Pig xenogeneic antigen modification with green coffee bean alpha-galactosidase.

Green coffee bean alpha-galactosidase can cleave the terminal alpha-galactose (alphaGal) on oligosaccharides that form the major antigen on pig endothelial cells recognized by primate-specific antibodies. Studies have been made of the conditions under which it is functional (e.g. temperature, pH) and of its biochemical and immunologic effects. Pig-to-rhesus monkey vein transplants were studied to identify the efficiency of the enzyme in delaying hyperacute rejection. When a graft became occluded, biopsies were taken for light microscopy (hematoxylin and eosin), scanning electron microscopy (SEM) and immunostaining with Griffonia simplicifolia IB4 lectin (GSIB4), and for IgM, IgG and C3. alpha-Galactosidase was stable for 72-96 h and was effective at 4 degrees C and pH 6.9 (conditions of human liver graft storage), although better function was obtained at 20 degrees C and pH 6.5. Using the porcine PK15 cell assay, the cytotoxicity of human serum was reduced after treatment of the pig cells with the enzyme. In vitro studies demonstrated that porcine veins treated with alpha-galactosidase lost endothelial expression of the Gal epitope within 30 min. SEM, however, demonstrated endothelial damage beginning within 2 h, probably caused by the alpha-galactosidase, as no damage was found in phosphate-buffered saline-treated veins, where the Gal epitope was preserved for >3 h. No change was found in either group on light microscopy. In vivo studies demonstrated that patency of the alpha-galactosidase-treated veins (mean 2.5 h) was longer than that of untreated veins (0.23 h) (P < 0.01). Biopsies showed no GSIB4 lectin staining for alpha-Gal epitopes and much less IgM and C3 deposition in the treated group. Light microscopy and SEM demonstrated more severe endothelial damage, hemorrhage, and fibrin formation in the untreated group. Galactosidase is effective in removing the terminal alphaGal and delays the onset of hyperacute rejection of pig veins transplanted into monkeys. However, its effect is temporary and, on its own, its use is unlikely to prolong survival of pig organs transplanted into primates sufficiently to be of clinical value.

Photodynamic therapy: a mitochondrial inducer of apoptosis.

Photodamage to the mitochondria of murine leukemia P388 cells resulted in immediate loss of the mitochondrial membrane potential together with the release of cytochrome c into the cytosol. This was followed by a rapid activation of caspase 3-like proteases, as indicated by a marked rise in DEVDase activity. There was no significant effect on WEHDase or VEIDase activities, suggesting that only the late-stage caspases had been effected. The apoptotic response to mitochondrial photodamage was abolished by the broad-spectrum caspase inhibitor zVAD-fmk, but this did not prevent loss of viability after mitochondrial photodamage. These studies indicate that the release of cytochrome c from photodamaged mitochondria is sufficient to directly initiate a caspase-dependent apoptotic response.

Calcitonin gene-related peptide upregulates melanogenesis and enhances melanocyte dendricity via induction of keratinocyte-derived melanotrophic factors.

It has recently been shown that cutaneous axon terminals and epidermal melanocytes make contact via chemical synapses in human skin and that calcitonin gene-related peptide (CGRP) induces melanocyte proliferation. To further clarify the effect of neuropeptides on the biology and morphology of melanocytes, especially with respect to melanogenesis and melanocyte dendricity, organ cultures of normal human skin and cultured melanocytes were exposed to various neuropeptides present in intraepidermal nerve endings. Of the neuropeptides examined, skin exposed to CGRP in organ culture showed increases in melanocyte number, epidermal melanin content, melanosome number, and degree of melanization. CGRP alone had no significant effect on melanogenesis of cultured melanocytes, whereas the addition of medium conditioned by CGRP-stimulated keratinocytes (CGRP-KCM) induced melanogenesis as indicated by biochemical assays of tyrosinase activity and melanin content. Furthermore, CGRP-KCM significantly enhanced melanocyte dendricity, a crucial factor affecting epidermal pigmentation. These findings suggest that keratinocytes produce and secrete some melanotrophic factors following stimulation with CGRP, which modulate growth, melanin synthesis, and dendricity of melanocytes. These data demonstrate intimate interactions between the cutaneous nervous system and melanocytes within the epidermal environment.

HICLAS: a taxonomic database system for displaying and comparing biological classification and phylogenetic trees.

MOTIVATION: Numerous database management systems have been developed for processing various taxonomic data bases on biological classification or phylogenetic information. In this paper, we present an integrated system to deal with interacting classifications and phylogenies concerning particular taxonomic groups. RESULTS: An information-theoretic view (taxon view) has been applied to capture taxonomic concepts as taxonomic data entities. A data model which is suitable for supporting semantically interacting dynamic views of hierarchic classifications and a query method for interacting classifications have been developed. The concept of taxonomic view and the data model can also be expanded to carry phylogenetic information in phylogenetic trees. We have designed a prototype taxonomic database system called HICLAS (HIerarchical CLAssification System) based on the concept of taxon view, and the data models and query methods have been designed and implemented. This system can be effectively used in the taxonomic revisionary process, especially when databases are being constructed by specialists in particular groups, and the system can be used to compare classifications and phylogenetic trees. Intrastriatal dopamine injection induces apoptosis through oxidation-involved activation of transcription factors AP-1 and NF-kappaB in rats.

ABSTRACT More and more evidence suggests that increases in dopamine (DA) in striata may participate in neurodegenerative processes during acute ischemia, hypoxia, and excitotoxicity. With a rat model of intrastriatal DA injection, we studied the molecular events involved in DA toxicity. Intrastriatal injections of DA in amounts from 1 to 2 micromol result in apoptotic cell death, as indicated by terminal deoxynucleotidyl transferase labeling of DNA strand breaks and Klenow polymerase-catalyzed [(32)P]deoxycytidine triphosphate-labeled DNA laddering. Injections of DA produce a strong and prolonged activated protein 1 (AP-1) activity that contains c-fos, c-jun, and phosphorylated c-jun protein. DA injections also stimulate the activity of nuclear factor-kappaB (NF-kappaB), an oxidative stress-responsive transcription factor. Injection of curcumin at a dose that selectively inhibits AP-1 activation without affecting NF-kappaB activity attenuates DNA laddering induced by DA. Preinjection with SN50, a specific permeable recombinant NF-kappaB translocation inhibitor peptide, reduces DA-induced NF-kappaB activation and apoptosis. Moreover, preinjection of the antioxidant GSH significantly inhibits both DA-induced activation of transcription factors AP-1 and NF-kappaB and subsequent apoptosis. Thus, our data suggest that DA-oxidative stress-induced apoptosis in vivo is mediated by activation of transcription factors AP-1 and NF-kappaB.

Functional magnetic stimulation of the abdominal muscles in humans.

Functional magnetic stimulation (FMS) of the thoracic nerve roots to simulate cough has been suggested as a treatment approach in patients unable to voluntarily activate the abdominal muscles. However, factors that could influence the efficacy of FMS in clinical use have not been evaluated. In the present investigation we studied train length, posture, and frequency to determine the optimal stimulation protocol. We also evaluated the use of a valve at the mouth to enhance glottic function and investigated whether lung volume at the time of stimulation would influence the tension generated by the abdominal muscles. Studies were performed using a Magstim rapid stimulator augmented by four booster packs in nine healthy subjects; we measured the change in gastric (DeltaPga(FMS)), esophageal (DeltaPes(FMS)), and mouth pressure and expiratory flow. With our apparatus pressure generation was maximized by having a train length of at least 300 ms and a frequency of 25 Hz. Posture and valve use were not important determinants of DeltaPga(FMS) or DeltaPes(FMS). Lung volume exerted only a minor influence on DeltaPga(FMS), but the ratio DeltaPes(FMS):DeltaPga(FMS) was increased at TLC compared with FRC. Expiratory flow was increased by adopting a seated posture and using an occlusion valve with an opening threshold close to the maximum DeltaPes(FMS) generated by the stimulus train; however, expiratory flow was susceptible to interference from glottic incoordination. Representative results (with train length 600 ms, 25 Hz, and 100% power, seated) were mean DeltaPga(FMS), 166 cm H(2)O; mean DeltaPes(FMS), 108 cm H(2)O; and mean expiratory flow, 311 L/min. We confirm that FMS of the abdominal muscles can generate a substantial positive intra-abdominal and intrathoracic pressure and, consequently, expiratory flow in normal subjects.

Mutated ras p21 as a target for cancer therapy in mouse transitional cell carcinoma.

PURPOSE: To establish an experimental mouse model for bladder cancer immunotherapy using mutated ras as a target. MATERIALS AND METHODS: A tumorigenic mouse bladder transitional cell carcinoma (TCC) line MB49 (C57BL/6 origin) was analyzed for its c-ras gene status by DNA cloning and sequencing. Aberrant expression of the ras gene was measured with Western blotting. 13-mer peptides corresponding to residues 5 to 17 of the ras protein were synthesized and tested for immunogenicity in syngeneic C57BL/6 mice. Induction of specific immune responses was evaluated by analyzing splenocyte activity in vitro and tumor suppression in vivo. RESULTS: MB49 cells were found to contain a single amino acid substitution of serine for glycine at codon 12 in K-ras loci and an abundant amount of cellular mutated ras p21 protein. C57BL/6 mice immunized with the 13-mer serine-containing ras peptide exhibited mutation-specific immune responses in splenocyte proliferation, cytokine production and cytotoxicity. Specific antitumor immunity in the form of tumor growth delay in vivo was observed in mice immunized with the same mutant peptide followed by subcutaneous MB49 tumor challenge and was enhanced by the addition of low dose interleukin-12. CONCLUSIONS: The mouse bladder TCC line MB49 contains a serine mutation at codon 12 of its K-ras gene that is sufficient to induce mutation-specific immune responses in vitro and specific protective immunity to MB49 tumor in vivo. Mutated oncoproteins may be ideal targets for the development of specific immunotherapy regimens for bladder cancer immunotherapy.

[The cDNA clone and sequence analysis of the coding region of human augmenter of liver regeneration (hALR) gene].

OBJECTIVE: To obtaining cDNA clone of the coding region of human augmenter of liver regeneration(hALR) as a basis for further study. METHODS: Total RNA of human fetal liver was extracted as a template. First strand of cDNA was synthesised by reverse transcription with olig(dT)-primer and then amplified by PCR with the primers designed by ourself. The size of PCR product was about 380 bp. it was subcloned into pUC19 and was sequenced by auto-sequencing instrument. The sequence of hALR was analysed with PCGENE program. RESULT: The cDNA of integrity coding region of hALR was obtained. Its size was 378 bp. The sequence homology of our hALR compared with rat ALR and human ALR recently reported were 86.5% and 99.2%, respectively. CONCLUSION: It was also suggested that hALR takes part in growth regulation of fetal liver during human fetus late development.

Clinical investigation of a high-strength glass ionomer restorative used with the ART technique in Wuhan, China: one-year results.

OBJECTIVE: To evaluate the safety and effectiveness of a new glass ionomer restorative, ChemFlex, and to compare its clinical performance and wear to another popular material, Fuji IX GP, when used with the atraumatic restorative treatment (ART) approach in posterior teeth in school children. METHODS: Ninety-two subjects aged between 6 and 14 years who had bilateral matched pairs of carious posterior teeth were selected. A split-mouth experimental design was used in which the two restorative materials were randomly placed on contralateral sides. The restorations were assessed directly and also indirectly from color transparencies and die replicas. RESULTS: After one year, the success rates of ART restorations in the primary teeth were 96.6% for ChemFlex restorations and 89.7% for Fuji IX GP restorations placed in the Class I cavity preparations, whereas only 46.2% (ChemFlex) and 61.5% (Fuji IX GP) of Class II restorations were assessed as clinically satisfactory. In the permanent dentition, the success rates were 94.6% and 98.2% for ChemFlex and Fuji IX GP, respectively; however, there was no statistically significant difference between the two restoratives for either the permanent or primary teeth. The mean occlusal wear after one year in the permanent teeth was 53.2 microns for ChemFlex and 56.3 microns for Fuji IX GP. Again, there were no statistically significant differences in wear between the two materials (P > 0.05). CONCLUSION: The clinical performance of both ChemFlex and Fuji IX GP over a 12-month period was highly satisfactory and completely adequate for the ART technique, particularly in Class I cavities.

[Measurement and clinical use of recoil pressure of the respiratory system].

Using the principle that the difference of MIP (maximal inspiratory mouth pressure) between RV (residual volume) and FRC (functional residual capacity) equals Prs (recoil pressure of the respiratory system) at RV, we performed the measurement of Prs at RV in 20 normal subjects, 90 patients with COPD (chronic obstructive pulmonary disease) and 15 patients with ILF (interstitial lung fibrosis). We also performed the correlation analysis of Prs at RV and RV/TLC (total lung capacity) %. The results indicated the Prs values at RV (-2.8 +/- 0.5 kPa) in normal subjects measured in this manner was in accord with the values reported by other authors abroad. The Prs in patients with emphysema decreased and it was highly and negatively correlated with RV/TLC% (r = -0.872), but the Prs in patients with ILF was increased. The results suggest that this method for measurement of Prs is simple, reliable, and clinically useful.

[Clinical analysis of 50 cases of transvaginal hysterectomy].

OBJECTIVE: To assess the advantage, indications, surgical management and the clinical value of transvaginal hysterectomy in patients without uterine prolapse. METHODS: From Oct. 1995 to Nov. 1998, 50 women undergone transvaginal hysterectomy for various benign gynecological lesions were consecutively included in this study. Their mean age was 45 years (range 36-50). There were 10 patients (20%) with normal uterine size, and 16 (32%) with uteri of > 12 gestational weeksize (range 14-16 weeks) and the remaining 24(48%) < or = 12 weeksize. 11 patients (22%) underwent salpingo-oophorectomy simultaneously, 6 patients (12%) had previous laparotomy. RESULTS: The mean operating time was 63 min with a range 40-110 min. Post-operative hospital stay averaged 4 days (range 2-8 days). The amount of blood loss during operation ranged 50-100 ml. The only postoperative complication was small residue hematoma in 1 case and cured with conservative treatment. CONCLUSIONS: Transvaginal hysterectomy was associated with remarkable advantages: shorter operating time and hospital stay, fewer complication, less discomfort, quicker recovery and less expenses. Enlarged uterus to a size of > 12 gestational weeks; benign adnexal mass or previous operation should no longer be considered as contraindication of transvaginal hysterectomy.

Clinicopathologic, immunophenotypic and ultrastructrual analyses of ATLL patients with cutaneous involvement.

OBJECTIVE: To study 4 cases of adult T-cell leukemia/lymphoma (ATLL) associated with cutaneous lesions for clinicopathology, immunophenotype, human T-cell leukemia/lymphoma virus type I (HTLV-I) provirus DNA and their ultrastructure. At the same time, HTLV-I provirus DNA of ATLL patients were also compared with 18 cases of cutaneous lymphoma (CL), two cases of actinic reticuloid as well as two cases of lymphocytic infiltration. METHODS: Immunohistochemistry studies were carried out on the infiltrating cells using monoclonal antibodies against CD45-RO, CD20, CD68 on paraffin-embedded sections by ABC method and using monoclonal antibodies against CD3, CD4 and CD8 with indirect immunofluorescence (IIF) on frozen sections. Skin biopsies were examined by electron microscope. Serum and bone marrow cells were tested for antibodies against HTLV-I-associated antigen by IIF, and HTLV-I provirus DNA was examined by PCR method. RESULTS: The research showed four patients with ATLL manifesting cutaneous lesions, were subsequently found with additional systemic symptoms, as extensively enlarged superficial lymph node, abnormal increased IL-2 receptor, flower-like cells in their peripheral blood and marrow. The HTLV-I provirus DNA was positive in the peripheral blood, bone marrow, cutaneous lesions and lymph node biopsy specimens by using PCR amplification of specific HTLV-I fragment while 18 cases of the CL were negative for HTLV-I. The special ultrastructure of skin lesions was also found in ATLL patients. CONCLUSIONS: The cutaneous involvement in ATLL is a type of cutaneous T cell lymphoma (CTCL) but shows some differential immunological markers for differential diagnosis. The examination of HTLV-I antibodies or HTLV-I provirus DNA is necessary for diagnosis of ATLL. The ultrastructural characteristics in skin lesions of ATLL were of atypical lymphocytes and mononuclear cells invading the epidermis, and the mononuclear cells are possessing the phagocytic function and phagocytizing the degenerated epidermic cells or lymphocytes.

[Characteristics and criteria for bronchial responsiveness measurement in cough variant asthma].

OBJECTIVE: To investigate the value of sGaw in the differentiation of CVA from COPD. METHODS: MEFV, airway resistance(Raw) and bronchial responsiveness were measured in 30 cases with CVA, 133 cases with typical bronchial asthma(BA) and 37 cases with COPD. RESULTS: (1) The expiratory-flow parameters (PEF, MMEF, FEV1/FVC) in MEFV curve in remission period of CVA were largely normal, but most patients with CVA (67%) have increased Raw and decreased sGaw, characteristics similar with typical bronchial asthma (BA). (2) If PC35sGaw < 8 g/L was set as the positive limit, 100% of CVA and BA patients showed bronchial hyperresponsiveness(BHR), the average positive MCH concentration being about 1.0 g/L. 5 cases with COPD (13.5%) also showed BHR, but the average MCH concentration was 5.24 g/L. (3) If PC35sGaw < 4 g/L was set as the positive limit for the diagnosis of CVA, the sensitivity was 96.7%, specificity 97.3%, confidence limit 99%. (4) A equation for PC35sGaw was deduced, and its calculation values showed the same results with the ones from diagrammatization. CONCLUSIONS: sGaw is a sensitive index for the measurement of bronchial responsiveness, and it is useful in the differentiation of CVA from COPD. It is suggested that PC35sGaw < 8 g/L be regarded as bronchial hyperresponsiveness, and PC35sGaw < 4 g/L as candidate for CVA.

[Study on relationship of different thickness and opaquing capacity of various opaques].

OBJECTIVE: To evaluate the relation between different opaque thickness and color difference. METHODS: Model made of the thin plastic paper was used to control the ceramic thickness. The data of chromaticity was measured by TC-PIIG auto color difference meter and its results were processed by computer. RESULTS: There are two thickness values which can be considered as ideal range of opaque thickness for color opaquing: one is the values of ceramic thickness which is based on the color difference values of the infinite chromaticity thickness (ICT) and the other is the values of ceramic thickness which is depended on the color difference values which is 1.5 compared with ICT. The ideal range for VITA is (0.207-0.152) mm, SHOFO is (0.190-0.160) mm, CARAT is (0.140-0.074) mm and EXCELCO is (0.169-0.144) mm. CONCLUSION: There is a relationship of non linear direct ratio between opaque thickness and its shadow capacity. However, no more increase of color difference values in respect to the thickness was found when the ceramic thickness reach a certain value. Different ceramic porcelain needs different ideal range of thickness.

[The detection of microsatellite instability in carcinoma and adenoma of large bowel and its significance].

OBJECTIVE: To explore the role of microsatellite instability (MSI) in colorectal carcinogenesis and the relationship between MSI, proliferation activity and prognosis. METHODS: PCR-SSLP and immunohistochemistry methods were used to detect MSI and the expression of PCNA in 56 cases of carcinomas, 9 cases of adenomas and 6 cases of adenomas with malignant changes. RESULTS: The total positive rate of MSI were 25/56 cases in colorectal carcinomas. The MSI positive cases were 3/4 in HNPCCs, 22/52 in sporadic colorectal carcinomas, 2/9 in adenomas and 2/6 in adenoma with malignant changes respectively. The PCNA labelling index of MSI positive tumors were significantly lower than that of MSI negative tumors (P < 0.01). The 3 and 5 year survival rates in patients with MSI positive colorectal carcinomas were higher than those of MSI negative tumors. CONCLUSION: MSI may be an early molecular alteration and another molecular mechanism in colorectal carcinogenesis. The MSI positive tumors have low proliferating activity and a better clinical outcome. The detection of MSI might be useful in predicting prognosis of colorectal carcinomas.

[A study on the loss of heterozygosity of multiple tumor suppressor genes in gastrointestinal metaplasia].

OBJECTIVE: To investigate the role of multiple tumor suppressor gene abnormality in gastrointestinal metaplasia (IM). METHODS: The loss of heterozygosity (LOH) of YNZ22, APC, MCC and DCC genes was examined by PCR-RFLP technique in 60 IM cases. RESULTS: LOH of YNZ22, APC, MCC and DCC genes were found in 19.4% (6/31), 12.0% (3/25), 13.0% (3/23) and 4.3% (2/47) respectively. IM was classified into types I, II and III. It was found that the LOH rates of each tumor suppressor gene of type III IM were higher than those of types I & II IM, but the difference was not significant (P > 0.05). Two cases of type III IM were found to also have 2 gene alterations (APC, MCC or YNZ22, MCC) respectively. All LOH were found in IM of adjacent carcinoma. None were found in IM of cancer-free patients. CONCLUSION: The alteration of tumor suppressor gene may play a role in the development and carcinogenesis of IM.

[Study on antifungal mechanism of alpha-pinene].

The antifungal mechanism of alpha-pinene was studied using electronic microscopy and incorporation of isotope-labelled precursor. The results demonstrated that alpha-pinene had significant roles in inhibiting and killing Candida albicans. After treatment with alpha-pinene the fungal morphology and ultrastructure showed obvious changes: their cell wall and cytoplasmic membrane ruptured; intracellular components released out and the cell residue fused to form irregular masses. In addition, the synthesis of DNA, RNA, polysaccharide of cell wall and ergosterol of cytoplasmic membrane was inhibited. It is indicated that these changes are related to antifungal mechanism of alpha-pinene.
