Performance improvement of hybrid membrane bioreactor with PAC addition for water reuse.

A study was carried out on a hybrid (AS-SBF) membrane bioreactor (HMBR) for the municipal wastewater reclamation and reuse at Chengfengzhuang WWTP in Daqing City, Heilongjiang Province. It was found that the effects of DO and water temperature on performance of the HMBR was significant. Under the conditions of water temperature in range of 10-14 degrees C, pH of 6.6- 7.0, DO of 4-6 mg/l and HRT of 7 h, the HMBR exhibited removal efficiencies for CODcr, BOD5, NH3-N and TN of 96.7%, 98.9%, 93.7% and 60.5% respectively. The turbidity of effluent from HMBR was below 1 NTU. The effluent of HMBR meets the standard of wastewater reclamation for oil exploitation. PAC was added into the bioreactor at the second operating stage, in order to further research parameters variation. The flux was improved by 53.2%, compared to the membrane without PAC-addition, due to formation of a PAC pre-coat layer on the membrane surface, with lots of advantages such as larger granules, higher porosity, non-compressibility, higher filterability and easy removal, compared with pure biomass layer. In addition, the performance of HMBR was further improved, due to adsorption and degradation of SMPs, the average removal of CODcr and TN was further improved by 5.1% and 13.5% respectively. Biomass in the HMBR was quantitatively measured, of which the biofilm played a major role in pollutants removal.

The effect of inlet velocity profile on the bifurcation COPD airway flow.

The effect of inlet velocity profile on the flow features in obstructed airways is investigated in this study. In reality, the inlet velocity distributions on such models, which are extracted from medial branches of natural human lung, should be neither uniform, nor symmetric parabolic, but skewed-parabolic due to having been skewed by the upper carina ridges. Four different three-dimensional three-generation models based on the 23 generations model of Weibel have been considered, respectively. The fully three-dimensional incompressible laminar Navier-Stokes equations and continuity equation have been solved using CFD solver on unstructured tetrahedral meshes. To reduce the complexity of the simulations, only one Reynolds number of 900 was used in this calculation. Four types of inlet boundary conditions, namely uniform, parabolic, positive-skewed parabolic (skewed to the positive x-direction), and negative-skewed parabolic, were imposed on the obstructed airway models, which were considered to be obstructed at either the second generation or the third generation airways, respectively. The results show that the inlet velocity profile has significant influence on the flow patterns, mass distributions, and pressure drops in either the symmetric model, or the three obstructed models. The three generation airways may not be enough to study the bifurcation flow in chronic obstructive pulmonary disease (COPD) airways, and a four-generation or more airway model is necessary to get better predictive results.

Presence of plastid and absence of mitochondrial DNA in male reproductive cells as evidence for cytoplasmic inheritance in Turnera ulmifolia and Zantedeschia aethiopica.

Epifluorescence microscopy of mature pollen grains of Turnera ulmifolia and Zantedeschia aethiopica stained with 4',6-diamidino-2-phenylindole demonstrated the presence of fluorescent cytoplasmic DNA aggregates in the male reproductive cells of both species. Double staining of the cells with 4',6-diamidino-2-phenylindole and 3,3'-dihexyloxacarbocyanine iodide in Technovit resin sections showed that the mitochondria of these cells did not correspond to the fluorescent cytoplasmic DNA aggregates. Electron microscopy studies revealed both plastids and mitochondria in the cells of these species. In addition, immunoelectron microscopy using an anti-DNA monoclonal antibody showed clear labeling of plastids but not mitochondria. These data provide cytological evidence for biparental plastid inheritance and maternal mitochondrial inheritance in these species.

Effect of temperature on the surface free energy of amorphous carbon films.

Diamond-like carbon (DLC) and tetrahedral amorphous carbon (ta-C) have attracted much attention recently for biomedical and antifouling applications due to their excellent biocompatibility and inherent nonstick properties. It has been demonstrated that the solid surface free energy is a dominant factor in cellular or fouling adhesion. However, few data for the surface free energy of DLC and ta-C coatings at temperatures in the range 37-95 degrees C are available. In this study DLC and ta-C coatings on stainless steel 304 sheets were prepared using an unbalanced magnetron sputtering system and a filtered cathodic vacuum arc system, respectively. The contact angles of water, diiodomethane and ethylene glycol on the coated surfaces at temperatures in the range 20-95 degrees C were measured using a Dataphysics OCA-20 contact angle analyzer. The surface free energy of the coatings and their components (e.g., dispersion, polar or acid/base portions) were calculated using various methods. The experimental results showed that the total surface free energy and dispersive surface free energy of the ta-C coatings, DLC coatings, stainless steel 304 and titanium decreased with increasing surface temperature, while the acid-base SFE component increased with increasing temperature.

Odd-parity superconductivity in Sr2RuO4.

Phase-sensitive measurements were made on Sr2RuO4 to establish unambiguously the odd-parity pairing in this material. The critical current of Au(0.5)In(0.5)-Sr2RuO4 superconducting quantum interference devices prepared on Sr2RuO4 single crystals was found to be a maximum for devices with junctions on the same side of the crystal and a minimum for devices with junctions on opposite sides, in the limit of zero magnetic flux; these findings indicate that the phase of the superconducting order parameter in Sr2RuO4 changes by pi under inversion. This result verifies the odd-parity pairing symmetry and the formation of spin-triplet Cooper pairs in Sr2RuO4.

Disgust and the insula: fMRI responses to pictures of mutilation and contamination.

Although previous functional brain imaging studies have found that the insula responds selectively to facial expressions of disgust, it remains unclear whether the insula responds selectively to disgust-inducing pictures. In this fMRI study, healthy volunteers viewed pictures of contamination, human mutilation, attacks and neutral scenes during scanning, and then rated pictures for the 'basic' emotions. The anterior insula responded to contamination and mutilation but not attacks, while the ventral visual areas responded to attacks and mutilations more strongly than contamination. The above activations were predicted by disgust and arousal ratings respectively. Additionally, mutilations uniquely activated the right superior parietal cortex. These results support selective disgust processing at the insula, and suggest distinct neural responses to contamination and mutilation.

Novel potentiometric immunosensor for hepatitis B surface antigen using a gold nanoparticle-based biomolecular immobilization method.

A novel potentiometric immunosensor for the detection of hepatitis B surface antigen has been developed by means of self-assembly to immobilize hepatitis B surface antibody on a platinum disk electrode based on gold nanoparticles, Nafion, and gelatin as matrices in this study. The modification procedure of the immunosensor was further characterized by using cyclic voltammetry and the enzyme-linked immunosorbent assay (ELISA) method. The detection is based on the change in the electric potential before and after the antigen-antibody reaction. In contrast to the commonly applied methods (e.g., the glutaraldehyde crosslinking procedure), this strategy could allow for antibodies immobilized with a higher loading amount and better retained immunoactivity, as demonstrated by the potentiometric measurements. A dynamic concentration range of 4-800 ng ml(-1) and a detection limit of 1.3 ng ml(-1) were observed. Analytical results of several human serum samples obtained using the developing technique are in satisfactory agreement with those given by ELISA. In addition, the technique presents some distinct advantages over the traditional sandwich format in that the analyzing performances are direct, rapid, and simple without multiple separation and labeling steps.

Dynamics of Rydberg electron transfer to CH3CN: velocity dependent studies.

The dynamics of free-ion production through electron transfer in K(np)/CH3CN collisions are examined through measurements using velocity-selected Rydberg atoms. The data show that Rydberg electron transfer leads to the creation of two groups of dipole-bound CH3CN- ions, one long lived (tau>85 micros), the other short lived (tau<1 micros). The velocity dependences associated with the production of both groups of ions are similar, the ion formation rate decreasing markedly with decreasing Rydberg atom velocity, principally as a consequence of postattachment electrostatic interactions between the product ions. The results are in reasonable accord with the predictions of a Monte Carlo collision model that considers the effect of crossings between the diabatic potential curves for the covalent K(np)/CH3CN system and the K+/CH3CN- ion pair. This model also accounts for the relatively small reaction rate constants, approximately 0.5-1.0 x 1.0(-8) cm(3) s(-1), associated with the formation of long-lived CH3CN- ions. No velocity dependence in the lifetime of the CH3CN- ions is observed.

Interrelation between HeLa-S3 cell transfection and hemolysis in red blood cell suspension using pulsed ultrasound of various duty cycles.

We have studied the in vitro transfection of a plasmid DNA with the lacZ gene to HeLa-S3 cells and hemolysis in a red blood cell (RBC) suspension under pulsed ultrasound with duty cycles gamma of 10, 20 and 30% using a digital sonifier at a frequency of 20 kHz and an intensity of 6.2 W/cm(2) on the surface of a horn tip. Cultured HeLa-S3 cells in suspension were exposed to pulsed ultrasound for an apparent exposure time t' from 0 to 60 s. HeLa-S3 viability decreased as a single exponential function of the total exposure time t = gammat' with a common time constant tau = 3.8 s for three duty cycles. Transfection was evaluated by counting the number of beta-galactosidase(beta-Gal)-positive cells relative to the total number of cells. Pulsed ultrasound provided an enhanced transfer of the beta-Gal plasmid to HeLa-S3 cells, 3.4-fold as compared with that in the case of the control. The optimal transfection efficiencies were 0.75, 0.80 and 0.74% near t = tau with gamma = 10, 20 and 30%, respectively. The number ratio of beta-Gal-positive cells to the surviving cells after exposure increased with t' according to a modified logistic equation. The degree of hemolysis also increased exponentially with t' at a time constant tau' = tau(0)/gamma for the RBC suspension in physiological saline at a hematocrit concentration of 0.5% with tau(0) = 0.9 s. Thus the total exposure time for the optimal transfection efficiency was tau, that is, nearly four times of tau(0). Hemolysis in the RBC suspension may be a useful model for determining optimal transfection by pulsed ultrasound of various duty cycles.

Influence of the arbuscular mycorrhizal fungus Glomus mosseae on uptake of arsenate by the As hyperaccumulator fern Pteris vittata L.

We report for the first time some effects of colonization by an arbuscular mycorrhizal (AM) fungus (Glomus mosseae) on the biomass and arsenate uptake of an As hyperaccumulator, Pteris vittata. Two arsenic levels (0 and 300 mg As kg(-1)) were applied to an already contaminated soil in pots with two compartments for plant and hyphal growth in a glasshouse experiment. Arsenic application had little or no effect on mycorrhizal colonization, which was about 50% of root length. Mycorrhizal colonization increased frond dry matter yield, lowered the root/frond weight ratio, and decreased frond As concentration by 33-38%. Nevertheless, transfer of As to fronds showed a 43% increase with mycorrhizal colonization at the higher soil As level. Frond As concentrations reached about 1.6 g kg(-1) (dry matter basis) in non-mycorrhizal plants in the As-amended soil. Mycorrhizal colonization elevated root P concentration at both soil As levels and mycorrhizal plants had higher P/As ratios in both fronds and roots than did non-mycorrhizal controls.

Opposite morphological responses of partially denervated cortical serotonergic and noradrenergic axons to repeated stress in adult rats.

We examined plastic changes in serotonin (5-HT) axons following repeated stress in the adult rat brain, and compared stress-induced changes between 5-HT and noradrenaline (NA) axons. We locally injected the specific neurotoxin to 5-HT axons or to NA axons into the frontal cortex to cause partial denervation. The animals were mildly restrained from 1 day after the neurotoxin injection and this stress was repeated daily for 20 min during the first 2 days and for 40 min during the next 11 days. On the fourteenth day after injection, the brains were removed to visualize 5-HT and NA axons by immunohistochemistry. Repeated stress did not significantly alter the denervation area of 5-HT or NA axons, but the density of 5-HT axons was increased whereas that of NA axons was decreased in cortical regions outside the denervation site. In addition, the expression of brain-derived neurotrophic factor (BDNF) was increased in cortical regions where the 5-HT axon density was increased in response to stress. These results suggest that repeated stress causes opposite changes in the morphology of partially denervated 5-HT and NA axons in the cerebral cortex. The stress-induced increase in BDNF expression may contribute to 5-HT axon sprouting following repeated stress.

Highly coherent light at 13 nm generated by use of quasi-phase-matched high-harmonic generation.

By measuring the fringe visibility in a Young's double pinhole experiment, we demonstrate that quasi-phase-matched high-harmonic generation produces beams with very high spatial coherence at wavelengths around 13 nm. To our knowledge these are the highest spatial coherence values ever measured at such short wavelengths from any source without spatial filtering. This results in a practical, small-scale, coherent, extreme-ultraviolet source that is useful for applications in metrology, imaging, and microscopy.

Determinants of isocyanate exposures in auto body repair and refinishing shops.

As part of the Survey of Painters and Repairers of Auto bodies by Yale (SPRAY), the determinants of isocyanate exposure in auto body repair shops were evaluated. Measurements (n = 380) of hexamethylene diisocyanate-based monomer and polyisocyanate and isophorone diisocyanate-based polyisocyanate were collected from 33 auto body shops. The median total reactive isocyanate concentrations expressed as mass concentration of the NCO functional group were: 206 microg NCO/m3 for spray operations; 0.93 microg NCO/m3 for samples collected in the vicinity of spray operations done on the shop floor (near spray); 0.05 microg NCO/m3 for office or other shop areas adjacent to spray areas (workplace background); 0.17 microg NCO/m3 for paint mixing and gun cleaning operations (mixing); 0.27 microg NCO/m3 for sanding operations. Exposure determinants for the sample NCO mass load were identified using linear regression, tobit regression and logistic regression models. For spray samples in a spray booth the significant determinants were the number of milliliters of NCO applied, the gallons of clear coat used by the shop each month and the type of spray booth used (custom built crossdraft, prefabricated crossdraft or downdraft/semi-downdraft). For near spray (bystander) samples, outdoor temperature >65 degrees F (18 degrees C) and shop size >5000 feet2 (465 m2) were significant determinants of exposure levels. For workplace background samples the shop annual income was the most important determinant. For sanding samples, the shop annual income and outdoor temperature >65 degrees F (18 degrees C) were the most significant determinants. Identification of these key exposure determinants will be useful in targeting exposure evaluation and control efforts to reduce isocyanate exposures.

Smallest carbon nanotube is 3 a in diameter.

Previous energetic considerations have led to the belief that carbon nanotubes (CNTs) of 4 A in diameter are the smallest stable CNTs. Using high-resolution transmission electron microscopy, we find that a stable 3 A CNT can be grown inside a multiwalled carbon nanotube. Density functional calculations indicate that the 3 A CNT is the armchair CNT(2,2) with a radial breathing mode at 787 cm(-1). Each end can be capped by half of a C12 cage (hexagonal prism) containing tetragons.

Charge separation of dense two-dimensional electron-hole gases: mechanism for exciton ring pattern formation.

We report on new experiments and theory that unambiguously resolve the recent puzzling observation of large diameter exciton emission halos around a laser excitation spot in two dimensional systems. We find a novel separation of plasmas of opposite charge with emission from the sharp circular boundary between these two regions. This charge separation allows for cooling of initially hot optically generated carriers as they dwell in the charge reservoirs for very long times.

Synthesis and characterization of a brush-like copolymer of polylactide grafted onto chitosan.

A brush-like poly(DL)-lactide grafted onto chitosan as the backbone was investigated. The graft copolymerization was carried out with triethylaluminum as catalyst in toluene at 70 degrees C. It was found that a greater lactide content in the feeding ratio results in a higher grafting percentage. FTIR spectrometry, (1)H NMR, DSC scanning, and wide-angle X-ray scattering, respectively, are used to characterize these branch copolymers. A copolymer has a definite melting point when the molar feeding ratio of lactide to chitosan is more than 10:1, and the deltaH of the copolymers increases with the feed ratio of lactide to chitosan in feeding.

Observation of the decay psi(2S)-->K0SK0L.

The decay psi(2S)-->K(0)(S)K(0)(L) is observed using psi(2S) data collected with the Beijing Spectrometer at the Beijing Electron-Positron Collider; the branching fraction is determined to be B(psi(2S)-->K(0)(S)K(0)(L))=(5.24+/-0.47+/-0.48)x10(-5). Compared with J/psi-->K(0)(S)K(0)(L), the psi(2S) branching fraction is enhanced relative to the prediction of the perturbative QCD "12%" rule. The result, together with the branching fractions of psi(2S) decays to other pseudoscalar meson pairs (pi(+)pi(-) and K+K-), is used to investigate the relative phase between the three-gluon and the one-photon annihilation amplitudes of psi(2S) decays.

Identification of molecular targets for dietary energy restriction prevention of skin carcinogenesis: an idea cultivated by Edward Bresnick.

Dietary energy restriction (DER) has long been known to strikingly inhibit carcinogenesis in many animal models. The animal data has been corroborated by recent and ongoing epidemiological studies demonstrating the importance of energy balance, physical exercise and obesity in human cancer. Dr. Edward Bresnick provided key insights into this important area of research and pivotal direction for the author's research while he served as Director of the Eppley Institute for Research in Cancer, Omaha, NE. These insights moved this research toward demonstrating that DER reduced the expression of key protein kinase C isoforms in mouse skin. More recent studies have uncovered downstream events that are inhibited by DER including blockage of tumor promoter activation of Raf-1, ERK 1,2 and AP-1 expression. Parallel studies have demonstrated the DER inhibition of these key cellular signaling events in mouse skin carcinogenesis are dependent upon an intact adrenal gland because adrenalectomized mice fed DER diet did not have reduced tumor burden or inhibited signaling and blocked AP-1 activation as was observed in DER mice with intact adrenal glands. In addition, the DER inhibition of tumorigenesis and AP-1 signaling was restored in adrenalectomized mice that were given corticosterone in the drinking water. This showed that in mice in the chemical carcinogenesis protocol glucocorticoid hormone plays a major role in mediating DER prevention of cancer. Studies are ongoing to further assess the mechanism of DER modulation of skin cancer by assessing impacts on transcriptional regulation and expression of genes that are critical in skin carcinogenesis.
