Detection of flail aortic valve in bacterial endocarditis with real-time two-dimensional echocardiography. A case report.

This report describes a case with flail right and noncoronary cusps due to bacterial endocarditis, diagnosed with real-time two-dimensional echocardiography. Real-time two-dimensional phased array sector scanner demonstrated 2 cugdel-shaped lesions in the region of the left ventricular outflow tract through the aortic root, pendulating upward and downward floating along the blood stream. This study shows that the real-time two-dimensional echocardiography is a very useful noninvasive tool in the early diagnosis of flail aortic valve.

The change in the left atrial dimension during diastole. Echocardiographic assessment of mitral stenosis.

The change in the left atrial dimension during diastole was examined in patients with mitral stenosis using echocardiography. The left atrial emptying fraction (LAEF), defined as the ratio of enddiastolic dimension to endsystolic dimension of the left atrium, was 0.65 +/- 0.03 (mean +/- SE) for normal subjects, 0.86 +/- 0.01 for low grade mitral stenosis, and 0.92 +/- 0.01 for high grade stenosis. The LAEF correlated well with mitral valve area index (r = -0.70), but the correlation between E-F slope of the anterior mitral leaflet and mitral valve area index was less significant (r = 0.48). In patients who received mitral valve replacement the LAEF and the left atrial dimension significantly improved. The left atrial emptying fraction is an easily obtainable and reliable index to estimate the severity of mitral stenosis, even in patients who have undergone mitral valvular surgery.

A study of kinesthetic illusion.

Subjects experience a mismatch between execution and appreciation (kinesthetic illusion) under certain conditions: lifting up a weight to a suggested height with instructed speed. To get the aspect of this kinesthetic illusion from the movement of the forearm, the measurement of forearm-trajectory was made. From the result, it was recognized that there was a linear correlation between the difference of weight (DW) and the difference of reaction time (DRT). This fact shows that there is a constant error between execution and appreciation of movement.

Reduced deformability of erythrocytes exposed to hypercapnia.

The effect of hypercapnia on the deformability of erythrocytes was studied by means of a nuclepore membrane filter method. A decrement of the deformability by 20--40% was observed when PCO2 was increased from 50 mm Hg to 200 mm Hg, accompanied with an increment of 5% in hematocrit value.

Interaction of int protein with specific sites on lambda att DNA.

We have studied the interaction of highly purified Int protein with DNA restriction fragments from the lambda phage attachment site (attP) region. Two different DNA sequences are protected by bound Int protein against partial digestion by either pancreatic DNAase or neocarzinostatin. One Int binding site includes the 15 bp common core sequence (the crossover region for site-specific recombination) plus several bases of sequence adjoining the core in both the P and P' arms. The second Int-protected site occurs 70 bp to the right of the common core in the P' arm, just at the distal end of the sequence encoding Int protein. The two Int binding sites are of comparable size, 30-35 bp, but do not share any extensive sequence homology. The interaction of Int with the two sites is distinctly different, as defined by the observation that only the site in the P' arm and not the site at the common core region is protected by Int in the face of challenge by the polyanion heparin. Restriction fragments containing DNA from the bacterial attachment site (attB) region exhibit a different pattern of interaction with Int. In the absence of heparin, a smaller (15 bp) sequence, which includes the left half of the common core region and the common core-B arm juncture, is protected against nuclease digestion by Int protein. No sequences from this region are protected by Int in the presence of heparin.

Nicking-closing activity associated with bacteriophage lambda int gene product.

Integrative recombination of bacteriophage lambda requires the action of the protein Int, the product of the phage int gene. In this paper we show that highly purified Int relaxes supercoiled DNA. The association of this nicking-closing activity with Int is shown by: (i) the cosedimentation of nicking-closing and recombination activities of purified Int, (ii) the parallel inactivation of the two activities in purified Int by both heat and a specific antiserum, and (iii) the alteration of both activities in crude extracts of a strain expressing a mutant int gene. The nicking-closing activity of Int functions in the absence of divalent cations and in the absence of an apparent source of chemical energy. The activity displays no obvious sequence specificity and is inhibited by Mg2+, spermidine, and single-stranded DNA. Int relaxes positive as well as negative supercoils. We present a model for the mechanism of strand exchange that describes how the nicking-closing activity of Int might be used during recombination.

Addition of short guanylyl blocks to oligonucleotide primers with a thermophilic polynucleotide phosphorylase. Its application to the synthesis of oligonucleotides containing guanylyl residues.

Polynucleotide phosphorylase from Thermus thermophilus catalyzed the addition of short guanylyl blocks from GDP to the 3'-hydroxyl termini of oligonucleotide primers at low temperature in a simple reaction mixture. Polyguanylic acid formation was inhibited at 37 degrees C, but the addition of one or two guanylyl residues to oligonucleotide primers proceeded in high yields. The reaction was applied to the synthesis of oligonucleotides containing guanylyl residues at the 3'-end. Using (Ap)2A and (Up)2U as primers, (Ap)3G, (Ap)3GpG, and (Up)3G were synthesized in yields of 25--52%. (Ap)2GpG was synthesized from ApA and GDP in a yield of 13%.

A new needle-type colloid osmometer for continuous determination of blood oncotic pressure.

For the purpose of facilitating measurement of colloid osmotic pressure of body fluids, a new needle-type colloid osmometer which uses a hollow fiber as a semipermeable membrane instead of a sheet ultrafilter was developed. The simple construction, easy production, and satisfactory performance of the osmometer are described in detail. Although hydrostatic pressure existing in the sample interfered a little with the accuracy of measurement, quickly interchangeable sensing probes brought great advantages in practice.

[Variations in electrolytes in the smooth muscle tissue of the alimentary canal (author's transl)].

Electrolytes in the smooth muscle tissue of the alimentary canal were measured and their behavior under various conditions were studied on the basis of values measured. The results are summarized as follows: 1. Organic changes due to edema and the like are more prominent in the region near the wound in cases of damaged intestine. Utmost care should therefore be exercised when surgical treatment including resection of the intestine is given. 2. Variations in electrolytes are slight in cases of mesenteric hematoma, and surgical treatment is not necessary. 3. Variations in electrolytes are rather prominent in cases of intestinal fistula, and full consideration should be given to the balance of electrolytes, particularly supplement of K. 4. In casees of intestinal anastomosis, the rate of variation is the highest near the site of anastomosis and on the third day after surgery. Therapeutically, therefore, attention should be given to prevention of complications such as incomplete suture. 5. In cases of intestinal obstruction, variations are more marked and rapid. It brings about a high degree of disequilibrium of intra-tissue electrolytes in the upper intestinal tract; hence the early and adequate treatments are needed.
