Kinetic study of transfer of 11-cis-retinal between rod outer segment membranes using regeneration of rhodopsin.

The present study demonstrates some important facts on the regeneration of rhodopsin in rod outer segment membranes. 11-cis-Retinal added to a rod outer segment membrane suspension did not react directly with opsin but was rapidly solubilized into membranes and then recombined with opsin in the membrane. It was also revealed that the regeneration of rhodopsin was perturbed by the formation of retinylidene Schiff base with phosphatidylethanolamine in rod outer segment membranes, which decreased with increasing temperature. The activation energy of rhodopsin regeneration in rod outer segment membranes was 18.7 kcal/mol, being smaller than the value of 22 kcal/mol in 1% digitonin solution. 11-cis-Retinal could be found to transfer relatively fast (tau-1/k(1) R 10(3) s) between rod outer segment membranes by using the regeneration of rhodopsin. It was demonstrated that the kinetic measurement for the transport of membrane-soluble molecules such as retinal between membranes could be perform ed with ease and precisely by the method described in this paper.

Primary failure of Hancock porcine bioprosthetic valves: two dimensional echocardiographic assessment.

To reveal the process of primary failure of the Hancock porcine bioprosthetic heart valve, a real-time two-dimensional echocardiography was undertaken. There were 75 cases (84 valves) with 26 in the aortic valve position, 39 in the mitral valve position, 9 in both the aortic and mitral valve positions, and 1 in the tricuspid valve position. The subjects comprised 35 males and 40 females, whose ages ranged from 18 to 65 years with a mean of 44.7 years. Valvular changes such as thickened valve cusps or abnormal valve movement were observed in 27 (32%) of the 84 valves; more specifically, in 5 (14.3%) of the 35 aortic valves and in 22 (45.8%) of the 48 mitral valves, indicating that valvular changes occurred most frequently in the mitral valve position (p less than 0.01). In the period between valve replacement surgery and the appearance of valvular changes, no significant difference was noted between aortic and mitral valve bioprostheses. The duration ranged from 14.5 to 74.5 months with a mean of 50.2 months. Valvular changes were evident in more than 50% of the valve bioprostheses implanted 6 or more years ago. In the mitral valve position, valve cusp changes appeared more frequently at the anterior cusp than at the inner and outer cusps. The frequency was 17 cases at the anterior cusp, 6 at the inner and 9 at the outer. In 7 of the 11 cases followed up after the appearance of valvular changes, an increase in the changes was recognized 7.5 to 29.5 months (a mean of 16.4 months) later.(ABSTRACT TRUNCATED AT 250 WORDS)

Current status and problems in long-term management of patients undergoing coronary artery bypass surgery in Japan.

We studied 272 patients undergoing coronary artery bypass surgery (CABS) to clarify the long-term effects of CABS and to identify problems in long-term management. Data on patients' pre and postoperative medical status were obtained from hospital files. A specially designed questionnaire was sent to all patients to assess residual angina, return to work, non-work physical activity and improvement in the quality of life after a mean follow-up of 29 months. Multivessel disease accounted for 89% of all patients and complete revascularization was achieved in 55%. A total of 131 events of complications associated with CABS occurred in 112 patients; post-transfusion hepatitis was the most frequent complication (26%). Graft patency rate was 89% at the time of hospital discharge. Bypass grafts were patent in 95% of 243 grafts which were restudied at a mean period of 19 months after the first study. There was no significant difference in the patency rate between patients with well controlled, poorly controlled or discontinued anticoagulant therapy. Patients who were relieved from angina, returned to gainful work and had no limitation in non-work physical activity accounted for 74%, 79% and 73%, respectively. By subjective assessment, the quality of life improved in 76% of all patients. Objectively, contributing factors preventing improvement in the life quality were residual angina and post-transfusion hepatitis. There were 13 patients who were readmitted because of cardiac events. Three patients died from these events including two from sudden death during follow-up. We conclude that the beneficial effects of CABS are actually attainable.(ABSTRACT TRUNCATED AT 250 WORDS)

Volatile anesthetics cause conformational changes of bacteriorhodopsin in purple membrane.

We examined the effects of volatile anesthetics on the structure of the bacteriorhodopsin in the purple membrane by measurements of the absorption spectrum and the visible circular dichroism (CD) spectrum and assay of the retinal composition. As the concentrations of halothane, enflurane and methoxyflurane were increased, the absorption at 560 nm decreased but that at 480 nm increased with an isosbestic point around 510 nm. These anesthetic-induced spectroscopic changes were reversible. The CD spectrum showed the biphasic pattern with a positive and a negative band. As the concentration of halothane was increased from 4 mM to 8mM, the negative band reversibly diminished more drastically than the positive band, and at 8 mM of halothane the positive band shifted to around 480 nm. These results show that halothane disturbed the exciton coupling among bacteriorhodopsin molecules. The retinal isomer composition was analyzed using high performance liquid chromatography. The ratio of 13-cis- to all-trans-retinal was 47:53, 34:66 and 19:81 at control, 7.4 mM and 14.9 mM enflurane, respectively. After elimination of enflurane, the ratio returned to the control value. These findings indicate that volatile anesthetic directly affect a bacteriorhodopsin in the purple membrane and induce conformational changes in it.

Chiral discrimination of 5,6-epoxy-3-dehydroretinal by aporetinochrome and cattle opsin.

The combination of a racemic all-trans 5,6-epoxy-3-dehydroretinal (EDR) with aporetinochrome formed a mixture of two diastereomeric pigments. One of the diastereomeric pigments which contained the all-trans EDR with a negative circular dichroic (CD) band, hereafter called EDR(-)-chrome, has its visible absorption maximum around 438 nm, while the other pigment, called EDR(+)-chrome, has its maximum at 464 nm. These were substantiated by measuring the optical activities of the two EDR isomers which were extracted from a mixture of racemic all-trans EDR and a smaller amount of aporetinochrome following exposure to orange light (greater than 530 nm) that irradiates EDR(+)-chrome selectively. The extracted all-trans EDR had a negative CD band around 240 nm and the extracted 11-cis EDR had a positive band in that region with two negative bands on either side of the main band. In the case of both pigments, the effect of alkalinization on the increase of absorbance in the near-ultraviolet and the decrease of absorbance in the visible region was proportionate, qualitatively, to that on the positive CD intensities in both regions. These results suggest that the chromophore EDR in each pigment binds to the same binding site via a Schiff base. The EDR(+)-chrome exhibited properties similar to those of retinochrome, but EDR(-)-chrome showed some different properties, i.e. its formation rate was slower than that of the former one and its absorption band in the near-ultraviolet appeared even at neutral pH. Moreover, by exposing EDR(-)-chrome to yellow light (greater than 480 nm), only a part of its prosthetic all-trans EDR was isomerized and resulted in the formation of 11-cis and 13-cis isomers. This variation in photoisomerizing activity was supposed to be due to the difference in conformation of the side chain between EDR(+) and EDR(-) in aporetinochrome. Combination of 11-cis EDR with cattle opsin was also shown to result in the formation of two diastereomeric pigments. The absorption maxima of the diastereomers containing 11-cis EDR(+) and EDR(-) were at about 446 and 474 nm, respectively.

A photoproduct with 13-cis retinal generated by irradiation with violet light in the octopus retina.

The photoconversion between rhodopsin (R) and metarhodopsin (MR) was investigated in the retina of Octopus ocellatus by measurements of the fast photovoltage (FPV) in conjunction with high pressure liquid chromatography (HPLC). Following conversion of most of R to MR by short-term exposure to an intense violet light, a large vitreous negative FPV due to photon absorption by MR was observed with an orange test flash. By continuing the exposure to the same violet light, however, the negative FPV became very small, though long-term irradiation with blue-green light produced a recovery. With the violet light exposure a substance other than R and MR is produced, which hardly contributes to FPV generation. Analysis of retinal isomers with HPLC show that such an exposure produces a substance with 13-cis retinal as its chromophore and that it was significantly reduced after exposure to blue-green light.
