RNA-protein interactions: involvement of NS3, NS5, and 3' noncoding regions of Japanese encephalitis virus genomic RNA.

The mechanism of replication of the flavivirus Japanese encephalitis virus (JEV) is not well known. The structures at the 3' end of the viral genome are highly conserved among divergent flaviviruses, suggesting that they may function as cis-acting signals for RNA replication and, as such, might specifically bind to cellular or viral proteins. UV cross-linking experiments were performed to identify the proteins that bind with the JEV plus-strand 3' noncoding region (NCR). Two proteins, p71 and p110, from JEV-infected but not from uninfected cell extracts were shown to bind specifically to the plus-strand 3' NCR. The quantities of these binding proteins increased during the course of JEV infection and correlated with the levels of JEV RNA synthesis in cell extracts. UV cross-linking coupled with Western blot and immunoprecipitation analysis showed that the p110 and p71 proteins were JEV NS5 and NS3, respectively, which are proposed as components of the RNA replicase. The putative stem-loop structure present within the plus-strand 3' NCR was required for the binding of these proteins. Furthermore, both proteins could interact with each other and form a protein-protein complex in vivo. These findings suggest that the 3' NCR of JEV genomic RNA may form a replication complex together with NS3 and NS5; this complex may be involved in JEV minus-strand RNA synthesis.

Preclinical evaluation of Fe-(5-C2H5-EHPG)- as a contrast agent in MR imaging of hepatobiliary system.

Iron(III)-N, N'-ethylenebis[2-(2-hydroxy-5-phenyl)glycine] [Fe-(5-C2H5-EHPG)-] is a paramagnetic complex designed for use as a hepatobiliary agent. Test procedures included synthesis, characterization, toxicity evaluation, biodistribution and experiments for animal MR images. The dose of LD50 in acute toxicity test of Fe-(5-C2H5-EHPG)- in mice was 3.49 mmol/kg. 111In-(5-C2H5-EHPG)- biodistribution studies revealed that the activities were (4.78 +/- 0.97, 5.34 +/- 0.91, 4.53 +/- 0.35)%ID and (0.88 +/- 0.18, 0.99 +/- 0.17, 0.84 +/- 0.06)%ID/gm in the liver at time intervals of 10, 30 and 60 minutes after injection; (5.76 +/- 0.15, 5.75 +/- 0.15, 4.00 +/- 0.04)%ID and (0.49 +/- 0.03, 0.49 +/- 0.05, 0.34 +/- 0.01)%ID/gm in the blood; (1.27 +/- 0.91, 1.46 +/- 1.00, 1.52 +/- 0.46) %ID and (0.89 +/- 0.17, 1.02 +/- 0.18, 1.06 +/- 0.08)%ID/gm in the kidneys, respectively. The results of image enhancement correlated well to the biodistribution. Analysis of the MR images showed degrees of maximal parenchymal % increase of signal to noise ratio (S/N) was 42.09 +/- 3.59% for normal liver at 30 minutes postinjection, which exceeded the value of pathologic liver with bile duct obstruction 16 hours 17.26 +/- 6.6 %, 1 week 19.80 +/- 6.46% and 4 weeks 32.20 +/- 9.01%, respectively, and acute hepatitis 16.50% +/- 4.02%. Persistent enhancement plateau was documented up to 60 minutes after injection and normalized to precontrast value within 22 hours. The common duct was opacified at 10-15 minutes after injection of contrast agent. These results indicated that the Fe-(5-C2H5-EHPG)- could be rapidly extracted from the blood stream by the hepatocytes and excreted into the bile duct. The initial evaluation of Fe-(5-C2H5-EHPG)- demonstrated that Fe-(5-C2H5-EHPG)- was well suited for enhancement of normal liver parenchyma and was compromised with deterioration of liver function. However, the increase of the liver intensities in the animal model of the total biliary obstruction group normalized to precontrast value within 22 hours, which indicated that renal clearance as an effective alternative pathway for biliary excretion. In conclusion, these results indicate that Fe-(5-C2H5-EHPG)- has the potential of becoming a safe and reliable magnetopharmaceutical for the hepatobiliary system.

[Seroprevalence of Helicobacter pylori infection among Yi and Han nationalities in Yunxian County, Yunnan Province].

Under cluster sampling, an early serum essay of helicobacterpylori (Hp) infection among 1084 healthy people, was carried out. Out of them, 325 among the Yi and Han Nationalities at Yunxian County of Yunnan Province were given a simultaneous test of anti-HAV IgG. The results were as follows: overall Hp infection rate was 51.1%; no difference of the infection was observed between the Yis and the Hans; the prevalence of Hp antibody went up with age; Higher rates were detected among those who drank river water or unboiled-water than among those who drank tap water, well water or boiled water, from poultry raisers than from non-poultry raisers and with drug abusers than with non-drug abusers. Hp infection showed a parallel relation to HAV infection. Finally, seven main epidemic factors were sifted from multi-factor logistic analysis namely source of drinking water, drinking habit, chicken-raising, pig-keeping, dog-breeding, drug-abusing and consumption of garlic. All these findings suggested that drinking water was an important vehicle of Hp infection. Our research data seems to have lend support to both fecal- oral transmission viewpoint and the hypothesis that Hp infection derives from animals. Frequent intake of garlic may serve as an agent in long-term prevention of Hp infection among people in this area.

LYS-49 phospholipase A2 homologs from venoms of Deinagkistrodon acutus and Trimeresurus mucrosquamatus have identical protein sequence.

The complete amino acid sequences of the Lys-49 PLA2s from the venom of Deinagkistrodon acutus (from Taiwan and China) and Trimeresurus mucrosquamatus (Taiwan habu) were solved by a facile cDNA cloning and sequencing method. The deduced amino acid sequences of the Lys-49 PLA2s of both venoms are identical, suggesting close phylogenic relationship between this two snake species of different genera. In addition, by cloning and cDNA sequencing, the mRNA coding for a Arg-49 PLA2 homolog of low expression level was also found in the venom gland of T. mucrosquamatus.

Molecular cloning and deduced primary structures of acidic and basic phospholipases A2 from the venom of Deinagkistrodon acutus.

This paper presents the nucleotide sequences of the cDNAs encoding three acidic phospholipases A2 and one basic phospholipase A2 from Deinagkistrodon acutus venom. The deduced primary structure of the basic enzyme is closest to that of the basic neurotoxic enzyme from Trimeresurus mucrosquamatus venom, while the acidic phospholipases from D. acutus have highest sequence similarity to that from Agkistrodon halys pallas. The phylogeny of this monotypic species is discussed.

Add-in macros for rapid and versatile calculation of non-compartmental pharmacokinetic parameters on Microsoft Excel spreadsheets.

We developed a package of macro programs (named PK_MOMENT) to automatically calculate non-compartmental pharmacokinetic parameters on Microsoft Excel spreadsheets. These macros include rigorous algorithms to execute moment calculations in a comprehensive manner. An optimum number of terminal data points for infinite-time extrapolation can be calculated with one of these macros so that automatic calculation of infinite moment parameters is possible. The moment calculation with PK_MOMENT provided satisfactory results using the hybrid (mixed linear-logarithmic) trapezoidal method rather than the conventional linear trapezoidal method. The macro-aided pharmacokinetic analyses turned out to be useful in that the macro-containing cells can be easily copied and pasted to analyze other data sets and that powerful tools of Excel can be utilized. The use of our macros will be significantly time-saving for routine pharmacokinetic analyses, considering that pharmacokinetic data are usually stored in a spreadsheet format, typically with Excel.

Optimization of the formulation design of chitosan microspheres containing cisplatin.

This study describes an orthogonal experimental design to optimize the formulation of cisplatin (CDDP)-loaded chitosan microspheres (namely, CDDP-DAC-MS) which were produced by an emulsion-chemical cross-linking technique. Seven factors and three levels for each factor that might affect the formulation of microspheres were selected and arranged in an L27(3(13)) orthogonal experimental table. A desirability function (df) calculated according to the trapping efficiency of CDDP, the drug content (%, w/w), and the size distribution of each batch of microspheres was introduced as an index of the microsphere formulation. The overall desirability functions (DF) were produced and treated by a statistic analytical system to optimize the formulation. Moreover, the contour maps were produced to analyze the influence of the seven factors on the size distribution, the drug content, and the drug trapping efficiency. The established optimum procedure was reproducible. Scanning electron micrographs showed that CDDP-DAC-MS were spherical with a coarse surface. The average diameter, drug content, and drug trapping efficiency of CDDP-DAC-MS were 74.8 microns, 20.8% (w/w), and 77.5%, respectively. The in vitro release of cisplatin from chitosan microspheres in saline was retarded compared with that from saline solution; the release of CDDP from chitosan microspheres was suggested to be controlled by the dissolution and diffusion of the drug from the chitosan matrix.

Preparation of bronchoalveolar lavage fluid with microscope slide smears.

The method of preparation of bronchoalveolar lavage fluid (BALF) for cytological examination can significantly affect the results of cellular quantitation. Investigations have shown that cytocentrifugation leads to an underestimation of the number of lymphocytes and membrane filter preparation to an underestimation of the number of neutrophils. As a simple alternative to these two techniques, BALF cells could be prepared by the microscope slide smear technique, which is familiar as the means for preparing peripheral blood for differential counts. In order to compare cell differentials determined by microscope slide technique with differentials resulting from cytocentrifugation, cells were isolated from 35 BALF samples using standard methods, and counted using a haematocytometer. Forty thousand cells in 200 microL were prepared by cytocentrifugation (3 min, 57 x g; Cytospin 2) and 5 x 10(5) cells in 5 microL by microscope slide smear. Both samples were air-dried, stained using May-Grünwald Giemsa stain, and 600 cells were counted to obtain differentials. To test the adequacy of sampling by the microscope slide smear technique, known quantities of lymphocytes or neutrophils were added to fixed numbers of BALF cells, microscope slide smears prepared, and differentials determined on 600 cells. The resulting differentials were compared to the calculated differentials. Preparation of BALF cells with the microscope slide smear technique yielded well-preserved cell morphology. Compared to cytocentrifugation, microscope slide smear preparations had significantly higher percentages of lymphocytes. The microscope slide smears for the samples with predetermined numbers of cells yielded lymphocyte and neutrophil percentages which did not differ from the calculated differentials (59.6 +/- 1.5 vs 59.6 +/- 5.2% and 54.6 +/- 6.0 vs 53.1 +/- 6.0%, respectively). Varying the number of cells counted from 100 to 800 confirmed the reproducibility of the counts for counting 600 cells. Using 5 x 10(5), 2.5 x 10(5), or 1 x 10(5) cells per preparation demonstrated that adequate specimens could be obtained from as few as 1 x 10(5) cells. Thus, microscope slide smear preparation is a simple and accurate method for the quantitation of bronchoalveolar lavage fluid cytology.

Pharmacokinetic study of taxol-loaded poly(lactic-co-glycolic acid) microspheres containing isopropyl myristate after targeted delivery to the lung in mice.

This study describes the pharmacokinetic behaviours of taxol after intravenous administration of taxol-loaded poly(lactic-co-glycolic acid) microspheres containing isopropyl myristate (namely, Taxol-IPM-PLGA-MS) and taxol saline solution to mice. Taxol-IPM-PLGA-MS were prepared using a solvent evaporation technique. The drug content and trapping efficiency of taxol in the microspheres were 5.09% (w/w) and 98%, respectively; the average diameter of the microspheres was 30.1 microns. Scanning electron microscopy showed that Taxol-IPM-PLGA-MS were spherical with a smooth surface. After administration of the drug saline solution (3 mg taxol/kg), taxol disappeared rapidly from plasma within 4-6 h and distributed extensively in various tissues. The tissue levels and AUCfinite of taxol in the lung were obviously higher than those in plasma but relatively lower than those in kidneys, bile, and liver. The biodistribution of taxol after administration of Taxol-IPM-PLGA-MS (3 mg taxol/kg), on the other hand, was altered significantly from the control (taxol solution) group. No taxol was detected in plasma or bile within 3 weeks, and only very low level of taxol was detected in the kidneys or liver within 48 h. However, taxol concentrations in the lung were increased significantly with the microsphere group; the peak concentration of taxol and AUCfinite in the lung was three times and 500 times higher than those with the taxol solution group, respectively. It was also noticed that the taxol levels in the lung were maintained at relatively high levels (> 10 micrograms/ml) for 3 weeks. Thus, the present study demonstrated the effective targeted delivery of taxol to the lung of mice using Taxol-IPM-PLGA-MS.

Preparation and characterization of poly(lactic-co-glycolic acid) microspheres for targeted delivery of a novel anticancer agent, taxol.

This study describes the preparation and characterization of poly(lactic-co-glycolic acid) microspheres containing a novel anticancer agent, taxol (namely, Taxol-PLGA-MS). A solvent evaporation technique was utilized to prepare Taxol-PLGA-MS. The trapping efficiency of taxol in the microspheres was greater than 90% and reproducible. The in vitro release rate of taxol from the microspheres was very low, and less than 15% of the initial amount of taxol was released in three weeks, irrespective of the drug loading level. When a chemical additive, isopropyl myristate (IPM), was introduced at the level of 30% (w/w), the release of taxol increased significantly; approximately 70% of the initial amount of taxol was released at a nearly constant rate for three weeks. Elevation of the loaded IPM level to 50% (w/w) produced a more rapid release of the drug. Scanning electron microscopy showed that Taxol-PLGA-MS were spherical with a smooth surface. More than half (55-65%) of the microspheres had a diameter of 20-45 microns. Incorporation of IPM had no significant influence on the particle size, surface morphology, or degradation behavior of the microspheres. It was strongly suggested that the release of taxol from the microspheres was dominated mainly by the drug diffusion in the matrix. As evaluated from the particle size, drug content, and in vitro release property, IPM-containing Taxol-PLGA-MS may be suitable for chemoembolization therapy of cancer diseases.

Detection of iron(III) chelate separated by reversed-phase high-performance liquid chromatography.

A reversed-phase high-performance liquid chromatographic method for the analysis of iron(III)-N, N'-trimethylenebis[2-(2-hydroxyl-5-ethyl-phenyl)glycine] (Fe(TM-5-ethyl-PHPG)-), which is potentially used as effective hepatobiliary magnetic resonance imaging (MRI) contrast agent, has been developed in this study. The effect of mobile phase, such as the varied pH values, ion-pairing agents and concentration of the ion-pairing agent on the capacity factor of the diastereomers and free ligand, were investigated to improve the separation and peak shape. HPLC in connection with a ion exchange column and a UV/VIS detector can be used to determine metal ion dissociated from this complex. This protocol can be successfully used in the quality assay of the synthesized potential MRI contrast agent of [Fe(TM-5-ethyl-PHPG)-].

Serological profiles of hepatitis B carrier patients in Singapore with special reference to the frequency and significance of concurrent presence of HBsAg and anti-HBs.

Hepatitis B serological markers were investigated in 1,132 consecutive Singaporean HBV carriers. Hepatitis B surface antigen (HBsAg) and hepatitis B surface antibody (anti-HBs) were found concurrently in 234 carriers (234/1132 or 21%). Serum anti-HBs levels were more than 10 mIU/mL in 80 of these carriers (80/234 or 34%). There were no difference in HBeAg positive status, as well as HBV-DNA positive status in concurrent HBsAg/anti-HBs carriers compared to carriers without anti-HBs. Our results suggested that concurrent HBsAg and anti-HBs is a common serologic pattern in Singaporean HBV carriers.

[Studies on preparation and characteristics of cisplatin chitosan microspheres].

In this paper, the preparation, drug content, size and size distribution, appearance and morphology, release characteristics in vitro and degradation characteristics of cisplatin chitosan microspheres (CDDP-DAC-MS) were studied. CDDP-DAC-MS were prepared by emulsion-crosslink technique. The CDDP-DAC-MS was shown to have rough spherical surface under scanning electron microscopy. The average diameter of the microspheres was 74.80 microns and CDDP content was 20.83% +/- 0.36%. CDDP-DAC-MS swelled slightly in saline after 1 h. Within the test period, the release of CDDP from CDDP-DAC-MS in saline solution could be described by first-order equation. The microspheres were sterilized by 60Co radiation. After 28 d of hepatic artery embolization with CDDP-DAC-MS in dogs, pathological photomicrograph showed that CDDP-DAC-MS could still be observed.

[Isolation and amplification of DNA from tortoise and turtle shells].

DNA was extracted from tortoise and turtle shells, two Chiense crude drugs which have been preserved for at least nine years. The cytochrome b sequence was amplified from the extracts using the polymerase chain reaction. Treatments with detergent and ultraviolet light were employed to destroy possible extraneous contamination. A control extract was set in parallel with the extracts of the specimens in order to detect contamination in solutions and reagents. The results show that significant amounts of genetic information can survive in the shells and may have important implications for their identification.

Effects of m-nisoldipine on aortic calcium accumulation in rats with vascular calcium overload.

AIM: To study the effects of a novel calcium channel blocker, m-nisoldipine, on vascular calcium overload (VCO) at both tissue and cellular levels. METHODS: VCO was induced in Wistar rats by treatment with colecalciferol (Col, 400,000 IU.kg-1, p.o.) and an aqueous mixture of ethanol and polyethyleneglycol-400 for 3 d. The tissue and subcellular calcium contents of aorta were determined by atomic absorption spectrometer and electron probe microanalysis, respectively. RESULTS: Chronic treatment with m-nisoldipine (m-Nis, 1-15 mg.kg-1, p.o., bid) only had mild inhibition on the elevation of total calcium in aorta, and the dose-response relationship of m-Nis displayed a bell shape, with inhibition ratio of 24% only for m-Nis 2.5 mg.kg-1. The effect of verapamil (12.5 mg.kg-1, p.o., bid) was a little better than that of m-Nis. The intracellular VCO in medial smooth muscle cells of aorta were remarkably inhibited by m-Nis (2.5 mg.kg-1), with inhibition ratios of 72% for cytoplasm and 76% for mitochondrion. The calcium accumulation in nucleus was reduced to a lesser degree than those in cytoplasm and mitochondrion. CONCLUSION: As for aorta in VCO rats, m-Nis mainly had conspicuous inhibition on intracellular VCO in medial smooth muscle cells, particularly in cytoplasm and mitochondrion, but with little effect on extracellular calcium deposition at tissue level.
