 0.001). In contrast, PGE2 content of the kidney and stomach and the production of PGE2, nitrite and TNF-alpha by spleen cells was not affected by treatment with TWHF. CONCLUSIONS: These results indicate that TWHF exerts a significant anti-inflammatory activity in vivo.

Conformations of Gly(n)H+ and Ala(n)H+ peptides in the gas phase.

High-resolution ion mobility measurements and molecular dynamics simulations have been used to probe the conformations of protonated polyglycine and polyalanine (Gly(n)H and Ala(n)H+, n = 3-20) in the gas phase. The measured collision integrals for both the polyglycine and the polyalanine peptides are consistent with a self-solvated globule conformation, where the peptide chain wraps around and solvates the charge located on the terminal amine. The conformations of the small peptides are governed entirely by self-solvation, whereas the larger ones have additional backbone hydrogen bonds. Helical conformations, which are stable for neutral Alan peptides, were not observed in the experiments. Molecular dynamics simulations for Ala(n)H+ peptides suggest that the charge destabilizes the helix, although several of the low energy conformations found in the simulations for the larger Ala(n)H+ peptides have small helical regions.
