Correlations between iron content and isoferritin profiles of normal and malignant tissues.

Isoelectric focusing profiles of ferritins isolated from livers of anemic rats and 3'-methyl-4-dimethylaminoazobenzene-induced hepatomas were more acidic than their normal counterparts, and exhibited patterns similar to those of heart ferritin. Iron administration induced a shift to more basic components for the normal and tumor ferritins. The isoferritin patterns reflected the subunit populations of the protein, as the more acidic components had greater proportions of H-type subunits and iron induced an increase in L subunits with concomitant increases in more basic isoferritins. For both normal and malignant tissues there was a direct correlation between iron content of the tissue and subunit composition. The apparently unique isoferritin patterns in the tumor tissue may, therefore, be ascribed primarily to the lower iron contents.

[Case of choriocarcinoma of a probable testicular origin with a good response to cis-diamminedichloroplatinum (CDDP)].

A case of choriocarcinoma responding well to cis-diamminedichloroplatinum (CDDP) was reported. A 24-year-old male was admitted to our hospital with chief complaints of upper abdominal discomfort and gynecomastia. Physical findings on admission included solid tumors in the upper abdomen and bilateral inguinal regions. The chest X-ray picture showed numerous coin lesions, and abdominal ultrasound examination revealed a widespread metastasis in the retroperitoneum. Blood chemical analysis pointed out the high levels of LH, HCG, and beta-subunit of HCG. The pathological diagnosis of the biopsied inguinal mass was choriocarcinoma. The patient was treated with CDDP, vincristine and bleomycin according to the protocol of Einhorn. After three courses of this therapy, serum HCG level was normalized and pulmonary metastatic lesions were diminished, although abdominal ultrasound examination showed remnants of the decreased tumor masses. The patient was transferred to the urological department for further cytoreductive therapy.

Differential turnover of rat liver isoferritins.

Rat liver ferritins consist of families of isoferritins fashioned from different proportions of two subunit types, H and L. Iron administration alters both the absolute and the relative amounts of these isoferritins through an increase in L-rich forms. This phenotype change represents preferential synthesis of L-rich isoferritins together with a faster degradation of H-rich isoferritins. The turnover rates of isoferritins in the same liver are markedly different and vary progressively with subunit composition. The H and L subunits in individual isoferritins appear to turn over at similar rates, suggesting that the various isoferritins are degraded as intact shells rather than through exchange and degradation of subunits.

Enzyme immunoassay for human ferritin.

We describe an enzyme immunoassay with use of beta-D-galactosidase for quantitation of ferritin in human serum. The minimum detectable ferritin concentration is 0.25 microgram/L of serum, which is comparable to results obtained by radioimmunoassay. The correlation coefficient between values determined by enzyme immunoassay and radioimmunoassay was 0.95 (n - 1 = 85, p less than 0.001).

Immunoreactive transferrin receptor in sera of pregnant women.

Immunoreactive transferrin receptors in sera of 90 pregnant women of various gestational periods were investigated. The mean concentrations of the serum transferrin receptor in normal males and females were 251 +/- 94 ng/ml and 256 +/- 99 ng/ml, respectively. Serum transferrin receptor levels in pregnant women did not show a significant increase in the early stage of gestation. However, a rapid elevation of the mean concentration of transferrin receptor was observed after 20 weeks of pregnancy. The apparent increase with gestation period suggests that this immunoreactive receptor in the sera of pregnant women is derived from the placental syncytiotrophoblast and reflects the activity of maternofetal iron transport.

The near-infrared electronic endoscope for diagnosis of esophageal varices.

We developed an electronic endoscope sensitive to light delivered by an infrared laser, which makes it possible to visualize submucosal vessels of the gastrointestinal tract. The system consists of a modified electronic endoscope and an image processing unit. A high output laser diode was chosen as the source of the infrared ray with a wavelength of 815 nm, an output of 200 milliwatts, and a band width of 30 nm. Using this instrument, esophageal varices were classified into three categories: venous dilation, dark spot, and diffuse dark area. The venous dilation and dark spot appearances correlated with early esophageal varices, which were rarely seen with visible light. On the other hand, the diffuse dark area appearance was considered to be a high risk sign of future bleeding. This system is useful not only for the early diagnosis of esophageal varices, but also for the localization of optimal puncture sites and the evaluation of the effect of sclerotherapy.

Expression of syndecan-1 is common in human lung cancers independent of expression of epidermal growth factor receptor.

In order to determine syndecan-1 expression in lung cancer, we examined 115 lung cancer specimens and 17 lung cancer cell lines. Syndecan-1 was immunohistochemically stained with a polyclonal antibody in 115 paraffin-embedded specimens; 84 cases out of 97 non-small cell lung cancer (NSCLC) and eight cases out of 18 small cell lung cancer (SCLC) were positively stained. Simultaneously, epidermal growth-factor receptor (EGFR) was stained; 47 cases out of 97 NSCLC and one case of 18 SCLC were positively stained. No significant correlation was shown between EGFR and syndecan-1 expression (P=0.68). Syndecan-1 mRNA was detectable in 16 of 17 lung cancer cell lines and EGFR mRNA in nine of 17. Eight cell lines had syndecan-1 mRNA as well as EGFR mRNA. PR-39 (1 microM) and 80 pM transforming growth factor-beta(1) (TGF-beta(1)), did not increase expressions of syndecan-1 mRNA and EGFR in five lung cancer cell lines. We concluded that lung cancer had detectable syndecan-1; however, expression of syndecan-1 protein did not correlate with survival time of lung cancer patients.

Immunochemotherapy in B-16-melanoma-cell-transplanted mice with combinations of interleukin-2, cyclophosphamide, and PSK.

The effect of combination immunochemotherapy using interleukin-2 (IL-2), PSK and cyclophosphamide (CY) was evaluated in a pulmonary metastasis model in BDF1 mice. B-16 melanoma cells were inoculated into a hind limb. On day 3 after inoculation, 20 mg/kg of CY was administered intraperitoneally, and IL-2 (3.75 x 10(4) BRM units/head) was injected into the tail vein on days 7, 8 and 9. PSK (1,000 mg/kg) was administered orally every day from day 1 to day 10 using a stomach tube. This treatment cycle was repeated three times. Using this combination therapy, the cytotoxicity of lymphokine-activated killer cells and tumor-infiltrating lymphocytes was enhanced. Pulmonary metastasis was remarkably suppressed and a prolongation of survival was obtained compared with the nontreated group and an IL-2+CY group. The effect was augmented by repeating the therapy protocol. By analyzing the killer activity and surface markers of tumor-infiltrating lymphocytes, it was recognized that increased numbers of Lyt-2-positive T cells with augmented cytotoxicity were obtained. This treatment modality should have clinical significance.

Mineralogical analysis of sputum lith from an apparently healthy subject.

The sputum lith, 1 to 3 mm in diameter, were examined by microanalyser and by the method of X-ray diffraction, which revealed that the lith was composed of calcium carbonate and calcite in crystalline style. A composition of bronchial liths is often associated with different underlying diseases so that these nondestructive analyses seem to provide useful information for differential diagnosis. The present case is the first one to expectorate bronchial lith without marked pulmonary diseases.

Plural immunoglobulin synthesis in a single cell: an ultrastructural study of two cases with three M-proteins.

Electrophoresis revealed two cases of malignant lymphoma that each contained three M-proteins (IgM lambda.lgG kappa.lgG lambda and IgM lambda.IgM kappa.lgG kappa) in the sera. To determine cellular origin of each M-protein, atypical lymphoid and plasmacytoid cells of both cases were examined by electron microscopy. Atypical lymphoid and plasmacytoid cells possessed rough endoplasmic reticula (RERs) in varying degrees, as seen by conventional electron microscopy, and showed double-stainability for plural antibodies against immunoglobulins following double stainings of immunoelectron microscopy using immunogold staining. Rabbit antibodies against human IgM, lgG, free kappa-light chain and free lambda-light chain were used for the immunoelectron microscopic staining. By the double staining method, plural immunoglobulins, IgM/IgG, IgM/free kappa, IgM/free lambda, IgG/free kappa, IgG/free lambda and free kappa/free lambda, were simultaneously detected in varying degrees in the Golgi area, RERs, and dense bodies of lymphoid and plasmacytoid cells. In conclusion, this study directly exhibited, through electron microscopy, that plural immunoglobulins were synthesized at the same time in a single cell, and that the process of immunoglobulin synthesis in the lymphoid and plasmacytoid cells was different from that in a normal B-cell.
