Synthesis of kanamycin A analogs containing a 6-amino-6-deoxyglycofuranose moiety.

Three kanamycin A analogs containing 6-amino-6-deoxyglycofuranoses have been prepared as candidates for potential activity against resistant bacteria producing 6'-N-acetyltransferase. They are 4-O-(6-amino-3,5,6-trideoxy-alpha-D-, -beta-D-, and -beta-L-erythro -hexofuranosyl)-6-O-(3-amino-3-deoxy-alpha-D-glucopyranosyl)-2,5-dideoxy-5-epi-5-fluorostreptamine. Structure-activity relationships of these compounds are discussed.

Macaque monkey retrosplenial cortex: I. three-dimensional and cytoarchitectonic organization.

This is the first in a series of reports on the neuroanatomic organization and connectivity of the macaque monkey retrosplenial cortex, i.e., areas 29 and 30. To elucidate the topographic configuration of the retrosplenial cortex and adjacent structures, we have made three-dimensional computer reconstructions of the posterior cingulate region that includes the retrosplenial cortex. The largest portion of the posterior cingulate gyrus is located dorsal to the corpus callosum. At the caudal limit of the corpus callosum, the gyrus curves around the splenium, turns laterally and forms a region called the isthmus that links the cingulate and parahippocampal gyri. The isthmus contains the caudomedial lobule, which is a rostrally oriented bulge that is made up, in part, of portions of the retrosplenial cortex. To delineate the subdivisions of the retrosplenial and adjacent cortices, we conducted a cytoarchitectonic analysis by using cerebral hemispheres that were cut at oblique angles and stained with a variety of techniques, including immunohistochemistry for nonphosphorylated neurofilament protein. The dorsal bank of the callosal sulcus and the rostral surface of the isthmus are covered by the retrosplenial cortical areas 29l, 29m, and 30, whereas most of the medial surface of the posterior cingulate gyrus and the ventral bank of the posterior cingulate sulcus consist of areas 23i and 23e. The most caudoventral portion of the cingulate gyrus is composed of an area (area 23v) that resembles the retrosplenial and posterior cingulate cortices but has a much more prominent layer IV. On the dorsal bank of the calcarine sulcus, we also defined a transitional zone, area 30v, located between the retrosplenial cortex and the prestriate visual cortex.

Identification and characterization of rat AILIM/ICOS, a novel T-cell costimulatory molecule, related to the CD28/CTLA4 family.

Activation-inducible lymphocyte immuno-mediatory molecule (AILIM) is an inducible cell surface glycoprotein expressed on thymocytes and activated lymphocytes. Specific monoclonal antibody to rat AILIM induced the cell aggregation of a rat thymoma cell line and ConA-activated splenocytes. In the present study, we identified the primary structure of two species of rat AILIM by expression cloning. We also cloned mouse and human AILIM homologues and the predicted amino acid sequences were identical to those of the inducible costimulator ICOS/CRP-1, which belongs to the CD28/CTLA4 family. Although the human and mouse AILIM/ICOS molecule is localized on T-cells, the major population of AILIM/ICOS-positive cells in rat splenocyte was CD45RA-positive B-cells. The expression level of AILIM/ICOS on T-cells was relatively low; however, its expression was drastically induced by the treatment with PMA plus Ca-ionophore or the engagement of CD3 and these costimulatory molecules. Almost all T-cells exhibited potency as to its expression. Functional analysis of AILIM/ICOS demonstrated that AILIM-mediated costimulation was relatively weak compared to that of human.

Protein expression and functional difference of membrane-bound and soluble receptor activator of NF-kappaB ligand: modulation of the expression by osteotropic factors and cytokines.

A variety of humoral factors modulate the osteoclastogenesis. Receptor activator of NF-kappaB ligand (RANKL) expressed on osteoblast/stromal lineage cells plays a pivotal role to transduce an essential differentiation signal to osteoclast lineage cells through binding to its receptor, RANK, expressed on the latter cell population; however, the difficulty to detect RANKL protein expression hampers us in investigating the regulation of RANKL expression by humoral factors. To determine protein expression of RANKL, we have established a new method, named as a ligand-receptor precipitation (LRP) Western blot analysis, which can specifically concentrate the target protein by the use of specific binding characteristic between RANKL and RANK/osteoprotegrin (OPG). RANKL protein expression in the postnuclear supernatant was not detected by common Western blotting, but LRP Western blot analysis clearly showed that RANKL is produced as a membrane-bound protein on murine osteoblasts/stromal cells, and cleaved into a soluble form by metalloprotease. Cytokines stimulating the osteoclastogenesis, such as IL-1beta, IL-6, IL-11, IL-17, and TNF-alpha, increased the expression of RANKL with decrease of OPG expression in osteoblasts/stromal cells. In contrast, cytokines inhibiting the osteoclastogenesis, such as IL-13, INF-gamma, and TGF-beta1 suppressed the expression of RANKL and/or augmented OPG expression. Functional difference between membrane-bound and soluble RANKL was demonstrated, which showed that membrane-bound RANKL works more efficiently than soluble RANKL in the osteoclastogenesis developed from murine bone marrow cell culture. The present study indicates the usefulness of LRP Western blot analysis, which shows that the modulation of osteoclastogenesis by humoral factors is achieved, in part, by regulation of the expression of RANKL and OPG in osteoblast/stromal lineage cells.

Bisphosphonate administration alters subcellular localization of vacuolar-type H(+)-ATPase and cathepsin K in osteoclasts during experimental movement of rat molars.

This study was designed to clarify the effects of bisphosphonate (BP) administration on structure and functions of osteoclasts in alveolar bone resorption during experimental movement of rat molars. To produce orthodontic force, elastic band was inserted between the upper first and second molars for 4 days, and dissected maxillae were then examined by means of light and electron microscopic immunocytochemistry for vacuolar-type H(+)-ATPase and lysosomal cystein proteinase, cathepsin K in osteoclasts. Vacuolar-type H(+)-ATPase and cathepsin K in osteoclasts are the most important enzymes for demineralization of apatite crystals and degradation of bone type-I collagen, respectively. At 1 day before elastic band insertion, BP was administered intraperitoneally. Control rats received the same volume of physiologic saline. In BP-administered rats, most osteoclasts exhibited either irregularly-formed ruffled borders and clear zones or only clear zones of various degrees of extension. Subcellular localization and expression of both vacuolar-type H(+)-ATPase and cathepsin K was significantly decreased in such osteoclasts with impaired ruffled borders and/or only clear zones by BP administration. In particular, cathepsin K secretion by osteoclasts towards resorption lacunae was markedly inhibited by BP administration. Our results indicate for the first time that BP administration significantly impair the osteoclast structure and reduces expression of both vacuolar-type H(+)-ATPase and cathepsin K in osteoclasts during tooth movement.

Mapping of the HLA-linked genes controlling the susceptibility to Takayasu's arteritis.

To further define the HLA-linked genes controlling the susceptibility to Takayasu's arteritis, polymorphisms in five microsatellites around the HLA-B and MICA genes, C1-2-A, MIB, C1-4-1, C1-2-5, and C1-3-1, were investigated in 91 Japanese patients with Takayasu's arteritis and 248 healthy Japanese controls. It was found that allele 238 of C1-2-A [60.4% in patients vs. 29.8% in controls, odds ratio (OR)=3.59, P(c)<0.000004], allele 332 of MIB (22.0% vs. 6. 1%, OR=4.32, P(c)<0.0003), allele 208 of C1-2-5 (47.3% vs. 24.6%, OR=2.75, P(c)=0.001), and allele 291 of C1-3-1 (62.6% vs. 44.8%, OR=2.07, P(c)<0.02) were significantly associated with the disease. Combined analyses of polymorphisms in the HLA-B and MICA genes with those in the microsatellites suggest that there are two different disease-susceptible loci for Takayasu's arteritis; one is mapped near the C1-2-A locus and the other is more closely linked to the HLA-B gene than to the MICA gene, because there are at least two different disease-associated HLA-B haplotypes, HLA-B*52 and -B*39.2 haplotypes, in which the disease-associated C1-2-A allele is shared in common.

Cloning and sequencing of the levansucrase gene from Acetobacter xylinum NCI 1005.

The levansucrase gene (lsxA) was cloned from the genomic DNA of Acetobacter xylinum NCI 1005, and the nucleotide sequence of the lsxA gene (1,293 bp) was determined. The deduced amino acid sequence of the lsxA gene showed 57.4% and 46.2% identity with the levansucrases from Zymomonas mobilis and Erwinia amylovora, respectively, while only 35.2% identity with that from Acetobacter diazotrophicus. The gene product of lsxA (LsxA) that was overproduced in E. coli coded for a polypeptide of molecular mass 47 kDa. The LsxA released glucose and produced polysaccharide from sucrose, the structure of which was analyzed by nuclear magnetic resonance spectroscopy and determined to be a beta-(2,6)-linked polyfructan.

Observation of nuclear excitation by electron transition in 197Au with synchrotron X rays and an avalanche photodiode

We have succeeded in observing nuclear excitation by electron transition (NEET) in 197Au by a new method. Monochromatic x-rays of synchrotron radiation were used to ionize the K shell of gold atoms in a target foil. The internal-conversion electrons emitted from excited nuclei were detected with a silicon avalanche photodiode. At a photon energy of 80.989 keV, the NEET probability in 197Au was determined to be (5.0+/-0.6)x10(-8) from a comparison of the event number per photon between NEET and the nuclear resonance at 77.351 keV.

A single theory explains two empirical laws applicable to plant populations.

Two empirical laws, formulated independently, are known to be well satisfied in even-aged plant monocultures. One relates yield to plant density in different plant populations, and the other relates cumulative plant mass to cumulative plant number from the largest individual within a population. In this paper, we construct a mathematical model of plant growth under asymmetric competition between individuals in a population, where large individuals grow larger than small individuals because they pre-empt resources, especially light. The model categorizes influences on the growth of a plant into those from individuals larger than itself, and those from all individuals in the population. We derive the two laws from our model. Thus, individual growth, determined by asymmetric interaction between individuals in a population, can explain the two different laws relating to plant populations.

Development of a competitive polymerase chain reaction assay for the ruminal bacterium Butyrivibrio fibrisolvens OB156 and its use for tracking an OB156-derived recombinant.

A competitive polymerase chain reaction assay targeting the 16S rDNA was developed for quantitating the rumen bacterium Butyrivibrio fibrisolvens OB156. A competitor DNA, serving as an internal control in the competitive polymerase chain reaction reaction, was constructed by polymerase chain reaction using a looped oligo longer than the normal primer. Coamplification of the target DNA with known amounts of the competitor DNA allowed quantitation of the target DNA in both pure culture and mixed culture systems, where minimum quantifiable level of OB156 was 1.7x10(2) and 5.6x10(4) cells, respectively. When an erythromycin-resistant recombinant derived from OB156 was inoculated into a rumen fluid culture, its numbers decreased with time. The rate of decrease measured by the competitive polymerase chain reaction assay was much slower than the rate determined by culture enumeration using erythromycin selection. The competitive polymerase chain reaction assay also showed 48 h persistence of the recombinant at 10(4) ml(-1) even after disappearance of culturable recombinant, suggesting maintenance of the target DNA from uncultivable cells. In an in vivo tracking trial, the recombinant became undetectable within 72 h with either assay, indicating rapid hydrolysis and/or outflow of the cells from the rumen.

A PML/RARA chimeric gene on chromosome 2 in a patient with acute promyelocytic leukemia (M3) associated with a new variant translocation: t(2;15;17)(q21;q22;q21).

We describe a patient with acute promyelocytic leukemia (APL) carrying a new complex variant translocation of t(2;15;17)(q21;q22;q21). The karyotypic interpretation was confirmed by fluorescence in situ hybridization (FISH) with the use of painting probes of chromosomes 2, 15, and 17 and a PML/RARA dual color DNA probe. FISH showed a PML/RARA fusion gene on the der(2) instead of the der(15). These results suggest that the critical event in the development of APL is the formation of a PML/RARA chimeric gene, regardless of its locus in the genome.

A macrophage receptor for apolipoprotein B48: cloning, expression, and atherosclerosis.

We have cloned a human macrophage receptor that binds to apolipoprotein (apo)B48 of dietary triglyceride (TG)-rich lipoproteins. TG-rich lipoprotein uptake by the apoB48R rapidly converts macrophages and apoB48R-transfected Chinese hamster ovary cells in vitro into lipid-filled foam cells, as seen in atherosclerotic lesions. The apoB48R cDNA (3,744 bp) encodes a protein with no known homologs. Its approximately 3.8-kb mRNA is expressed primarily by reticuloendothelial cells: monocytes, macrophages, and endothelial cells. Immunohistochemistry shows the apoB48R is in human atherosclerotic lesion foam cells. Normally, the apoB48R may provide essential lipids to reticuloendothelial cells. If overwhelmed, foam cell formation, endothelial dysfunction, and atherothrombogenesis may ensue, a mechanism for cardiovascular disease risk of elevated TG.

Cell-type specificity of l-leucyl l-leucine methyl ester.

l-Leucyl l-leucine methyl ester (LeuLeuOMe) is a lysosomotropic agent which is converted to a membranolytic compound by dipeptidyl peptidase I and kills human leukocytes such as CD8+ T cells and monocytes but not B cells. The reagent has also been used in mice on the assumption that the cell-type specificity to murine leukocytes is the same as that to human leukocytes. During study on the effect of LeuLeuOMe on antigen-driven IL-2 production using murine splenocytes as antigen-presenting cells, however, we noticed that murine B cells were sensitive to LeuLeuOMe. We therefore examined the cell-type specificity using murine splenocytes and peritoneal macrophages. Flow cytometric analysis revealed that the most sensitive cells to LeuLeuOMe were CD8+ cells and that CD19+ cells (B cells) were as sensitive as CD3+ cells (T cells). Murine splenic B cells, which were either positively or negatively sorted with a cell sorter, were also sensitive to LeuLeuOMe, whereas human peripheral blood B cells, which were positively sorted, were not. Peritoneal macrophages were the most insensitive to LeuLeuOMe. Thus, this study demonstrated that the cell-type specificity to murine leukocytes is different from that to human leukocytes.
