Amino acid substitution (Ile194----Thr) in exon 5 of the lipoprotein lipase gene causes lipoprotein lipase deficiency in three unrelated probands. Support for a multicentric origin.

Studies on the molecular biology of lipoprotein lipase (LPL) deficiency have been facilitated by the availability of LPL gene probes and the recent characterization of gene mutations underlying human LPL deficiency. Typically, missense mutations have predominated and show a preferential localization to exons 4 and 5. This distribution supports earlier studies attributing functional significance to residues encoded by these exons. We now report a further missense mutation within exon 5 of the LPL gene in three unrelated patients. Amplification of individual exons by the polymerase chain reaction and direct sequencing revealed a T----C transition at codon 194 of the LPL cDNA which results in a substitution of threonine for isoleucine at this residue. The catalytic abnormality induced by this mutation was confirmed through in vitro mutagenesis studies in COS-1 cells. Transfection with a LPL cDNA containing the codon 194 transition resulted in the synthesis and secretion of a catalytically defective protein. The Thr194 substitution was associated with two different DNA haplotypes, consistent with a multicentric origin for this mutation.

Antitumor activity of some polysaccharides isolated from a Chinese mushroom, "huangmo", the fruiting body of Hohenbuehelia serotina.

Polysaccharides were extracted from a Chinese mushroom, "Huangmo" with hot water (FI), 1% ammonium oxalate solution (FII), and 5% sodium hydroxide (FIII) and 20 more polysaccharides were isolated by ethanol precipitation, ion-exchange chromatography, gel filtration, and affinity chromatography. Of the six polysaccharides obtained, FI0, FA-1, FA-2, FII-2, FIII-1-b, and FIII-2-b were selected on the basis of the results of the antitumor activity test (the Sarcoma 180/mice, i.p. method). Six fractions which showed relatively high activities were examined: FI0, a mixture of galactose-containing heteroglycan and beta-D-glucan; FA-1 and -2, beta-D-glucan with galactose-containing hetero-glycan and protein; FII-2, beta-D-glucan; and FIII-1-b and FIII-2-b, (1----6)-beta-D-glucosyl branched (1----3)-beta-D-glucan.

[Study on relationship between human ABO blood groups and type A behavior pattern].

Investigation of the relationship between ABO blood groups (ABO BG) and Type A behavior pattern (TABP) was performed. No significant correlation was found between human ABO BG and TABP (P less than 0.05). However, CH and TH + CH scores in individuals with blood type A and type A personality were significantly lower (P less than 0.05). The values of TH-CH scores among tested people were of no statistical significance (P greater than 0.05). Moreover, nearly half of the examinees (41.32%) had higher CH score and the distribution of the individuals was obviously different in statistics (P less than 0.005). In addition, L score of examinees with blood type A and type B personality was relatively lower.

[Determination of cholic acid in man-made bezoar by derivative spectrometry].

Cholic acid in man-made bezoar can be determined by the suggested first order derivative spectrometric method. The peak amplitude within 310-290 nm is independent of the other components and only varies linearly with cholic acid concentration. The method is simple and accurate with good reproducibility.

[Clinical and CT correlative researches in deep infarction of cerebral hemispheres].

Clinical and CT correlative researches in 172 patients with deep infarction of cerebral hemisphere were reported. The lesions are classified into eight types according to their location. Lateral of the central part of lateral ventricle was most frequently, and putamen, posterior limb of internal capsule, lateral of the anterior horn of lateral ventricle were subsequently. Only a few deep infarctions were watershed infarction possibly. The question of low perfusion in the peripheral of the lesion were discussed.

[Neolignans from Piper polysyphorum C.DC].

Piper polysyphorum C.DC (Piperaceae) is indigenous to the southern part of China. In the course of screening for inhibitors of platelet activating factor (PAF), the nonpolar fraction was found to exhibit PAF inhibitory activity. One new neclignan named polysyphorin and three new enantiomeric forms of (+)-virolongin, (+)-grandisin and (+)-lancifolin D were isolated. Two known neolignans, wallichinine and hancinone D were also obtained from the same sources. The structure determination was based upon spectroscopic analysis (UV, IR, CD, MS, 1HNMR, 13CNMR, COSY) and derivative preparation. The structure for polysyphorin was established as threo-delta 7'-7-hydroxy-3,4,5,3',5'-pentamethoxy-8-O-4' -neolignan. It is a racemic enantiomer. The PAF inhibitory activities were reported.

[Studies on ultraweak luminescence of bacteria].

Ultraweak luminescence of Escherichia coli, Bacillus subtilis and Brevibacterium ammoniagenes was measured with high sensitive single photon counting equipment (Made in China). The results obtained from ultraweak luminescence of as above three bacterial strains were as follows: spectral distribution curves, photon emission kinetic curves or emission intensity and its qualitative relationship between intensity and bacterial counts.
