Analysis of the 5' sequence, genomic structure, and alternative splicing of the presenilin-1 gene (PSEN1) associated with early onset Alzheimer disease.

Mutations in the human presenilin genes (PSEN1 and PSEN2) are associated with early onset familial Alzheimer disease. The presenilin genes encode integral membrane proteins with similar structures, which suggests that they may have closely related, but as yet unknown functions. Analysis of the 5' upstream sequence and the structure of the PSEN1 gene reveals that the 5' sequence contains multiple putative transcription regulatory elements including clusters of STAT elements involved in transcriptional activation in response to signal transduction. The first four exons contain untranslated sequences, with Exons 1 and 2 representing alternate initial transcription sites. The function of these alternate initial exons is unclear. Exon 4 bears the first ATG sequence. The last 12 bp of Exon 4 is used as an alternative splice donor site. Exon 9 is alternately spliced in leukocytes, but not in most other tissues. Splicing of Exon 9 is predicted to cause significant structural changes to the protein. The majority of transcripts expressed in most tissues are polyadenylated 1127 bp from the TAG stop codon in Exon 13. A small proportion of transcripts contain the same 5'UTR and ORF but are polyadenylated 4435 bp from the stop codon. The longer polyadenylated transcripts contain three additional palindromes and at least one additional stem-loop structure with stabilities greater than -16 kcal/mol.

Cloning and characterization of the Drosophila presenilin homologue.

Mutations in two genes, PS1 and PS2, coding for the presenilins, have been linked to the early onset form of familial Alzheimer's disease (AD). Here we report the identification of a Drosophila melanogaster homologue of human PS genes, Dps, which maps to band 77B-C on chromosome 3 and is expressed at multiple developmental stages. The predicted amino acid sequence of the Dps product is 53% identical to human presenilins, with the greatest similarity in the putative transmembrane domains, the hydrophobic domains at the beginning and the end of the cytoplasmic TM6-TM7 loop and the C-terminus. Analysis of Dps in a genetically tractable model system such as Drosophila may provide insight into the mechanisms of Alzheimer's disease (AD) necessary for the development of rational therapeutic approaches.

Human inner ear OCP2 cDNA maps to 5q22-5q35.2 with related sequences on chromosomes 4p16.2-4p14, 5p13-5q22, 7pter-q22, 10 and 12p13-12qter.

Mouse Ocp2-rs2 maps to chromosome 11 and encodes an 18.6 kDa peptide abundantly expressed in the organ of Corti. We show that sequences similar to murine Ocp2-rs2 are found on human chromosomes 4p16.2-4p14, 5p13-5q35.2, 7pter-q22, 10 and 12p13-12qter as revealed by Southern blot analyses of human/rodent somatic cell hybrids. A fetal human inner ear cDNA library was screened with a cloned 254 bp PCR product of murine Ocp2-rs2. One of two human cDNA clones (CM1) was sequenced from the 5' end that begins with murine Ocp2-rs2 codon 14 through the stop codon and 258 nucleotides of 3-UTR and was found to have the identical deduced amino acid sequence to Ocp2-rs2. Based on the sequence in the 3'-UTR of CM1, a PCR primer pain was synthesized and used to confirm that a human homologue of Ocp2-rs2, designated OCP2 and expressed in the developing human inner ear, is localized to 5q22-5q35.2. Other OCP2-like sequences located on chromosomes 4p16.2-4p14, 7pter-q22 and 12p13-12qter (but not the chromosome 10 OCP2-like sequence) will PCR amplify the expected size product at a lower annealing temperature using the OCP2 3'-UTR PCR primers indicating that there may be a human OCP2 gene family.

Chronic effect of fatty acids on insulin release is not through the alteration of glucose metabolism in a pancreatic beta-cell line (beta HC9).

Hyperinsulinaemia in the fasting state and a blunted insulin secretory response to acute glucose stimulation are commonly observed in obesity associated non-insulin-dependent diabetes mellitus. Hyperlipidaemia is a hallmark of obesity and may play a role in the pathogenesis of this beta-cell dysfunction because glucose metabolism in pancreatic beta cells may be altered by the increased lipid load. We tested this hypothesis by assessing the chronic effect of oleic acid on glucose metabolism and its relationship with glucose-induced insulin release in beta HC9 cells in tissue culture. Our results show: (1) A 4-day treatment with oleic acid caused an enhancement of insulin release at 0-5 mmol/l glucose concentrations while a significant decrease in insulin release occurred when the glucose level was greater than 15 nmol/l; (2) Hexokinase activity was increased and a corresponding left shift of the dose-dependency curve of glucose usage was observed associated with inhibition of glucose oxidation in oleic acid treated beta HC9 cells, yet the presumed glucose-related ATP generation did not parallel the change in insulin release due to glucose; (3) The rate of cellular respiration was markedly increased in oleic acid treated beta HC9 cells both in the absence of glucose and at all glucose concentrations tested. This enhanced oxidative metabolism may explain the increased insulin release at a low glucose level but is clearly dissociated from the blunted insulin secretion at high glucose concentrations. We conclude that a reduction of oxidative metabolism in pancreatic beta cells is unlikely to be the cause of the dramatic effect that high levels of non-esterified fatty acids have on glucose-induced insulin release.

GLUT1 is adequate for glucose uptake in GLUT2-deficient insulin-releasing beta-cells.

GLUT2 may play an important role in pancreatic beta-cell glucose metabolism. A decrease in glucose uptake due to underexpression of GLUT2 has been considered as the cause of beta-cell dysfunction in diabetes with different pathogenesis. However, this view has been challenged by recent studies, in which the underexpression of GLUT2 was not accompanied by a decrease in glucose uptake. Our present aim is to evaluate the presumed importance of GLUT2 in maintaining the efficiency of beta-cell glucose uptake. We studied the kinetic characteristics of 3-O-methylglucose uptake in two beta-cell lines. One of these is the beta TC3 cell line which expresses GLUT1 and the other is the beta HC9 cell line which expresses both GLUT1 and GLUT2. Under equilibrium exchange conditions, 3-O-methylglucose transport in these two cell lines showed similar values of K(m) and V(max). The apparent IC50 of cytochalasin B for inhibiting 3-O-methylglucose transport in beta HC9 cells was nine times as high as in beta TC3 cells, indicating that GLUT1 is the critically important glucose transporter in the beta TC3 cell line and GLUT2 in the beta HC9 cell line. In both cell lines, the rates of glucose uptake were at least three times as fast as that of glucose phosphorylation. Our results suggest that GLUT1 is able to compensate for GLUT2 loss as it occurs in beta TC3 and maintains a commensurately high capacity of glucose uptake to sustain glucose metabolism in pancreatic beta-cells.

Measuring anteversion and femoral neck-shaft angle in cerebral palsy.

Femoral anteversion was measured by computed tomography (CT) in 40 children with cerebral palsy, however an accurate measurement was possible in only 59 of the 80 hips. Flat-surface measurement of anteversion was possible in 72 of 80 hips by CT, and in 36 of 40 hips by ultrasound. A high neck-shaft angle correlated with an inability to measure anteversion by CT. Measuring by ultrasound allowed subjects to be positioned in internal rotation and in this way accurate measurements of neck-shaft angles were obtained.

[Immunohistochemical change of endothelin-1 in acute lung injury and the role in its pathogenesis].

On the rat acute lung injury model produced by intraperitoneal cavity injection of zymosan showed that the respiratory rate became slow at first time (1st day), subsequently became rapid. Pulmonary histology showed that there was an accumulation of neutrophils in pulmonary microvessels, neutrophils infiltration in interstitial tissue, interetital and olveolar edema, hyaline membrane, atelectasis. Positive expression of ET-1 in experimental rats was stronger than than contral rats. Image analysis showed that OD value was elevated, especially at the fist day. The quantity of ET-1 was corresponding to the degree of pathologic change of acute lung injury. It suggests that the ET-1 might play an important role in the mechanism of acute lung injury.

[An experimental study of cellular response on implanted intraocular lens].

OBJECTIVE: To study the pathological changes and morphological features of cellular response on the implanted posterior chamber intraocular lens (IOL) in rabbits and discuss the mechanism of postoperative intraocular inflammatory response. METHODS: 9 adult pigmented rabbits were divided into three groups. IOLs were extracted on days 1, 7 and 14 after IOL implantation. Pathological changes on IOLs were observed with light microscopy and scanning electron microscopy, and the number of inflammatory cells on IOLs were counted. The data were analyzed by using analysis of variance of SAS software. RESULTS: There were inflammatory cells distributed over the entire IOL surface. In the mean time this study had clearly shown that morphological transformation of macrophages into spindle-shaped fibroblast-like cells took place on IOL, and phagocytic particles were in the cytoplasm of fibroblast-like cells, epithelioid cells and macrophages on the IOL. CONCLUSIONS: The phagocytic ability of fibroblast-like cells, epithelioid cells and macrophages is relatively strong, and besides macrophages and eosinophils, there were many lymphocytic clusters on the IOL, suggesting that an active immune response exist on the IOL surface.

[Color Doppler imaging analysis of ocular hemodynamics in low-tension glaucoma].

OBJECTIVE: To investigate ocular hemodynamics with color Doppler imaging, in low-tension glaucoma. METHODS: Nineteen patients with low-tension glaucoma (LTG) and 15 normal subjects were studied. The peak systolic velocity (Vmax), the end diastolic velocity (Vmin), the mean-enveloped velocity (Vmean), the pulse index (PI), the resistance index (RI) and S/D (Vmax/Vmin) of the ophthalmic artery (OA), the short posterior ciliary arteries (SPCA) and the central retinal artery (CRA) were compared in the two groups. RESULTS: (1) Compared with the normal subjects, the patients with LTG showed a decreased Vmax and Vmin in SPCA in bilateral eyes and Vmean in the right eye (P < 0.05), and a decreased RI and S/D in the left eye (P < 0.05). (2) Vmax and Vmean of CRA in LTG were lower than that in normal subjects (P < 0.01). (3) All six blood flow parameters of OA in the two groups were not significantly different from each other. CONCLUSIONS: Blood flow velocity of SPCA in LTG was lower than that in the normal subjects. The results support the theory that LTG is induced by the perfusion impairment of optic nerve head.

[The anti-inflammatory effects of superoxide dismutase].

This paper reports the anti-inflammatory effects of superoxide dismutase (SOD) from pig blood on inflammatory animal models. The experimental results have shown that SOD has significant anti-inflammatory effects. It inhibited carrageenin-induced foot-edema and croton oil induced granulation tissue edema of rats. It also inhibited arthritis induced by egg serum and Freund's adjuvant in rats. The effects were significantly dose-dependent.

[Effect of Yang-warming and Qi-tonifying natural products on neuroendocrine of deficiency-cold rats].

The rat fed with the compound drug Zhi Mu and Shi Gao was made into an animal model of deficiency cold syndrome. The deficiency cold state was kept for more than 7 days after stopping the drug. So the animal model is good for studing the therapeutic effect of products of heat nature. After 3 therapeutic days with Fu Jiang and Shen Qi compounds, the D beta H activity in serum, cortisone in adrenal gland and NE, DA amount in brain were increased, but serotonin was decreased, and after 7 days the effect was increased significantly. Both compounds could excite the sympathetic nerve and endocrine system in the peripheral and central nervous systems. Shen Qi compound acts quickly, but does not last as long as Fu Jiang. The effect will be better if the two compounds are used together.

[Significance of endocrine cells and their hormones in colorectal cancer].

OBJECTIVE: To analyse the expression and clinical pathologic significance of endocrine cell (EC) and their hormones in colorectal cancer (CC). METHODS: Chromograin A (CGA) and five kinds of hormones in 117 cases of CC were detected by immunohistochemistry with 13 observed by electron microscopy. RESULTS: CGA positive cells (EC) occurred in 46 cases (39.3%). 69.2%(27/39) expressed more than one hormone. The lymph node metastasis rate (68.0%) in the EC-positive CC was higher than EC-negative CC (43.7%) (P < 0.05). The incidence of EC(+2) in CC with morderative and poor differentiated (25.5%) was higher than well differentiated (P < 0.05). EC-positive CC gave shorter survival. It's an independent factor for evaluating prognosis in CC. The expression of 5-HT, GLU, GAST and beta-HCG occurred more offen in tumor with moderate and poor differentiation giving poor prognosis (P < 0.05). Cancer cells containing endocrine granules were found in 8 cases of CC by electron microscopy. The two methods were comformable in 92.3%. CONCLUSION: EC-positive CC has poor histological differemtiation, giving rise to more lymph mode metastasis and poor prognosis. Some hormones secreted by EC could partly account for the growth and metastasis of CC.

[Minor Bupleurum decoction in the treatment of viral myocarditis models].

A myocarditis model of Balb/c mice established by means of coxsackie virus type B3 was treated with the Minor Bupleurum Decoction. The results indicate that the decoction can regulate T lymphocyte subsets, influence anti-cardiac antibody production, help the organism to get clear of viruses, and thus plays an important role in treating viral myocarditis.

[Effects of n-butanol extract from Cortex Ilicis rotundae on blood pressure and heart rate in rate].

It was studied that effects of n-butanol extract from Cortex Ilicis Rotundae on blood pressure and heart rate in differedt state. Results showed that the drug elicited a sgnificant depressor effect on both normal pressure and artery pressure to rise for common carotid artery blocked in rats. Of the two latter decline of percent of blood presure and active time and slowing heart rate were more significan than the former. It was same depressor effect that injected the drug into duodenum but not act to hypotensive rats. These results suggested that the drug induce significantly depressor effect and slow heart rat with the exception of hypotension.

[The detection of human immunodeficiency virus type 1/2 antibody using synthetic peptides].

Four peptides of human immunodeficiency virus (HIV) located in the regions of gp41, gp120, p24 of HIV-1 and gp36 of HIV-2 have been synthesized according to the published amino acid sequences and the positions of antigenetic determinants of viruses by a solid-phase method. An indirect ELISA for detecting antibody against HIV 1/2 was established using these synthetic peptides as the coating antigens. We detected 41 control sera provided by National Institute for the Control of Pharmaceutical and Biological Products. The results indicated that the general coincident rate were 100%, the variant coefficient was less than 10%. A comparison of our reagent with Huayi, Pasteur and Jinhao reagents in detection of 90 positive sera and 140 normal sera for HIV-1/2 showed that the coincident rate was 100% with Pasteur and Jinhao, 99.57% with Huayi. All of 186 sera from patients with other diseases were negative. The reagent are stable at 37 degrees C for 4 days. The results indicated that our reagent is highly specific, sensitive and stable, it can be used for detection of HIV-1/2 infections.

Effects of antisense oligodeoxynucleotides to NR1 on suppression of seizures and protection of cortical neurons from excitotoxicity in vivo and in vitro.

OBJECTIVE: To investigate the effects of NR1 subunit on the initiation and development of seizures and protection of cortical neurons from excitotoxicity by using antisense oligodeoxynucleotides (ODN) to NR1 in vivo and in vitro. METHODS: Intracerebroventricular injection, temporal cortex slices discharge, cerebral cortical neuronal culture, induction of neurotoxicity and [3H]MK-801 binding were used in this study. RESULTS: After an antisense ODN for NR1 was administered intracerebroventricularly (i.c.v. 100 micrograms in 10 microliters) once daily, for three days in genetically epilepsy-prone rats (GEPR, P77PMC), the animals did not develop any clonic and tonic convulsions and their seizure scores were significantly lower compared to the control groups. The frequency and amplitude of early seizure-like events (SLEs) and late recurrent discharges (LRD), induced by lowering Mg2+, were reduced in entorhinal cortex (EC) of the temporal slice treated by antisense ODNs. Pretreatment with antisense ODN (2 microM) protected more than 52% of glutamate-sensitive neurons and reduced the [3H]MK-801 binding to 50% in cultured cerebral cortical neurons. CONCLUSIONS: N-methy-D-aspartate-receptors (NMDAR), specifically the NR1 subunit, may participate and play important roles in the initiation and propagation of epilepsy in the P77PMC rat.
