[Analysis of telomerase activity and telomere length in acute myelogenous leukemia].

OBJECTIVE: To study the changes and significance of telomerase activity and telomere length in acute myelogenous leukemia. METHODS: TRAP-ELISA-PAGE was used to detect telomerase activity, Southern blot to estimate the length of telomere. RESULTS: Telomerase activity was significantly higher in AML(Absorption(A): 2.298 +/- 1.059) than in normal control(A: 0.387 +/- 0.598) and the mean telomere length of AML [(7.6 +/- 2.1) kb] was significantly shorter than that of normal control[(9.3 +/- 1.9) kb]. The alteration of the telomere length was detected mainly in telomerase-positive AML. CONCLUSIONS: It suggested that there was a close relationship between telomerase activity and telomere length in AML. The activation of telomerase might play an important role in the genesis and development of AML and telomere length changes may correlate with the activation of telomerase.

[E-cadherin associated protein expression and its significance in invasive lobular carcinoma and invasive ductal carcinoma of breast].

OBJECTIVE: To study E-cadherin associated proteins alpha-, beta-, gamma-catenin expression and their significance in invasive lobular carcinoma (ILC) and invasive ductal carcinoma (IDC) of breast. METHODS: Immunohistochemistry staining (labeled streptavidin-biotin method) was used to detect alpha-, beta-, gamma-catenin expression in invasive breast carcinoma (19 cases of ILC and 32 cases of IDC ). RESULTS: The loss and reduction rate of alpha-, beta-, gamma-catenin expression in ILC was 78.9% (15 cases), 52.6% (10 cases) and 84.2% (16 cases), respectively. The rate of alpha-, beta-, gamma-catenin expression showing loss and reduction in IDC was 75.0% (24 cases), 43.8% (14 cases) and 81.3% (26 cases), respectively. The staining intensity of alpha-, beta-, gamma-catenin expression in invasive carcinoma of breast was weaker than that in foci of carcinoma in situ (semiquantitative grading). There was a positive relationship between the expression of alpha-catenin and beta-catenin in invasive breast carcinoma. No significant association was seen between reduced protein expression of alpha-, beta-, gamma-catenin and the development of lymph node metastasis. CONCLUSION: alpha-, beta-, gamma-catenin expression was almost identical in ILC and IDC of breast, showing significant loss and reduction of protein expression in the carcinoma cells of invasive breast carcinoma, suggesting that alpha-, beta-, gamma-catenin may lose their normal adherent ability in the pathogenesis of invasive breast carcinoma.

[The influence of recombinant human growth hormone on the metabolism of glucose and protein in severely burned patients].

OBJECTIVE: To investigate the influence of recombinant human growth hormone (rhGH) on the metabolism of glucose and protein in severely burned patients. METHODS: The serum levels of total protein and albumin and the incidence of hyperglycemia in 24 cases of severely burned patients treated by rhGH were determined and compared with those in patients without rhGH treatment (control group). RESULTS: The serum levels of total protein (TP) and albumin in burned patients after being treated by rhGH were much higher than those before treatment and those in control group. The blood level of glucose increased within 2 weeks, especially 1 week postburn. The incidences of hyperglycemia were 52.38%, 50.24% and 20% on 0 similar 3, 4 similar 7 and 8 similar 14 postburn days (PBDs), respectively. And the incidences of hyperglycemia were 100%, 50% and 0% when the administration of rhGH started within 1, 2 and after 2 weeks respectively. CONCLUSION: The systemic protein synthesis in severely burned patients could be accelerated by the use of rhGH. But early administration of rhGH in burned patients could affect blood sugar level obviously. And the optimal time to give rhGH is 2 weeks after burn injury.

[Affect of As(2)O(3) on the sister chromatid exchanges in human blood lymphocytes].

OBJECTIVE: To assess the potential of As(2)O(3) in inducing malignant transformation at its therapeutic concentrations. METHODS: The sister chromatid exchange (SCE) in human peripheral blood lymphocytes (PBL) treated with different concentrations of As(2)O(3) was detected by the staining technique for sister chromatid differentiation. RESULTS: At 2 x 10(-6)mol/L and 1 x 10(-6) mol/L of As(2)O(3), the SCE frequency in PBLs was significantly increased as compared with the control (P < 0.01). Most of the lymphocytes were arrested at the first cell cycle (M1) and the mitotic index was significantly decreased in a dose dependent manner. At 1 x 10(-7) mol/L or less of As(2)O(3), the SCE frequency, the cell proliferation kinetics and the mitotic index in PBLs were similar to the control (P > 0.05). CONCLUSION: It seemed that As(2)O(3) might induce transformation and affect the function of lymphocytes when used within the therapeutical concentration range.

[Yi lin cuo yao (Essentials of medical works) and its historical materials of medical exchanges between China and Korea].

Yi lin cuo yao, including 13 chapters and 128 categories, was written by Zheng Zhengxian and collated by Yang Lishou, both Korean imperial physicians in the 16 -- 17th centuries. With notation of a number of the Chinese and Korean medical books and materials in the first decade of the Li dynasty, this book is a comprehensive prescription work, very useful to clinical application. Especially with several points of the formulas from Zhong chao zhi wen fang (Sino -- Korean Prescriptions of Questioning) and zhong chao chuan xi fang (Sino -- Korean Prescriptions of Questioning) recorded in this book connected with Ren zhai zhi zhi fang lun (Renzhai's Comprehensive Prescriptions) in the Song dynasty. This book is an important record of the exchanges on medical ideas between China and Korea. However, it has nothing to do with Yi xue yi wen (Questions on Medicine) by Fu Maoguang, which records the questions and answers between Chinese and Korean officials.

[The recombinant vaccinia viruses expressing HPV type 16 wild or mutant E7 protein elicit immunity against tumor cells in mice].

OBJECTIVE: To select mutants of HPV 16 E7 suitable for vaccine development. METHODS: Several recombinant vaccinia viruses (VwE7, VmE7-1, and VmE7-2) expressing wild E7 or its mutants, ME7-1 (24G26G) and ME7-2 (24G26G91G) respectively, were employed to study their antigenicity and anti tumor activity. RESULTS: Both VmE7-1 and VwE7 could elicit cytotoxic T lymphocytes (CTL)and protect mice against tumor cells challenge, but VmE7-2 could not induce the immune reactions compared with VmE7-1 and VwE7. CONCLUSIONS: It suggested that ME7-1 could be used to develop a therapeutic vaccine for cervical cancer.

[The management of posterior capsular in cataract operations of high myopia].

OBJECTIVE: To study the management of posterior capsular in cataract operation of high myopia in order to avoid posterior capsular opacity(PCO). METHODS: Twenty-two cases(30 eyes) of high myopia with an average axial length of 31.62 mm were analyzed. Cataract phacoemulsification were performed in all cases, among which 12 eyes were operated with continuous curvilinear capsulorhexis (group I) of posteror capsular and 18 eyes were operated with capsulorectomy combined with anterior segment vitrectomy (group II). No artificial lenses were implanted in all cases. RESULTS: The post-operative naked vision were improved in both groups. Group I: The VA > or = 0.1 in all 12 eyes(100%), > or = 0.3 in seven eyes (58.3%) and no complication happened. Group II: The VA > or = 0.1 in 16 eyes(88.9%), > or = 0.3 in nine eyes (50%), and one eye with hemmorhage choroid detachment in the operation and three eyes with retina tear after the operation. CONCLUSION: Continuous curvilinear capsulorhexis of posteror capsular in treating PCO of high myopia with cataract is a safe effective and economic way compared with capsulorectomy combined with anterior segment vitrectomy.

[Three lethal cases of Angiostrongylus cantonensis infected children].

OBJECTIVE: To offer approaches to diagnosis and treatment of Angiostrongylus cantonensis infection on the basis of analyzing three lethal cases of the disease. METHODS: Clinical manifestations, pathological changes and species identification were pursued. RESULTS: Angiostrongyliasis is usually neglected in medical departments. The three cases here reported were all misdiagnosed and had not receiced anthelmintic treatment hence fatality ensued. CONCLUSION: Early examination of CSF, bronchial lavage fluid and feces is urged so as to secure accurate diagnosis and conduct efficacious anthelmintic therapy to cure the patients.

[Expression and significance of interleukin-6,8 in human nasal polyps].

OBJECTIVE: To explore the role of interleukin-6,8 in nasal polyp formation and to search into the effect of allergy in the pathogenesis of nasal polyps (NP). METHODS: The expression and significance of interleukin-6,8 were studied in 36 nasal polyps and 36 serum samples of NP patients by radioimmunoassay. RESULTS: The mean value of IL-6 and IL-8 was (2.7658 +/- 0.3797) ng/L and (4.1877 +/- 0.1758) ng/L in all nasal polyp tissue homogenates. As compared with serum of NP patients, IL-6 and IL-8 were over expressed in nasal polyp tissue homogenates. No relation was found between the expression of IL-6/IL-8 and patients' gender, age and clinical stage. The expression of IL-6 and IL-8 in patients' serum, cord blood and normal serum showed no significant difference. CONCLUSION: IL-6 and IL-8 are strongly correlated with the formation of nasal polyp. Neither allergy nor infection play a role in the pathogenesis of nasal polyps.

Long-term results of ammonia removal and transformation by biofiltration.

In this paper, long-term (>8 month) results of ammonia removal in biofilters was studied. Compost was used as the biofilter medium and activated carbon as an added material. The ammonia removal was normally >95% at influent ammonia concentrations of 20-500ppmv. According to the test results, the influent ammonia concentration should be <200ppmv (0.1570g ammonia/kg media per day) so that the effluent concentration of ammonia is <1.0mg/m(3) (the emission standard of China), and the biofiltration system can achieve good long-term performance. In the biofiltration system utilized in this study, the shortest retention time that the system could attain was 0.532min. However, the retention time can be decreased further without decreasing the ammonia removal efficiency. Countercurrent flow is favorable, as it enhances the moisture retention ability of the media. In the bioreactors, ammonia can be converted into the nitrate. The bioreactors have a stratification phenomenon for ammonia removal over the biofiltration depth, which implies that different parts of the bioreactor play different roles in the ammonia removing process.

Regulation of cyclooxygenase-2 and periostin by Wnt-3 in mouse mammary epithelial cells.

Wnt family members are critical in developmental processes and have been shown to promote carcinogenesis when ectopically expressed in the mouse mammary gland. The gene expression pattern mediated by Wnt is pivotal for these diverse responses. The Wnt pathway has been conserved among different species. Genetic studies have shown that Wnt effects are mediated, at least in part, by beta-catenin, which regulates transcription of "downstream genes." Wnt stimulation inactivates glycogen-synthase kinase-3beta (GSK-3) with subsequent stabilization of beta-catenin, which after heterodimerizing with lymphocyte enhancer factor-1/T-cell factor cofactors stimulates transcription. To establish whether Wnt-stimulated transcription is mediated solely by beta-catenin, a comparison was made of gene expression profiles in response to Wnt-3, overexpression of beta-catenin, and inhibition of GSK-3. Infection of cells with Wnt-3 and inhibition of GSK-3 regulate a set of genes that include cyclooxygenase-2 and periostin. Interestingly, overexpression of beta-catenin or reducing beta-catenin levels with antisense oligonucleotide transfection did not have any effect on cyclooxygenase-2 or periostin expression, thereby defining a Wnt pathway, which cannot be mimicked by beta-catenin overexpression.

Mutation of the conserved N-terminal cysteine (Cys92) of human presenilin 1 causes increased A beta42 secretion in mammalian cells but impaired Notch/lin-12 signalling in C. elegans.

The presenilin proteins are involved in the proteolytic processing of transmembrane proteins such as Notch/lin-12 and the beta-amyloid precursor protein (betaAPP). Mutation of a conserved cysteine (Cys60Ser) in the C. elegans presenilin sel-12 has a loss-of-function effect on Notch/lin-12 processing similar to that of null mutations in sel-12. In contrast, in mammalian cells, most missense mutations increase gamma-secretase cleavage of betaAPP. We report here that mutation of this conserved cysteine (Cys92Ser) in human presenilin 1 confers a loss-of-function effect in C. elegans, but causes increased A beta42 secretion in mammalian cells. These data suggest that the role of presenilins in Notch/lin-12 signalling and betaAPP processing are either separately regulated activities or independent activities of the presenilins.

Nicastrin modulates presenilin-mediated notch/glp-1 signal transduction and betaAPP processing.

Nicastrin, a transmembrane glycoprotein, forms high molecular weight complexes with presenilin 1 and presenilin 2. Suppression of nicastrin expression in Caenorhabditis elegans embryos induces a subset of notch/glp-1 phenotypes similar to those induced by simultaneous null mutations in both presenilin homologues of C. elegans (sel-12 and hop-1). Nicastrin also binds carboxy-terminal derivatives of beta-amyloid precursor protein (betaAPP), and modulates the production of the amyloid beta-peptide (A beta) from these derivatives. Missense mutations in a conserved hydrophilic domain of nicastrin increase A beta42 and A beta40 peptide secretion. Deletions in this domain inhibit A beta production. Nicastrin and presenilins are therefore likely to be functional components of a multimeric complex necessary for the intramembranous proteolysis of proteins such as Notch/GLP-1 and betaAPP.

Importance of arginines 63 and 423 in modulating the bile salt-dependent and bile salt-independent hydrolytic activities of rat carboxyl ester lipase.

Previous studies using chemical modification approach have shown the importance of arginine residues in bile salt activation of carboxyl ester lipase (CEL) activity. However, the x-ray crystal structure of CEL failed to show the involvement of arginine residues in CEL-bile salt interaction. The current study used a site-specific mutagenesis approach to determine the role of arginine residues 63 and 423 in bile salt-dependent and bile salt-independent hydrolytic activities of rat CEL. Mutations of Arg(63) to Ala(63) (R63A) and Arg(423) to Gly(423) (R423G) resulted in enzymes with increased bile salt-independent hydrolytic activity against lysophosphatidylcholine, having 6.5- and 2-fold higher k(cat) values, respectively, in comparison to wild type CEL. In contrast, the R63A and R423A mutant enzymes displayed 5- and 11-fold decreases in k(cat), in comparison with wild type CEL, for bile salt-dependent cholesteryl ester hydrolysis. Although taurocholate induced similar changes in circular dichroism spectra for wild type, R63A, and R423G proteins, this bile salt was less efficient in protecting the mutant enzymes against thermal inactivation in comparison with control CEL. Lipid binding studies revealed less R63A and R423G mutant CEL were bound to 1,2-diolein monolayer at saturation compared with wild type CEL. These results, along with computer modeling of the CEL protein, indicated that Arg(63) and Arg(423) are not involved directly with monomeric bile salt binding. However, these residues participate in micellar bile salt modulation of CEL enzymatic activity through intramolecular hydrogen bonding with the C-terminal domain. These residues are also important, probably through similar intramolecular hydrogen bond formation, in stabilizing the enzyme in solution and at the lipid-water interface.

micro-Terephthalato-bis[bis(1,10-phenanthroline)copper(I)] diperchlorate.

The title compound, [Cu(2)(C(8)H(4)O(4))(C(12)H(8)N(2))(4)](ClO(4))(2), was prepared from the hydrothermal reaction of CuCl(2), 1,4-dicyanobenzene, 1,10-phenanthroline and water at 443 K. The compound is a dimer in which the cation lies about an inversion center. The terephthalate moiety acts as a bridging ligand and the phenanthrolines as terminal ligands. The unique Cu atom is coordinated by two O and four N atoms in a distorted octahedral geometry, with Cu-O distances of 1.955 (2) and 2.815 (2) A, and Cu-N distances of 2.008 (2) to 2.216 (2) A.

Histamine immunocytochemistry: a new method for detection of basophils in peripheral blood.

We report that basophils in peripheral blood can be stained using histamine immunocytochemistry. The staining is based on the fixation of leucocytes with 1-ethyl-3-(3-dimethylaminopropyl)-carbodiimide (CDI) and the subsequent incubation of these cells with antisera raised against histamine conjugated to different carrier proteins using CDI. The staining appears to be specific for basophils and stained cells can be examined using both fluorescence microscopy and flow cytometry. In addition, histamine immunocytochemistry can be combined with conventional immunocytochemistry by incubating leucocytes with antibodies to cell surface antigens prior to or following fixation of the cells with CDI. Thus, histamine immunocytochemistry may be a valuable tool in future studies of human basophils.
