The structures of the N-linked oligosaccharides of miraculin, which is a taste modifying glycoprotein isolated from miracle fruits, berries of Richadella dulcifica, are reported. Asparagine-linked oligosaccharides were released from the protein by glycopeptidase (almond) digestion. The reducing ends of the oligosaccharide chains thus obtained were aminated with a fluorescent reagent, 2-aminopyridine, and the mixture of pyridylamino derivatives of the oligosaccharides was separated by high performance liquid chromatography (HPLC) on an ODS-silica column. More than five kinds of oligosaccharide fractions were separated by the one chromatographic run. The structure of each oligosaccharide thus isolated was analyzed by a combination of sequential exoglycosidase digestion and another kind of HPLC with an amidesilica column. Furthermore, high resolution proton nuclear magnetic resonance (1H NMR) measurements were carried out. It was found that 1) five oligosaccharides obtained are a series of compounds with xylose-containing common structural core, Xyl beta 1----2 (Man alpha 1----6) Man beta 1----4-GlcNAc beta 1----4 (Fuca1----3)GlcNAc, 2) a variety of oligosaccharide structures are significant for two glycosylation sites, Asn-42 and Asn-186, and 3) two new oligosaccharides, B and D, with unusual structures containing monoantennary complex-type were characterized. A silver staining method was used to analyze the distribution of nucleolar organizer regions (Ag-NORs) on chromosomes of 45 wild mice (Mus musculus). The four subspecies represented were M. m. musculus, M. m. molossinus, M. m. castaneus, and M. m. bactrianus. Ag-NORs were observed near the centromeric regions of 11 chromosomes (4, 8, 9, 10, 11, 12, 15, 16, 17, 18, and 19), indicating a preponderance of Ag-NORs on smaller chromosomes. The first five loci have not been observed previously. It is suggested that a correlation may exist between the specific features of mouse Ag-NORs and the events involved in intra- and interchromosomal homogenization of rDNA.
Drooling is a significant problem demonstrated by individuals of all ages with a variety of etiologies such as neurological disorders, cerebral palsy, mental retardation and others. Persistent drooling not only creates troublesome hygienic problems for patients, teachers, nurses, and playmates because of the constant soiling of clothes, toys, and work materials, but also causes an odor from their clothing and bibs. The older patient with normal cognition is disturbed by the drooling and may become depressed and reclusive. Successful management of drooling alleviates these problems, improves appearance and self-esteem and significantly reduces the time involved in the care of the sufferers. The author, who had studied at the University of Toronto for 2 years, acquired the technique of submandibular duct relocation under the guidance of Dr. Crysdale. We carried out the surgical procedure of submandibular duct relocation for drooling on six patients who had cerebral palsy. The surgical procedure resulted in a dramatic decrease in drooling and odor levels. All of them improved in appearance, and the time involved in the care of the suffers was significantly reduced. The complication of ranula, however, appeared in two cases, which suggested a much higher frequency than that in the report of Crysdale. It seems that a detailed explanation to parents and teachers about the advantages and disadvantages of submandibular duct relocation is important.
The Hallermann-Streiff Syndrome has been characterized and established according to 7 positive and 5 negative signs, which were described by François. We encountered an 11 year, 5 months old boy who had 7 positive symptoms of this syndrome in our clinic. In this study, we reported on this typical patient focusing on his dental view. 1) Prolonged retention of the primary teeth which involved microdontia were noted. Congenitally missing teeth were also seen. 2) The occlusal relationship indicated open bite, and also the mandibular function was impaired. 3) The measurements of the length and width of the dental arch were smaller than that of a normal subject, and the dental arch of the maxilla was V-shaped. 4) According to X-ray cephalometric analysis, (a) the dental calculus and the alveolar bone absorption were very evident. (b) abnormal morphologic of the glenoid fossa, mandibulars condyle and the neck of mandibula were seen. These conditions were very evident on the left side. 5) The growth obstade of the maxilla and mandibla and the left shift of the mandible were found. 6) According to histological study, enamel hypoplasia was noted.
It has been reported by BALAKRSHANN and ASHTON that about 20-30 bands of human salivary protein separated by polyacrylamide gel electrophoresis (PH 8.0) were detected by staining with Coomassie brilliant blue R. Employing the same method, MATSUSHIMA discovered five main regions (I-V) in mouse salivary protein (Msp) separated by poly-acrylamide gel electrophoresis (PH 8.0). He also discovered distinctive features of biochemical genetic markers among each of the five regions. In this experimental study, we investigated biochemical genetic markers of Msp of C3H/HeJ, BALB/c, BALB/k and NOD/shi strains. The following were the results of the present study. 1) The results of C3H/HeJ and BALB/c mice were in accord with those of MATSUSHIMA's. 2) The BALB/k mouse showed the same results as the BALB/c the difference in sex of Msp-1 seemed to have no relation to H-2 complex. 3) With the NOD/she strain, Msp-3 showed the b-type, Msp-2 the a-type, Msp-1 the o-type showed no difference based on sex, that is to say, F-type in both male and female. Amylase-1 showed the b-type.
The purpose of this paper was to investigate the dental situation of children who visited the pedodontics clinic of Nihon University at Dental-Hospital during the period from Jan. 1983 to Dec. 1987. The subject is the awareness in parents of 68 children who carried out the administration of medical care. The results were as follows. 1) Recently we observed that the ratio of physically handicapped children was was on the increase among the first visit patients. 2) The principal diseases of the physically handicapped children were cyclothymic, allergy and blood dyscreasia. 3) The majority of the patients visited for receiving caries treatment. 4) We presumed that the parents of the physically handicapped children showed deep concern for oral diseases.

The taste-modifying protein, miraculin, has the unusual property of modifying sour taste into sweet taste. The complete amino acid sequence of miraculin purified from miracle fruits by a newly developed method (Theerasilp, S., and Kurihara, Y. (1988) J. Biol. Chem. 263, 11536-11539) was determined by an automatic Edman degradation method. Miraculin was a single polypeptide with 191 amino acid residues. The calculated molecular weight based on the amino acid sequence and the carbohydrate content (13.9%) was 24,600. Asn-42 and Asn-186 were linked N-glycosidically to carbohydrate chains. High homology was found between the amino acid sequences of miraculin and soybean trypsin inhibitor.

ATP-dependent profiles of phosphofructokinase (PFK) activity were determined in both crude and Sephadex G-25-filtered fractions from rabbit dental pulp. ATP had a dual effect on PFK as an activator and an inhibitor, according to its concentration. Gel-filtered PFK showed a similar profile to that of crude PFK, indicating a lack of low-molecular-weight effector(s) for PFK in rabbit dental pulp. For complete inhibition of the PFK in rabbit dental pulp, 5 mM ATP was required. This level of ATP is much higher than that required for other typical isozymes of PFK from liver, skeletal muscle or brain. It is postulated that differences in the properties of PFK isozymes are due not only to the subunit structure but also to the presence of other effectors.
