Impact of CO2 addition to milk on selected analytical testing methods.

The addition of CO2 to raw milk and dairy products controls the growth of psychrotrophic bacteria at refrigeration temperatures. The objective of this study was to determine the effects of dissolved CO2 in milk on the performance of four important routine testing methods: antibiotic residue test, freezing point test, infrared milk component analysis, and alkaline phosphatase test. Raw or pasteurized whole milk was carbonated at <4 degrees C to contain approximately 0 (control), 200, 400, 600, and 1000 ppm of CO2. The addition of CO2 to raw milk up to 1000 ppm had no effect on the performance of the three antibiotic (beta-lactams) residue tests: IDEXX SNAP, Charm II Sequential Tablet, and Delvo-P Ampule. Milk freezing point decreased linearly with increasing concentration of dissolved CO2, from -0.543 degrees H (control) to -0.595 degrees H (1000 ppm). Carbonation to 1000 ppm decreased milk pH (measured at 38 degrees C) from 6.61 (control) to 6.15 (1000 ppm). The effects of CO2 on milk freezing point and pH were reversible upon removal of dissolved CO2. Increased CO2 levels in milk changed the infrared absorption spectrum of milk and caused the corrected lactose readings to decrease and the corrected fat B readings to increase. For the alkaline phosphatase tests, 0 (none), 0.05, 0.1, and 0.2% raw milk were deliberately added to pasteurized milks of six levels of carbonation (0 to 1000 ppm). The addition of CO2 did not influence the ability of Fluorophos, Charm PasLite, and Scharer Modified Rapid tests to differentiate between a pasteurized milk and a pasteurized milk with raw milk contamination.

The effect of Bcl-2 adenovirus against murine hepatocyte apoptosis caused by tumor necrosis factor alpha and D-galactosamine.

OBJECTIVE: To evaluate the role of Bcl-2 family proteins in hepatic apoptosis caused by TNF-alpha and D-galactosamine. METHODS: We induced mouse liver injury with TNF-alpha and D-galactosamine, and detected hepatic apoptosis, the expression of Bcl-2, Bax, and Bak proteins on hepatocytes by using TUNEL or immunohistochemistry, respectively. We also observed the expression of Bcl-2 protein on hepatocytes infected with Bcl-2 adenovirus vector and its protection against hepatocyte apoptosis. RESULTS: Hepatocyte apoptosis was induced in BalB/c mice pretreated with TNF-alpha plus D-galactosamine, accompanying the enhanced expression of Bax, Bak proteins in hepatocytes. Bcl-2 protein was expressed in murine hepatocytes and lasted at least 1 month after injection of Bcl-2 adenovirus vector, which also lowered ALT level from (1372.9+/-251.4)U/L to (796.5+/-78.7)U/L and reduced hepatocyte apoptosis caused by TNF-alpha and D-galactosamine. CONCLUSIONS: The enhanced expression of Bax, Bak proteins may play a role in hepatocyte apoptosis induced by TNF-alpha and D-galactosamine. D-galactosamine adenovirus vector can partially reduced hepatocyte apoptosis induced by TNF- alpha and D-galactosamine.

[Studies of a new adsorbent cross-linked agar beads entrapped attapulgite clay for hemoperfusion].

The preparation and properties of a new adsorbent cross-linked agar beads entrapped attapulgite clay (CAA) are reported in detail. Medicinal attapulgite clay made in China was encapsulated with agar, shaped in organic solvent, and cross-linked by epichlorohydrin. The products withstood autoclaving at 121 degrees C for half an hour with no breakage, and had a good selective adsorption ability for some model compounds and medicaments such as methylene blue and phenothiazine drugs. Preliminary investigations and SEM photographs indicate that the CAA as an adsorbent is relatively hemocompatible for hemoperfusion.

[A new briquetting method for biomass coal and its influencing factors].

Regarding to the limitation and low economical efficiency of conventional biomass coal briquetter determined by its briquetting method, a new briquetting mechanism using the linkage function of biomass fiber was established. The effect of different coal, briquetting pressure, biomass shape and content, water content on new brequtting mechanism was studied preliminarily. The results showed that the new mechanism was suitable for varied kinds coal and biomass. The briquette mechanical performance was improved as biomass content increased with a reasonable low limitation of about 15%. Pure biomass also could be briquetted. The briquetting pressure which could consolidate the linkage of biomass fiber was no more than 120 MPa. There would be a negative effect at too high pressure. Increasing biomass content and decreasing pressure at certain degree could offset the negative effect of water content significantly, with a permissive limitation over 10% for Datong coal.

Optimization of control parameters for petroleum waste composting.

Composting is being widely employed in the treatment of petroleum waste. The purpose of this study was to find the optimum control parameters for petroleum waste in-vessel composting. Various physical and chemical parameters were monitored to evaluate their influence on the microbial communities present in composting. The CO2 evolution and the number of microorganisms were measured as the activity of composting. The results demonstrated that the optimum temperature, pH and moisture content were 56.5-59.5 degrees C, 7.0-8.5 and 55%-60%, respectively. Under the optimum conditions, the removal efficiency of petroleum hydrocarbon reached 83.29% after 30 days composting.

[Characteristic analysis on different thermal active surfaces in canopy gap of tropical secondary forest].

Microclimatic measurements were conducted in the canopy gap of tropical secondary forest in Xishuangbanna in fog-cool and dry-hot season. The daytime thermal effect of different thermal active surface in the canopy gap was discussed, and the variations of trunk surface temperature near gap edge and of surface temperature on the gap were analyzed. The result shows that the woody-wall surface is new thermal active surface on the vicinage of canopy gap, with the exception of the forest canopy surface, soil surface in the forest gap, and soil surface of the interior. Because of the influence of season, situs and time, the thermal effect of different thermal active surface of gap was significant different. Being subject to fog, the soil surface on the center of canopy gap is an important thermal active surface in the morning; while in the midday and afternoon, because of the influence of incident radiation, the woody wall and soil surface of edge on the east and the woody wall surface of edge on the north are the key thermal active surface of the canopy gap. In the midday, the thermal active was salient on the surface of gap, and in the afternoon, it was salient on the woody wall. The thermal variations of canopy gap of forest are controlled by the thermal characteristics of different thermal active surfaces and by their interaction, affecting the growth of plants.

[Effect of soil moisture and wetting-drying on Mn release in Ca(NO3)2-amended soils].

A study on the effects of two moisture levels (200 and 400 g.kg-1) and wetting-drying on the solubility of Mn in Ca(NO3)2-amended soils showed that water regime was a factor affecting Mn availability. Easily reducible Mn was transformed into soluble and exchangeable Mn, when soil water content was high. Salt had a definite effect on the transformation of easily reducible Mn. NO3- could decrease soil redox potential, and inhibit the transformation of easily reducible Mn. Wetting-drying significantly(alpha < 0.05) decreased the content of soil water-soluble and exchangeable Mn, while Ca(NO3)2 increased the insolubility of Mn, which might eventually lead to the deficiency of soil Mn.

[Effects of human immunodeficiency virus infection on pregnancy outcome].

OBJECTIVE: To investigate the effects of human immunodeficiency virus (HIV) infection on the pregnancy outcome and the incidence of vertical transmission in HIV-positive pregnant women. METHOD: 86 cases of HIV-positive pregnant women and their infants were evaluated retrospectively, and HIV antibody in peripheral blood was detected using enzyme linked immunosorbent assay (ELISA). RESULTS: Compared with normal control group, the incidences of abortion, preterm birth, low birth weight and small for gestational age babies were 9.3%, 14.0%, 16.3% and 10.5%, respectively (P < 0.05); the occurrence of conditional infections of reproductive tract was also higher (P < 0.05); and the incidence of vertical transmission was 12.8% (P < 0.01). CONCLUSIONS: HIV infection in pregnant women markedly increases the risks of abortion, preterm birth, low birth weight and small for gestational age babies, and elevates the incidence of conditional infections of reproductive tract. The incidence of vertical transmission was 12.8%. These data support that it is very important to provide prenatal care to HIV-infected pregnant women and prevent pregnant women from HIV infection.

[The expression of human telomerase reverse transcriptase gene in cord blood hematopoietic stem cells and its significance].

OBJECTIVE: To investigate the expression and significance of human telomerase reverse transcriptase (hTERT) gene in cord blood hematopoietic stem cells. METHODS: Using in situ hybridization techniques, we detect the expression of hTERT gene in cord blood hematopoietic stem cells in different condition during different culture time. RESULTS: hTERT gene was lowly expressed in freshly isolated cord blood CD34+ cells; the positive rate was 13%. It could be increased in 5-7 days when the cells were cultured in vitro, especially in the presence of stem cell factor, interleukin-3(IL-3), IL-6 and Flt-3 Ligand together; the positive rate reached 48%. Transform growth factor-beta 1 and all-trans retinoic acid could repress hTERT gene. CONCLUSION: hTERT gene was lowly expressed in the cord blood CD34+ cells; it could be upregulated in the culture in vitro along with optimal cytokines, but was downregulated by negative regulator and induction differentiation.

[Association between renin-angiotensin system gene polymorphism and type 2 diabetics with stroke in China].

OBJECTIVE: The aim of this study was to explore the association between gene polymorphisms of renin-angiotensin system (RAS) and type 2 diabetes mellitus (DM2) complicated with stroke. METHODS: Angiotensin 1-converting enzyme (ACE) genotypes and type 1 angiotensin II receptor(AT1R) genotypes were investigated with PCR and PCR/DdeI enzyme digesting techniques in 152 cases of DM2. RESULTS: No significant differences in genotype frequencies of AT1R gene were found between DM2 complicated with stroke and that without stroke (P> 0.05). The C allele frequency of AT1R gene in DM2 with stroke was higher than that in DM2 without stroke (6.8% vs 1.9% P< 0.05; OR=3.693, 95% CI: 0.977-13.704). The ACE DD genotype and D allele frequencies were higher in DM2 with stroke than in DM2 without stroke(51.4% vs 33.3%, P<0.05; 69.6% vs 54.5%, P<0.05). There was interaction between AT1R gene A1166C polymorphism and ACE gene I/D polymorphism in the development of stroke in DM2 patients (P<0.05; OR=4.00, 95% CI: 0.733-21.838). Logistic regression analysis showed that ACE DD genotype, systolic blood pressure and serum uric acid level were risk factors of DM2 complicated with stroke. CONCLUSION: It is suggested that ACE gene I/D polymorphism is a susceptible gene for stroke in DM2 patients in China. AT1R gene A1166C polymorphism contributes to the development of stroke in DM2 patients, but it is not an independent risk factor. There is synergistic effect of AT1R A1166C and ACE I/D gene polymorphisms on the occurrence and development of stroke in type 2 diabetics in China.

[Role of tumor necrosis factor receptor 1 and Fas antigen in hepatocellular apoptosis of viral hepatitis B patients].

OBJECTIVE: To evaluate the role of tumor necrosis factor receptor 1 (TNFR1) and Fas antigen in hepatocellular apoptosis and necrosis in viral hepatitis B. METHODS: Seventy paraffin sections from patients with HBV infection were studied. Five cases of normal liver tissues were studied as control. Apoptosis was detected by terminal deoxynucleotidyl transferase mediated dUTP nick end labeling (TUNEL). TNFR1 and Fas antigen were determined by immunohistochemistry. RESULTS: Hepatocellular apoptosis and the expression of TNFR 1 and Fas antigen on hepatocytes were not detected in normal liver tissues. In contrast, there was a strong reaction in TUNEL assay and TNFR1 or Fas antigen expressed on hepatocytes in liver tissues infected with HBV. The degree of apoptosis and the expression of TNFR 1 or Fas antigen on hepatocytes had distinct difference in different hepatitis (P<0.005). There was a positive correlation between degrees of the expression of Fas and hepatic apoptosis (P<0.005), but the similar result was not observed in the expression of TNFR 1 and hepatocellular apoptosis. Our observations also showed that TNFR1 was detected in cytoplasm and cell membrane similarly, and Fas antigen was mainly detected in cytoplasm. There was not positive correlation between degrees of the expression of TNFR1 and Fas antigen on hepatocytes. It was showed that 60.9% (28/46) hepatocytes with strong reaction in TUNEL assay expressed both TNFR1 and Fas antigen. CONCLUSIONS: The degrees of hepatocellular apoptosis and necrosis induced by Fas antigen are stronger than that induced by TNF. The expression of Fas and TNFR 1 on hepatocytes simultaneously may enhance hepatocellular apoptosis in hepatitis B patients.

Analysis of the T cell receptor V delta region gene repertoire in bronchoalveolar lavage fluid (BALF) and peripheral blood of asthmatics.

OBJECTIVE: To explore the role of gamma delta T cells in the airway of asthmatics and to identify the forces which induce and maintain the inflammatory process. METHODS: Peripheral blood (PB) and bronchoalveolar lavage fluid (BALF) were obtained from 7 asthmatic subjects and 7 nonsmoker control subjects. The percentage of gamma delta T cells in the PB and BALF was measured by immunofluorescent staining and flow cytometry. The frequency of usage and the clonality of V delta subfamilies (V delta 1-V delta 3) were assessed by RT-PCR and gene scanning. RESULTS: A higher proportion of gamma delta T cell was detected in the BALF of asthmatic subjects (7.8% +/- 4.7%) than that from control subjects (3.3% +/- 3.0%, P = 0.04). No selective usage for a particular V delta subfamily was found, but the relative expression level of V delta 1 was significantly higher in the asthmatic airway (44% +/- 13%) than in the control (19% +/- 5%, P = 0.0002). In asthmatic subjects, the monoclonal or oligoclonal expansion of gamma delta T lymphocytes was predominant in the BALF, especially V delta 1+ T lymphocytes. CONCLUSIONS: Antigenic specific gamma delta T cells might play an important role in the inducement and maintenance of airway inflammation. Persistent antigenic stimulation may be the key factor that maintains chronic airway inflammation in asthma.

[Characterization of rpoB mutation in rifampin - resistant clinical Mycobacterium tuberculosis isolates from China].

OBJECTIVE: To elucidate the characterization of rpoB mutation in rifampin - resistant clinical isolates of Mycobacterium tuberculosis isolates from China. METHODS: 588 bp DNA fragment of rpoB gene including 81 bp code region (rifampin resistance determination region, RRDR) was sequenced. 242 strains of Mycobacterium tuberculosis, including 193 rifampin - resistant strains and 46 rifampin - susceptible strains and 3 artificially induced rifampin - resistant strains were sequenced. RESULTS: 89.1% (172/193) rifampin - resistant strains had rpoB gene mutations in RRDR, no mutation was found in rifampin - susceptible strains. 46.1% rifampin - resistant strains had mutations located at 531-Ser; 17.1% strains had mutations located at 526-His; Combinative mutation rate was 12.4%; 4 strains had synonymous mutations; No deletion or insertion mutation was found among all strains. High level rifampin resistant strains (250 microgram/ml rifampin resistant) had higher mutation frequency at 531 - Ser than low level rifampin resistant strains (50 microgram/ml rifampin resistant) (P < 0.05). CONCLUSIONS: About 90% rifampin resistant clinical strains of Mycobacterium tuberculosis from China had rpoB mutations; 531-Ser and 526 - His were the most common positions to be substituted, the added mutation rate was about 46%; High level rifampin resistant strains had a higher frequency of 531-Ser mutation than low level rifampin resistant strains; No insertion or deletion mutation was found in these specimens; DNA sequencing is significant in drug choosing for tuberculosis treatment.

[Glutathione-S-transferase M1 genotype in patients with hepatocellular carcinoma].

OBJECTIVE: To study the glutathione-S-tranterase M1 (GSTM1) genotype in patients with hepatocellular carcinoma (HCC) with aflatoxin B1(AFB1) in a high risk region in Guangxi. METHODS: Specific GSTM1 primers and PCR technique were used for the detection of GSTM1 genotype using the peripheral leukocytes. A total of 379 samples was examined including 162 HCC patients whose lesions had been confirmed by pathology, 177 adults of local residents without cancer and 40 young men coming from other places with incidence of HCC. RESULTS: The frequency of GSTM1-null genotype in HCC patients was 102/162(63%), in local residents it was 92/177(52%) and in young men from outside it was 13/40(33%), which were significantly different. CONCLUSION: GSTM1 is one of the pivotal phase II detoxicated enzymes for AFB1. GSTM1 genetic deletion predisposes the individuals to HCC. However, the development of a HCC not only requires a genetic susceptibility, but also a AFB1 highly contaminated environment. The synergism of carcinogenic viruses such as HBV and HCV is also needed. These, when happen to be present together in an area in Guangxi, would lead to a high incidence of HCC.

[A research of infant refraction in Kunming Municipality].

OBJECTIVE: To study the infant refraction, detect ametropia in mass screening refraction and correct the ametropia properly for the treatment and prevention of amblyopia and strabismus as early as possible. METHODS: The refraction status of 1 146 children (2 291 eyes, aged 1 month -3 years) were determined with retinoscopy after tropicamide cycloplegia. RESULTS: The results of statistics showed that 89 eyes (3.88%) had emmetropia, 2 139 eyes (93.37%) hypermetropia, 38 eyes (1.66%) myopia, 560 eyes (24.44%) astigmatism and 25 eyes (1.09%) mixed astigmatism. In cases with ametropia, most of them were mild, and 97 eyes (4.23%) were moderate and severe. Anisometropia occurred in 34 infants (2.97%), and 7 infants (11 eyes, 0.48%) had strabismus. The degree of hypermetropia decreased with the increase of age. The percentage of astigmatism decreased with the age increase (chi(2) = 7.46, P<0.01), and the degree of astigmatism also decreased with the age increase (chi(2) = 26.1l, P < 0.01). Myopia increased with the age increase (chi(2) = 4.06, P < 0.05). CONCLUSIONS: The prevention of amblyopia and strabismus in children should begin at the infant period, and the cases with moderate and severe ametropia should wear eye glasses as early as possible.

[Effects of Rholida on the free radical metabolism and serum creatine kinase after exercise at plateau].

Objective. To study the effect of Rholida on free radical metabolism and serum creatine kinase CK) after exercise at plateau. Method. After staying at high altitude (4100 m) for 20 d, 40 healthy young men were divided into 4 groups randomly (Rholida, Acetazolamide, Xi' s capsule and control, 10 men each group). And their SOD, MDA, GSH-Px CK, and CK-MB were determined respectively. Before, after taking drugs and after finishing the 5 min-stair-exercise. Result. Before taking drugs and after exercise, MAD GSH-Px, CK, CK-MB, increased as compared with quiet state (P<0.05, P<0.01), but SOD showed no significant chang (P>0.05). After taking drugs for 6 d, those who took Rholida, Acetazolamide and Xi's capsule, their MAD, GSH-Px CK, CK-MB increased after exercise as compared with quiet state (P<0.05). In Rholida group SOD increased and had significant change (P<0.05); but there was no significant change in Acetazolamide, Xi' s capsule group, SOD increased, MDA decreased (P<0.05), CK, CK-MB had no significant change (P>0.05), GSH-Px increased in Xi's group (P<0.05), but not in Acetazolamide group (P>0.05). SOD, GSH-Px increased, MDA, CK-MB decreased in Rholida group after taking drugs and the changes were significant (P<0.01). In Acetazolamide and Xi's capsule group, GSH-Px increased significantly, MDA, CK, CK-MB decreased significantly (P<0.05), but SOD didn't (P>0.05). Conclusion. Rholida, Acetazolamide, Xi's capsule could regulate the disorder of free radical metabolism at plateau and Rholida had advantage over the others.
