[Chemical constituents of fruits of Gardenia sootepensis Hutch].

OBJECTIVE: To demonstrate the chemical constituents of the fruits of Gardenia sootepensis. METHOD: Column chromatographic technology was employed to separate the water soluble part, and the compounds obtained were elucidated by spectral and chemical analysis. RESULT: Ten compounds have been isolated, seven of which elucidated respectively as D-mannitol, beta-sitosterol, deacetylasperulosidic acid methyl ester, geniposidic acid, geniposide, scandoside methyl ester and dimeric molecule of quinide. CONCLUSION: All the compounds were obtained from this plant for the first time.

[Protective effect of Radix Salvia Miltiorrhizae on radiation damage of the cochlea].

In order to evaluate the preventive and therapeutic effects of Radix Salvia Miltiorrhizae on radiation damage of the cochlea, guinea pigs were divided into 3 groups. Group 1 was treated by radiation added with Radix Salvia Miltiorrhizae. Group 2 was radiated alone, and Group 3 was control. Group 1 and Group 2 were radiated with a single dose of gamma radiation(60 Gy). Morphological and functional observation of the cochleae was performed in two weeks after radiation. The result showed that the changes of complex action potential(CAP) and the cochlear structure were slight in Group 1, but obvious changes were found in Group 2. There was a significant difference in CAP response threshold and incidence of the cochlear hair cell loss between Group 1 and Group 2 (P < 0.01). These results suggest that Radix Salvia Miltiorrhiza may prevent radiation-induced cochlea damage. Its protective mechanisms may be cleaning free radicals, blocking calcium channel and improving microcirculation of the cochlea.

[Study of the immunological tolerance induced by peripheral intravenous infusion of donor-specific antigens].

In the experiment, donor specific tolerance was induced by DST, DSSL and DSBM through peripheral intravenous transfusion in rat cervical cardiac transplantation model. The results showed there were significant differences in graft survival time between the blank control group and the other groups (P < 0.05). The mean graft survival time days of the DST, DSSL and DSBM groups were 28.9 +/- 13.7, 42.9 +/- 6.6 and 40.3 +/- 11.6 days, respectively. The effect of component infusion was obviously better than that of DST (P < 0.05), but no statistical difference was observed between DSSL and DSBM. CsA gave a noticeable synergistic effect to the induction of immunological tolerance. MLR and CD8+ T cell were remarkably reduced(P < 0.05) in the cases of component transfusion. This study suggested that components transfusion may be of clinical use for donor-specific tolerance inducing.

[Ameloblastic fibrosarcoma: an immunohistochemical and ultrastructural study of the mesenchymal component].

To study the characteristics of the mesenchymal cells of ameloblastic fibrosarcoma (AFS), three cases of AFS were studied immunohistochemically and ultrastructurally. The results showed that the mesenchymal component of AFS consisted predominantly of fibroblastic cells with a small number of undifferentiated cells, a few histiocytes and occasionally myofibroblastic cells under electron microscope. The fibroblastic cells were Vimentin positive only, and myofibroblastic cells were positive for Vimentin, HHF35 and alpha-SMA. The histiocytes were positive both for kp1 and PG-M1, suggesting that these cells were infiltrating cells from peripheral blood rather than histiocytic differentiation of tumor cells. Compared with ameloblastic fibroma, AFS showed much higher PCNA labeling index, suggesting higher proliferation activity of AFS.

[An immunohistochemical and morphometric study of myoepithelial cells in salivary glands of human and experimental animals].

OBJECTIVE: To investigate whether differences exist between myoepithelial cells of human beings and experimental animals. METHODS: Myoepithelial cells(MeCs) of 8 kinds of experimental animals were studied immunohistochemically using muscle-acting-specific monoclonal antibody HHF35 and morphometrically using image-analysis. RESULTS: There were differences in mean proportional volumes of MeCs between human beings and experimental animals. The mean proportional volumes of mucous acini were more than those of serous acini except salivary glands of rodents. CONCLUSION: The mean proportional volume of MeCs in salivary glands associates with animal species and secretion of salivary glands.

[Traumatic perilymphatic fistula of round and oval windows (four cases reported)].

OBJECTIVE: To explore the diagnosis and treatment to traumatic perilymph fistula (PLF) of round and oval window. METHOD: Traumatic PLF was diagnosed by the traumatic history of head, neck and ear, the examinations of auditory and vestibular function, and the exploratory tympanotomy. The PLF of round and oval windows were repaired by fascia graft or tragus perichondrium and gelform. Four cases with traumatic PLF of round and oval window were reported in this paper, which included 2 cases hitten by hand, 1 by brick, 1 insulted by middle ear surgery. Exploratory tympanotomy was performed from one and half to nine months after injury. One case was misdiagnosed as Meniere's disease before confirming PLF. Exploration and repair of PLF underwent in all cases. RESULT: The symptom of vertigo relieved in all cases after surgery, while the hearing recovery was not evident. CONCLUSION: 1. Traumatic PLF is unrare, hence, traumatic PLF should be alerted if patients suffer from vertigo and hearing impairment after head and ear injury. 2. The features with vertigo attacks, fluctuating hearing loss and tinnutis should be distinguished from Meniere's disease. 3. Early exploratory tympanotomy and repair of PLF are effective for relieving vertigo and improving hearing.

[The determination of F-, Cl-, NO3- and SO(4)2- in the total suspended particles in air by ion chromatography].

In this paper the method used for the determination of F-, Cl-, No3- and SO(4)2- in the total suspended particles in air (TSP) by ion chromatography was established. The contrast between the blank values of filter membranes of ultra-thin glass fiber and chlorinated PVC, the contrast between extraction methods by supersonic extraction with 70 degrees C water and by just stand for 24 h, and the contrast between extraction solutions with deionized water and the eluent (2.2 mmol/L Na2CO3-2.8 mmol/L NaHCO3) were involved. The determination of the uniformity of filter membrane and the typical samples were also carried out. This paper adopted ethylene tetrachloride filter membrane, deionized water and supersonic for extraction. The average recoveries of this method for F-, Cl-, NO3- and SO(4)2- were 98.7%, 103%, 101% and 99.8% respectively and the RSD's were 0.29%, 0.32%, 0.30% and 0.25% respectively. This method possesses better recovery and precision.

[Cloning and sequencing analysis of rice stripe virus genome segment 4 of Chinese isolate Y].

The cDNA fragment covering full-length sequence of RStV RNA4 of Yunnan isolate in China was obtained by RT-PCR. The PCR-derived fragment was then cloned into vector pCRII. The cloned cDNA was sequenced. Comparison of the nucleotide and deduced amino acid sequences with those of the Japanese isolate T was made. The results showed that at the nucleotides level, vORF, vcORF and the intergenic region had 94.9%, 94.1% and 86.1% identity respectively, the 5'-untranslational region was exactly the same as that of Japanese isolate T, while the 3'-terminal sequence had 96.1% identity, differing by two nucleotides; at the amino acid level, vORF and vcORF had 99.4% and 98.3% identity respectively. Therefore, as well as being exactly the same size for the two isolates, the amino acid sequences of the coding regions and the 5'- and 3'-terminal sequences were well conserved. Our results indicated that the Chinese isolate is closely related to the Japanese isolate T.

[Studies on biosorption of Pb2+ by Phanerochaete chrysosporium].

The biosorption of Pb2+ from solution by Phanerochaete chrysosporium was studied. The effects on biosorption capacity of pH, Pb2+ concentration in solution, co-ion and temperature, time have been examined. The adsorption capacity was compared among the biomass harvested at various growth stages. Pretreatment of biomass by NaOH solution improved its adsorption capacity. The uptake could reach 108.4 mg/g at optimal conditions of biosorption using pretreated biomass.

[The morphological structure of classical swine fever virus and some characteristics of its multiplication].

Some characteristics of the multiplication of classical swine fever virus(CSFV) Thiverval strain were studied by means of the immunofluorescence technique. Under optimum culture conditions, the concentration of CSFV in the culture liquid multiplicated by MPK cells is ten times higher than by PK-15 cells. The half-life period of CSFV at 37 degrees C is 3 hour. The location of CSFV's replication in host cells is detected by the monoclonal antibodies of the structural protein E2 and the non-structural protein p120 of CSFV. Based on the above results, the ultrastructure of CSFV particles inside host cells was further studied using electron microscope and the changes of the ultrastructure of CSFV-infected cells are shown. The possible reasons for lower viral titre are also discussed in this paper.

[The outer membrane protein (OMP) patterns of Escherichia coli isolates of predominant O serogroups originated from chichens in different regions in China].

The purpose of this study is to determine the outer membrane protein (OMP) patterns of avian Escherichia coli isolates with predominant serogroups originated from 18 provinces, autonomous regions and municipal cities in China. Total of 204 isolates belonging to O18, O78, O2, O88, O11 and O26 serogroups respectively, were tested. The outer membrane proteins of these isolates were isolated with the improved N-lauroylsarcosine method and analyzed by SDS-PAGE. 4 different OMP patterns were identified with these isolates of predominant serogroups. 3 different OMP patterns with 56 isolates of O18, 4 with 54 isolates of O78, 2 with 28 isolates O2, 1 with 26 isolates of O88, 3 with 22 isolates of O11 and 1 with 18 isolates of O26 respectively, were found. Isolates with OMP pattern 1 were discovered in 6 different serogroups, and OMP-3 pattern was also shared by O18, O78 and O11 serogroups isolates. These results indicated that the OMP patterns of avian pathogenic Escherichia coli isolates of O18, O78, O2, O11 serogroups which were isolated from different regions in China were heterogeneous, and all of O88 and O26 serogroups isolates just belonged to OMP pattern 1. Moreover, the OMP pattern 1 was presented in isolates of six different predominant O serogroups.

[Differences in haemozoin production and pathogenicity between chloroquine-sensitive and chloroquine-resistant strains of Plasmodium berghei].

AIM: To better understand the differences in haemozoin formation and pathogenicity between chloroquine-sensitive(N) and chloroquine-resistant(RC) strains of Plasmodium berghei. METHODS: IRC mice were grouped as follows: group Xba I (Normal control, NC), group II (control treated with chloroquine alone, CC), group III (mice infected with N strain), group IV (mice infected with RC strain, RC) and group V (mice infected with RC strain and treated with chloroquine, RCC). Morphologic features of the parasites, parasitaemia, histological and ultrastructural changes of livers among the groups were compared. RESULTS: Severe damages of the hepatic cells of the N group including increased lysosomes and swollen and fused mitochondria were detected. On the contrary, the prominent features in liver section of the RC group were inflammatory cell (notably mononuclear) infiltration and Kupffer cell activation. Numerous trophozoites and schizonts were sequestrated in hepatic sinusoids and less degeneration of parenchymal cells was found except for some swollen and vacuolated mitochondria. Internal food vacuoles containing haemozoin were found in the parasites of the N group, whereas external food vacuoles without haemozoin granules were arranged in foamy appearance within the parasitized cell of the RC group. CONCLUSION: P. berghei RC strain may modify the mode of ingestion and degradation of hemoglobin in the parasites, resulting in impeding haemozoin formation. The difference in virulence between the N and RC strain of P. berghei is probably attributed to the significant differences in the induction of immune response of the host.

[Hair cell apoptosis and hearing loss of perilymphatic fistula].

OBJECTIVE: To investigate the effect of hair cell apoptosis on hearing loss of perilymphatic fistula (PLF) in guinea pig. METHOD: Twenty-five guinea pigs with light microscope and TdT mediated biotin dUTP nick-end labelling (TUNEL) techniques. The ECochG and ABR were measured and the data analyzed with statistics. RESULT: 1. The apoptosis of hair cell was not revealed in 0-hour-group and 2-hour-group of PLF. Following the time of PLF was longer, the apoptosis of hair cell was more increased. The apoptosis of hair cell was demonsted in 1-day-group (1 case, 17%), 2-day-group (4 case, 67%) and 7-day-group (4 case, 80%); 2. The amplication of AP was reduced after operation induced PLF. The CAP of experimental ear was significantly higher than that of control ear (P < 0.01) in 1-day-group, 2-day-group and 7-day-group. CONCLUSION: 1. Apoptosis of hair cell was appeared in PLF; 2. The apoptosis of hair cell may be one of morphological evidence in hearing loss of PLF.

[Structural disorder and conformation difference in the organic molecular crystals].

OBJECTIVE: Investigating multiple configuration nature of natural organic molecules through study on atom disorder in crystal state molecules. METHODS: Three-dimensional determination for four natural organic compound samples by using single crystal X-ray diffraction. RESULTS: Through disorder analysis on molecules in crystal state, we have found that the disorder of some atoms may cause conformation difference for molecules, bring about multiple conformation in crystal state hence reducing the symmetry of molecules in crystal cells. CONCLUSIONS: There can be two or three molecules, even two different compounds in an asymmetrical unit of structure cell because of partial disorder or configuration difference in organic molecule crystals.

[Experimental study on antitumor activity of the root of Eurphorbia helioscopia in vivo].

Previous studies showed EWE could strongly inhibit the proliferation of tumor cells in vivo. Experimental results also showed that EWE had antitumor effect in S180-bearing mice and H22-bearing mice and prolonged life-span in S180-bearing mice in this paper, Furthermore, it was found that EWE could improve the immune function of S180-bearing mice. Therefore EWE can be considered as a potent antitumor herb.

[The development of intelligent multi-function instrument of treatment and rehabilitation for disablement of limb].

Intelligent multi-function limb treatment and rehabilitation instrument based on IBM-PC is introduced in this paper. It is mainly used for treatment and rehabilitation in the disablement of limb. The range speed and training time can be controlled and monitored. The instrument has the advantages of high reliability, easy operation and compact structure. It is a new kind of intelligent limb rehabilitation instrument.

[Surgical treatment of laryngotracheal stenosis].

OBJECTIVE: To evaluate the surgical treatment results of laryngotracheal stenosis. METHODS: Forty-five patients with laryngotracheal stenosis, atresia or defects who had been treated surgically were reviewed. Different operative reconstructions were used accordingly, such as simple scar resectoin and dilatation, mucosa resuturing, Z-plasty, flap reconstruction, sleeve resection. RESULTS: Forty-three patients had been treated successfully with good airway patency, although 37 of them had hoarseness of voice. One patient lost to follow-up and another had compromised airway after operation. CONCLUSION: The different methods of surgical reconstruction used here for the laryngotracheal stenosis proved to be effective.

Phase-1 studies of malariotherapy for HIV infecton.

OBJECTIVE: To determine whether malariotherapy (an old therapy for treatment of neurosyphilis) improves some clinical and laboratory parameters of HIV-positive patients without iatrogenic complications. METHODS: Total 8 asymptomatic HIV-1 positive subjects whose CD4 cell counts were over 250 x 10(6) cells/L were selected for the phase-1 studies of malariotherapy and were intravenously injected Plasmodia vivax to induce artificial malaria. Malaria was terminated with chloroquine after 10 to approximately 20 malarial fever episodes. Cell-bound CD4 levels were measured by APAAP (a solid-phase enzyme essay) and levels of neopterin (NPT), beta-2-microglobulin (B2M), soluble tumor necrosis factor receptor-2 (sTNF-RII), interleukin-2 (IL-2) and HIV P24 antigen were measured by ELISA. Patients were followed up to 24 to approximately 30 months. RESULTS: CD4 levels increased in 5, NPT decreased in 7 of 8 patients; IL-2 increased in 5 of 6 patients after malariotherapy. The total trends of B2M and sTNF-RII basically remained stable. HIV P24 antigen remained undetectable in 6, remained detectably low level in 1 and experienced increase in 1 of 8 patients after malariotherapy. No any severe complications occurred in all 8 patients. CONCLUSIONS: The results indicate that malariotherapy basically is safe for HIV infection and it seems that the therapy improves some immunological parameters of HIV patients.
