Histopathological changes of Macaca mulatta infected with Plasmodium knowlesi.

OBJECTIVE: To study the histopathological changes of relevant internal organs of Macaca mulatta infected with Plasmodium knowlesi (P. knowlesi). METHODS: Histopathological examination of 3 monkeys who died of P. knowlesi infection, 2 P. knowlesi infected monkeys who died of treatment failure with artesunate suppository and 1 P. knowlesi infected monkey that was cured by piperaquine phosphate (PQP) but died of trauma and necrosis of the fore limb. RESULTS: The heart, liver, spleen, lung, kidney, brain, pancreas, parathyroid, pituitary and lymph nodes showed severe pathological changes in 3 monkeys (No. 1, 7 and 12) who died of P. knowlesi infection and 1 infected monkey (No. 72) who died of treatment failure with artesunate suppository. Red blood cells containing malarial parasites and pigments were concentrated in the capillaries of these organs. Malarial pigments were deposited in many organs or phagocytized by macrophages in 1 monkey (No. 131), it was cured by piperaquine phosphate but died of trauma and necrosis of the fore limb; cellular atrophy and disappearance of pancreatic islets, parathyroid and pituitary cells were also observed. One monkey (No. 33) treated with artesunate suppository, showed that blood parasites became negative but recrudesced and pituitary later died from a gavage accident. Its organs showed a significant difference to those of the infected monkeys receiving no treatment. Only the liver Kupffer cells and cerebral matrix contained malarial parasites and pigments; many relevant internal organs showed repair. CONCLUSION: The pathological changes of relevant internal organs of Macaca mulatta infected with P. knowlesi were examined in detail, especially cellular atrophy and the disappearance of pancreatic islets, parathyroid and pituitary cells and myolysis of cardiac muscles. These changes have not previously been reported elsewhere.

[Multivariate logistic regression analysis on the risk factors of type 2 diabetes mellitus].

OBJECTIVE: To explore the relationship between multivariate factors and type 2 diabetes mellitus. METHODS: Polymorphisms of microsatellite markers in uncoupling protein 3 (UCP3) gene, hormone-sensitive lipase (HSL) gene and protein tyrosine phosphatase-1B (PTP-1B) gene were tested in 106 patients with type 2 diabetes and 102 control subjects by performing polymerase chain reaction (PCR), polyacrylamide gel electrophoresis and silver staining. Multivariate logistic regression analysis was performed by all factors. RESULTS: Through univariate analysis, type 2 diabetes had significantly positive associations with age, systolic blood pressure (SBP), fasting insulin (FINS) level, cholesterol, triglyceride, low-density lipoprotein, apolipoprotein B (ApoB), alpha lipoprotein, UCP3 gene allele 6, UCP3 gene allele 7, HSL gene allele 9, and negative associations with UCP3 gene allele 1 and 3, HSL gene allele 5, high-density lipoprotein. The results of multivariate logistic analysis showed that UCP3 gene allele 6, UCP3 gene allele 7, SBP, ApoB, alpha lipoprotein were still positively related to type 2 diabetes, while HSL gene allele 5, high-density lipoprotein were still negatively related to type 2 diabetes. CONCLUSION: Our data showed that UCP3 gene allele 6, UCP3 gene allele 7, SBP, abnormity of plasma ApoB and alpha lipoprotein might play a role in the development of type 2 diabetes. HSL gene allele 5, high-density lipoprotein might play some protective role in the development of type 2 diabetes.

[Effect of photoperiod and temperature on fertility alteration of short photoperiod-low temperature induced male sterile rice].

Short photoperiod-low temperature induced male sterile rice is a new germplasm, whose fertility alteration characteristic is contrary to that of long photoperiod and high temperature induced male sterile rice. Compared with that of Yid1s sowed under natural conditions by stages, the fertility alteration of Yid1s was observed under simulated short photoperiod treatment. The results showed that the photoperiod-sensitive stage of Yid1s occurred from differentiation of rachis primordia (phase I) to secondary rachis-branch and spikelet primordia (phase III). The panicles induced by appropriate short daylight treatment at photoperiod-sensitive stage tended to be completely sterile under normal temperature, but the sterile effect was weakened under high temperature. The pollen fertility of Yid1s was reduced under high or low temperature. The temperature-sensitive stages of Yid1s based on the analysis of the effects of high and low temperature on fertility were basically identical, i.e., from pollen mother cell formation (phase V) to single nucleus (phase VII).

[Relationship between the expression of connexin 43 and bystander effect of suicide gene therapy in ovarian cancer].

OBJECTIVE: To explore the relationship of connexin 43(Cx43) and bystander effect in ovarian tumor cells in herpes simplex virus thymidine kinase/ganciclovir (HSV-TK/GCV) gene therapy in vitro, and to investigate the effection of all-trans retinoic acid (RA) on expression of Cx43 and bystander effect. METHODS: Cx43 expression was examined with flowcyto-metry, Western Blot, and immunofluorescence in two ovarian tumor cells OVCAR3, CAOV3 before and after RA treatment. Bystander effect was determined by the cells growth inhibitory rate with methyl thiazolyl tetrazolium. RESULTS: Following exposure to ganciclovir, there was much greater bystander killing in OVCAR3 than in CAOV3 (P < 0.05). The expression of Cx43 was detected in OVCAR3 with flowcytometry and Wstern Blot, but it could not be detected in CAOV3. The expression of Cx43 in both cell lines could be induced by RA. Immunofluorescence staining showed that OVCAR3 Cx43 protein is located in membrane surface, whereas CAOV3 is in cytoplasm. RA could not change the location of Cx43 protein in both cell lines. CONCLUSIONS: There is relationship between Cx43 expression and HSV-TK/GCV bystander effect. HSV-TK/GCV bystander effect can be inhanced by RA in ovarian cancer.

[Suppression subtractive hybridization in the cloning of gene fragments in relation to lung cancer metastasis].

OBJECTIVE: In order to provide important information about early diagnosis and treatment of tumor metastasis, a study on the biological regulation of metastasis at the molecular level was carried out by isolating and identifying human metastatic suppressor gene or related DNA sequences from a pair of cell clones with different metastatic phenotypes. METHODS: Suppression subtractive hybridization technique (SSH), gene cloning and analysis by bioinformatics were applied to this study. Two cell clones of pulmonary giant cell carcinoma originating from the same patient were used. The PLA-801C cell clone which shows lower metastatic potentiality was used as tester and the PLA-801D as a driver. RESULTS: Five cDNA clones so obtained were expressed on much higher levels in the poorly metastatic cancer cell clone than the highly metastatic one. Homology analysis of all five cDNA fragments by BLASTN through GenBank showed high homology with the known human genes. CONCLUSION: Through using SSH, we have identified five cDNA fragments which are expressed at much higher levels in the poorly metastatic cancer cell clone than the highly metastatic variety. Therefore, it is supposed that they might play an important role in the suppression of metastatic tumor cells and may very well have potential application in the diagnosis and treatment of metastasis.

[Insertion/deletion polymorphsim of the angiotensin I converting enzyme gene and pulmonary thromboembolism in Chinese population].

OBJECTIVE: To investigate whether there is an association between the insertion deletion polymorphsim of human angiotensin I converting enzyme gene (ACE) and pulmonary thromboembolism and whether D allele increases the risk of pulmonary thromboembolism in Chinese population. METHODS: Seventy-two patients with pulmonary thromboembolism and 72 sex and age matched healthy controls were recruited in this study, all patients were diagnosed by lung ventilation/perfusion scan and/or ultrafast CT as well as medical history. Risk factors of venous thrombosis and pulmonary thromboembolism were inquired. Genome DNA was extracted from whole blood using phenol-chloroform. Subjects were genotyped for the I/D polymorphism of ACE gene using polymerase chain reaction based restriction fragment length polymorphism analysis. RESULTS: (1) The incidences of trauma, operation, phlebitis and venous varicose were significant higher in the patient than in controls. No significant difference were found in the family history of pulmonary thromboembolism and cardiovascular diseases, personal history of oral contraceptive, cigarette smoking and alcohol drinking between the patient and controls. (2) Frequencies of allele I and D in the controls were 0.66 and 0.34 respectively, the distribution of genotypes met the in Hardy-Weinberg equilibrium. No significant differences were found in the frequencies of genotype II, ID and DD between patients and controls. (3) The recessive allele model was informative and the odds ratio of DD genotype was 2.51 (P < 0.05) compared with the other two genotypes. (4) After further stratification, we found DD genotype was bound to be associated with an 2.64-fold risk of pulmonary thromboembolism for those individuals without existing traditional environmental risk factors and 3.36-fold risk for individuals with medical history of venous thrombosis (P < 0.05). CONCLUSION: This study shows that I/D polymorphism of ACE gene is associated with pulmonary thromboembolism in Chinese population and allele D is recessive. Our results indicate that DD genotype increases the risk of pulmonary thromboembolism for individuals who have no traditional risk factors of venous thrombosis or pulmonary thromboembolism and also for those with personal history of venous thrombosis.

[A clinical report on hydrophilic acrylic foldable lens implantation].

OBJECTIVE: To investigate the therapeutic effects of hydrophilic acrylic foldable lens implantation. METHODS: This prospective randomized placebo-controlled study comprised 214 eyes of 190 patients after phacoemulsification. The hydrophilic acrylic foldable lens was implanted in group 1 (114 eyes), and the PMMA intraocular lens (IOL) implanted in the control group 2 (100 eyes). Within the 3-6 month follow-up period, the changes of visual acuity, corneal astigmatism and endothelial cells, the location of IOL and the degree of posterior capsular opaqueness postoperatively between the two groups were compared. RESULTS: The visual acuity of group 1 was significantly better than that of group 2 within 3 days postoperatively (P < 0.05), but there was no statistically significant difference after 3 months. The surgically induced astigmatism, incidents of posterior capsular opaqueness and subluxation of IOL of group 1 were significantly lower than that of group 2 after 3 months postoperatively (P < 0.05). The comparison of the endothelial cell loss ratio between the two groups was not significantly different (P > 0.05). CONCLUSION: The hydrophilic acrylic foldable IOL is easy to handle during implantation, highly biocompatible in the eyes, with less postoperative inflammatory reactions and shorter recovery period, and may provide satisfactory therapeutic effects.

[Meta-analysis on the association of ACE/ID polymorphism and essential hypertension in Chinese population].

OBJECTIVE: To study on the association of the insertion(I)/deletion(D) polymorphism of the angiotensin-converting enzyme gene intron 16 (ACE/ID) and essential hypertension(EH) in Chinese population by the means of meta-analysis. METHODS: Odds ratios of ACE/ID genotype distributions in EH patients against healthy control were analysed. All the relevant studies were identified, poor-qualified studies were eliminated, and the risk of publication bias was excluded. The meta-analysis software, REVMAN3.1, was applied for investigating heterogeneity among individual studies and summarising effects across studies. RESULTS: A total of 1,612 cases and 1,710 controls from 18 studies were included. No heterogeneity among the studies has been found. The frequencies of the ACE DD, ID and II genotypes were 23%, 41%, and 36% in cases and 19%, 46% and 35% in controls respectively. The pooled odds ratio (with 95% CI) of DD vs ID + II is 1.37(1.15-1.63) (P < 0.01), while the pooled odds ratio of II vs ID + DD is 0.96 (0.83-1.12) (P > 0.05). CONCLUSION: In Chinese population (mainly the Hans), DD genotype is associated with the increased risk of EH.

[The influence of transforming growth factor beta 1 (TGF beta 1) on fibroblast proliferation and collagen synthesis].

OBJECTIVE: To investigate the significance and the influence of transforming growth factor beta 1 (TGF beta 1) on fibroblast proliferation and collagen synthesis. METHODS: The fibroblasts derived from healthy human dermis and pathologic scar were harvested and cultured in vitro with TGF beta 1. The effects of TGF beta 1 on the proliferation and collagen synthesis of the fibroblasts were investigated during the early phase (24 h) of cell confluence by means of nonradioactive cellular proliferation detection, 3H thymidine and 3H proline incorporation and DNA quantification analysis. RESULTS: After the co-culture with TGF beta 1, the incorporation of 3H thymidine and 3H proline and the proliferation were enhanced significantly in the fibroblasts from pathologic scar (P < 0.01) but remained at normal level in those from healthy human dermis(P > 0.05). CONCLUSION: The cell proliferation and collagen synthesis of scar-derived fibroblasts could be positively modulated and controlled by TGF beta 1, which might be very important in the scar formation.

[The effect of viper venom on the attachment, migration and proliferation of Tenon's capsular fibroblasts].

OBJECTIVES: To observe the effect of viper (Ahylysantipfarciasi) venom on attachment to collagen, migration and proliferation of rabbit Tenon's capsular fibroblast (TFs) in tissue culture. METHODS: Anti-adhesion experiment: The 3 -- 5 passage TFs suspended in DMEM medium were pre-incubated for 30 minutes in the presence of various concentrations of viper venom (0, 2.5 x 10(-4), 5.0 x 10(-4), 1.0 x 10(-3) and 5.0 x 10(-3) U/ml) at 37C, 5% CO(2), then inoculated in 24-well plate coated with rat-tail collagen. After incubation for 90 minutes, the floating cells were removed. The attached cells in each well were enumerated microscopically and determined by the value of absorption (A) of 3-(4, 5-dimethylthiazolzyl)-2, 5-diphenyl tetrazodium bromide (MTT). Anti-migration experiment: A denuded area was made when the 3rd passage of TFs was confluent into a monolayer. Cells were then exposed to viper venom (0 -- 5.0 x 10(-3) U/ml) at 37C, 5% CO(2), and the cells having migrated into the denuded area were enumerated every 6 hours. Anti-proliferative experiment: Two hours after the 3rd passage of TFs was incubated in the 24-well plate at 37C, 5% CO(2), they were exposed to different concentrations of viper venom drug (0 -- 5.0 x 10(-3) U/ml). Twenty-four and 48 hours later, the number of cells was determined by MTT method. RESULTS: Viper venom inhibited TFs from attaching to collagen in a dose-dependent manner and the ID(50) was 1.0 x 10(-3) U/ml. Only did 5.0 x 10(-3) U/ml of viper venom show significant difference from the control during 6 -- 48 hours. The difference of A value was not significant among all groups at 24 and 48 hr. CONCLUSIONS: In vitro, viper venom (> 1.0 x 10(-3) U/ml) can significantly inhibit the attachment of TFs to collagen, and 5.0 x 10(-3) U/ml can inhibit the migration, but can not affect their proliferation.

[Study on the association of pathologic myopia with HLA-DQB1 gene].

OBJECTIVE: To investigate the association of pathologic myopia (PM) with HLA-DQB1 alleles in order to study the pathogenesis of PM. METHODS: The genome DNA of 66 patients with PM was extracted from the blood. The second exon of the HLA-DQB1 gene was amplified by polymerase chain reaction (PCR), and individual PCR products were digested by allele specific restriction enzymes of Hae III, BssH II, Apa I, BsaH I, Hae II, Hpa II, Rsa I, Bsp 1286 I for typing. Genotype was determined by restriction fragment length polymorphism (RFLP) pattern. The frequency of 16 alleles of HLA-DQB1 was detected and compared with that of the healthy control. RESULTS: The frequencies of HLA-DQB1 * 0301 and * 0303 were significantly higher in the patients with PM (P < 0.0001), while that of HLA-DQB1 * 0601 and * 0602 were significantly lower (P < 0.0001) than that of the control group. CONCLUSION: HLA-DQB1 * 0301, * 0303 alleles are susceptible alleles, and possibly they are the pathogenic genes, * 0601, * 0602 are resistant alleles, and may be they possess the protective property.

[Protective effect of huyin decoction on acute hepatic injury induced by CCl4 in mice].

OBJECTIVE: To observe the protective effect of Huyin decoction on acute hepatic injury induced by CCl4 in mice. METHODS: The model of acute hepatic injury induced by CCl4 was used. The levels of ALT and AST in serum were detected. And histopathology of liver was observed. RESULTS: Huyin decoction could decrease ALT and AST in serum and hepatic histopathological injury of the acute hepatic injury mice. CONCLUSION: Huyin decoction possessed protective effect on acute hepatic injury in mice.

[Regulatory effect of Bcl-2 on the cortical neurons of primary cultured mice with HSV-1 infection].

OBJECTIVE: To study the effect of Bcl-2 on the cortical neurons of primary cultured mice with HSV-1 infection. METHODS: Analysis of the expression of Bcl-2 on the cortical neurons of primary cultured fetal mice in vitro infected with HSV-1 for 11 hours or exposed to sorbitol for 5 hours was made by flow cytometry and Western blotting. RESULTS: The Bcl-2 of the cultured neurons undergoing HSV-1 infection expressed upregulating compared with the mock untreated neurons. The Bcl-2 protein of HSV-1 infected and exposed to sorbitol neurons expressed upregulating compared with the control group. The Bcl-2 protein of the primary cultured 3 day cells expressed higher than that of the primary cultured 7 day cells. The higher neurovirulence, the higher expression of Bcl-2. CONCLUSIONS: Bcl-2 might protect the primary cultured cortical neurons of fetal mice from apoptosis where infected with HSV-1, and thereby the lifespan of host cells may be prolonged.

[Study of blood dynamics of ophthalmic artery in patients with sudden sensorineural hearing loss using color Doppler flow imaging].

OBJECTIVE: To investigate the relationship between the pathogenesis of sudden sensorineural hearing loss (SSHL) and the disorder of blood circulation in inner ear. METHOD: Blood dynamics of the ophthalmic artery were studied quantitatively using color doppler imaging in 34 patients with SSHL. RESULT: Compared with 34 self-controls and 15 normal controls, 28 patients (82.4%) with SSHL had significantly lower blood flow velocities and higher resistance indices (P < 0.05), and there was no significant difference between the self-control group and the normal control group (P > 0.05). CONCLUSION: The study suggested that the blood situations--the decreased blood flow velocities and perfusion and increased resistance of ophthalmic artery in patients with SSHL maybe play a role in the pathogenesis of SSHL.

[The application of mastoid cavity plombage in open method tympanoplasty].

OBJECTIVE: The role mastoid cavity plombage played in restoring physiological function of external meatus in open method tympanoplasty was studied. METHOD: 85 cases of cholesteatoma otitis media were performed open method tympanoplasty with the broken bits of their own mastoid process cortex, homological costal cartilage or U shape periosteum connective tissue valve. And plombage was performed at the same time. The reconstruction of ossicular chain (type 0 tympanoplasty): 10 ears did not undergo reconstruction due to increase ment of bone conduction threshold or were in preparation for the second phase operation; 40 ears underwent type III tympanoplasty; 35 ears underwent type IV tympanoplasty. RESULT: The total improvement rate of hearing was 76%, among which 80% were performed type III tympanoplasty and 71.4% underwent type IV tympanoplasty. After operation, the time waited till dry-ears was 19.8 +/- 65.31 days and the dry-ears rate was 97.6% (83/85). Six months after operation, 89.4% (76/85) had smooth external meatus and fine self-cleaning function and no recurrence was found. CONCLUSION: As to cholesteatoma otitis media that could not be performed closed method tympanoplasty, U shape periosteum connective tissue valve and mastoid process cortex were used. Mastoid cavity plombage with homological costal cartilage could effectively overcome the shortcomings of destruction of physiological structure of external meatus caused by open method tympanoplasty, the self-cleaning function of skins of external meatus was remained and the life quality of patients after operation was increased.

[Selective isolation of anethole from volatile oil of Foeniculum vulgare Mill by inclusion crystalline with chela-shape host].

AIM: To isolate the components from the volatile oil of Foeniculum vulgare Mill. METHODS: According to the function of molecular recognition of supramolecular chemistry, chela shape molecule, trans-1, 2-biphenyl-1, 2-acenaphthendiol was used as host molecule and the volatile oil of Foeniculum vulgare Mill as guest molecule. Trans-1, 2-biphenyl-1, 2-acenaphthendiol can recognize the components that endowed with interactional complementarity and form inclusion compound as crystals. RESULTS: The anethole in the volatile oil was selectively included as trans-1,2-biphenyl-1,2-acenaphthendiol which was obtained in pure state from the inclusion compound by Kugelrohr vacuum technology. The formation of inclusion compound was confirmed by means of IR and powder XRD. The structure of the selectively isolated component was elucidated as trans-anethole by means of IR, 1HMMR and MS. CONCLUSION: The experimental results showed that the method is simple, rapid and selective for isolation anethole from volatile oil of Foeniculum vulgare Mill.

[Study on the structure of epimerides of cycloclausenamide].

AIM: To study the structures of the epimerides of cycloclausenamide. METHODS: The structures of compound I, extracted from Clausena lansium (Lour.) Skeels, and synthesized compound III were determined by single crystal X-ray diffraction analysis. The stereo-structures of compound II and IV were also built up through Tripos force field based on crystal structures of compound I and III. RESULTS: The molecular formula and molecular weight were found to be C18H17O2N and 279.34 respectively. Compound I crystallized in monoclinic system, space group P2(1) with a = 0.5928(1), b = 1.5014(1), c = 1.6190(1) nm, V = 1.4410(3) nm3, Z = 4, Dx = 1.288 g.cm-3, Rf = 0.075, Rw = 0.073(w = 1/sigma 2|F|), S = 3.983; compound III crystallized in triclinic system, space group P1 with a = 0.5667(1), b = 1.2934(1), c = 2.1119 (1) nm, alpha = 102.17(1), beta = 90.25(1), gamma = 102.65(2) degrees, V = 1.4770(5) nm3, Z = 4, Dx = 1.224 g.cm-3, Rf = 0.047, Rw = 0.051(w = 1/sigma 2|F|), S = 0.467. CONCLUSION: These results showed that compound I and III both are cycloclausenamide except that the directions of the phenyl group on C6 are different. Cycloclausenamide can form 4 pairs of epimerides but the directions of the phenyl group does not affect their energy in free state.
