[Tumor growth inhibitory effects of SMANCS, a poly (styrene-maleic acid) conjugated derivative of neocarzinostatin, on various tissue culture cells].

Growth inhibitory effects of neocarzinostatin (NCS), and its conjugated derivative with poly (styrene-maleic acid), SMANCS, were examined on various tissue culture cells consisting of 13 lymphatic and 7 non-lymphatic cells (15 cell lines, 5 primary culture cells; 14 in suspension culture cells, 6 in monolayer cells). Both agents showed almost similar concentrations for the growth inhibition against 5 non-lymphatic cell lines (an average minimum growth inhibitory concentration of NCS, 0.076 microgram/ml, and SMANCS, 0.094 microgram/ml, a ratio of 1 : 1.24 (W/W), or 1.0 : 0.9 (mol/mol]. However, SMANCS showed a much less growth inhibitory effects against lymphatic 10 cell lines than NCS. Namely, an average 50% growth inhibitory concentration of NCS was 0.019 microgram/ml whereas that of SMANCS was 0.091 microgram/ml (a ratio of 1 : 4.79). From the consideration of each molecular weight, this ratio is 1 : 3.6. Similar results were obtained in experiments in which 2 primary non-lymphatic cells and 3 primary lymphatic cells were used. Although the largest drawback with NCS was known to be leukopenia, present results indicated the less toxicity of SMANCS than NCS on lymphatic cells and may provide a positive aspect of the new derivative, such as less incidence of leukopenia.

[A case of pseudoxanthoma elasticum showing various heteroptics due to infarction of right parieto-occipital lobe].

A case of pseudoxanthoma elasticum showing various heteroptics due to infarction of the right parieto-occipital lobe was reported. The patient was 48-year-old male. Angiography revealed an obstruction of the right internal carotid artery, aneurysmal dilation of the right posterior communicating artery, and obstructions of the right parieto-occipital artery and calcarine artery. A low density area was found in right occipital lobe by CT. MRI clearly identified lesions in the right cuneus and precuneus. Pseudopsia in the left homonymous quadrantanopsia, various types of visual illusion in the remaining visual field, and moreover, palinopsia were existed. Visual illusion was recognized in the early stage after the onset, while the pseudopsia was recognized about one month after the onset. Both abnormalities tended to improve from the peripheral region with the improvement of the defect in the visual field. Administration of phenytoin resulted in an improvement in the pseudopsia but it resulted in an aggravation in the visual illusion. Thus, the mechanisms of development of this phenomena were thought to be different from each other. It was found by dynamic CT that the blood flow in the primary visual cortex was rather increased in the affected site in comparison with that in the healthy site. It was speculated that the pseudopsia in the defective region of the visual field reflected a pathological repair process represented by a development of the collateral circulation. On the other hand, the visual illusion was thought to originate from visual unstability due to incomplete damage of the visual pathway at the affected site.

[A report of a resected case of malignant lymphoma combined with follicular carcinoma of the thyroid].

A 66-year-old female administered with complaining of cervical mass. All of the thyroid function tested were within normal limits. A diagnosis of Malignant lymphoma was done by fine aspiration cytology. Total thyroidectomy and dissection of bilateral regional lymph nodes were performed. And additional chemotherapy is continued. The histological findings revealed diffuse lymphoma, mixed type (LSG), but there is no invasion of the lymphoma cell to the lymph node. There are also two follicular carcinomas with metastasis to the dissected regional lymph nodes. In the residual thyroid tissue, the findings of chronic thyroiditis was shown histopathologically.

[An ultrastructural study of the rabbit intraovarian lymphatic capillaries after ovulatory stimulus].

Fine distribution and ultrastructural changes in intraovarian lymphatics were studied at accurately timed intervals after the injection of human chorionic gonadotropin (hCG). Four and six hours after hCG injection, edema around the lymphatic capillaries in the theca externa followed that around the blood capillaries. The lymphatic capillaries were markedly dilated and displayed wide openings between neighboring endothelial cells in them. At these stages, lysosome disappeared from the lymphatic endothelium, while macrophages with numerous lysosomes appeared around the lymphatic capillaries and some of them entered the lumen. Eight hours after hCG injection, the ultrastructure of the lymphatic capillaries resumed the appearance of their pre-injection stage. Three days after hCG injection, lymphatic capillaries were observed among the theca lutein cells, but not among the granulosa lutein cells. Edema around the lymphatic capillaries and openings between neighboring endothelial cells reappeared as shown 4 and 6 hours after hCG injection. Five and seven days after hCG injection, macrophages and degenerated lutein cells were observed in the lymphatic capillaries. Fourteen days after hCG injection, lymphatic endothelium began to degenerate, but that of the blood capillaries showed no degenerative signs. These results suggest that the functions of the lymphatic capillaries are absorption and transport of excess fluid, waste products and hormones.

A 59-kilodalton protein associated with progestin, estrogen, androgen, and glucocorticoid receptors.

Previous studies of the anti 8.5S progestin receptor monoclonal antibody KN 382/EC1 showed that it was specific for nontransformed progestin receptors. However, with different methods of tissue disruption and the use of protease inhibitors, we found that other nontransformed steroid receptors formed immune complexes with KN 382/EC1. Binding of the antibody to rabbit uterine estrogen, progestin, and androgen and liver glucocorticoid receptor systems was characterized by sucrose density gradient centrifugation, high-pressure liquid chromatography (HPLC), immunoadsorption, and immunoblotting. Immobilized KN 382/EC1 adsorbed both Mr 59,000 and Mr 92,000 proteins. The Mr 92,000 protein appeared to be bound to the antigenic Mr 59,000 protein, and the two proteins were present in apparently the same stoichiometric relationship in several tissues. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis of immunoadsorbed material revealed appreciable amounts of both proteins in testis, stomach, lung, liver, uterus, and kidney. Only trace amounts were found in skeletal or heart muscle, and none was found in blood serum. Cleveland digestion of isolated Mr 59,000 and 92,000 proteins revealed dissimilar peptide constituents. Immunoblots of material from uterus and liver resulted in staining of the Mr 59,000 protein but not the Mr 92,000 protein. We conclude that similar antigenic determinants reside in components of several nontransformed steroid receptors and they reside on an Mr 59,000 protein. It is likely, therefore, that there are common components present in nontransformed steroid receptors.
