Somatic PKD2 mutations in individual kidney and liver cysts support a "two-hit" model of cystogenesis in type 2 autosomal dominant polycystic kidney disease.

An intriguing feature of autosomal dominant polycystic kidney disease (ADPKD) is the focal and sporadic formation of renal and extrarenal cysts. Recent documentation of somatic PKD1 mutations in cystic epithelia of patients with germ-line PKD1 mutations suggests a "two-hit" model for cystogenesis in type 1 ADPKD. This study tests whether the same mechanism for cystogenesis might also occur in type 2 ADPKD. Genomic DNA was obtained from 54 kidney and liver cysts from three patients with known germ-line PKD2 mutations, using procedures that minimize contamination of cells from noncystic tissue. Using intragenic and microsatellite markers, these cyst samples were screened for loss of heterozygosity. The same samples were also screened for somatic mutations in five of the 15 exons in PKD2 by single-stranded conformational polymorphism analysis. Loss of heterozygosity was found in five cysts, and unique intragenic mutations were found in seven other cysts. In 11 of these 12 cysts, it was also determined that the somatic mutation occurred nonrandomly in the copy of PKD2 inherited from the unaffected parent. These findings support the "two-hit" model as a unified mechanism for cystogenesis in ADPKD. In this model, the requirement of a somatic mutation as the rate-limiting step for individual cyst formation has potential therapeutic implications.

Shedding of feline immunodeficiency virus in semen of domestic cats during acute infection.

OBJECTIVE: To examine shedding of cell-free and cell-associated feline immunodeficiency virus (FIV) in semen of domestic cats during acute infection. ANIMALS: 7 specific-pathogen-free sexually intact male cats. PROCEDURE: 6 cats were inoculated IV with 5 x 10(6) 50% tissue culture infective doses of FIV-NCSU1, and 1 cat served as an uninfected (control) cat. Infection was confirmed in the 6 cats. Periodically for up to 16 weeks after inoculation, cats were anesthetized and ejaculates obtained by use of electroejaculation. Virus was isolated from filtered seminal plasma and washed seminal cells by co-cultivation with a feline CD4+ T-cell line. Seminal cell lysates were also examined for a 582-base pair segment of FIV gag provirus DNA, using a nested polymerase chain reaction amplification. RESULTS: During the acute phase of FIV infection, virus was evident in semen of 5 inoculated cats. Five cats had virus-positive seminal plasma and 3 had virus-positive cellular constituents during the study. Virus was isolated from 8/22 (36%) seminal plasma samples and 2/17 (18%) seminal cell specimens. Provirus DNA was detected in 5/24 (21%) seminal cell lysates. Cell-free virus was isolated as early as 6 weeks after inoculation, whereas cell-associated virus was isolated as early as 12 weeks after inoculation. Provirus DNA was detected in seminal cells from one cat as early as 1 week after inoculation. CONCLUSIONS AND CLINICAL RELEVANCE: Cell-free and cell-associated FIV are shed in semen of cats early during the course of infection. Samples obtained before seroconversion may contain virus. Virus shedding in ejaculates varies between and within cats during acute infection.

Intraepidermal nerves in human skin: PGP 9.5 immunohistochemistry with special reference to the nerve density in skin from different body regions.

The intraepidermal nerves of normal adult human skin were demonstrated by employing a powerful marker of neuronal elements, protein gene product (PGP) 9.5. There were two types of epidermal nerves, free nerve endings and nerves in the Merkel cell-neurite complex. The free nerve endings distributed to, and terminated in, all the strata basale, spinosum and granulosum, and they appeared as thin fibers, mostly varicose, branched or single processed, straight or bent. They existed at every site of the human body, including face, trunk and extremities. However, the densities of these nerves varied in different body parts and areas. The number of nerves decreased from the trunk to the distal parts of the limbs, and small denser 'innervation patches' showed up in the epidermis which were identified in confocal microscopy as one morphologic terminal field coming from the same dermal nerve bundle. This study has confirmed the existence of epidermal nerves in normal adult human skin, and presented a more clear picture than earlier. The difference between densities of epidermal nerves at different body areas implies area-specific functions of the intraepidermal nerve terminals. The observed intraepithelial nerve fibers may have a pain-perceiving role, however, also trophic or immunoregulatory roles can not be excluded.

Dendritic mast cells in prurigo nodularis skin.

Mast cells are traditionally recognized as round or oval connective tissue cells containing many specialized cytoplasmic granules. During recent years, more and more mast cell functions and properties have been clarified, and it is now evident that the mast cells are of different subtypes. The present study, utilizing chymase and tryptase immunofluorescence double-labelling and conventional electron microscopy techniques, has identified a kind of mast cells with obvious dendritic features in the lesional dermis of prurigo nodularis skin. This group of mast cell have enlarged cell bodies and contain fewer cytoplasmic granules, especially within certain dendrites. The morphological identification of such subgroups of mast cells could contribute to the understanding of mast cell heterogeneity.

Light and electron microscopic radioautographic studies on RNA synthesis in aging mouse adrenal gland.

Ultrastructural observation and radioautographic studies were carried out to detect aging changes of RNA synthesis in the adrenal glands of mice from fetal day 19 to postnatal 12 months by light and electron microscopic radioautography after 3H-uridine incorporation. The ultrastructural features of the cells at embryonic stages were not developed. The cell organelles appeared more developed at early postnatal stages. The organelles such as endoplasmic reticulum, ribosomes, Golgi apparatus, mitochondria with tubular or vesicular cristae and lipid droplets were more frequently observed in the cytoplasm of the cells in the 3 zones of the cortex and the medulla at later postnatal and senescent stages. The activity of RNA synthesis of adrenal glands, as expressed by grain counting per unit area as well as per cell, was the highest in the cortex and medulla at fetal day 19, and then gradually decreased with aging. The silver grains were electron microscopically localized over the nucleoli, euchromatin, endoplasmic reticulum, ribosomes and mitochondria of the cells in the 3 zones of the cortex and the medulla. The number of mitochondria per cell, the number of labeled mitochondria per cell and the mitochondrial labeling indices increased along with aging. These results demonstrated the aging changes of RNA synthesis in the adrenal glands of mice.

Properties of transient outward potassium current and inward rectifier potassium current in immature human atrial myocytes.

AIM: To study the properties of transient outward K+ current (Ito) and inward rectifier K+ current (IKl) in immature human heart. METHODS: Ito and IKl were recorded using whole-cell patch-clamp technique in atrial myocytes isolated from 12 immature (aged from 6 months to 5 a) human hearts. RESULTS: Ito was voltage-dependent, activated and inactivated rapidly. The IC50 (95% confidence limits) of 4-AP on Ito was 0.64 (0.48-0.87) mmol.L-1. 4-AP 1 mmol.L-1 shifted V1/2 of activation from (6.6 +/- 2.0) mV to (19.8 +/- 3.0) mV (n = 4-10, P < 0.01). 4-AP 0.3 mmol.L-1 changed V1/2 of inactivation from (-49 +/- 4) mV to (-61.4 +/- 2.1) mV (n = 3, P < 0.01), but there were no obvious influence on voltage-dependent activation of Ito (P > 0.05). At the same concentration, the recovery time constant (tau value) was prolonged from (108 +/- 16) ms to (220 +/- 67) ms (n = 3-12, P < 0.01). IKl was also voltage-dependent. Its reverse potential was -40 mV. CONCLUSION: Both Ito and IKl are important K+ channel currents in immature human atrial myocytes. 4-AP can affect the inactivation and recovery of Ito at low concentration (0.3 mmol.L-1) and affect its activation at high concentration (1 mmol.L-1).

[Study on the electrothermal tissue effects during transcervical resection of endometrium].

OBJECTIVE: To investigate the tissue electrothermal effects and to seek the appropriate setting of operative wattage during transcervical resection of endometrium (TCRE). METHODS: The power setting were 60 W, 80 W, 100 W for cutting and 30 W, 60 W, 80 W, 100 W for coagulation when performing endometrial resection in vitro or in vivo with operating time 3-5 seconds or 6-8 seconds respectively. After operation the uterine specimens were stained by hemaloxylin-eosin, and respiratory enzyme dehydronicotinamide adenine dinucleotide phosphate diaphorase methods for evaluating the tissue damage. RESULTS: (1) The depth of endometrial damage under cutting was considerably less severe than those under coagulation with the same power setting. (2) When the power of cutting and coagulation were 80 W, 100 W and 30 W, 60 W respectively and operating time prolonged more than 5 seconds, the thermal damage covered endometrium and 2-3 mm of myometrium in depth. (3) At such wattage and operating time the deepest destruction was only 16.27% of uterine wall. CONCLUSION: (1) There is significant difference in the depth of thermal damage between cutting and coagulation during hysteros copic surgery. (2) The appropriate power setting should be 80-100 W for cutting and 30-60 W for coagulation.

PCR assay for the inversion causing severe Hemophilia A and its application.

OBJECTIVE: To develop a new technique based on long distance polymerase chain reaction (LD-PCR) to replace Southern blotting method to detect Factor VIII (FVIII) gene inversion leading to severe Hemophilia A (HA) and carrier. METHODS: Four primers P, Q, A&B were designed and synthesized. P&Q is specific for 5' and 3' flanking regions of F8A1 respectively. A&B is specific for 5' and 3' flanking regions of F8A2/F8A3 respectively. LD-PCR with 3 primers and 3 temprature was set up, optimized and used to detect the inversion. RESULTS: The LD-PCR with primers P, Q, A&B, P, Q&B and P, Q&A can be used to detect the gene inversion and discriminate carrier from wild type. A blind analysis of 53 DNA samples from HA families was carried out by the LD-PCR and Southern blotting respectively. Two sets of the results were completely identical. They were 23 cases of inversion, 27 cases of wild type and 3 cases of carriers. The sensitivity and specificity of LD-PCR are both 100%. Three inversion hemizygotes and 4 female carriers were identified from 5 HA families by the LD-PCR technology. CONCLUSIONS: The LD-PCR with primer P, Q&B or P, Q, A&B can be used to detect the gene inversion and the carrier of inversion. Compared with Southern blotting, this technique is simple, rapid, inexpensive, more sensitive, accurate and non-isotopic.

[Optic nerve neuropathy by ethambutol toxicity].

OBJECTIVE: To investigate the clinical manifestations, treatment and prevention of optic nerve damage by ethambutol toxicity. METHODS: Retrospective analysis was done on 17 patients of tuberculosis who had been treated with ethambutol and shown ocular symptoms. RESULTS: Changes in visual acuity, fundal appearance, visual field and color sensation were major abnormal findings. According to the neural damage, the patients were diagnosed as axial neuritis 7 cases, periaxial neuritis 2 cases, mixed type 3 cases and there were no significant changes on fundus, color sensation or visual field in 5 cases. In 17 cases, none of the patient showed the appearance of retinitis. The impairment of visual acuity and the duration of damage were related to the daily dosage significantly. After stop of ethambutol and vasodilators or neurotrophic drugs were given, the function of optic nerves were recovered. CONCLUSIONS: To prevent the damage of optic nerves by the toxicity of ethambutol, the appropriate small dosage of ethambutol is recommended, regular examination and monitoring of visual function is necessary. The physicians should be fully aware of the diagnosis and clinical manifestations of ocular toxicity of ethambutol, as soon as the diagnosis is made, ethambutol should be stopped immediately and appropriate therapy should be given.

[The relationship between hypoxic pulmonary hypertension and changes of HO-1 and its mRNA].

OBJECTIVE: To study the relationship of HO-1 and HPH. METHODS: The expression of HO-1 and HO-1 mRNA were observed in pulmonary vessles of rats using the technique of immunohistochemistry and hybridization in situ. The average value of integral light density of HO-1 in three different diameters of arteries was detected by means of the image analysor and considered as the relative content in pulmonary arteries. RESULTS: Hypoxia induced the expression of HO-1 and HO-1 mRNA and the relative content of HO-1 was significantly increased in pulmonary arteries corresponding with the period of hypoxia. CONCLUSIONS: In the condition of HPH, relative content of HO-1 increase obviously. It suggests that HO-1 plays certain role in development of HPH. The change of HO-1 mRNA expression is the basis of molecular change of HO-1.

[Effect of compound Chinese drug bailong on the expression of tumor suppressor genes and relationship with prekallikrein activator signal pathway in human gastric carcinoma BGC82-3 cell line].

OBJECTIVE: To study the effect of compound Chinese drug Bailong on the transcription of Cyclin Dependent Kinase Inhibitor (CKI) p16INK4a, p21 and Rb, c-myc genes, and the relationship between gene expression and cAMP-PKA pathway. METHODS: Using the traditional molecular biology methods (cell synchronization, molecular hybridization--Western blotting, Northern blotting, etc.) examine the gene expression. RESULTS: Bailong promoted the expression (both mRNA and protein) of p16INK4a obviously in G1 phase cells. When prekallikrein (PKA) inhibitor was added in the cells which were treated by Bailong, the mRNA and protein level of p16INK4a decreased. It was shown that the inhibited proliferation of BGC82-3 cell by Bailong may come from the enhanced p16INK4a gene expression in G1 phase. Being same as p16INK4a, tumor suppressor genes Rb, p21 and oncogene c-myc expression were all affected by Bailong. When PKA inhibitor was added, the results were reversed. CONCLUSION: Bailong can affect many anticancer genes (including p16INK4a, p21 and Rb genes) and oncogenes (including c-myc) transcription by regulating cAMP-PKA pathway.

[Effect of caoguo zhimu decoction on N-methyl-D-aspartate receptor gene expression of epileptic kindling model in rats].

OBJECTIVE: To explore the effect of Caoguo Zhimu decoction (CGZMD) on N-methyl-D-aspartate (NMDA) receptor gene expression of epileptic model on rat brain. METHODS: Adopting the pentylenetetrazol (PTZ) kindling model to observe the NMDAR1 mRNA gene expression by situs hybridization. RESULTS: After 28 days of CGZMD treatment, the NMDAR1 mRNA gene expression on CA1, CA2, CA3 part of rat hippocampus reduced obviously, the comparison between experimental group and the model control group showed significant difference (P < 0.05). CONCLUSION: CGZMD decreased NMDAR1 mRNA gene expression, it may be caused the NMDA receptor decline, hence revealed its anti-epileptic effect.

[Effect of bailong recipe on proliferation phenotype of human gastric carcinoma BGC82-3 cell line].

OBJECTIVE: To observe the reversive effect of Composite Bailong recipe (BR), a preparation of Chinese herbal medicine, on proliferation phenotype of human gastric carcinoma BGC82-3 cell line. METHODS: Observation was conducted by using growth curve, colony on soft agar and nude mice oncogenesis experiment. RESULTS: Growth curve showed the proliferation of BGC82-3 cell was obviously inhibited by BR, the inhibiting rate reached 86% in the 5th day. The colony was few and small on soft agar after treated with BR, only accounted for 52% as compared with that of nontreated. BR obviously inhibited the oncogenesis of BGC82-3 cells in nude mice, the inhibition rate being 50%. CONCLUSION: Composite BR has obvious inhibiting action on malignant proliferation of human gastric BGC82-3 cell line.

[Expression and regulation of p16 in osteosarcoma].

OBJECTIVE: To study the expression and regulation of p16 and related factors in osteosarcoma. METHODS: The levels of p16 protein and related factors were explored by Western blot analysis in 12 cases of osteosarcoma tissues and in cells of different phases of osteosarcoma cell line OS732. The status of p16 protein in osteosarcoma cells was revealed by immunofluorescence analysis. RESULTS: p16 protein was detected in 10 of 12 osteosarcoma tissues. pRb was found in all of the specimens, but 8 of them were hyperphosphorylated. p16 protein was the least in G1 phase. Localized in cytoplasm during G1 phase, p16 protein gathered around nuclear envelope in S phase and entered the nucleus in G2 phase. CONCLUSIONS: The expression rate of p16 protein was 83.3% in osteosarcoma tissues. The expression of p16 protein was unable to inhibit the phosphorylation of pRb in most osteosarcomas; p16 protein was extremely low in G1 phase, presumably it was sufficient to inhibit the phosphorylation of pRb, causing abnormal proliferation of the cells.

[A pathological study on indirect optic nerve injury in rabbit eyes].

OBJECTIVE: To study the pathological changes after indirect optic nerve injury. METHODS: Animal models (72 rabbits, 144 eyes) for intracanalicular optic nerve injury was developed by keeping the optic canal intact. The pathological changes of optic nerve were observed under light microscope and transmission electron microscope and the number of myelinated axons was enumerated by computer-aided image analyzer. The data were analyzed by using variance of SAS software. RESULTS: The model contributed much valuable information regarding nerve edema, loss of myelin, myelin regeneration and changes of axons. The edematous area in the optic nerve injury was reduced (P < 0.01) by treating with prednisone. The counted number of myelinated fibers showed more in the group treated by prednisone than that in the group without treatment. CONCLUSIONS: Significant changes of optic nerve after indirect injury are edema, demyelination and the change of axon amount. The effects of prednisone on reducing these pathological changes are demonstrated in this study.

[A study on producing areas of Chinese flos Dendranthematis].

OBJECTIVE: To probe into the cultural environment and changes of producing areas of different species of Chinese Flos Dendranthematis. METHOD: On-the-spot investigation and literature study. RESULTS: A clearer idea has been gained on the geographical distribution, growing environment and changes of producing areas of Gongju, Hangju, Chuju, Boju, Huaiju and Jiju. CONCLUSION: Different environments fit for the growth of different species of Flos Dendranthematis. The present study provides some valuable information on the cultivation and development of Chinese Flos Dendranthematis.

[Species evolution of Chinese flos Dendranthematis].

OBJECTIVE: To probe into the history and current situation and main producing areas of different species of Chinese Flos Dendranthematis. METHOD: On-the-spot investigation, cultivation observation and literature study. RESULTS: Morphological characteristics of different species of Flos Dendranthematis were clarified, and the historical changes of these species were studied. CONCLUSION: Congju is related to Hangju. Boju is related to Huaiju. Jiju and Qiju, and Chuju is a distinctive species. These findings are important for the cultivation and processing of Chinese Flos Dendranthematis.
